Detekce a monitoring potenciálně toxických sinicových lipopeptidů

The aim of this study was to design and optimize new PCR primers for detection of potential cyanobacterial producers of cytotoxic lipopeptides puwainaphycins and minutissamides in environmental samples. Samples from two distinct localities were tested, as suggested based on preliminary data. The first set of samples consisted of cyanobacterial soil biofilms from sheep pastures affected by Alveld illness in Norway. The other one contained samples of planktic cyanobacaterial blooms from Protected Landscape Area Třeboň and its vicinity. Three different approaches were used for evaluation of the presence of cyanobacterial lipopeptide producers: microscopy, PCR with the designed primeres, and liquid chromatography-mass spectrometry analysis. Results of this study confirmed the specificity of the newly designed PCR primers. The presence of producers of puwainaphycins/minutissamides was proven at both tested localities.

Dynamika nadzemní biomasy ostřicového slatiniště / Dynamics of aboveground biomass of a sedge fen

This thesis is part of Project of the Grant Agency of the Czech Republic No P504/11/1151, focused on the role of plants in the balance of carbon dioxide and the other greenhouse gases produced in the ecosystem of a sedge fen, which is situated on the study site Wet Meadows near Třeboň. The thesis deals with the growth dynamics of the dominant sedge, Carex acuta. The samples were taken using the method of successive harvests near the automatic meteorological station of Czech Globe, Academy of Sciences of the Czech Republic. During the growing season nine harvests were accomplished on 25.3, 15.4, 6.5, 5.6., 9.7., 5.8., 9.9., 21.10., 25.11.. On each date eight replicates were taken, i.e., a total of 72 samples within the vegetation season. The seasonal dynamic of the above ground biomass was compared between C. acuta and the other plant species on the experimental area. Both live and dead biomass, was harvested and subsequently sorted to single botanic species, dried, weighed and the values were processed using the MS Excel tables. The seasonal maximum of aboveground biomass of all plant species (both live and dead parts) was found on 9.7. (1452,72 g.m-2). Among plant species the highest values of aboveground biomass were attained by Calamagrostis canescens (1257,93 g.m-2) and C. acuta (1163,49 g.m-2). C. acuta displayed the highest density of all shoots on 15.4 (300 m-2),and the highest average weight of one shoot. The maximum length of the longest live shoot of Carex acuta was very consistent among the measurememnts.

This thesis is part of Project of the Grant Agency of the Czech Republic No P504/11/1151, focused on the role of plants in the greenhouse gas budget of a sedge fen. The thesis deals with the growth dynamics of the dominant sedge, Carex acuta L., on the study site, Wet Meadows near Třeboň, Czech Republic. The seasonal changes in aboveground biomass were followed using successive harvests during vegetation seasons of 2010 and 2011. The seasonal maximum of aboveground biomass of Carex acuta L. (both live and dead parts) was 321,44 g.m-2 and 357,97 g.m-2 in 2010 and 2011, respectively. The seasonal maximum of total aboveground biomas sof all species was 558,22 g.m-2 and 522,38 g.m-2 in 2010 and 2011, respectively. The seasonal maximum of shoot density was 485 ks.m-2 and 435 ks.m-2 in 2010 and 2011, respectively.

Druhé bydlení na Třeboňsku jako předstupeň amenitní migrace / Second home in Třeboň area as the first stage for amenity migration

The diploma thesis deals with two contemporary phenomena, second home and amenity migration. These phenomena often overlap in the conditions of the Czech Republic, so it is difficult to find a clear border between them. Second home can be understood as a complex of phenomena and processes related to a real estate, which serves as a temporary residency for its owner or user, who uses this real estate especially for recreation purposes. Amenity migration can be defined as a migration of inhabitants with the aim to achieve a better quality of life, better health, relaxation or potentially entertainment as well. It concerns migration of people for non-economic reasons and the direction of amenity migration is predominantly from cities to the countryside. Unlike second home, it is not any form of a temporary residency, but a permanent accommodation. The main goal of the diploma thesis was to explore the phenomenon of second home in the region of Třeboňsko and to evaluate its potential for amenity migration. The goal of the work was realised on the basis of an analysis of the data of the Czech Statistical Office and the Czech Office for Surveying, Mapping and Cadastre, by means of both field research and a questionnaire survey with the subsequent evaluation.

The thesis deals with se the usage of animation method for medieval art mediation for children´s audience. The aim of the animation of the Resurrection painting by Master of the Třeboň Altarpiece is to effectively encourage a continued interest in art and history in children at a given age group. The content of the theoretical part is an expected scope of knowledge needed for an ideal realization of the animation. The practical part describes the creative art mediation for children and the concluding reflections are also an integral part of it. It is a big challenge for all museum, gallery and pedagogical professionals to present cultural heritage. The thesis also tries to point out the fact that preliminary animation work is for authors very demanding. It is necessary to completely avoid a close specialization, to work interdisciplinary and to combine all specialized theoretical and practical knowledge in an effective way.

The objective of this Diploma Thesis is to draft a project of cultural tourism development in Třeboň region. Quantity of ponds and water courses are typical for this territory, and it also parades with a number of water mills that are architecturally or technologically valuable in terms of historic monuments. Despite their value they are not used as touristic sites and that is why to draft a project of Museum of miller's craft Mill of Spolí is a constituent of the Thesis. The sense and the objective of the project are to provide public access into the existing building of the Mill of Spolí as a technical monument. Museum exposition and practical demonstrations of mill machinery should improve the visitors' knowledge and it should inform them about the history of miller's craft and about the history of the Mill of Spolí itself. By these means the fame of Czech miller's craft as one of the traditional elements of country settlement should be reminded.

Spa in the Czech Republic With a focus on spa Jeseník, Luhačovice, Třeboň and Františkovy Lázně As the subject of my work I chose the Czech Republic spa. I look at the spa environment as a social phenomenon that affects many areas of social life and has a great tradition in Czech culture. With its unique atmosphere and the composition of the clients who visit them, they are more than just popular places by tourists or health facilities but they have also influence on the life of the living there and spa visitors. The work is intended to serve as a general base that could be suitable for further exploration of this area, and therefore tries to cover as many areas of the issue as possible. I try to work with examples from history to point out the role of the spa in the past and how they gradually transformed into what they are today. I also look at services that are offered at the spa and the structure of clients who come to the spa. My field of interest was also the formation of spa towns and life in them. This area is distinguished from others for a large number of changing spa clients. The dissertation is dealing with work of Viennese architect and builder from the first half of the 18th century Antonio Ehorto Martinelli in the Schwarzenberk's service in the years 1722-1732. It contains an index of buildings, that Martinelli was working on for Schwarzenberks, including contentious and mistakenly ascribed buildings. The collected information is based on study of extant archival collections, on field research of separate buildings and on study of the relevant literature. The main goal was an evaluation of separate Martinelli's architectural fruitions relating to history of art, and furthuremore an attempt to compare them both within the bounds of the whole architect's work and within the bounds of contemporary Czech and Austrian architecture. The knowledge of work of A. E. Martinelli in the Shwarzenberk's service was markedly extended on the occasion of dealing with the settled topic and moreover a few little and even a few bigger new pieces of knowledge appeared, e.g. information about concerned gangers, building development of the studied objects out of the settled period etc.
