Diversidade genética de enterobactérias endofíticas de diferentes hospedeiros e colonização de Catharantus roseus por endófitos expressando o gene gfp. / Genetic diversity of endophytic enterobacteria from different hosts and colonization of Catharantus roseus by endophytes expressing gfp gene.

Bactérias endofíticas são aquelas que habitam o interior de tecidos vegetais, sem causar dano aparente aos mesmos, além de desempenhar funções importantes no processo de adaptação das plantas. Especial interesse tem sido dado a tais bactérias devido ao seu potencial no controle biológico. Por isso, é muito importante estudar a diversidade genética de endófitos, além de avaliar o impacto da introdução de endófitos geneticamente modificados no ambiente. Estudos vêm sendo feitos nesse sentido, mas não com bactérias endofíticas da família Enterobacteriaceae. Desta forma, o presente trabalho teve como objetivos estudar a diversidade genética de bactérias endofíticas da família Enterobacteriaceae isoladas de plantas de cacau, cana-de-açúcar, citros, eucalipto e soja, utilizando diferentes técnicas moleculares. Análises por ARDRA e seqüenciamento do rDNA 16S identificaram 20 haplótipos e revelaram que os isolados pertenceram aos gêneros Enterobacter, Erwinia e Pantoea, sendo este último o mais freqüente. Tais estudos revelaram ainda que a diversidade dos isolados variou de acordo com a planta hospedeira. A técnica de BOX-PCR foi também utilizada para avaliar a diversidade dos isolados. Um total de 23 diferentes OTUs (operational taxonimic units) obtidas indicaram que o total de isolados avaliados compreenderam pelo menos 23 espécies diferentes. Dois isolados foram transformados com pPAgfp, um plasmídio contendo os genes de resistência ao antibiótico ampicilina e o gene gfp, que codifica a proteína verde fluorescente. Tais isolados foram inoculados em plântulas de Catharantus roseus (vinca) e foi feito reisolamento de bactérias endofíticas em dois períodos diferentes após a inoculação. O impacto desta inoculação na diversidade da comunidade microbiana natural de vinca foi avaliado utilizando-se a técnica de ARDRA, a qual mostrou que os endófitos expressando gfp colonizaram as raízes e caules das plantas inoculadas, sem causar qualquer sintoma de doença. Além disso, os colonizadores endofíticos não levaram à diminuição da diversidade da população microbiana natural de vinca. Os resultados obtidos poderão contribuir para a compreensão sobre a interação entre Enterobacteriaceae endofítica e planta hospedeira, além de ajudar a responder questões sobre o papel ecológico dos endofíticos e seu potencial biotecnológico. / Endophytic bacteria have been defined as those that reside within living plant tissues, or extracted from inner plant parts without causing apparent damage to them. They also are able to play an important role in the process of plant adaptation. There is an increasing interest to endophytic bacteria and its potential in the biological control and many studies has been done in order to evaluate the diversity and the impact of endophytes and genetically modified endophytes (GME) released into environment. In this way, information about the diversity of endophytic bacteria has been obtained, except for bacteria exclusively from Enterobacteriaceae family belonged to different host plants. Thus, the aim of the present work was study the diversity of Enterobacteriaceae bacteria isolated from citrus, cocoa, eucalypti, soybean and sugar cane by different molecular approaches. The 16S rDNA of each isolate was amplified by PCR and the isolates were grouped into 20 clusters by analysis of restriction patterns of the PCR-amplified 16S rDNA (ARDRA). These analysis showed a variety of organisms, with 5 different genera encountered: Pantoea was most frequently encountered followed by Enterobacter and Erwinia, which isolates presented the great bacterial diversity according to host plants. Through cluster analysis of the BOX-PCR technique profiles, 23 different OTUs (Operational Taxonomic Units) were distinguished, the presence of 23 OTUs could indicate that isolates comprised at least 23 different species. Two isolates were transformed with pPAgfp, a plasmid harboring the ampicillim resistance gene and the gfp gene, which encodes for the green fluorescent protein. These two isolates were inoculated in seedlings of Catharantus roseus (vinca) and re-isolation of endophytic was performed in two times after inoculation. The impact of endophytes inoculation was evaluated by using the ARDRA technique. It showed that endophytes expressing gfp colonized roots and shoots of inoculated plants without causing any symptom of disease. Besides, the endophytes colonizers do not decreased the diversity of the vinca’s natural microbiota. The results obtained here provided important insights into the endophytic Enterobacteriaceae-host relationship that will be useful for further answer basic questions about the ecological role of the endophytes and its biotechnological potential.

V. Hojgrová: Alkaloids of Vinca minor L. and their biological activity VIII. Diploma thesis, Charles University, Faculty of Pharmacy in Hradec Králové, Department of Pharmaceutical Botany. Number of pages 69. This diploma thesis deals with the isolation of alkaloids from Vinca minor L. from the family Apocynaceae. Separation of alkaloids from the selected fraction (VM 215-258) or from their subfractions (VM 34-41, VM 86, VM 87-113) was performed by preparative TLC. Two pure alkaloids were isolated from the subfraction (VH 34-41). The first VH-1 alkaloid not yet isolated and the second VH 2 alkaloid: (-)-raucubainin. Alkaloids were identified by EI-MS, LC-MS, NMR and optical rotation and were compared with data in the literature. Isolated alkaloids were tested for acetylcholinesterase (AChE), butyrylcholinesterase (BuChE) and prolyloligopeptidase (POP) inhibitory activity and for cytotoxicity. Both substances did not show significant cholinesterase inhibitory activity IC50 against AChE after measurement, only (-)-raucubainin showed a slight activity against BuChE (IC50 = 94 ± 7 μM), VH-1 was found to be inactive (IC50 > 100 μM). Etude des mécanismes moléculaires de résistance différentielle du mélanome malin aux vincalcaloïdes / Study of the molecular mechanisms of malignant melanoma differential resistance to vinca alkaloids

Le mélanome malin (MM) est un cancer très réfractaire aux thérapies anticancéreuses, dont les vincalcaloïdes (VAs). Afin d'étudier le rôle de la GSTM1 (glutathion S-transférase 1) et la MRP1 (multidrug resistance protein 1) dans la résistance acquise du MM aux VAs, nous avons établi 4 modèles cellulaires de résistance à la vincristine (CAL1R-VCR), à la vindésine (CAL1R-VDS), à la vinorelbine (CAL1R-VRB) et à la vinflunine (CAL1R-VFL), par exposition continue de cellules du MM (CAL1-wt), pendant un an, à ces anticancéreux. L'expression d'ne GSTM1 fonctionnelle est spécifiquement observée (RT-PCR, western blot, activité GST totale) dans les cellules résistantes. Le curcumin (inhibiteur de GSTM1), la BSO (inhibiteur de synthèse de glutathion) et le MK571 (inhibiteur de MRP1), réduisent considérablement le résistance acquise à la VCR et à la VDS mais pas à la VRB ou à la VFL. Toutefois, tous ces VAs réduisent spécifiquement l'activité GSTM1. Ces données montrent l'implication différentielle de GSTM1 et MRP1 dans la résistance aux VAs. Pour déterminer les mécanismes moléculaires de cette chimiorésistance, nous avons réalisé une étude pangénomique (biopuces Affymetrix HG-U133 Plus 2.00) sur les lignées CAL1 (wt et R). Le regroupement hiérarchique (par Cluster et TreeView) des données des puces a révélé une similarité entre les profils d'expression génique de CAL1R-VRB et CAL1-wt mais aussi entre ceux de CAL1R-VCR et CAL1R-VDS. L'analyse bioinformatique (par IPA) des transcrits les plus différemment exprimés entre les lignées cellulaires, a mis en évidence 6 réseaux géniques connus pour leur rôle dans la chimiorésistance tumorale. Le programme FatiGO a révélé 3 termes biologiques sur-représentés (> 60%) dans CAL1R (ribosome, filaments intermédiaires du cytosquelette, récepteurs olfactifs) tandis que l'étude fonctionnelle (invalidation génique par siRNA, test de viabilité) de GPR143, KIT et SLC45A2 (gènes interagissant avec NF-κB et CCND1 (facteurs de la chimiorésistance tumorale), très exprimés dans CAL1-wt et muets dans CAL1R) a montré la faible tendance des deux premiers à être impliqués dans la résistance aux VAs. / Malignant melanoma (MM) is a very refractory tumor to anticancer therapies, including vinca alkaloïds (VAs). To investigate the role of GSTM1 (glutathione S-transferase μ1) and MRP1 (multidrug resistance protein 1) in MM acquired resistance to VAs, we established 4 cellular models of resistance to vincristine (CAL1R-VCR), to vindesine (CAL1R-VDS), to vinorelbine (CAL1R-VRB) and to vinflunine (CAL1R-VFL), by continuous exposure of MM cells (CAL1-wt), for one year, to these anticancer agents. The expression of a functional GSTM1 is specifically observed (RT-PCR, western blot, total GST activity) in resistant cells. Curcumin (GSTM1 inhibitor), BSO (glutathione synthesis inhibitor) and MK571 (MRP1 inhibitor), considerably reduce the acquired resistance to VCR and VDS but not that to VRB or VFL. However, all these VAs specifically reduce GSTM1 activity. These data show the differential involvement of GSTM1 and MRP1 in resistance to VAs. To determine the molecular mechanisms of this chemoresistance, we performed a pangenomic study (Affymetrix HG-U133 Plus 2.00 microarrays) on the CAL1 lines (wt and R). The hierarchical clustering (by Cluster and TreeView) of array data revealed a similarity between the gene expression profiles of CAL1R-VRB and CAL1-wt, but also between those of CAL1R-VCR and CAL1R-VDS. The bioinformatic analysis (by IPA) of the most differentially expressed transcripts between cell lines, highlighted 6 gene networks known for their role in tumor chemoresistance. FatiGO program revealed 3 biological terms overrepresented (>60%) in CAL1R (ribosome, intermediate filaments of cytoskeleton, olfactory receptors), while functional study (gene invalidation by siRNA, viability test) of GPR143, KIT and SLC45A2 (genes interacting with NF-kB and CCND1 (tumor chemoresistance factors), highly expressed in CAL1-wt and mute in CAL1R) showed the weak trend of the two formers to be involved in resistance to VAs.

It was obtained from the previous processing of an alkaloidal extract from the Vinca minor L at the Department of Pharmaceutical Botany as a part of elaboration of diploma thesis of Aneta Vítavcová.[78] The fraction VM 323-327 was separated by column chromatography on silica gel and a totally, of 7 subfractions were obtained. Subsequent repeated processing of the selected sub-fraction 3 (15 - 36) by preparative TLC on silica gel resulted in the isolation of (-)-vinoxine and its racemate (±)-vinoxine. Identification of their structure was determined based on MS, NMR and optical rotation. The inhibitory activity against acetylcholinesterase, butyrylcholiesterase and prolyl oligopeptidase were determined for the isolated substances. Inhibitory activity against selected enzymes was measured by spectrophotometric methods. Vinca minor L. is a species of species of flowering plant, native mainly to central and southern Europe, which containst more than 50 indole alkaloids. During screening of potential plant inhibitors against human acetylcholinesterase (hAChE) and butyrylcholinesterase (HBChE) at our department, an alkaloidal extract from dried aerial parts of Vinca minor demonstrated strong and selective hBChE inhibitory activity with an IC50 value of 13.60 ± 0.83 μg/mL, however, against hAChE was inactive (IC50 value >100 μg/mL). The fraction VM 323 - 327 (4,72 g) was separated by column chromatography on silica gel again with stepwise elution by using chloroform and ethanol and overall 7 joined fractions were obtained.Subsequently, repeated preparative TLC on silica gel led to isolation of three compounds; the newly isolated substance SP-1, (-)-picrinine (SP-2) and deacetylakuammiline (SP-3).
