Title: Effects of short-term parkour training programme on selected components of physical fitness in adolescents Objective: The aim of this work was to assess the influence of parkour on selected components of physical fitness - body composition, muscle fitness, aerobic fitness, flexibility - in a short-term programme. Methods: The ten-week experimental study dealt with the possibilities of using parkour to influence the parameters of physical fitness in boys age 16±2 years (N=10). Subjects went through measurement of bioelectrical analysis of body composition, muscle strength and endurance field tests, cardiopulmonary testing, sit-and-reach test. One-way analysis of variance with repeated measures (ANOVA) was used for statistical analysis. The strength of relationship between dependent and independent variables was expressed as unbiased effect size measure omega squared (ω2 ). Pearson two-tailed correlations were used to determine associations between measured parameters. Results: No significant changes in any body composition variables were observed with the parkour intervention. Fat mass increased by 0.18 %, fat free mass (FFM) decreased by 0.07 kg and the extracellular mass-body cell mass ratio (ECM/BCM) decreased by 0.02. Human immunodeficiency virus (HIV) is globally spread virus without available cure. Since its life-long presence, virus is carefully monitored as well as patient's immunological status. Replicative fitness of the virus is one of important aspects which can be taken into account, when monitoring HIV. Here, we are measuring HIV replicative fitness of gag recombinant viruses and comparing the results with replicative fitness of primary isolates. Further, we are comparing our findings of replicative fitness change over time with disease progression in the patient. We found that gag can be major contributor to overall fitness, although not in all cases. Additionally, we observed a correlation of replicative fitness development and slope of patient's CD4+ T cells. Moreover, this relation was even more noticeable in patients with slow disease progression or in carriers of protective alleles. In summary, our results extend the understanding of replicative fitness and its role in disease progression; and pave the way to use the recombinant HIV for replicative fitness measurement in clinical practice. Consultant: PhDr. Pavel Hráský, Ph.D. Aims of the thesis: To compare the level of basic ability in selected motor tests at athletes and modern pentathletes of the elementary school age. Methodology: Performance of athletes and modern pentathletes of elementary school age were compared in the following general tests: 12-minute run, one- way jump, stability tests, tests jump. At the same time, the body composition was monitored throughout the test group. Conclusions: Testing of base performance showed that there are no major differences between athletes and modern pentathletes. Despite slightly better average results of athletes, we can only register a statistically significant variance in "one-way jump" where athletes significantly outperformed pentathletes. Another variance was recorded in stability tests, namely standing on the right feet but only within boys category. Even in this case athletes achieved better results. Objective: The main goal of the thesis is to find out whether or not the seniors that participated in the project EXODYA back in 2017 and 2018, managed to maintain the level of physical fitness and adherence to physical aktivity (PA). The secondary goal was to determine the level of current PA and also how the level of PA was affected by the Covid-19 pandemic. Method: This is an empirical-theoretical study with a qualitative-quantitative research design. The research group I (N=30, average age 72 ± 4,02 years) took part in a questionnaire survey that included the International Physical Activity Questionnaire IPAQ and supplementary questions about influencing PA by the pandemics, using Google form. The research group II was made by 7 women that were also part of the research group I and that took part in the EXODYA project in 2018. The testing of these women consisted of 4 items of the Senior Fitness Test, hand grip strength using a digital hand dynamometer and maximum oxygen demand based on a stress test on a bicycle ergometer. Title: The effects of extreme physical and mental stress on the human body Goals: The main goal of this work was to evaluate the effect of physical stress load in a five-kilometer-long race in cross-country skiing on physical and mental functions of high school students. Methods: The investigated group consisted of 10 boys of the average age of 16 years. To evaluate their level of physical fitness and circulatory responses during cross-country stress, the maximal treadmill stress test was used. Circulatory response before, during, and after the race was monitored by a Sport-tester. To evaluate the mental background and the actual condition under the stress load, standardized questionnaires on ability to cross-country skiing, stress management strategy, degree of physical discomfort and current mental state were used. Results: The reference file tested by a stress test up to the maximum proved an above-average aerobic fitness. The extreme stress situation was a five-kilometer-long race in cross-country skiing. The results were processed and evaluated with a statistical and factual relevance. A number of correlations were made. Title: Selected fitness aspects of professional firefighters competing in fire sport and TFA Objectives: The aim of the thesis was to find out the level and the difference in the parameters of body constitution, postural stability and muscular and explosive force of lower limbs with groups of professional firemen competing in fire sport and TFA Methods: The research sample was represented by two groups of professional firemen (17 fire sportsmen and 17 TFA competitors). We assessed chosen parameters of body constitution (Tanita MC-980MA), postural stability (RS Footscan), explosive force (Kistler) and muscular force (Cybex Humac Norm). Assessed parameters of body constitution were percentage of body fat and fatless matter. In the tests of postural stability we assessed total travel way of the centre of pressure (TTW), narrow stand (open and closed eyes) and one-leg stand (right, left). When testing explosive force, overall produced maximal force and height of the leap were assessed. Muscular force was assessed with the help of muscular force moment in concentric muscle activity with angular velocity 60ř·s-1. Title: Effects of water-based aerobic programme on health-related physical fitness and health-related quality of life level in elderly females Objectives: The aim of this study was to asses the effect of shallow water aerobic programme on changes of health-related physical fitness and health- related quality of life level in elderly females. Methods: 33 women (mean age 67 ± 5,2 years) were eligible for participation for the study. Participants were divided into experimental and control group. The experimental group (n = 17) participated in a 6month shallow water aerobic programme once a week 60 minutes. The control group did not participate in the physical program. Body composition, low-back and hamstring flexibility, hand grip strength, upper body and lower body strength, aerobic capacity, dynamic postural stability and health-related quality of life were determined before and after completion. Results: This study has shown that the training programme caused a significant increase in aerobic capacity (p = 0,01; r = 0,62) and in general health perception (p = 0,04; r = 0,36). Title: Subjectively perceived health fitness and physical activity of the elderly compared to a fitness walk test Objectives: The aim of this paper is to compare the subjectively perceived health (fitness) and physical activity with an actual fitness walk test done by students of the University of the Third Age, using the LTEQ and SF-12 questionnaires. Methods: There were 64 seniors who participated in this study (age 67,36 ± 3,55, height 1,67 ± 7,04 cm, weight 69,80 ± 9,9 kg). All of them students of the University of the Third Age, Faculty of Physical Education and Sport, Charles University. Two types of Questionnaires were used: The Leisure Time Exercise Questionnaire and A 12-Item Short-Form Health Survey, both of which are typically used to study the population of the elderly. A 2 km fitness walk test by Stejskal was used to gather data about aerobic fitness. The test took place at the athletic oval on the grounds of Faculty of Physical Education and Sport, Charles University. Polar S610i sporttester was used to measure heart rate. This thesis deals with the impact of the specific physical activity on the physical fitness of children measured by UNIFITTEST (6-60). Final results were processed based on the UNIFITTEST standarts and the information about physical activity of the children were taken from the questionnaire which have been created specifically for this research. The stated research questions were compared with results from the practical part of the thesis and were discussed in the discussion chapter. By this research we have found out that children form seventh and eighth grade have reached an average value in motoric tests. Due to this work children have found out their physical fitness and so they could compared it with their classmates. Also they have got to know with the physical tests with which they can be part of during physical lessons. This work was made by research form realised at standard nonspecific elementary school Na Výsluní in Brandýs nad Labem. The total amount of the participants were 194 from 12 to14 years.
