Charakterizace magnetických nanostruktur pomocí mikroskopie magnetických sil / Characterization of magnetic nanostructures by magnetic force microscopy

The thesis deals with magnetic force microscopy of soft magnetic nanostructures, mainly NiFe nanowires and thin-film elements such as discs. The thesis covers almost all aspects related to this technique - i.e. from preparation of magnetic probes and magnetic nanowires, through the measurement itself to micromagnetic simulations of the investigated samples. We observed the cores of magnetic vortices, tiny objects, both with commercial and our home-coated probes. Even domain walls in nanowires 50 nm in diameter were captured with this technique. We prepared functional probes with various magnetic coatings: hard magnetic Co, CoCr and soft NiFe. Hard probes give better signal, whereas the soft ones are more suitable for the measurement of soft magnetic structures as they do not influence significantly the imaged sample. Our probes are at least comparable with the standard commercial probes. The simulations are in most cases in a good agreement with the measurement and the theory. Further, we present our preliminary results of the probe-sample interaction modelling, which can be exploited for the simulation of magnetic force microscopy image even in the case of probe induced perturbations of the sample.

This diploma thesis implements methods for a measurement of rotational speed. It is implemented on the Compact RIO platform from National Instruments. Corresponding SW is implemented using the graphical programming language G in LabView environment. Developed system uses two different sensors – incremental encoder and tacho sensor for measurements of rotational speed. Thesis further analysis and implements a method for an encoder nonlinearity determination and for its on-line correction. For used methods, effects adding errors into the measurements are evaluated and quantified.

In the beginning of this thesis is a short introduction into plasmid DNA which is in the form of a vector used in molecular biology. Plasmids can be used in the form of fluorescent probes to measure changes in membrane potential. Into their structure is added a dye called fluorophore. As an important representative of this thesis is a fluorescent probe ASAP1 which contains green fluorescent protein whose response to the membrane potential change is the decrease in the intensity of emitted light. The aim of this thesis was to make chemical transfection of this plasmid into the HEK293 cell line and carry out its characterization. In the work is also described the design of a method for the analysis of the time course of changes in fluorescence depending on the cell membrane depolarisation. In the end of this thesis is also desribed realized experiment including the discussion of aquired results.

BioSealing is an innovative method of soil body reconstruction, which uses stimulation of soil bacterial activity to seal the leakage. The method was developed in the Netherlands and it was used in conditions of the Czech Republic in years 2015 and 2016 on the Hornice water work. This earth dam was monitored by electrical impedance spectrometry by apparatus with device Z-meter IV in this period and the data are processed and assessed in this diploma thesis.

(english): In the theoretical part, I deal primary with defining the concepts of doll production and it is history first in a global context and than specifically in the Czech Republic. I also cover historic all production technques by artisans, the revival of there traditional technques as well as various doll produstion techniques. I highlight some of today's craftsmen as well as their personal stories about getting started in the handmade doll production. Next, I delve into rules and guidelines for doll production as well as doll selection by the consumer. A the end of theoretical part, I deal with the different types of gameplay with dolls along wth gender aspect. The first part of the project deals with the relationship between the person hand making the doll and the doll itself. It also includes a small making project with childern. The second part is the project goes over a collection of dolls I have handmade in the last three years divided according to production technique and further subdivided need on material, labor and suitable age. The third part is the research, Which has determine whether the observation of the making process of dolls can influence the kids. Středověké osídlení středního Poohří / Medieval settlement of the middle River Ohře region

The present work is focused on the medieval settlement in the river Oh e region (Czech Republic), with emphasis on the problem of development and the settlement process at the turn of the early and the high Middle Ages. The studied region, with the axis of the river Oh e, is occupied the area roughly from town Kada to Louny and it is corresponded to the parts of Chomutov and Louny districts. The definition includes geomorphologically different regional parts, providing diverse conditions for the development of settlement. At the same time, it is considered the cultural- historical and archaeological value of today's known archaeological sites. The basis of this thesis is the selection of several landscape probes with an area of several square kilometres, to which the detailed archaeological study is concentrated. These probes are defined in relation to the regional unit so that they can become representative samples that allow for the solution of major settlements of historical issues. The basis for defining probes is the characteristics of natural conditions, the list of settlement units and the catalogue of archaeological sites. The doctoral thesis deals with an experimental study of several diagnostic techniques intended for plasma diagnostics and diagnostics of thin films during reactive sputter deposition. A relatively novel probe diagnostic technique called Floating harmonic probe for measurement of the ion density and electron tem- perature in technological low-temperature plasma is studied. A Langmuir probe is commonly used, however, its application in conditions where non-conducting films are being deposited can be problematic or unreliable. The floating harmonic probe measurement technique deals with this inherent problem of the Langmuir probe. The Floating harmonic probe results are compared to those obtained by a classical Langmuir probe in non-reactive DC continuously driven discharge, and its applicability in reactive regime during deposition of iron oxide thin films is proved. The work deals also with a modification of the Floating harmonic probe called Phase Delay Harmonic Analysis Method which is intended for diagnostics of pulsed driven discharges. The second part of the thesis is devoted to a new proposed method for in-situ diagnostics of thin films.
