Reproductive performance of Holstein cows treated with prostaglandin F2a, gonadotropin releasing hormone, and recombinant bovine Somatotropin

The objective of this study was to examine the effects of presynchronization and recombinant bovine somatotropin (rbST) on conception rates following a timed insemination (TAI) protocol in lactating dairy cows. A further objective included the evaluation of the efficacy of the Early Conception Factor (ECF) test kit. Recombinant bST may offer some benefit when used in conjunction with estrus synchronization and TAI. Presynchronization treatment consisted of two injections of PGF2α given 14 d apart, with the second dose administered 14 d prior to the initiation of a TAI protocol. A total of 216 lactating Holstein cows were presynchronized with PGF2α and then received GnRH (100μg) at 67 ± 7 d post partum (PP), administration of PGF2α (25 mg) 7 d later, another GnRH (100μg) administration 2 d after PGF2α, and were inseminated 8-18h later (OvSynch). First service conception rate (CR) was determined by rectal palpation at 42 ± 7 d after artificial insemination (AI). Treated cows (n=113) received rbST 67 ± 7 d PP whereas control cows (n = 113) were presynchronized without rbST. The cycling status of all cows was determined by paired milk P4 levels at 53 and 67 ± 7 d PP. No differences (P > 0.10) in conception rate were observed between control and rbST treated cows (44.7 and 40.7% respectively), nor was there any interaction of cyclicity and rbST. Milk samples were collected 7 d following AI for use in ECF test kit evaluation. Samples were stored at -20ºC (n=216) and at 5ºC (n=113) until assayed. Test results for frozen and refrigerated samples were compared to conception rates determined by rectal palpation at 42 ± 7 d after AI. The rate of false positive and negative results for frozen milk samples were 36.1 and 14.8% respectively, and 40.7 and 7.1% for refrigerated milk samples. Treatment with rbST at the time of the first GnRH injection of an OvSynch protocol did not significantly alter first service conception rates. Additionally, an acceptable 92.9% accuracy of the ECF test for the detection of open cows 7 d after AI using milk samples stored at 5ºC was obtained. / Master of Science

Roles of IL-6, TNF-α and IL-1β in regulating growth hormone signaling and FGF19 signaling in the liver.

Evidence suggests that during inflammation, the liver becomes resistant to growth hormone (GH) actions, leading to downregulation of the anabolic gene IGF-I and the activation of catabolic processes. Decades of studies demonstrated that pro-inflammatory cytokines IL-6, TNF-α and IL-1β are involved in the pathogenesis of hepatic GH resistance. However, the exact mechanisms used by these individual cytokines to regulate GH signaling are not defined. Using Huh-7 human hepatoma cells and mouse models of chronic and acute inflammation, we show that TNF-α and IL-1β but not IL-6 inhibited hepatic GH receptor (GHR) expression, and that IL-6 but not TNF-α and IL-1β stimulated expression of suppressor of cytokine signaling-3 (SOCS3). TNF-α/IL-1β and IL-6 acted primarily at GHR and SOCS3 respectively to inhibit the GH-IGF-I pathway. While TNF-α/IL-1β exerted a tonic inhibition on hepatic GH signaling, IL-6 activity is dependent on the active GH pathway. IL-6 lost its inhibition on the GH-IGF-I pathway when GHR expression was blocked as the inflammation progressed. These results reveal previously undefined distinct mechanisms used by TNF-α/IL-1β and IL-6 to inhibit the hepatic GH pathway. Our results may provide a new guidance for clinical practice in treating pediatric infammation-induced GH resistance. / Fibroblast growth factors (FGFs) play critical roles in many physiological processes by binding to and activating FGF receptor (FGFR) family. FGF19 belongs to FGF15/19 subfamily of FGFs that includes FGF15/19, FGF21 and FGF23. FGF19 has been shown to regulate bile acid homeostasis, and protein and glycogen synthesis in the liver. FGF19 binds FGFR4 and the co-receptor β-klotho to initiate signaling. Studies have shown that proinflammatory cytokines such as TNF-α can impair FGF21 signaling in adipose cells by repressing the expression of β-klotho. However, little is known about the effects of IL-6, TNF-α and IL-1β on regulating hepatic FGF19 signaling. In the present study, we found that IL-1β inhibited β-klotho expression both in vitro and in vivo, and this inhibition required JNK and NF-κB pathways. This study employs duality theory to model the dairy industry. Supply and demands for milk, cull cows, feed, labor and veterinary services were simultaneously estimated using Weighted Least Squares. Elasticities and partial adjustments were obtained for the Nation and the following regions, Appalachia, Cornbelt, Northeast, Pacific, Southern Plains and Upper-Midwest. Predictions for the change in quantity of goods demanded and supplied were made assuming a parallel shift in the supply of milk and demand for feed. In conclusion, predictions on the impact of bovine Somatotropin are made assuming all results are correct.

The influence of rest-interval duration on the growth hormone response to resistance exercise / Influence of rest interval duration on the growth hormone response to resistance exercise

The purpose of this study was to establish an exercise protocol that demonstrated a relationship between rest-interval duration and the exercise induced human growth hormone (hGH) response. Ten recreationally trained male subjects (23 ± 0.9 yrs) performed three leg extension trials on an Eagle — Cybex leg extension machine that consisted of 4 sets of 10 repetitions. The workload and volume was kept constant, but each trial had different rest-interval durations. Rest-interval durations between sets were at: 30 sec intervals (T-30), 60 sec intervals (T-60), or 120 sec intervals (T-120). Blood samples were obtained pre- and 0 — 30 minutes post-exercise and analyzed for lactate and hGH. All blood lactates rose significantly above baseline after exercise, with no differences in time of occurrence between trials. Blood lactates were significantly greater after the T-30 trials, compared to that of the T-60 and T-120 trials. There was no significant difference in hGH concentrations between trials. However, the data did suggest a relationship between rest-interval duration and the variability of hGH responses. The T-30 trials yielded significantly greater variation in hGH concentrations than the T-120 trials, and the T-120 trials showed significantly less variation than both the T-30 and T-60 trials. Although significant differences were found in these variations between trials, they did not prevent any significant differences in concentrations between trials from being found. In summary, the results of this study demonstrated an exercise related increase in lactic acid that had an inverse relationship to the length of the rest-intervals. hGH data on the other hand, did not show a relationship between rest-interval duration and the hGH concentrations. The connection between rest-interval duration and variability of hGH responses could possibly suggest that hGH values may have been significantly different if an exercise protocol higher in volume were utilized. Additionally, the results indicate that there is no direct relationship between blood lactate and hGH concentrations, as others have suggested. / School of Physical Education, Sport, and Exercise Science
