Alternative techniques for the production and manipulation of ultracold atomsBruce, Graham D. January 2012 (has links)
This Thesis contains details of the construction and characterisation of a compact apparatus for the cooling of ultracold atoms to quantum degeneracy, and their manipulation in flexible holographic optical traps. We have designed and built two iterations of this apparatus. The first version consists of a stainless steel single-cell vacuum chamber, in which we confine ⁸⁷Rb and ⁶Li or ⁷Li in a Magneto-Optical Trap. We characterise the alternative methods of pulsed atomic dispenser and Light Induced Atomic Desorption (LIAD) to rapidly vary the background pressure in the vacuum chamber with the view to enabling efficient evaporative cooling in the single chamber, loading MOTs of up to 10⁸ atoms using pulsed dispensers. The LIAD is found to be ineffective in loading large MOTs in this setup, while the pulsed dispensers method gradually increases the background pressure in the chamber over time. Based on the results of this first iteration, we designed and built a second single-chamber apparatus for cooling of ⁸⁷Rb to quantum degeneracy. The LIAD technique was used to successfully load MOTs containing 8x10⁷ atoms in this single pyrex cell with a rapidly-varying background pressure. The lifetime of an atomic cloud loaded from the MOT into a magnetic trap increased by a factor of 6 when LIAD was used. The holographic optical traps for cold atoms are generated using a Spatial Light Modulator, and we present our novel method for improving the quality of holographic light patterns to the point where they are suitable for trapping ultracold atoms using a feedback algorithm. As demonstrations of this new capability, we show power-law optical traps which provide an efficient, reversible route to Bose-Einstein Condensation and a dynamic ring trap for the investigation of superfluidity in cold atoms.

RECOVERY OF THE EASTERN WOODRAT IN SOUTHERN ILLINOIS: DISTRIBUTION, DEMOGRAPHICS, AND NONINVASIVE MONITORINGGooley, Aaron Clinton 01 May 2019 (has links)
Woodrats (Neotoma spp.) are imperiled in the north-central and north eastern United States. In Illinois, eastern woodrats (N. floridana) experienced range reductions and population bottlenecks over the past century. Hypothesized reasons for the decline of many woodrat populations that inhabit rock outcrops in the eastern United States include parasitism by raccoon roundworms (Baylisascaris procyonis), hard mast shortages, owl predation, and reductions in crevice availability for nest construction. During 2004-2005, the isolated remnant populations along the Mississippi bluffs in southwestern Illinois were genetically augmented with 47 eastern woodrats from Arkansas and Missouri resulting in 40% admixture within the largest population. In 2009, a strong windstorm created canopy gaps and woody debris throughout this area, potentially improving habitat for eastern woodrats. During 2003–2009, 422 eastern woodrats were reintroduced to 5 sites in the southeastern Illinois, and 172 eastern woodrats to 2 southern Illinois state parks during 2013–2014. These reintroductions are the only woodrat reintroductions to date with >50 individuals released per site. Most previous woodrat reintroduction attempts have released small numbers of individuals (10–15 per site and 10–54 total) and either failed to establish populations or required frequent management for populations to persist. My objectives were to (1) investigate the status of augmented eastern woodrat populations in southwestern Illinois, (2) evaluate the success of the southern Illinois reintroductions, (3) investigate whether eastern woodrats demographics within a reintroduced metapopulation could be predicted by factors underlying hypothesized reasons for woodrat declines, and (4) develop and evaluate noninvasive alternatives to live-trapping and sign surveys for monitoring woodrat populations.

Photochromism has been investigated extensively during recent years. The large interest for information storage in memory applications is associated with the bi-stable character of the photochromism phenomena. In molecular photochromics, two isomers with different absorption spectrum can be obtained according to the specific wavelength of the light exposure. This reversible transformation process can be considered as optical writing/erasing step of a memory. Here we first report the absorption spectra of solid-state films based on the blends consisting of PC molecules, the spirooxazine 1,3-dihydro-1,3,3-trimethylspiro[2H-indole-2,3’-[3H]phenanthr[9,10-b](1,4)oxazine] (PIII, Sigma-Aldrich, 32,256-3) and a polymer matrix host, poly(2-methoxy-5(2’-ethylhexyloxy)-1,4-phenylenevinylene) (MEH-PPV). The bi-stability in conjugated polymer matrix is studied by following the time evolution of the optical properties of the blends. Thereafter, the electrical performance of PC-polymer diodes is characterized and reported. While the PIII molecules in the blend bulks are switched to their low energy gap state, forming external energy levels above the valence band of MEH-PPV, the injected charges (hole-dominated) will be trapped by the low energy gap isomer of PIII and that leads to current modulation. PIII molecules can be switched between two energy gap states upon the photo-stimulation, and the I-V characteristics of the device can also be controlled reversibly via the photoisomerization. The retention time of the diode’s electrical switching fits quite well with the absorption characteristics of the blend films; this correspondence builds a good link between the film property and the device behavior. Furthermore, we observed a two-trap system in the blend diodes from the I-V curves, and a model is proposed which can explain the schematic concept of the trap-limited current modulation. To combine the knowledge and information from the investigations above, we tested a novel device design based on a bi-layer of the PC and polymer materials, and the promising result for future work is presented in the end.

The Double-TOP trap is a new type of magnetic trap for neutral atoms, and is suitable for Bose-Einstein condensates (BECs) and evaporatively cooled atoms. It combines features from two other magnetic traps, the Time-averaged Orbiting Potential (TOP) and Ioffe-Pritchard traps, so that a potential barrier can be raised in an otherwise parabolic potential. The cigar-like cloud of atoms (in the single-well configuration) is divided halfway along its length when the barrier is lifted.
A theoretical model of the trap is presented. The double-well is characterised by the barrier height and well separation, which are weakly coupled. The accessible parameter space is found by considering experimental limits such as noise, yielding well separations from 230 [mu]m up to several millimetres, and barrier heights from 65 pK to 28 [mu]K (where the energies are scaled by Boltzmann�s constant). Potential experiments for Bose-Einstein condensates in this trap are considered.
A Double-TOP trap has been constructed using the 3-coil style of Ioffe-Pritchard trap. Details of the design, construction and current control for these coils are given. Experiments on splitting thermal clouds were carried out, which revealed a tilt in the potential. Two independent BECs were simultaneously created by applying evaporative cooling to a divided thermal cloud.
The Double-TOP trap is used to form a linear collider, allowing direct imaging of the interference between the s and d partial waves. By jumping from a double to single-well trap configuration, two ultra-cold clouds are launched towards a collision at the trap bottom. The available collision energies are centred on a d-wave shape resonance so that interference between the s and d partial waves is pronounced. Absorption imaging allows complete scattering information to be collected, and the images show a striking change in the angular distribution of atoms post-collision. The results are compared to a theoretical model, verifying that the technique is a useful new way to study cold collisions.

Rac2 is Required for Formation of Extracellular Traps in NeutrophilsLim, Byung Hyun 25 August 2011 (has links)
Recently, it was found that pathogens are trapped and killed by neutrophil extracellular traps (NETs). The role of Rac small GTPases is explored in the formation of NET using neutrophils lacking Rac1, Rac2 or both isoforms. NET formation was observed in both wild-type and Rac1-null neutrophils. In contrast, NET formation was markedly impaired in cells lacking either Rac2 or both Rac2 and Rac1. The defect in NET formation in Rac2-null cells was rescued in the presence of exogenous reactive oxygen species sources, suggesting that Rac2-mediated ROS generation is required. In addition, the role of nitric oxide in NET formation is assessed. Blocking NO production with the nitric oxide synthase inhibitor L-NAME significantly reduced NET formation. Moreover, Rac2-null cells produced significantly less NO than Rac1-null cells or their wild type counterparts. Our data suggest that Rac2 is essential for NET formation via pathways involving both ROS and NO.
