Previous issue date: 1999 / Resumo: Estudos morfo-funcionais são imprescindíveis para compreensão da patogenia das diferentes doenças que acometem a articulação temporomandibular, mas há poucas publicações na literatura sobre histoquímica da membrana sinovial. Este trabalho teve por objetivo estudo histoquímico da membrana sinovial da articulação temporomandibular de coelhas da raça Norfolk inglesa. Para tanto, desenvolveu-se técnica de dissecção das partes moles sobrepostas à região da articulação temporomandibular. A remoção óssea do arco zigomático foi etapa importante para exposição ampla da região póstero-superior da cápsula articular, local da biópsia. As biópsias foram congeladas em n-hexana, resfriada a - 70 °C com auxílio de nitrogênio líquido, transferidas para criostato e submetidas a cortes histológicos para processamento pela histoquímica, nas reações tricrômico de Gomori modificado, nicotinamida-dinucleotídeo-tetrazolium-redutase, desidrogenase succínica, esterase inespecífica, fosfatase ácida, ácido periódico de Schiff, fosfatase alcalina, alcian blue no pH 2.4 e alcian-ponceau. Também foram processadas pela microscopia eletrônica de transmissão. A membrana sinovial mostrou positividade nas diferentes reações histoquímicas acima descritas, excetuando-se desidrogenase succínica e fosfatase alcalina. Os estudos de ultra-estrutura permitiram clara diferenciação entre as células A, semelhantes a macrófagos, e células B, semelhantes a fibroblastos. A positividade da membrana sinovial nestas diferentes reações histoquímicas indica a existência de complexos mecanismos bioquímicos envolvidos na produção e reabsorção do líquido sinovial / Abstract: Morpho-functional studies are very important in understanding the pathogenesis of different diseases which affect the temporomandibular joint, however there are very few published studies about the histochemistry of the synovial lining cells. This work analyses the histochemistry of the synovial lining cells of the temporomadibular joint of adult female English Norfolk rabbits. We have developed techniques for dissection around the temporomandibular joint, including skin, subcutaneous tissue, and the masseter muscle. The removal of the zygomatic arch was an important step in sufficiently exposing the posterior region of the joint socket area for biopsy. The biopsies were chilled in n-hexane, frozen to . 70°C using liquid nitrogen, transferred by cryostat and sectioned in preparation for histochemical processing, using modified Gomori trichrome stain reaction, nicotinamide-dinucleotide-tetrazolium-reductase, dehydrogenated succinic, nonspecific sterase, acid phosphatase, Schiff periodic acid, alkaline phosphatase, alcian blue in pH 2.4 , and alcian-ponceau. Ultrastructural studies were also performed. The synovial lining cells showed activity in the different histochemical reactions listed above except for dehydrogenated succinic and alkaline phosphatase. The ultrastructural examination permitted clear differentiation between the A cells, similar to macrophages, and the B cells, similar to fibroblasts. The activity of the synovial lining cells to these different histochemical tests indicates the existence of complex biochemical reactions involved in the production and reabsorption of synovial fluid / Tese (livre-docencia) - Univer / Anatomia / Livre Docente em Biologia Buco-Dental
