Design of a wall-less proportional counter for microdosimetry in nanometer dimensions

Knowledge of energy deposition in biological cells at nanometer dimensions is essential to understand the biological effects of radiation. This work has resulted in the development of a practical tool to study such energy deposition experimentally, at nanometer dimensions. The main contribution of this research is the design of a cylindrical wall-less proportional counter of 1mm height by 1mm diameter. A wall-less detector (also called grid-walled detector) overcomes the so-called “wall effect”, an experimental artifact that introduces distortions in the radiation energy measurements. An important feature of this detector that distinguishes it from other detectors is its modular design. This allows the detector to be repaired or modified, when necessary, without having to completely disassemble it. Novel design techniques were adopted resulting in a functional detector that can simulate cellular sites as small as 10 nanometers, approximately the size of many molecules in the cell. The detector was tested with a 1 microcurie sealed Am-241 source, which primarily emits monoenergetic alpha particles of energy 5.57 MeV. Microdosimetric spectra analysis for alpha particles and its delta rays from Am-241 were performed for simulated site sizes ranging from 500nm to 10nm. Initial studies to validate the detector design have confirmed good detector performance. We believe this work will serve as a vital platform for bridging the experimentally measured energy spectra to the biological effects of alpha and delta radiations.

Os feixes de nêutrons rápidos foram emitidos com três diferentes valores de potência. 0,5, 1,0, e 2,0 kVV, durante 1 hora para alto ganho e o mesmo tempo para baixo ganho, totalizando 2 horas para cada potência, com 0,0083 Gy/min de taxa de dose. O neutron foi filtrado usando o sistema de irradiação de néutrons rápidos fortemente nitrados (FNIS). do Nuclear Science Center, para obter uma redução da contaminação da radiação de neutron por radiação gama e assim obter espectros microdosimetricos de neutrons como, distribuição de freqüência de energia lineal e distribuição de dose de energia lineal, com boa precisão, e outras grandezas como, freqüência média de energia lineal, dose média de energia lineal, dose absorvida, dose equivalente e fator de qualidade médio de neutron rápido. Os resultados obtidos foram satisfatórios, com os espectros microdosimetricos de nêutrons mostrando uma contaminação de radiação gama abaixo de 5 %, especialmente para distribuição de dose de energia lineal. Identificação de diversidade genética em fitoplasmas com base na análise molecular dos gene 16S rRNA, SecY e Proteína Ribossomal / Identification of genetic diversity in phytoplasma based on molecular analysis of the 16S rRNA, SecY and ribosomal protein genes

Os fitoplasmas são organismos procariotos sem parede celular, que habitam as células do floema das plantas e são transmitidos, principalmente, por cigarrinhas sugadoras de floema. Este patógeno é responsável por causar doenças em diversas espécies vegetais, provocando a expressão de diversos sintomas, entre os quais podem ser destacados a redução do tamanho foliar, amarelecimento das folhas, superbrotamento de ramos e enfezamento da planta hospedeira. Entre os anos de 2013 e 2014, plantas de três espécies vegetais com sintomas característicos de infecção causada por fitoplasmas foram observadas no estado de São Paulo. O DNA total foi extraído a partir de plantas de couve-flor com sintomas de nanismo, malformação da inflorescência e necrose dos vasos do floema; de plantas da ornamental conhecida por árvore-da-felicidade com sintomas amarelecimento e redução do limbo foliar; e de plantas de Alho-poró com sintomas de enfezamento. Por meio do teste de PCR conduzido em sua maioria com os primers P1A/16S-SR foi possível detectar fitoplasmas nas três espécies vegetais testadas. Os fragmentos genômicos correspondentes ao 16S rRNA dos fitoplasmas foram sequenciados e identificados. Nas plantas de couve-flor e da árvore-da-felicidade foram identificados fitoplasmas afilados ao grupo 16SrVII-B, através da análise virtual de RFLP, cálculo do coeficientes de similaridade (F) e análise filogenética. Para ambas as espécies, este é o primeiro relato da ocorrência de representantes deste grupo, nas condições brasileiras. Nas plantas de Alho-poró foram identificados fitoplasmas afiliados ao subgrupo 16SrIII-J e ao grupo 16SrXIII, com base na análise dos genes 16S rRNA, SecY e proteína ribossomal, conduzidas através de análise virtual de RFLP, valores de coeficientes de similaridade e análise filogenética. Para o fitoplasma membro do grupo 16SrXIII foram identificadas quatro estirpes, uma delas afiliada ao subgrupo 16SrXIII-E e as demais como prováveis representantes de novos subgrupos. O presente estudo se constitui em uma contribuição ao conhecimento de novos patossistemas envolvendo fitoplasmas, bem como à caracterização molecular de fitoplasmas baseadas em diferentes genes marcadores, atualmente usados para a classificação de representantes deste grupo emergente de agentes causais de doenças de plantas. / The phytoplasmas are prokaryotic organisms without cell walls, which inhabit the phloem cells of plants and are transmitted mainly by leafhoppers sucking the phloem. This pathogen is responsible for causing diseases in various plant species, leading to expression of many symptoms, among which can be highlighted the reduction of leaf size, leaf yellowing, shoots proliferation and stunting of the plant host. Between the years 2013 and 2014, three plant species with characteristic symptoms of infection caused by phytoplasma were observed in São Paulo. Total DNA was extracted from cauliflower plants with symptoms of stunting, malformation of the inflorescence and necrosis of the phloem; ornamental plants known for Ming Aralia with symptoms yellowing and little leaves; and leek plants with symptoms of stunting. Through the PCR test conducted mostly with the primers P1A/16S-SR was possible to detect phytoplasmas in the three species of plants. The genomic fragments corresponding of 16S rRNA gene of the phytoplasma were sequenced and identified. In plants of cauliflower and Ming Aralia were identified phytoplasmas belonging to 16SrVII-B group through virtual RFLP analysis, calculation of similarity coefficients (F) and phylogenetic analysis. For both species, this is the first report of the occurrence of representatives of this group, in Brazilian conditions. In leek plants were identified phytoplasmas affiliated with 16SrIII-J subgroup, and the 16SrXIII group, based on the analysis of the 16S rRNA, SecY and ribosomal protein genes, conducted through virtual analysis of RFLP, similarity coefficient values and phylogenetic analysis. For the phytoplasma member of group 16SrXIII were identified four strains, one affiliated with 16SrXIII-E subgroup and the others as probable representatives of new subgroups. Life and worship : a practical theological enquiry into the activities of the Perth Asian Christian community

The focus of this practical theological study is the analysis of the life and worship of the Perth Asian Christian Community in the New Life City Church, where we note fast maturation of Christians with a desire to complete the Great Commission. As participant observer using the qualitative method, I was able to identify how the NLCC is wall-less by interviewing different groups within the church and two Western sample groups. To accomplish this, Chapter two analyses the Western and Asian churches’ understanding of the concepts ‘church’ and ‘church growth’. The evaluations showed a difference in understanding. This prompted an inquiry in chapter three into their understanding of the Great Commission from the Western and Asian viewpoint against the Biblical understanding thereof. The intention of NLCC groups interviewed in Chapters four to eight was to detect what they perceived as helping them mature as Christians, considering many are first-generation Christians. The groups interviewed included founder members, first-generation Christians, youth group leaders, returned NLCC missionaries and the pastor. By Chapter nine it was established that the Asian Christian understood worship in a broader context. Chapter ten summarises the interpreted data of the groups interviewed and identifies stimulants for maturation of individual Christians in a post-Christian environment. Electric field determination and magnetic topology optimization in Hall thrusters / Détermination du champ électrique et optimisation de la topologie magnétique dans les propulseurs de Hall

La propulsion électrique est à un tournant de son histoire. La récente mise en oeuvre de satellites de télécommunication "tout-électrique" se traduit par l’ouverture d’un nouveau marché, et par une évolution des besoins opérationnels. L’identification de ces nouvelles missions nécessite l’amélioration des performances de nos moteurs. La première partie de ces travaux rapporte l’utilisation d’un outil de spectroscopie laser destiné à caractériser la distribution en vitesse des ions. Les différentes méthodes de calcul du champ électrique dans les propulseurs de Hall ont été exposées. Deux instabilités ioniques ont été observées et analysées. Les sources d’erreurs liées aux mesures par sondes électrostatiques ont été analysées, et un exemple de sonde plane étudié. Une analyse paramétrique de l’influence des paramètres de champ magnétique sur les performances des propulseurs de Hall a été conduite. Le concept de moteur "sans parois" est présenté. L’identification de ce concept constitue une avancée porteuse de promesses pour l’avenir de la propulsion électrique. / Electric propulsion is facing new challenges. Recently, the launch of "all-electric" satellites has marked the debut of a new era. Going all-electric now appears as an interesting alternative to conventional systems for telecom operators. A laser spectroscopy technique was used during this research to investigate the ion velocity distribution dynamics. The different methods for determining the electric field in Hall thrusters were exposed. Two unstable ion regimes were identified and examined. Measurement uncertainties using electrostatic probes were assessed. Planar probed have been designed and tested. A thorough investigation of the influence of the magnetic field parameters on the performance of Hall thrusters was performed. The wall-less Hall thruster design was presented, and preliminary experiments have revealed its interest for the electric propulsion community.
