The main purpose of this diploma thesis was to evaluate the functioning of the project "School milk and School fruit and vegetables" in the selected region, to find out its possible shortcomings and to propose further direction of this program. For the purposes of this diploma thesis the district of Tábor was selected. As part of the practical part of this diploma thesis, the primary research in this district first carried out through an online questionnaire survey. Subsequently, a survey of parents' opinion was conducted in the selected elementary school through all nine years of schooling. At the end there are mentioned problem areas of the project and suggested concrete recommendations. There is also verified the possibility of using organic food and regional foods in the project, and determined the attitude of schools and parents to the "Dainty" Decree.

Příprava vzorků pro analýzu DNA v potravinách rostlinného původu / Sample preparation for DNA analysis from foods of plant origin

The isolation of high quality DNA is nessecary for many molecular biology applications. However, plant DNA contains high amonts of polysaccharides, polyphenols and various secondary metabolites, which decrease yield and quality of isolated DNA. The aim of this study was preparation of samples and different food matrices for DNA isolation DNA by magnetic particles. It was about 5 species of vegetable and 10 species of processed plant food. Homogenization of samples was performed in CTAB buffer. Isolation of plant DNA was performed by magnetic particles covered with carboxyl groups. All DNAs were isolated in conventional PCR qualities using primers for 700 bp amplicons, in the case of heat processed products for 220 bp ampilicons and for real time PCR. The efficiancy of separation of magnetic particles with DNA by magnetic separator and magnetic needle was compared. It was find out that DNA of higher purity was isolated using magnetic needle. The micromethod of isolation of plant DNA from homogenates with CTAB with magnetic particles is suitable for different processed food.

Pěstování microgreens v mateřské škole / Growing Microgreens in Nursery Schools

The diploma thesis focuses on the topic of cultivating microgreens in nursery school. Microgreens are young plants of various species of vegetable, that are grown in soil and are harvested approximetely after ten days. The simplicity of growing microgreens makes it an appropriate activity for preeschool-age children. Besides rewarding the children with nutricious food, it brings children the oppoturnity to develop key competences and pre- literacies. The theoretical part is an information base for growing microgreens in kindergarten conditions. It describes cultivating plants in kindergarten, it deals with historicaly, legislatively and didactic point of wiew and also describe kinds of microgreens, possibilities of use and cultivation procedures suitable for kindergarten. The aim of the diploma thesis is to present the possibility of growing microgreens in nursery school with preschool children. The empirical part focuses on the design and implementation of a set of activities in which children in kindergarten grow microgreens. In addition to cultivation, other activities are also included, in which children use the topic of microgreens in individual stages of growth.
