Single-Step Analysis of Metabolites by Capillary Electrophoresis Using On-Line Sample Preconcentration with Chemical Derivatization

New strategies for integrating sample pretreatment with chemical analyses under a single format is required for rapid, sensitive and enantioselective analyses of low abundance metabolites in complex biological samples. Capillary electrophoresis (CE) offers a unique environment for controlling analyte/reagent band dispersion and electromigration properties using a discontinuous electrolyte system allowing for highly efficient separations to be achieved. A fundamental study of the thermodynamic and electrokinetic parameters influencing enantioselectivity in chiral CE separation is first examined. A new strategy for single-step analysis of low abundance metabolites via online sample preconcentration with chemical derivatization by CE (SPCD-CE) is then detailed within. In-capillary sample preconcentration serves to enhance concentration sensitivity via electrokinetic focusing of long sample injection volumes for lower detection limits, whereas chemical derivatization by zone passing of single or multiple reagents is used to expand detectability and selectivity, notably for enantiomeric resolution of metabolites lacking intrinsic chromophores. Together, on-line SPCD-CE can provide over a 100-fold improvement in concentration sensitivity, shorter total analysis times, reduced sample handling and improved reliability for a variety of biologically relevant amino acid and amino sugar metabolites, which is also amenable to automated high-throughput screening. The basic method development and optimization parameters relevant to SPCD-CE, including applications to bacterial metabolite flux and biomarker analyses are discussed. Insight into the mechanism of analyte focusing and labelling during electromigration by SPCD-CE is also presented, as well as future directions for continued research using this unique integrated analytical platform. / Thesis / Doctor of Philosophy (PhD)

Main goal of my thesis is evaluation of strategical management at examined enterprise and suggest a strategy for its future activity on a market with barreled water based on acquired findings. Theoretical part is dedicated to basic terms and selected analyses from area of strategical management. Practical part contains STEP analysis, Porter's model, analysis of results in particular functional areas, SWOT analysis applied to the chosen enterprise. Closing part of my thesis is dedicated to evaluation of present state of the enterprise and recommendation which steps should the enterprise take to gain a better competitive position on the market.

Aim of this study is to analyze the range of underwear, thermal underwear and hosiery and then to propose optimization measures. The theoretical part is devoted to methods which focus on analyzing the marketing environment and product mix. In the practical part, internal company data of EVONA a.s. served to analyze the product range by using Boston Consulting Group matrix and General Electric Business matrix. To illustrate the situation a survey was carried out and it helped in the final recommendations of the product range.

The aim of this diploma thesis is to elaborate an analysis of the external environment of a chosen company on bases of a special literature, own pieces of knowledge and gained information about the market. I have reached the results by the help of the Step analysis, the Porter{\crq}s model, the Competition analysis and the SWOT analysis. By the help of the Step analysis I tried to define factors that could influence the company even though they are outside the enterprise. On bases of the Porter{\crq}s model I have analyzed five competitive forces that influence our company namely new competitors, suppliers, customers, substitutes and a competition ring. By the help of the Competition analysis I have reviewed which companies concerned with the metal production and locksmithery are direct competitors of the company. Through the SWOT analysis method I tried to define inner strong and weak points and external opportunities and jeopardy. I have found convenient factors afterwards I have determined their importance by the help of the method of the Geminate comparison. In conclusion I have evaluated data. On their bases I have drawn up recommendations for the future development and I have suggested convenient strategies. Those strategies were strategy of the Market Development, Market Penetration, Concentric Diversification and strategy of Creation of the competitive advantage.

The main goals of the thesis were to prepare an analysis of activities of strategic management, to improve the synergy between strategic and operational management, to enhance the quality of strategic decisions in selected company. The first step was to study and comparison of Czech and foreign professional literature and make literary summary that gives the reader into the issue of strategic management. The practical part is performed by first analysis of the current state of strategic and operational management in selected company through the following analyzes: STEP analysis, industry analysis, financial analysis, guided interview with the director of the company. Based on the results of previous analyzes were determined strengths and weaknesses and the opportunities and threats of the company. Their influence is compared in the SWOT analysis. The last part presents proposals for measures that would lead to improving the coordination of strategic and operational management and an overall improvement in key areas of the company.

The main target of the thesis was to elaborate a project documentation that would focus on the solution of the conflicts caused by the recreational exploitation of the area. However, the results of preceding bachelor thesis (guestioning of visitors and situation analysis), interviews and of SWOT, PEST analyses pointed to the existence of several conflict issues. Based upon the current situation of the area, strategic targets, measures and tourism development plan were presented. The plan itself then offers an overview of the solutions and decisions that should contribute to the coordinated tourism development in the area. Presented project is dealing with the issue of insufficient quality of tourism infrastructure and it covers campsite reconstruction.
