Previous issue date: 2001-08-10 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Avaliou-se o efeito da eliminação genética das três lipoxigenases de sementes de soja sobre sua qualidade fisiológica e sanitária. Sementes das variedades CAC-1, Cristalina, Doko-RC e UFV-16 e de suas respectivas linhagens sem lipoxigenases nas sementes (triplo-nulas) foram multiplicadas em campo e a colheita efetuada no estádio R 8 de maturação e aos 15, 30 e 45 dias após este estádio. A qualidade fisiológica das sementes foi avaliada pelos testes de germinação, primeira contagem do teste de germinação, emergência em leito de areia, envelhecimento acelerado, porcentagem de emergência de plântulas em campo e índice de velocidade de emergência de plântulas em campo. Foram realizados também o teste de sanidade e a determinação do teor de aldeídos totais pelo método colorimétrico do MBTH. A quarta época de colheita (R 8 + 45 dias) evidenciou as maiores diferenças entre os materiais com ou sem lipoxigenases. As sementes das linhagens sem lipoxigenases derivadas das variedades Doko-RC e UFV-16, apresentaram maior susceptibilidade à deterioração no campo, provocada pelo atraso da colheita. Não houve efeito da retirada de lipoxigenases sobre a qualidade de sementes das linhagens triplo-nulas derivadas das variedades CAC-1 e Cristalina. As características de germinação e vigor correlacionaram -se, significativamente, com a incidência de fungos, sugerindo ser este o motivo dos baixos valores obtidos nesses testes para as linhagens triplo-nulas de Doko-RC e UFV-16. Não foi possível estabelecer relação direta entre retirada de lipoxigenase e qualidade de sementes, nas linhagens estudadas. / The effect of genetic elimination of the three lipoxygenases from the seeds of soybean on their physiological and sanitary quality was evaluated. Seeds of varieties CAC-1, Cristalina, Doko-RC and UFV-16 and of their respective lines without lipoxygenases in the seeds (triple-null) were multiplied in the field and were harvested at the R8 stage and 15, 30 and 45 days latter. The physiological quality of the seeds was evaluated by the germination test, first counting on the germination test, emergence in sand bed, accelerated aging, and percentage and rate of emergence in the field. Data on sanitary quality of the seed and total aldehyde content evaluated by the MBTH method were also obtained. The largest differences between the materials with or without lipoxygenases were evidenced on the fourth harvesting time (R8+45 days). The seeds of the lines lacking lipoxygenases, derived from Doko-RC and UFV-16, presented greater susceptibilidade to field decay with harvest delaying. The genetic elimination of lipoxigenases from the seeds did not affect the quality of seeds of the triple-null lines derived from CAC-1 and Cristalina. Germination and vigour were significantly correlated to fungi incidence, suggesting that this was one of the reasons for the lower values observed for the seeds of triple-null lines of Doko-RC and UFV-16. It was not possible to establish a direct relationship between lipoxigenases elimination and seed physiological and sanitary quality of the studied lines. / Dissertação importada do Alexandria
