Thesis has been focused on the development of corpus luteum and embryos in Holstein cows. Corpus luteum and embryos was detected and measured by ultrasonography. The aim of thesis was determine the effect of corpus luteum and other factors on the size of embryos. Results showed that the size of corpus luteum has no influence on the size of embryos. This research area is still scarce and further experiments will be needed to deepen understanding of the factors affecting embryo development.

Regrese koronární aterosklerózy při hypolipidemické terapii / The coronary atherosclerosis regression during hypolipidemic therapy

Background: There is no study focusing on changes of coronary atherosclerosis during dual hypolipidemic therapy with statin and ezetimibe. Methods: 107 patients with stable angina were enrolled and the final analysis was performed in 89 patients. Randomization was 1:1 to the group A (atorvastatin 80mg and ezetimibe 10mg) and to the standard group S. Treatment period was 12 months. Results: Changes of percent atheroma volume (PAV) were -0,4% in group A and + 1,4% in group S, p=0,014. Combine atherosclerosis regression (increase of lumen volume together with decrease of PAV) was found more frequent in group A (40,5%) than the group S (14,9%), p=0,007. The target LDLc level < 2mmol/l, presence of at least four of five atherosclerotic risk factors, and decrease of VCAM level were independent predictors for plaque regression. There were no significant differences in plaque composition between the two groups over the duration of the study. However during analysis the two groups together, fibrous and fibro-fatty tissues decreased and dense calcification and necrotic core increased during follow-up. Conclusion: The dual hypolipidemic therapy starts atherosclerosis regression. The function of the machine is relatively difficult and analysis of it is complicated in time. That is why I am introducing only evolution of design and ergonomy. The aspire was to approach medical diagnostic machines to patients and raise the positive thinking to them. The colour alternation is choose with the respect to psyche of examine human at the same time.

This master's thesis is concerned with the basis of ultra-acoustics and it explains the application of Doppler effect in medicine as a diagnostic method used for measuring blood flow velocity in blood vessels. The thesis also presents Doppler systems used for diagnostic purposes. It describes the principles of these systems, shows accompanying flowcharts and explains the function of these systems. There is also included the system design of the whole ultrasonic blood flow velocity meter. The system has been designed for the specified parameters with continuous 4 MHz operating frequency, generated intensity of ultrasound 100 mW/cm^2 and diameter D-shaped transmitting transducer 8 mm. In the design of personal solution there are described all the function blocks of the ultrasonic meter. There is also described the choice of appropriate components and designed all the required supply voltages. All the function blocks of the system are experimentally verified. The system has been designed with acoustic intercept and the resultant velocity curves are displayed on software oscilloscope on PC.

Porovnání účinnosti metod extrakce tropických dřev na základě analýzy získaného extraktu a jeho vlivu na dřevokazné houby

The thesis compares the efficiency of two extraction methods: using the FexIKA apparatus and using ultrasound. To compare the extraction methods, extracts from merbau (Intsia bijuga O.Ktze.) and jatoba (Hymenaea courbaril L.) were created. These tropical woods were ground and extracted. From the extracts, three concentrations of protective substances were created. The protective substances were tested to determine the content of extractive compounds and polyphenolic compounds. The obtained protective substances were impregnated into the wood of common beech (Fagus sylvatica L.) and pine (Pinus sylvestris L.). The impregnated beech and pine samples were tested to determine the protective effectiveness against Trametes versicolor L. and Coniophora puteana P. Karst. From the acquired data, the loss of mass of the wood was assessed. The results showed that only samples impregnated with the protective substance with concentration of 3 showed adequate protection. Analysis of the protective substances showed a higher yield when using FexIKA on Merbau wood. Jatoba wood extraction showed a slightly higher yield when extracting using ultrasound. In general, the samples impregnated with extracts obtained using FexIKA showed lower mass loss than samples impregnated with extracts obtained using ultrasound.

Využití cytogenetických a molekulárně cytogenetických metod v prenatální diagnostice / Application of cytogenetic and molecular cytogenetic methods in prenatal diagnosis

Foetal anomalies found on ultrasound increase the probability of occurrence of chromosomal abnormalities. They cause about one quarter of all abortions and stillbirths and many of inborn defects in newborns. Karyotype analysis is number one method in prenatal diagnosis whereas array CGH is often used as a verification and supplemental method. The aim of this work was to prove that array CGH gives additional chromosomal findings to karyotypes and could substitute conventional karyotyping as a primary examination method in foetuses with ultrasound findings. We examined 45 prenatal samples using both methods. These samples were referred for invasive examination because of abnormal ultrasound findings. Karyotype analyses found two abnormalities in two (4,4 %) patients and array CGH identified aberrations in five (11,1 %) foetuses whereas both anomalies detected by karyotypes were discovered by array CGH too. This means that array CGH identified about 6,7 % more aberrations than karyotype. Our results correspond with scientific articles which refer that array CGH should replace karyotype not only in postnatal examinations but even in prenatal diagnosis.
