Genetic Determinants of the Acute Effects and Withdrawal Symptoms of Caffeine

The mechanisms underlying caffeine’s acute effects and withdrawal symptoms are not entirely understood. The purpose was to determine whether these effects or symptoms co-exist in clusters, and whether they are associated with polymorphisms in β1- and β2-adrenoceptors. Subjects (n=1271) were from the Toronto Nutrigenomics and Health Study. The acute effects and withdrawal symptoms clustered into 4 and 6 factors, respectively. Subjects with the ADRβ2 Gly16Arg Gly/Arg genotype were more likely than Gly allele homozygotes to report “fatigue” withdrawal symptoms. Among >200 mg/d caffeine consumers, ADRβ2 Gly allele carriers had a greater risk, compared to Arg allele homozygotes, of reporting ‘flu-like somatic’ withdrawal symptoms. Among subjects with the CYP1A2 -163 A>C A/A genotype and 100-200 mg/d caffeine consumers, ADRβ1 Arg389Gly Gly allele carriers had a greater risk, compared to Arg allele homozygotes, of reporting “dysphoric mood” withdrawal symptoms. The findings suggest that β1- and β2- adrenoceptors play a role in caffeine withdrawal.

This diploma thesis deals with aspects of breaking the habit of smoking. My thesis consists of theoretical and practical part. The theoretical part is divided into three main chapters: smoking, impact of smoking and breaking of the smoking habit. I also defined the history of smoking, type of tobacco consuming, addiction to tobacco, treatments of the addiction, negative aspects of smoking etc. Practical part consists of quantitative research through questionnaires. The aim of my thesis was to find out what is tobacco addiction, gather the information and experience from the smokers who had experience with breaking their habit, analyze the methods of breaking the smoking habit and explain their main motivations for quitting the smoking. I have also included statistic data of my research.

Genome-Wide Association Study Identifies 5q21 and 9p24.1 (KDM4C) Loci Associated With Alcohol Withdrawal Symptoms

Several genome-wide association (GWA) studies of alcohol dependence (AD) and alcohol-related phenotypes have been conducted; however, little is known about genetic variants influencing alcohol withdrawal symptoms (AWS). We conducted the first GWA study of AWS using 461 cases of AD with AWS and 408 controls in Caucasian population in the Collaborative Study on the Genetics of Alcoholism (COGA) sample. Logistic regression analysis of AWS as a binary trait, adjusted for age and sex, was performed using PLINK. We identified 51 SNPs associated with AWS with p<10 -4. The first best signal was rs770182 (p = 3.65 × 10 -6) at 5q21 near EFNA5 gene which was replicated in the Australian twin-family study of 273 families (p = 0.0172). Furthermore, three SNPs (rs10975990, rs10758821 and rs1407862) within KDM4C gene at 9p24.1 showed p<10 -4 (p = 7.15 × 10 -6, 2.79 × 10 -5 and 4.93 × 10 -5, respectively) in the COGA sample while one SNP rs12001158 within KDM4C with p = 1.97 × 10 -4 in the COGA sample was replicated in the family sample (p = 0.01). Haplotype analysis further supported the associations of single-marker analyses of KDM4C in the COGA sample. Moreover, two SNPs (rs2046593 and rs10497668) near FSIP2 at 2q32.1 with moderate associations with AWS in the COGA sample (p = 2.66 × 10 -4 and 9.48 × 10 -5, respectively) were replicated in the family sample (p = 0.0013 and 0.0162, respectively). In addition, several SNPs in GABRA1, GABRG1, and GABRG3 were associated with AWS (p<10 -2) in the COGA sample. In conclusion, we identified several loci associated with AWS. These findings offer the potential for new insights into the pathogenesis of AD and AWS.

Genes Associated with Alcohol Withdrawal

Worldwide, alcohol is the third leading risk factor for disease burden, while its harmful use leads to 2.5 million deaths every year. Alcohol dependence (AD) is a complex disease, with devastating effects on individuals, families, and society. It is estimated that 76.3 million people worldwide have suffered from alcohol use disorders (AUD), including alcohol abuse and AD. Alcohol withdrawal or alcohol withdrawal symptom (AWS) refers to a cluster of symptoms that may occur when a heavy drinker suddenly stops or significantly reduces their alcohol intake. These symptoms can start as early as 2 h after the last drink, persist for weeks, and range from mild anxiety and shakiness to severe complications, such as seizures and delirium tremens. Family, twin, and adoption studies have indicated that genetic and environmental factors and their interactions contribute to the development of AD and related phenotypes, with a heritability coefficient of more than 0.5 for AD. Whole-genome linkage and candidate gene association studies have successfully identified several chromosome regions and genes that are related to AD and AWS. Furthermore, gene expression analysis, epigenetic studies, and genome-wide association studies (GWAS) have provided regions and loci for AWS. This chapter reviews the recent findings in genetic studies of AWS.

The aim of this thesis is to draw attention to the serious problem of drug addiction among adolescents who are abusing addictive substances or marijuana. This increasingly important issue is discussed a lot, but very often neglected by the society. This thesis provides definitions and characteristics of individual drugs and their effects on adolescents. The need for prevention and treatment of adolescent clients is emphasized, alongside with the family support, which is often crucial. In the practical part, the author of the thesis focuses on organizations that are critically important in this field. These organizations focus on prevention, counseling services, social inclusion and treatment facilities (inpatient and outpatient services), etc. Their characteristics and methods of client treatment are described. Furthermore, in this work the author presents a survey that confirms the findings of some detailed quantitative research papers. The main findings are that drug abuse is a major problem of this century and also a burden on society, especially on the adolescents themselves. The solution is very complicated and time consuming, but everyone can contribute by not being indifferent to this problem.
