Φασματοσκοπία χρονικής ανάλυσης και διφωτονικής απορρόφησης οργανικών ενώσεων παράγωγων της βενζοδισθιαζόληςΚοτσιάς, Δημήτριος 26 April 2012 (has links)
Στην παρούσα μεταπτυχιακή αυτή εργασία μελετήσαμε την συμπεριφορά για πρώτη φορά ενώσεων που είχαν σαν βάση την βενζοδισθιαζόλη. Συγκεκριμένα οι ενώσεις αυτές μελετήθηκαν με την χρήση των τεχνικών της φασματοσκοπίας διφωτονικής απορρόφησης της φασματοσκοπίας σταθερής κατάστασης και της φασματοσκοπίας φθορισμού χρονικής ανάλυσης.
Αρχικά όσον αφορά την φασματοσκοπία διφωτονικής απορρόφησης, μπορέσαμε να οδηγηθούμε στα εξής συμπεράσματα: οι καλύτερες ενώσεις που παρουσιάζουν αρκετά μεγάλη διφωτονική απορρόφηση είναι τα γραμμικά μόρια (PK-439 και PK-452) σε σχέση με τα U-shaped μόρια με μέγιστη ενεργό διατομή ΔΦΑ ~2000GM. Επιπλέον παρατηρήσαμε ότι η χρήση της βενζοδισθιαζόλης σαν κεντρικός πυρήνας προκαλεί σημαντική αύξηση της διφωτονικής απορρόφησης, σε σχέση με την βενζοθιαζόλη.
Τέλος, με την τεχνική της φασματοσκοπίας φθορισμού χρονικής ανάλυσης μπορέσαμε να οδηγηθούμε σε κάποιες διαπιστώσεις: συγκεκριμένα παρατηρήσαμε ότι όσο το μήκος κύματος καταγραφής μεγαλώνει, τόσο οι καμπύλες αποδιέγερσης γίνονται πιο αργές. Ακόμα διαπιστώσαμε ότι από την σύγκριση μορίων στο μήκος κύματος του μεγίστου, σε εκείνα τα μόρια που αποδιεγείρονται γρήγορα, ευνοούνται οι μη-ακτινοβολητικές διεργασίες και ταυτόχρονα παρουσιάζουν μικρή ενεργό διατομή ΔΦΑ. / --

INVESTIGATING THE PHOTOPHYSICAL PROPERTIES OF POTENTIAL ORGANIC LEAD SENSORSCarlos Quinones Jr (17015838) 03 January 2024 (has links)
<p dir="ltr">LeadGlow (<b>LG</b>) was reported in 2009 for its ability to both sensitively and selectively detect Pb<sup>2+</sup> in aqueous solutions. Utilizing the synthetic approach of <b>LG</b>, it is possible to generate a class of novel fluorophores. A derivative of first-generation <b>LG </b>was synthesized and reported here for the first time, intuitively named <b>LG2</b>. Both compounds contain interesting photophysical properties that have not been extensively researched prior to this work. Because of this, photophysical properties of both <b>LG</b> and <b>LG2</b> are unveiled here for the first time. These properties were investigated by determinations of quantum yield (QY), average fluorescence lifetime, and DFT calculations. <b>LG</b> was found to have a higher QY (0.057) than <b>LG2</b> (0.011); however, <b>LG2</b> displays an average fluorescence lifetime (3.186 ns) 5x greater than that of <b>LG</b>. Both <b>LG </b>and <b>LG2</b> are synthesized via Hg<sup>2+</sup>-facilitated desulfurization of their respective thiocarbonyls, resulting in a turn-on fluorescence feature. The thiocarbonyl-containing fluorophores (<b>SLG </b>and <b>SLG2</b>) display quenched fluorescence compared to their oxo-derivatives (<b>LG </b>and <b>LG2</b>), this work attempts to investigate the mechanism(s) responsible.<b> </b>A whole class of LeadGlow compounds can be synthesized and could be potentially used as fluorescence-based sensors.</p>

A compreensão dos caminhos de desativação não radiativa em polímeros conjugados é fundamental para o uso desses materiais em dispositivos luminescentes, células fotovoltaicas e sensores. Nesta tese, os processos não radiativos em filmes automontados de polímero luminescente foram investigados via análise da supressão de intensidade de fotoluminescência, utilizando a técnica de fluorescência estacionária, e através da medida do tempo de vida do estado excitado com fluorescência resolvida no tempo em arranjo de contagem de fótons. A camada emissiva constituída por poli(p-fenileno vinileno) (PPV) obtido da conversão térmica do precursor poli(cloreto de xilideno tetraidrotiofeno) (PTHT) apresentou tempos de vida entre 150 e 250 ps, e fraca dependência com o comprimento de onda de detecção da emissão. Na presença de uma monocamada contendo azocorante Vermelho do Congo (CR) ou ftalocianina tetrassulfonada de níquel (NiTsPc) atuando como receptor de energia foi observada supressão da fluorescência e encurtamento do tempo de vida (50-100ps). À medida que se aumenta a distância entre as camadas de doador e receptor via interposição de bicamadas de espaçador, a dinâmica do sistema tende à dinâmica de decaimento do estado excitado do PPV na ausência de receptor. A dependência da eficiência com a distância 1/r2 ou 1/r3 apontam para transferência de energia ressonante (RET) típica da interação plano-plano ou plano-camada. Os processos não radiativos foram também estudados em função da diluição de segmentos PPV dispersos em matriz de polieletrólito poli(cloreto de dodecildimetilamônio) PDAC. A diluição dos segmentos conjugados aumenta a eficiência quântica de emissão, torna o mecanismo de desativação mais lento (500 ps) e diminui a dependência do tempo de vida com o comprimento de onda de detecção, devido à menor eficiência dos mecanismos de desativação não radiativa. Na presença de uma camada receptora, a eficiência de transferência de energia é maior para amostras com segmentos mais diluídos indicando que esse mecanismo compete com os processos internos à própria camada emissiva. O tempo de vida aumenta em filmes com mais baixo grau de conjugação e a dependência com o comprimento de onda de emissão é mais forte. Em resumo, a técnica de automontagem se mostrou adequada para obter filmes com propriedades fotofísicas controláveis no nível molecular, que puderam ser estudadas com espectroscopias de fluorescência. / Understanding the pathway of excitation in conjugated polymers is important for using these materials as active layer in devices, photovoltaic cells and sensors. In this thesis, non-radiative processes in layer-by-layer (LbL) films were investigated with measurements of fluorescence quenching in steady-state fluorescence spectroscopy and of the singlet excited state lifetime using time-resolved fluorescence spectroscopy in a single photon counting (SPC) apparatus. The emissive layer of poly(p-phenylene vinylene) (PPV) obtained via thermal conversion of the soluble precursor poly(xylylidene tetrahydrothiophenium) chloride (PTHT) showed lifetime 150-250 ps and weak dependence on the emission wavelength. In the presence of an acceptor layer of either Congo Red (CR) or nickel tetrasulfonated phthalocyanine (NiTsPc) the PL intensity was quenched and the lifetime was shorter (50 -100 ps). Upon increasing the distance between the energy donor (PPV) and acceptor layers by interposing bilayers of inert polyelectrolytes the dynamics tended to the PPV dynamics. The distance-dependence of the energy transfer efficiency (1/r2 or 1/r3) points to a resonant process (RET) with a plane-to-plane or plane-to-slab interaction. The non-radiative processes were also studied for varying dilutions of the PPV segments in a matrix of the polyelectrolyte poly(dodecyl methylamonium chloride) (PDAC). This dilution increased the quantum efficiency and led to longer lifetimes (500 ps) owing to a less effective non-radiative energy transfer. When an energy acceptor layer is present, energy transfer is more efficient for the most diluted systems, indicating that such transfer competes with internal processes in the emissive layer. The lifetime increased in films with low degree of conjugation, for which there was a stronger dependence on the emission wavelength. It is concluded that the LbL methodology was suitable to obtain photophysical film properties that could be controlled at the molecular level, and investigated with varied fluorescence spectroscopies.

Este estudo reúne os principais resultados de fluorescência estática e resolvida no tempo sobre a interação da porfirina meso-tetrakis (4-metilpiridil), na forma de base livre (TMPyP) e complexada com Zn2+ (ZnTMPyP), com vesículas de fosfolipídio. Adicionalmente foram utilizadas as técnicas de potencial zeta e espalhamento de luz dinâmico (DLS, do inglês \"dynamic light scattering\"). As vesículas de fosfolipídio foram formadas por dois conjuntos de fosfolipídios: saturados e insaturados. O primeiro grupo é formado pela mistura dos fosfolipídios zwiteriônico 1,2-dipalmitoil-sn-glicero-3-fosfocolina (DPPC) e aniônico 1,2-dipalmitoil-sn-3-glicero-[fosfo-rac-(1- glicerol)] (DPPG), a diferentes razões molares. Os estudos utilizando tais sistemas foram realizados abaixo (25oC) e acima (50oC) da temperatura de transição de fase gel-líquido cristalino destes fosfolipídios (~ 41oC). O segundo grupo é formado pela mistura dos fosfolipídios zwiteriônico 1-palmitoil-2-oleoil-sn-glicero-3-fosfocolina (POPC) e aniônico 1-palmitoil-2-oleoil-sn-glicero-3-fosfo(1-rac-glicerol) (POPG). Como a transição de fase destes dois fosfolipídios ocorre a temperaturas negativas, todos os experimentos foram realizados a 25oC (vesículas no estado líquido cristalino). Todos os sistemas foram preparados através do método de extrusão para a obtenção de vesículas grandes unilamelares (LUV, do inglês \"large unilamellar vesicles\"). As análises dos dados de fluorescência indicaram que a atração eletrostática entre os substituíntes (positivamente carregados) das porfirinas TMPyP e ZnTMPyP e o grupo das cabeças polares (camada de Stern) das vesículas de fosfolipídio desempenha um papel fundamental na associação da porfirina. A distribuição da TMPyP entre o meio aquoso (tampão) e as vesículas de fosfolipídio foi evidenciada pela coexistência de um tempo de vida de fluorescência mais curto (~ 5 ns) e outro mais longo (~ 9-11 ns), respectivamente. Baseado nos valores das constantes pré-exponenciais, estudos adicionais mostram que a distribuição acima é afetada pela concentração de sal na solução. Os resultados de supressão de fluorescência com o supressor iodeto de potássio (KI) indicaram que ambas porfirinas estão localizadas, preferencialmente, na região da camada de Stern. The fluorescence data analyses indicated that the electrostatic attraction between the porphyrin substituents (positively charged) and the polar head groups of the phospholipid vesicles (Stern layer) plays an important role on the porphyrin binding affinity. The distribution of TMPyP between the aqueous medium (buffer) and the phospholipid vesicles was characterized by the coexistence of a shorter (~ 5 ns) and a longer (~ 9-11 ns) fluorescence lifetimes, respectively. Based on the pre- exponential values, additional time-resolved experiments showed a redistribution of the porphyrin at increasing salt concentration. The quenching studies, using potassium iodide (KI) as quencher, indicated that both TMPyP and ZnTMPyP are preferentially located at the Stern layer region. This result is in agreement with the zeta potential and DLS findings, which demonstrated a partial neutralization of the negative charges at the vesicle surface due to the porphyrin association.

The epilepsies are a heterogenous group of symptom complexes, whose common features is the recurrence of seizures. There is no certain therapy for epilepsy. In order to promote new advances for the prevention of epilepsy the molecular mechanism of epileptic activities should be clarified.
In the present study the goal is to obtain information for molecular mechanism of epilepsy. To achieve this, molecular alterations from pentylenetetrazol (PTZ)-induced epileptic activities on rat brain tissue and cell membranes were investigated by Fourier Transform Infrared (FTIR) spectroscopy, Fourier Transform Infrared Microscopy and Atomic Force Microscopy (AFM). Moreover, the therapeutic role of an antiepileptic agent vigabatrin (VGB) on epileptic rat brain membranes were examined at molecular level.
For better understanding of the action mechanism of PTZ and an antiepileptic drug VGB in cell membranes we firstly studied at model level using multilamellar liposomes (MLVs). We investigated PTZ-DPPC MLVs interactions
in terms of lipid phase behavior, order and dynamics and nature of hydrogen bonding around its polar part, using Fourier Transform Infrared (FTIR) spectroscopy, Differential Scanning Calorimetry (DSC), Electronspin Resonans Spectroscopy (ESR) and Steady State Fluorescence Spectroscopy. According to our data, PTZ has no ability to interact with membrane lipids. On the other hand, the results of VGB-DPPC interactions showed that VGB strongly interact with the head group and/or the region near the head of membrane phospholipids.
The molecular investigation of PTZ-induced epileptic activities revealed that PTZ-induced seizures cause a decrease in the lipid and protein content, membrane fluidity and glycogen level. They stimulate alterations in membrane packing and the secondary structure of proteins as well as lipid peroxidation. In addition, our results show the transcription of early genes following high dose PTZ administration. All these molecular alterations variatins are only resulted from the consequences of epileptic activities not from convulsant agent PTZ itself.
The important finding is that, VGB restored some of the alterations by PTZ-induced epileptic activities on brain cell membrane. For instance, it restored membrane fluidity, lipid peroxidation, phospholipid degradation and changes in membrane organization. However, it was found that VGB has no significant effects on the changes in protein secondary structure.

Based on the pre- exponential values, additional time-resolved experiments showed a redistribution of the porphyrin at increasing salt concentration. The quenching studies, using potassium iodide (KI) as quencher, indicated that both TMPyP and ZnTMPyP are preferentially located at the Stern layer region. This result is in agreement with the zeta potential and DLS findings, which demonstrated a partial neutralization of the negative charges at the vesicle surface due to the porphyrin association.

This diploma thesis is oriented to analysis of physiological effect of synthetic estrogen ethinylestradiol (EE), which represents the main component of steroid-based substance used in hormonal contraception. From wide range of physiologically important protein molecules, which might be effected by this steroid, thesis focuses to the study of the sodium plus potassium activated, magnesium dependent adenosinetriphosphatase (Na+, K+ - ATPase), which is selectively inhibited by cardiac glycosides such as ouabain (g strophantine). Na+, K+ - ATPase represents an important plasma membrane bound enzyme, which catalyzes the active transport of sodium and potassium across plasma membrane. In the first part of this work, Na+, K+ - ATPase was determined by binding of radioactively labeled selective inhibitor of this enzyme [3H]ouabain, used for this purpose. In the second part of this work, plasma membrane fluidity was analyzed by steady-state fluorescence anisotropy of DPH. The effect of EE on [3H]ouabain binding was studied first under in vitro conditions by using human embryonic kidney cells (HEK293) which were cultivated for 24 hours in the presence of EE in tissue culture medium. The rise in the world population can be correlated with an increase in energy need. Fossil fuels are not going to able to cover this need in energy because not only are they limited, they also have a negative effect on the environment. A reason the more to switch renewable energy. One of the most popular renewable energy source is solar energy. The organic solar cell could be a low-cost, light-weight and flexible option for photovoltaics. This thesis will discuss the morphology of the active layer of an organic solar cell. The polymer poly(9,9-dioctylfluorenyl-2,7-diyl) and the fullerene derivate [6,6]-phenyl C61-butyric acid methyl ester were used as model components for the active layer. These two components were processed in different solvents, different ratios, different total concentrations and were either dip- or spin-coated on glass substrates. These samples were analyzed with atomic force microscopy, steady state and time resolved fluorescence and UV/Vis spectroscopy. The analysis show that the morphology of the films processed in chloroform and tetrahydrofuran would react very similar in α-phase and β-phase by dip- and spin-coated samples. Xylene would react the opposite as tetrahydrofuran and chloroform while ethylbenzene would react little with different samples. / De stijging in wereldpopulatie kan gelinkt worden met een stijging in energieverbruik. Het is niet aan te raden om fossiele brandstoffen te gebruiken voor deze energiestijging want niet alleen zijn ze beperkt aanwezig op aarde ook zijn ze niet goed voor het milieu. Een reden te meer om naar duurzame energie over te schakelen. Één van de meeste populaire energiebronnen is zonne-energie. Hierbij zou de organische zonnecel een goedkope, lichte en flexibele optie zijn. Deze thesis zal de morfologie van de actieve laag van een zonnecel bespreken. Het polymeer poly(9,9-dioctylfluorenyl-2,7-diyl) en het fullereen derivaat [6,6]-fenyl C61-butylzuur waren de twee model componenten voor de actieve laag. Deze twee componenten werden in verschillende oplosmiddelen, verschillende verhoudingen en verschillende totaal concentraties bereidt en werden vervolgens gedipcoated of gespincoated op glazen substraten. De stalen werden vervolgens geanalyseerd door atomic force microscopy, steady state en time resolved fluorescence en UV/Vis spectroscopy. De analyse toont dat de morfologie van de films bereidt in chloroform en tetrahydrofuraan gelijkaardig reageren in α- fase en β-fase bij gedipt- en gespincoaten stalen. Terwijl xyleen net omgekeerd reageert als chloroform en tetrahydrofuraan. Bij ethylbenzeen zou de fases maar heel weinig veranderen bij de verschillende stalen.
