Express?o Imuno-histoqu?mica de COX-2, EMMPRIN, HIF-1? e GLUT-1 no tecido gengival normal e inflamado

Previous issue date: 2012-02-24 / The aim of this study was determine whether an association exists in the gum tissue between the expression of markers of tissue hypoxia (HIF-1? and GLUT-1) with a marker of inflammatory activity (COX-2) and a marker of collagen degradation (EMMPRIN). Was performed immunohistochemistry with antibodies specific for these markers on 60 samples of gingival tissue divided into two groups: gums (n = 26) and
gingivitis (n = 34) and expression was analyzed in the epithelial tissue and connective tissue . The reactivity epithelial for COX-2 was observed in only two cases as the HIF-1?, GLUT-1 and EMMPRIN was strongly expressed in the epithelial basal
layer and the immunostaining was gradually decreased as the cells away from this layer, and negative in the region suprabasal in most specimens. In connective tissue, and HIF-1? EMMPRIN were strongly positive for most cases analyzed as GLUT-1
was negative in most cases. Immunostaining for COX-2 showed an association with gingival inflammatory infiltrate. The expression of EMMPRIN, HIF-1? and GLUT-1 in normal gums confirms the physiological role of these markers, however there was no association with tissue inflammation. Given the findings we can conclude that the inflammatory changes installed in frames of chronic gingivitis may not be sufficient to
activate the factors of hypoxia to levels that can be quantified by immunohistochemical analysis, in addition, the findings are not conclusive in relationship to involvement of EMMPRIN in the secretion of MMPs to degrade collagen in the frames of gingivitis. We suggest the use of technical analysis and quantification of RNA of EMMPRIN and MMPs in order to determine whether
collagen degradation observed in gingivitis suffers or not, significant influence of EMMPRIN for secretion and activation of MMPs / Este estudo pretendeu verificar se no tecido gengival normal e em quadros de gengivite existe associa??o entre a express?o dos marcadores de hip?xia tecidual (HIF-1? e GLUT-1) e os marcadores da atividade inflamat?ria (COX-2) e da degrada??o das fibras col?genas (EMMPRIN). Foi realizada a t?cnica imunohistoqu?mica com anticorpos espec?ficos para os referidos marcadores em 60 amostras de tecido gengival divididas em dois grupos: gengivas saud?veis (n=26) e inflamadas (n=34) e a express?o foi analisada no tecido epitelial e no tecido
conjuntivo. A reatividade epitelial para a COX-2 foi observada em apenas dois casos enquanto o HIF-1?, GLUT-1 e EMMPRIN estavam fortemente expressos na camada basal do epit?lio e a imunomarca??o foi gradualmente diminuindo ? medida que as
c?lulas afastavam-se desta camada, sendo negativa na regi?o suprabasal na maioria dos esp?cimes. No tecido conjuntivo, EMMPRIN e HIF-1? foram intensamente positivos para maioria dos casos analisados enquanto o GLUT-1 foi negativo na maioria dos casos. A imunomarca??o para a COX-2 revelou associa??o
com o infiltrado inflamat?rio gengival. A express?o do EMMPRIN, HIF-1? e GLUT-1 em gengivas normais, confirma o papel fisiol?gico destes marcadores, entretanto n?o houve associa??o com a inflama??o tecidual. Diante dos achados pode-se
concluir que as modifica??es inflamat?rias instaladas em quadros de gengivite cr?nica podem n?o ser suficientes para ativar os fatores de hip?xia a n?veis que possam ser quantificados atrav?s da an?lise imuno-histoqu?mica. Al?m disso, os achados n?o s?o conclusivos em rela??o ? participa??o do EMMPRIN na secre??o
das MMPs para degrada??o do col?geno nos quadros de gengivite. Sugere-se a utiliza??o de t?cnicas de an?lise e quantifica??o de RNA do EMMPRIN e das MMPs com o intuito de verificar se a degrada??o do col?geno observada nos quadros de
gengivite sofre, ou n?o, influ?ncia significativa do EMMPRIN para secre??o e ativa??o das MMPs
