The diploma thesis devotes to the process of ensuring functional safety of products in Bosch Rexroth Brno, which deals with industrial hydraulic systems. The first part introduces the basic legislative regulations concerning product safety and describes selected standards relevant to this thesis. The second part focuses on Bosch Rexroth and the current state in the field of ensuring functional safety of their products. It also includes a description of the functional safety solution of a specific project – a hydraulic try-out press. At the end of the thesis, improvements to the process of ensuring functional safety in response to the identified shortcomings are proposed.

This diploma thesis describes Rapid Prototyping methods. It introduces their overview, principles, advantages and disadvantages. This project also deals with aplication and comparison of modifications of samples made by Fused Deposition Modeling. Findings from tests of mechanical features (pulling test and bending test) are used for comparison.

This work deals with methods of web page classification. It explains the concept of classification and different features of web pages used for their classification. Further it analyses representation of a page and in detail describes classification method that deals with hierarchical category model and is able to dynamically create new categories. In the second half it shows implementation of chosen method and describes the results.

Genetic influence of Trypanosoma brucei brucei infection in mice The African trypanosomes are zoonotic parasites transmitted by Tse-Tse flies. Two of the three subspecies, Trypanosoma brucei gambiense and Trypanosoma brucei rhodesiense, cause sleeping sickness in humans whereas the third subspecies, Trypanosoma brucei brucei is not infective to humans. These parasites are members of Kinetoplastida. Trypanosomes are extracellular parasite witch have complex life cycles involving both insect and mammalian hosts. African trypanosomes after infection penetrate mainly vascularized organs and get into brain where cause serious pathology. Parasite can manipulate with immune system of mammal host in wide spectrum of interactions witch are not clearly understood so far. Discovering of a new immune mechanisms, whitch participite in reaction on african trypanosomes, can reveal some general characteristics of immune system. The results of these studies can help to prepare effective drugs and vaccines against this disease. The thesis deal with the soil sealing and the rehanding with the uncovering of soil cultural layers. This problem is describe on the example of the industrial zone Kolín - Ovčáry. In the thesis there is the evaluation of the soil characteristics and their changes in areas where due to the trensfer of the cultural layer was created completely new structure of soil cover compact of soil units with artificial stratigraphy of profile. For this area the new classification subsumtion is suggested. The overall impacat of the soilsealig and implementation of the uncovering of soil cultural layers, despite of local incerase of soil fertility parameters, we evaluate as the degradation of soil cover.

Monoclonal antibody MEM-57 recognizes CD3 antigen expressed on peripheral blood T-lymphocytes. CD3 surface glycoprotein complex associates with T-cell receptor and is responsible for the transduction of activation signal. Antibody MEM-57 has, therefore, a large diagnostic and therapeutic potential. It could be used in autoimmune diseases diagnostics, for classification of T-cell leukemias and, as an immunosuppressant, in transplantation. The most promising therapeutic use of MEM-57 antibody would be the construction of a "Bispecific T-cell Engager" (BiTE) antibody format with potential application in cancer therapy. In this format, single-chain variable fragment (scFv) of MEM-57 would be fused with an anti-tumor antigen scFv. The thesis is focused on biochemical and biophysical characterization of MEM-57 antibody scFv fragment. Recombinant antibody fragment scFv MEM-57, equipped with the pelB leader sequence, c-myc tag and His5 tag, was produced from a pET22b(+) vector into the periplasmic space of E. coli BL21 (DE3). The goal of this thesis is to get familiarized with software component Microsoft .NET Framework and the problems of methods of interest point detection. On the basis of this knowledge, it is required to develop a method for the evaluation of used detectors and then implement a console application that is simply able to evaluate the results of interest point detection methods. Such evaluation is important in the process of development of the algorithms for the detection using projected method of evaluation. The interest points are searched in several images which represent the same scene from different angles of view. The requirements also inculde creation of graphic user interface that allows an easy way to setup the evaluation conditions.

This dissertation desribes the behavior of cellulose fiber reinforced concrete. In the theoretical section one can find basic information on cellulose fiber and its methods of production and usage in concrete structures. One can find the information about the effect of high temperature on the mechanical and physical attributes of cellulose fiber reinforced concrete, and also a comparison with other fibers used in concrete based on the research done using scientific literature. Furthermore the details about particular cellulose fibers are provided. In the experimental section the formulas for CHRYSO and GREENCEL concrete are designed. The influence of the addition of various types of cellulose fibers into both liquid and solid states of concrete was researched. Tests were also done on the effect of high temperatures (600 °C and 800 °C) on the firmness and densitiy of concrete, its deformations and visual changes. Furthermore, the resistance of cellulose fibers was monitored by monitoring the development of the pH environment (from pH 4 to pH 12) with long-term deposition of different types of cellulose fibers in these environments.

This diploma thesis deals with surface forming methods, especially accuracy and measurement of sheet metal bending on Van Mark brakes. The aim of the work was to make bends of samples in the range of 30 °, 60 °, 90 ° and 120 °. The size of the bend of the examined samples is influenced by the suspension of the sheet, which is not taken into account in the currently used type of protractor. The samples were then analyzed on two types of measuring devices - tactile and optical. After measuring, results were evaluated and plotted in the appropriate graphs. Based on the measurement results, a new type of protractor was designed, which will make the user's work easier and more efficient.
