Meta: Este trabalho foi focado em avaliar a expressão de RNA-m das neurotrofinas (Fator de Crescimento de Nervo - NGF, Fator Neurotrófico Cerebral - BDNF e neurotrofina 3 - NT-3) no nervo mediano de ratos Wistar submetidos à sutura término-terminal imediata após neurotmese. Material e métodos: Ratos Wistar foram usados no experimento. O nervo mediano foi seccionado e os animais receberam três pontos de sutura epiperineural após a neurotmese. O estudo do RNA-m para BDNF, NT-3 e NGF no nervo mediano foi analisado nos períodos de 2, 7, 14, 30 e 60 dias após o procedimento cirúrgico usando RT-PCR. Também foi feita a análise histológica do nervo mediano usando coloração por hematoxilina-eosina - HE. Análise estatística: A diferença estatística entre os grupos foi conduzida com a análise de variância (ANOVA), seguida por teste t de "Student's" e/ou Dunnett's ou teste de correlação de Pearson. Para todos os testes foi utilizada análise bicaudal. Valores de p menor que 0,05 foram considerados estatisticamente significativos. Resultados: Os resultados mostraram um aumento da expressão do nível de RNA-m para BDNF e NT-3 60 dias após o procedimento cirúrgico (P=0,03). Houve também aumento na expressão de RNA-m para NGF, que foi detectado aos 7 dias e manteve-se elevado até 60 dias após o procedimento cirúrgico (p=0,0004). Conclusão: Estes resultados confirmam e aumentam a evidência de que a sutura imediata término-terminal é eficiente na recuperação funcional do nervo mediano lesado, uma vez que esta técnica induz a liberação de RNA-m para neurotrofinas que estão relacionadas à regeneração do nervo. / Neurotrophins regulate the adult nervous system plasticity promoting the survival of neurons and stimulating nerve regeneration after injury. Aim: This work aimed to evaluate the neurotrophins (nerve growth factors (NGF), brain derived nerve factor (BDNF) and neurotrophin-3 (NT-3)) in the timing of RNAm expression in the median nerve in Wistar rat submitted to primary end-to-end repair after neurotmesis. Material and Methods: Wistar rat were used thought the experiments. The rat median nerve was sectioned and the animals received primary end-to-end sutures with three epiperineurial stitches after neurotimesis. The study of RNAm to BDNF, NT-3 and NGF in the median nerve was analised 2 to 60 days after surgical procedure using the RT-PCR. Also histological analysis of the median nerve was performed using hematoxilin-eosin (HE) Statistical analysis. Statistical differences between groups were conducted with the analysis of variance (ANOVA), following by Dunnett's and/or Student's t-tests or Pearson rank correlation test. P-values of less than 0.05 were considered statistically significant, and all tests were two-tailed. Results: The results showed an enhancement of mRNA expression levels to BDNF and NT-3 at 60 days after surgical procedure (P = 0.03). Also an increase of RNAm for NGF was detected at 7 to 60 days (p = 0.0004). Conclusion: These results confirm and extend the evidence that the immediate end-to-end suture procedure is efficient in the functional recovery of the injured median nerve, because this technique induces the neurotrophins RNAm release that are related to the nerve regeneration.
