EVALUATION OF CELLULOLYTIC ENZYMES FROM A NEWLY ISOLATED BREVIBACILLUS SP. JXL; AND OPTIMIZATION OF COSLIF PRETREATMENT VARIABLES OF SWEET SORGHUM BAGASSE USING A RESPONSE SURFACE METHOD

The first part of the dissertation presented a potentially novel aerobic, thermophilic, and cellulolytic bacterium identified as Brevibacillus sp. Strain JXL which was isolated from swine waste. Strain JXL can utilize a broad range of carbohydrates including: cellulose, carboxymethylcellulose (CMC), xylan, cellobiose, glucose, and xylose. In two different media supplemented with crystalline cellulose and CMC at 57°C under aeration, strain JXL produced a basal level of cellulases as FPU of 0.02 IU/ml in the crude culture supernatant. When glucose or cellobiose was used besides cellulose, cellulase activities were enhanced ten times during the first 24 h, but with no significant difference between the effects caused by these two simple sugars. After the end of the 24 hour period, however, culture with glucose demonstrated higher cellulase activities compared with that from cellobiose. Similar trend and effect on cellulase activities were also observed when glucose or cellobiose served as a single substrate. The optimal doses of cellobiose and glucose for cellulase induction were 0.5 and 1%. These inducing effects were further confirmed by scanning electron microscopy (SEM) images, which indicated the presence of extracellular protuberant structures. These cellulosome-resembling structures were most abundant in culture with glucose, followed by cellobiose and without sugar addition. With respect to cellulase activity assay, crude cellulases had an optimal temperature of 50°C and optimal pH range of 6-8. These cellulases also had high thermotolerance as demonstrated by retaining more than 50% activity after 1 h at 100°C. In summary, this is the first study to show that the genus Brevibacillus may have strains that can degrade cellulose. In the second part of the dissertation, the effect of Cellulose- and Organic-Solvent based Lignocellulose Fractionation (COSLIF) (Zhang, Y.-H. P.; Ding, S.-Y.; Mielenz, J. R.; Elander, R.; Laser, M.; Himmel, M.; McMillan, J. D.; Lynd, L. R. Biotechnol. Bioeng.2007, 97 (2), 214−223) pretreatment conditions on sweet sorghum bagasse (SSB) feedstock was studied using Response Surface Methodology (RSM). Batch experimental matrix was set up based on response surface method's central composite design in two factors to determine the effects of reaction time and temperature on the yield of simple sugars after a sequential pretreatment-enzyme hydrolysis process. Accordingly, changes in delignification, total reducing sugar (TRS) yield, glucan retention, digestibility and overall sugar yields resulting from various combinations of reaction times and temperatures were determined. The results suggested that both pretreatment temperature and reaction time were significant factors, although temperature was more so than reaction time. COSLIF pretreatment conditions of 50°C and 40 min were found to be the optimum pretreatment conditions for the saccharification of SSB. At the end of pretreatment and enzymatic hydrolysis, maximum values of 51.4% delignification, 85% overall glucose yield, and 44% overall xylose yield at an ACCELERASE®1500 loading of 0.25 mL/g sweet sorghum bagasse were achieved. Optimum ACCELERASE®1500 dosage of 0.1 mL/g of sweet sorghum bagasse was identified which resulted in an overall glucose yield of 82.2%±1.05. An effort has also been made to prescribe predictive models which represented the correlation between independent variables (reaction time and temperature), and dependent variables (delignification, and overall glucose yield) using RSM. The significance of the correlations and adequacy of these models were statistically tested for the selected objective functions. The outcomes suggested very competent and statistically adequate regression models which provided quantitative information both for delignification and overall glucose yield for the batch experiments studied.

Preharvest ethylene and postharvest curing effects on baked sweet potato (Ipomoea batatas L. Lam) quality and prevention of sprouting on cured sweet potatoes

The effect of preharvest foliar applications of ethephon and postharvest curing on baked sweet potatoes (Ipomoea batata) were analyzed. Ethylene had negative effects on appearance of baked roots while curing increased the total phenolics in sweet potatoes. Panelists preferred the non-ethylene treated and green or cured over ethylene treated sweet potatoes with respect to texture, color and flavor. No differences in chemical or color properties were found between treatments. The effect of curing time (0, 7, 14 days) on baked sweet potatoes was studied. Curing caused more browning on flesh of sweet potatoes. Moreover, panelists preferred the texture of the green over the cured roots but did not find any differences in flavor sensory quality. The effect of hot water treatment on sprouting and spoilage of cured sweet potatoes was investigated. The most effective treatment was at 53-56 °C for less than 10 min, resulting in less than 60% (control=80%).

Generation of Biodiesel and Carotenoids from Rhodotorula Glutinis using Sweet Sorghum Juice

The growth of Rhodotorula glutinis in sweet sorghum juice in three levels of three factors of temperature, carbon to nitrogen ratio and pH was evaluated. Accompanying of this growth was the generation of lipids converted to fatty acid methyl ester (FAMEs) and carotenoids. The optimized condition for maximum biomass and carotenoid accumulation was determined to be at 25C, pH of 5.5 and carbon to nitrogen ratio of 10. This condition yielded 22.7 g/L biomass with specific growth rate of 0.213 hr-1. At this condition the carotenoids generation was also maximum with 2.6 mg/gram biomass, comprising of torularhodin, beta-carotene and torulene. The accumulation of lipids following generation of biodiesel was highest at same temperature and pH but carbon to nitrogen ratio of 70, generating 96.3 mg of FAMEs/gram of biomass containing methyl ester of oleic acid, linoleic acid, palmitic acid, stearic acid and linolenic acid.

Impact of daily sweet potato ingestion on alterations to the gut microbiome, cholesterol, and exercise performance

The importance of the gut microbiome is being explored in relation to multiple facets of health and performance. The purpose of this study was to explore the impact of frequent sweet potato ingestion on alterations of the gut microbiome which might in turn alter cholesterol levels and exercise performance. Thirteen recreationally trained males had fecal and blood samples collected and exercised to fatigue with and without exogenous carbohydrate ingestion prior to and following 42 days of daily sweet potato ingestion. Bacterial communities were extracted from fecal samples and bacterial DNA were sequence. Blood samples were analyzed for total cholesterol, low-density lipoprotein-C, high-density lipoprotein-C, and triglycerides. Performance was analyzed comparing changes in time-to-volitional fatigue with and without carbohydrate ingestion. Changes were observed in microbial abundance following 42 days of sweet potato ingestion but no differences in cholesterol or cycling fatigue with and without carbohydrates.

Pearl Millet Nutritional Quality and Fertilization of Sweet Corn in Senegal

Agricultural production is the main source of income and major employer in many countries in Africa, including Senegal. Commercial sweet corn (Zea mays L. ssp. saccharata) production in Senegal is increasing in response to global marketing opportunities and offers producers the ability to increase income and diversify the cropping base. Production of optimum sweet corn yield and quality depends on adequate nutrient supply, particularly, nitrogen (N). Current N recommendations are based on recommendations specific to corn for grain. This study aimed to identify tools to estimate sweet corn N status and determine the most appropriate fertilizer dosage for sweet corn. Non-destructive remote sensing tools and ion exchange resin membranes (IEMs) were used to evaluate the effect of fertilizer dose. No differences in ear yield or yield components, normalized difference vegetation index (NDVI) values, biomass, N uptake or leaf N concentration due to fertilizer dose were detected at Ndiol. However, significant relationships existed between NDVI values and yield, biomass, and N uptake at the V9 growth stage. Only yield was affected by fertilizer dose at Sangalkam, and no consistent relationships were found between chlorophyll meter readings and others measured parameters. Treatment differences due to fertilizer dose for available NH4+ at V9 (Ndiol), and NO3- at V5 (Sangalkam) were found, however further research is needed to fully evaluate the usefulness of IEMs to measure available soil N. Based on these studies, sweet corn fertilizer rates should likely be based on 75% of the dose applied to field corn, however more work is needed to confirm this finding. Pearl millet (Pennisetum glaucum (L) R. Br.) is the most widely grown staple crop in Senegal. Introduction of drought tolerant millet genotypes has helped mitigate the effect of increased water shortage in the region, but little is known about the nutritional composition of these genotypes. Our objective was to compare millet grain nutritional composition among and between putative drought tolerant and drought sensitive pearl millet lines under drought stress and well-watered conditions. One field experiment was conducted in 2014 at the National Center for Agronomic Research (CNRA) of Bambey, Senegal (16°30' and 16° 28' N; 15o44' and 15o42' W). The experiment utilized a split-plot design with four replications. Water regime was the main plot experimental factor while genotype, a total of 20 was the sub-plot. Pearl millet genotypes were divided into three contrasting groups based on drought tolerance for comparisons. Water stress did not affect 100-grain weight, test weight, protein, soluble protein, starch, sugars, amino acids or vitamin B2 content of grains among VPD-groups. Accumulation of these constituents of pearl millet grain appear to be genetically controlled and are probably not affected by late drought stress. However, differences were noted among genotypes as the sensitive VPD-group accumulated greater soluble protein, starch and soluble sugars (except sucrose) than the tolerant and medium VPD-groups. The tolerant VPD-group, however, accumulated greater protein and vitamin B2 content. Arginine, proline and serine content was greater in the sensitive VPD-group, while lysine, aspartic acid, and glutamic acid were greater in the tolerant VPD-group. Glycine, histidine, threonine, alanine, tyrosine, valine, methionine, leucine, isoleucine, and phenylalanine were relatively equal in tolerant and sensitive VPD-groups. Calcium and Na levels were affected by water stress in the sensitive VPD-group, but differently. Calcium content was greatest for the sensitive group under drought stress, while sodium was the lowest. Iron accumulation in sensitive VPD-group increased under water stress. Potassium decreased for all VPD-groups under stress, while across water regime, K levels in the drought-sensitive group were lower. Selection for drought appears to effect many of the nutritional constituents of pearl millet grain, however many of these differences appear to be directly related to parameters known to effect plant water relations. / Master of Science
