Previous issue date: 2013-03-02 / The objective of this research was to measure the concentration of glutamine (GLN) plus glutamate (GLU) in milk and blood of dairy cows, milk composition ([Lactose], [Fat], [Protein], [Total solids]) and somatic cell count (SCC) in different periods of lactation, and the concentration of these amino acids in the blood of their calves Samples of blood and milk were collected from 21 Holstein cows during five months of lactation (first, second, third, fifth and tenth) and serum samples from 28 calves in first three months of life. The results were analyzed by ANOVA, with 5% of significance and the Tukey test, with the same level of significance. There was not statistical difference (P>0,05) in [GLN + GLU] in the blood of the cows and calves, neither in the concentration of these amino acids in the milk; [Lactose], [Fat] and CCS also were not affected (P>0,05). It was observed higher protein content (P<0,05) in the fifth and tenth months of lactation (~ 3,61% in both) relative to the first, second and third months; the total solids was higher in the tenth month (~12,92%) related to the first and second months of lactation (~ 11,83% and ~ 11,81%, respectively). The period of lactation did not affect [glutamine] plus [glutamate], neither in the concentration of these amino acids in milk. SCC, [lactose] and [fat] did not change, and only [protein] and [total solids] were significant different. There was no correlation with age of calf and [glutamine] plus [glutamate]. / A glutamina, aminoácido condicionalmente essencial, é considerada como fator limitante na produção leiteira. Assim, Objetivou-se mensurar a concentração de glutamina (GLN) mais glutamato (GLU) no leite e sangue de vacas leiteiras, a composição do leite e a contagem de células somáticas (CCS) nos diferentes períodos de lactação, além da concentração destes aminoácidos no sangue de seus respectivos bezerros. Foram colhidas amostras de sangue e leite de 21 vacas holandesas durante cinco meses da lactação e amostras sanguíneas de 28 bezerros nos três meses iniciais de vida. No sangue determinaram-se [GLN+GLU] e no leite verificaram-se a concentração de GLN+GLU, Lactose, Gordura, Proteína, Extrato Seco Total (EST) e CCS. Os resultados foram analisados pelo ANOVA, com significância de 5% e pelo teste de Tukey, com mesmo nível de significância. Observou-se que não houve diferença estatística (P>0,05) nas [GLN+GLU] no sangue de fêmeas e bezerros, nem na concentração destes no leite; a [Lactose], [Gordura] e CCS também não alteraram (P>0,05). Contudo, constatou-se maior teor proteico (P<0,05) no quinto e no décimo mês de lactação (~3,61%, em ambos), em relação ao primeiro, segundo e terceiro mês; o EST foi maior no décimo mês (~12,92%) relacionado ao primeiro e ao segundo mês de lactação (~11,83% e ~11,81%, respectivamente). Concluí-se que o período de lactação não interferiu na concentração sanguínea de glutamina mais glutamato, nem na concentração destes aminoácidos no leite, bem como não alterou a contagem de células somáticas e o teor de lactose e gordura, promovendo alterações apenas no nível de proteína e EST. Percebeu-se, ainda, que a idade do bezerro não influenciou a concentração sanguínea de glutamina mais glutamato.
