Memoria para optar al título de Ingeniero Civil / Este trabajo de título tiene por objetivo estudiar el comportamiento sísmico de perfiles
de suelos de espesores menores a 30 metros y perfiles que poseen una inclusión de
alta o baja velocidad de onda de corte. Para ello, se analizaron 19 estaciones
sismológicas de la red japonesa KiK-net, de las cuales 4 poseen una inclusión de alta o
baja velocidad, 11 tienen espesores menores a 30 metros y 4 cumplen con ambas
características. Además, se consideró dentro del estudio la estación de Llolleo,
estudiada por Sáez et al. (2018). Estas estaciones se caracterizan por tener un perfil de
velocidades conocido a partir de ensayos downhole y por tener un acelerómetro en
superficie y otro más profundo que en basamento rocoso. En cada una de estas
estaciones, se obtuvieron razones espectrales a partir de registros sísmicos, calculadas
como la razón entre las componentes horizontales en superficie y las horizontales en la
base (HS/HB) y razones espectrales entre las componentes horizontales y la vertical en
superficie (HS/VS). Estas razones espectrales fueron comparadas con la función de
transferencia teórica obtenida mediante la teoría unidimensional de propagación de
ondas de corte SH, concluyendo que las frecuencias predominantes obtenidas a partir
de HS/HB y HS/VS difieren de la obtenida teóricamente. Al evaluar el coeficiente de
correlación r de Pearson, se concluye que las diferencias en forma entre la función de
transferencia teórica y las razones espectrales son mayores en los depósitos que
poseen una inclusión de velocidad. Por otro lado, las frecuencias predominantes
obtenidas con las razones espectrales HS/HB y HS/VS coinciden en 13 de las 19
estaciones analizadas, a pesar de que el sensor en la base no se encuentra
exactamente en la interfaz entre el suelo y la roca. En los depósitos donde se presentan
similitudes entre las razones espectrales HB/VB y HS/VS, se encontró un patrón que
consiste en tener un estrato de suelo rígido, seguido de uno flexible el cual coincide
con la profundidad que requiere tener el depósito para que la frecuencia predominante
coincida con la teórica y bajo este un estrato de suelo más rígido, siendo más común
este patrón en los perfiles de suelo con inclusión de alta velocidad. Por otro lado, se
evaluó la clasificación sísmica que utiliza la norma NCh433 y se comparó con la
propuesta por Idini et al. (2016), observándose que depósitos de suelo que tienen la
misma clasificación según la norma NCh433, presentan clasificaciones distintas según
la propuesta de Idini et al. (2016).

Thermophilic proteins : stability and function / Les protéines thermophiles : stabilité et fonctionKatava, Marina 14 October 2016 (has links)
La température est un paramètre crucial dans le fonctionnement du monde vivant, notamment de la machinerie moléculaire (les protéines) dont la stabilité et l’activité en dépendent sensiblement. Celles-ci sont souvent considérées comme étant équivalentes : si une protéine fonctionne, c’est qu’elle est stable, et vice-versa. Cependant, les protéines des organismes thermophiles, qui prolifèrent dans de températures élevées, sont stables à température ambiante, mais y présentent une faible activité. Cette dernière est optimale à la température de croissance de l’organisme hôte. Lorsqu’on parle de stabilité et d’activité protéique, la rigidité mécanique est souvent utilisée comme paramètre pertinent, offrant une explication simple et attractive à la fois pour la stabilité thermodynamique à haute température et au manque d’activité à des températures plus modérés. La réalité s’avère souvent plus complexe, et les mécanismes moléculaires reliant rigidité/flexibilité avec la stabilité et l’activité sont encore mal compris. Dans ce travail, nous abordons le problème au travers de trois systèmes. Nous avons examiné l’activation thermique des modes fonctionnels du domaine G de la protéine EF ainsi que les homologues mésophiles et thermophiles de la déshydrogénase Lactate/Malate. Par ailleurs, nous avons mis en évidence l’existence d’un paramètre unique (la moyenne des fluctuations atomiques) permettant d’expliquer la dynamique de la protéine lysozyme près de son point de fusion, et ce quelle que soit la nature de l’environnement autour de la protéine (qui décale le point de fusion). Nos conclusions se basent principalement sur une approche in silico où la dynamique moléculaire et des techniques d’échantillonnage améliorées sont utilisées et sont complémentées par des expériences de diffraction de neutrons / Temperature is one of the major factors governing life as demonstrated by the fine tuning of stability and activity of the molecular machinery, proteins in particular. The structural stability and activity of proteins have been often presented as equivalent. However, the thermophilic proteins are stable at ambient condition, but lack activity, the latter recovered only when the temperature increases to match that of the optimal growth condition for the hosting organism. In discussing the protein stability and activity, mechanical rigidity is often used as a relevant parameter, offering a simple and appealing explanation of both the extreme thermodynamic stability and the lack of activity at low temperature. The reality, however, illustrates the complexity of the rigidity/flexibility trade off in ensuring stability and activity through intricate thermodynamic and molecular mechanisms. Here we investigate the problem by studying three study cases. These are used to relate the thermal effects on mechanical properties and the stability and activity of the proteins. For instance, we have probed the thermal activation of functional modes in EF G-domain and Lactate/Malate dehydrogenase mesophilic and thermophilic homologues and verified a “universal” scaling of atomistic fluctuation of the Lysozyme approaching the melting in different environmental conditions. Our conclusions largely rest on an in silico approach, where molecular dynamics and enhanced sampling techniques are utilized, and are often complemented with neutron scattering experiments

Efecto de los depósitos de gas somero del fondo marino en las comunidades macrobentónicas del estuario de Bahía BlancaBravo, María Emilia 20 March 2019 (has links)
Esta tesis tiene como finalidad caracterizar los depósitos de gas somero de la
zona interna del estuario de Bahía Blanca y evaluar el efecto de los sedimentos
gasíferos sobre las comunidades bentónicas submareales asociadas. Para tal fin, se
mapeó la distribución del gas somero en el estuario mediante el análisis de registros
sísmicos de alta resolución. En base a este mapa, se seleccionaron dos sitios de
estudio (con gas y control) que sólo difirieron en la presencia de gas en contacto con
la superficie del fondo marino. Mediante métodos acústicos se determinó la
estratigrafía y morfología del sitio con gas y el área adyacente. Además, se evaluaron
los sedimentos gasíferos por medio de los siguientes análisis: granulometría,
contenido de materia orgánica, rayos X y medición con HPLC head space. Los
organismos bentónicos de ambos sitios fueron comparados en términos de
abundancia, biomasa, diversidad, equitatividad, riqueza específica, estructura
taxonómica y funcional. El gas se encontró ampliamente distribuido en el estuario,
tanto cerca como en contacto con la superficie del fondo marino de la zona interna
del estuario de Bahía Blanca. Se evidenció en forma de turbidez acústica y
apantallamiento acústico. La columna sismoestratigráfica presentó cinco secuencias
sísmicas del Pleistoceno tardío-Holoceno. Se estableció que el origen del gas es
biogénico, asociado a un depósito del Holoceno Medio, desde el cual migró hasta
estar en contacto con la superficie del fondo gracias a la permeabilidad de los
estratos suprayacentes. El gas en la superficie del fondo fue evidenciado en
sonogramas por medio de la atenuación de la señal acústica, lo que evidencia una
baja cohesión de los materiales por la disminución en el esfuerzo de corte generada
por las burbujas de gas. La granulometría de los sedimentos fue similar en ambos
sitios, caracterizada como fango arenoso. Los sedimentos gasíferos presentaron un
mayor contenido de materia orgánica. Los análisis de rayos X demostraron mayor
homogeneidad en la columna de sedimentos gasíferos, respecto a los del sitio
control. La medición cualitativa del gas con HPLC probó la presencia del gas metano.
La riqueza específica, diversidad, equitatividad y abundancia total de organismos
fue menor en el sitio con gas. Entre sitios con gas y control hubo marcadas
diferencias en la estructura taxonómica. Se encontraron taxa caracterizando los sedimentos gasíferos como Stylatula darwini, Aricidea sp. y nematodes, y otros
ausentes como Polydora cornuta y Monocorophium insidiosum lo que indicaría una
alta sensibilidad de estas especies al gas. La diferencia en la estructura comunitaria
entre sitios fue explicada por el contenido de materia orgánica de los sedimentos.
Además, la estructura funcional de la comunidad bentónica en el sitio con gas
presenta una mayor diversidad de roles ecológicos que en el sitio control. Se
concluye que la baja estabilidad y cohesividad, así como el alto contenido de materia
orgánica de los sedimentos gasíferos afecta a especies sensibles comunes en el área
de estudio. Resultando en un hábitat dominado por especies oportunistas, así como,
por algunos taxa que podrían utilizar indirectamente el metano como fuente de
energía. / The aim of this thesis is to characterize the shallow gas deposits of the inner
area of the Bahía Blanca estuary and to evaluate the effect of the gas-bearing
sediments on the associated subtidal benthic communities. For this purpose, the
distribution of shallow gas in the estuary was mapped through the analysis of highresolution
seismic records. Based on this map, two study sites were selected (with
gas and control) that only differed in the presence of gas in contact with the surface
of the seabed. Acoustic methods were used to determine the stratigraphy and
morphology of the site with gas and the adjacent area. In addition, the gas sediments
were evaluated by means of the following analyses: grain size, organic matter
content, X-rays and HPLC head space measurement. The benthic organisms of both
sites were compared in terms of abundance, biomass, diversity, evenness, species
richness, taxonomic and functional structure. The gas was found widely distributed
in the estuary, both near and in contact with the surface of the seabed of the inner
area of the Bahía Blanca estuary. It was evidenced in the form of acoustic turbidity
and acoustic blanking. The seismo-stratigraphic column presented five seismic
sequences of the late Pleistocene-Holocene. It was established that the gas origin is
biogenic, being associated with a Middle Holocene deposit, from which it migrated
until it was in contact with the surface of the seabed due to the permeability of the
overlying strata. The gas in the surface of the seabed was evidenced in sonograms
by means of the attenuation of the acoustic signal, which evidences a low cohesion
of the materials by the decrease in the shear stress generated by the gas bubbles.
The grain size of sediments was similar in both sites, characterized as sandy mud.
The gas-bearing sediments presented a higher content of organic matter. X-ray
analysis showed greater homogeneity in the gassy sediments column, compared to
the control site. The qualitative measurement of the gas with HPLC proved the
presence of methane gas. The species richness, diversity, evenness and total
abundance of organisms was lower in the gas site. There were marked differences
in the taxonomic structure between gas and control sites. There were found taxa
characterizing the gas-bearing sediments as Stylatula darwini, Aricidea sp. and
nematodes, and others absent in this site, such as Polydora cornuta and
Monocorophium insidiosum, which would indicate a high sensitivity of these species
to gas. The difference in community structure between sites was explained by the
organic matter content of the sediments. In addition, the functional structure of the
benthic community in the gas site presents a greater diversity of ecological roles
than in the control site. It is concluded that the low stability and cohesiveness, as
well as the high organic matter content of the gas sediments affects common
sensitive species in the study area. Resulting in a habitat dominated by opportunistic
species, as well as by some taxa that could indirectly use methane as an energy
source.
