Environmental Impacts to Stream Acidification and Brook Trout Populations in the Great Smoky Mountains National Park

This research supports development of aquatic resource management strategies to address acid deposition in the Great Smoky Mountain National Park (GRSM) by 1) developing relationships between baseflow and stormflow chemical constituents and examining effects of elevation, area, geology, soil, and vegetation on stream chemistry; 2) evaluating physiological condition in brook trout in relation to changes in stream chemistry during stream acidification episodes, and 3) evaluating brook trout metrics with respect to stream chemistry, basin characteristics, and ecologically relevant hydrologic parameters. (1) Stream chemistry was monitored in eight GRSM streams considering basin area, site elevation, Anakeesta geology, soil, and vegetation. Following precipitation events, pH was significantly reduced and aluminum concentrations increased, while the concentration response of ANC, nitrate, sulfate, and base cations varied. Higher pH and ANC concentrations were observed in large and low-elevation streams. (2) Caged brook trout were exposed to two acid episodes during in situ bioassays conducted in three GRSM streams. Stream pH decreased (>0.7 pH units) and total dissolved aluminum increased (>0.175 mg/L) at all three sites during acid episodes. Whole-body sodium concentrations were significantly reduced (10-20%) when preceding 24-h time weighted average pH values (4.88, 5.09, 4.87) and corresponding 24-h aluminum concentrations (210, 202, 202 µg/L). Lower whole-body sodium concentrations were correlated with elevated proton and aluminum concentrations indicating physiological distress. (3) Water chemistry, hydrology and physical basin factors influenced brook trout distributions and densities in 16 collocated fish and water quality sampling sites (1990-2009). Higher concentrations of ANC, pH, sodium, and soil cation exchange capacity, and higher fall flows were associated with the presence of brook trout. Trout densities were higher in streams with higher concentrations of sodium, suggesting that sodium may ameliorate the effects of acid toxicity. These relationships provide useful information where GRSM managers can prioritize conservation and restoration efforts.

Characterizing thermal refugia for brook trout (Salvelinus fontinalis) and Atlantic salmon (Salmo salar) in the Cains River, New Brunswick, Canada

Anthropogenic influences and climate change are warming rivers in New Brunswick and threatening the cold water habitats of native salmonids. When ambient river temperatures in summer exceed the tolerance level of Atlantic salmon and brook trout, individuals behaviourally thermoregulate by seeking out cold water refugia. These critical thermal habitats are often created by tributaries and concentrated groundwater discharge. Thermal infrared imagery was used to map cold water anomalies along a 53 km reach of the Cains River on 23 July 2008. Although efficient and useful for mapping surface temperature of a continuous stream reach, the fish did not use all identified thermal anomalies as refugia. Overall, 100 % of observed large brook trout >35 cm in length were found in 30 % of the TIR-mapped cold water anomalies. Ninety eight percent of observed small brook trout 8 – 30 cm in length were found in 80 % of the mapped cold water anomalies and their densities within anomalies were significantly higher than densities outside of anomalies. Fifty nine percent of observed salmon parr were found in 65 % of the mapped anomalies; however, they were dispersed within study sites and their densities were not significantly different within anomalies compared to outside of the anomalies. No brook trout were observed at the seven noncold water study sites that were investigated. Preference curves for various habitat variables including velocity, temperature, depth, substrate, and deep water availability near cold water anomalies were developed based on field investigations during high temperature events (ambient river temperature >21 oC). Combined with thermal imagery, managers can use the physical descriptions of thermal refugia developed here as a tool to help conserve and restore critical thermal refugia for Atlantic salmon and brook trout on the Cains River, and potentially similar river systems.

Relations between metabolic rate, migration and behaviour in Atlantic salmon (Salmo salar) and brown trout (Salmo trutta)

ABSTRACT   Migration is common among populations of brown trout (Salmo trutta) and Atlantic salmon (Salmo salar). However, not all individuals in the same population migrate, a phenomenon referred to as partial migration. The aim of this thesis was to investigate if an individual’s behaviour and metabolic rate influences its decision to migrate and how such knowledge may be used when trying to produce hatchery-raised smolts with as high a proportion of migrating individuals as possible. In paper I the influence of reduced food ration on the proportion and swimming speed of migrating brown trout and Atlantic salmon smolts was investigated. Furthermore, the standard metabolic rate (SMR) of migrating and non-migrating individuals was compared. In paper II, a laboratory experiment, SMR was correlated to the behaviour of individual brown trout and Atlantic salmon. Dominant fish of both species had a higher SMR than subordinates (paper II). In addition, migrant brown trout had a higher SMR than non-migrant trout when given a normal food ration, whereas no difference in SMR between migrating and non-migrating salmon could be seen (paper I). When administered low food rations, smolts of both species migrated faster than smolts given a normal food ration, and the proportion of migrating smolts was higher for salmon given less food when the size difference for smolts from the two feeding regimes was large (paper I). Other factors that influenced migration speed were the degree of smolt development and water temperature (paper I). SMR was not correlated with aggressiveness, or with different measurements of boldness. Moreover, aggression and boldness were not correlated with each other (paper II). Trout showed a higher level of aggressiveness and acclimated more rapidly to laboratory conditions than salmon (paper II). In summary, there was no support for the existence of coping styles in migratory Atlantic salmon and brown trout. Instead, metabolic rates were related to both migratory behaviour and social status. Furthermore, an individual’s decision to migrate was influenced by ration size.

Forest-stream linkages : Experimental studies of foraging and growth of brown trout (Salmo trutta L).

Riparian vegetation along streams and rivers affects the aquatic community in numerous ways and often operates as a link for energy flux between forest and streams. The studies presented in this licentiate thesis focus on light and terrestrial invertebrates, two factors influenced by riparian zone structure, which potentially affect stream ecosystems and thus also brown trout (Salmo trutta). Paper I is a laboratory experiment where I study size dependent foraging behavior on surface-drifting terrestrial invertebrates and benthic invertebrates by brown trout. The results show a size-dependent difference in foraging ability with large trout being better able to use terrestrial surface prey than small trout. I argue that such ontogenetic foraging differences are due to both morphological constraints (eg. gape limitation) and size dependent behavioral differences related to predation risk. Paper II consists of a 5 month-long 2x2 factorial design field experiment where my objective was to examine the effects of terrestrial invertebrate input and solar radiation (PAR) on different trophic levels in a boreal headwater stream. More specifically, I followed the effects of increased light and decreased terrestrial invertebrate subsidies on periphyton, benthic macroinvertebrates and two size classes of the top fish predator, brown trout. The results showed that the reduction of terrestrial invertebrate input had size- and seasonal-dependent effects on trout, where large trout had lower growth rates than small trout, mainly in summer. Diet analyses of trout supported growth differences in that large trout in unmanipulated enclosures consumed relatively more terrestrial prey than large trout living in enclosures with reduced terrestrial inputs. A higher reliance on terrestrial prey subsidies by large trout compared to small may be explained by ontogenetic differences in foraging and habitat choice. Despite a 2.5-fold increase in PAR, light did not have an effect on chlorophyll a biomass, nor was there an effect on the density or composition of benthic macroinvertebrates. The lack of effects on primary production may be explained by very low nutrient levels in the stream.

Roles of Arginine-Vasotocin and Corticotropin-Releasing Hormone in Stress Responses and Agonistic Behaviour of Rainbow Trout

The neuropeptides arginine-vasotocin (AVT) and corticotropin-releasing hormone (CRH) are involved in the hypothalamic-pituitary-interrenal (HPI) axis. During stress, the HPI axis is activated and cortisol is released into the blood. In addition to their role in the HPI axis, AVT and CRH also have behavioural effects. The roles of AVT and CRH in stress responses and agonistic behaviour were studied in this thesis, using two different models. In the first model, two strains of rainbow trout (Onchorhynchus mykiss) divergent in stress-induced release of cortisol were investigated. This was done by observing behaviour and stress responses under different conditions. These strains were found to have divergent stress coping strategies based on the observed behaviour and levels of plasma cortisol. This divergence in behaviour could be associated with the CRH system, since the mRNA levels of CRH differed between the strains during stress. However, no differences between strains were observed in AVT or its receptor expressions. In the second model, non-selected rainbow trout were paired and the effect of intracerebroventricular (icv) injections of an active substance (AVT, CRH or the CRH related peptide Urotensin-I (UI)) on fights for dominance was investigated. One fish of the pair received the active substance icv and the other received saline icv. Fish receiving AVT became subordinate in accordance with the suggestion that AVT attenuates aggression in territorial vertebrates. Fish receiving CRH became subordinate whereas UI showed no effect on fights for dominance. Further, both CRH and UI induced an anxiety-related behaviour similar to non-ambulatory motor activity in rats. In addition, CRH appeared to affect the dopaminergic and serotonergic systems. In this thesis, it is suggested that CRH is involved in the behavioural modulation of the stress coping strategies in teleost fish. Further, AVT and CRH seem to act inhibitory on aggressive behaviour.

Syftet med denna uppsats är att undersöka huruvida fiskvägar förbi vattenkraftverket vid Finsjö i Emån fungerar eller inte. För att finna svar på den frågan har två olika metoder använts, dels telemetrisändare för att se om fiskar hittar fram till fiskvägarna (attraktionseffektivitet) och dels PIT-tags för att mäta om fiskarna lyckas passera genom fiskvägarna (passageeffektivitet). Undersökningen fann att det var en högre passageeffektivitet, 89 % vid nedre Finsjö och 100 % vid övre Finsjö, i Emån jämfört med andra liknande studier. Attraktionseffektiviteten var 75 % vid nedre Finsjö och 59 % vid övre Finsjö, vilket tyder på att det finns något som stör fisken innan de når fram till fiskvägen, särskilt vid övre Finsjö. / The purpose of this paper is to determine the extent to which trout used fishways to swim past two power plants at Finsjö in River Emån. Two different methods of  collecting data were used, radiotelemetry to determine if fish find the fishways (attraction efficiency) and PIT-tags to measure the number of fish that passed through the fishways (passage efficiency). The study showed that passageefficiency, being 89 % at lower Finsjö and 100 % at upper Finsjö, was high compared to other similar studies. Attraction efficiency was 75 % at lower Finsjö and 59 % at upper Finsjö, which indicates that the fish have difficulty finding the fishway entrances, particularly at upper Finsjö.

A Multi-Scale Investigation of Factors Limiting Bull Trout Viability

Effective conservation strategies for imperiled species require an understanding of processes that influence fitness throughout the organism's life cycle and across the range of habitats needed to complete that cycle. I evaluated factors that affect population viability of bull trout Salvelinus confluentus, a threatened freshwater char species, throughout individual life stages and over the entire life cycle. I assessed the relationship between bull trout egg incubation success and environmental variables. Egg survival was negatively related to the percent of fine sediment in redds and positively related to hydraulic conductivity and the strength of downwelling. Next, I quantified juvenile bull trout survival rates and described movement patterns for this life stage. Juvenile bull trout emigrated from natal headwaters into larger rivers throughout the entire year and across a range of sizes. Estimates of juvenile survival rates improved dramatically when emigration was incorporated into the analysis. I integrated my observations of bull trout survival, growth, and movement to create a life-cycle model, which I used to better understand how populations respond to changes in specific demographic rates. Bull trout populations were particularly sensitive to changes in juvenile growth and survival. The relative effect of changes to fertility rates and adult survival varied depending upon whether a population was composed primarily of large, migratory, or smaller, resident individuals. Dispersal helped to lower the probability of extinction for small or declining populations when neighboring populations were stable. My research demonstrates that bull trout require access to habitats throughout entire watersheds to maintain population viability. My results suggest that limiting anthropogenic sources of fine sediment and maintaining areas of channel complexity that promote downwelling can be important for bull trout embryo survival. Management decisions should also consider the diverse behavior of juvenile bull trout and the wide range of habitat they use. Additionally, connectivity between populations is likely to be important for declining populations to persist. The diversity of life-history strategies expressed by bull trout helps maintain demographic stability within and among populations. As such, preservation of habitat integrity and full life-history diversity is imperative for conservation and recovery of bull trout populations range-wide.

Energetic Costs of AhR Activation in Rainbow Trout (Oncorhynchus mykiss) Hepatocytes

Aquatic organisms in response to toxic insults from environmental pollutants activate defence systems including the aryl hydrocarbon receptor (AhR) in an attempt to metabolize and excrete these toxicants and their metabolites. These detoxification mechanisms however may come with certain energetic costs. I hypothesize that the activation of the AhR by β-Naphthoflavone (β-NF), a model AhR agonist, results in increased energetic costs requiring metabolic reorganization in rainbow trout hepatocytes. While the results obtained suggest that there are no significant energetic costs of AhR activation, analysis of enzyme activities suggests possible metabolic reorganization. This study also showed significant changes in cellular processes in hepatocytes over the incubation periods which previously were not reported. Furthermore, for the first time in fish hepatocytes, metabolic flux analysis (MFA) was used to examine intra-cellular metabolism, the applicability of which is discussed.

Corticosteroidogenesis as a Target of Endocrine Disruption for the Antidepressant Fluoxetine in the Head Kidney of Rainbow Trout (Oncorhynchus mykiss)

Fluoxetine (FLX), the active ingredient of Prozac™, is a member of the selective serotonin reuptake inhibitor (SSRI) class of anti-depressant drugs and is present in aquatic environments worldwide. Previous studies reported that FLX is an endocrine disruptor in fish, bioconcentrating in tissues including the brain. Evidence implicates that serotonin influences the activity of the hypothalamo-pituitary-interrenal (HPI) stress axis, thus exposure to FLX may disrupt the teleost stress response. This study examined in vitro cortisol production in rainbow trout (Oncorhynchus mykiss) head kidney/interrenal cells exposed to FLX and 14C-pregnenolone metabolism in head kidney microsome preparations of FLX-exposed trout. Results indicated that cells exposed in vitro to increasing concentrations of FLX had lower cortisol production and cell viability (versus control) and microsomes isolated from trout exposed to 54 μg/L FLX had higher pregnenolone metabolism versus those of control and low FLX-exposed (0.54 μg/L) trout.

Effects of ibuprofen on Rainbow Trout (Oncorhynchus mykiss) following acute and chronic waterborne exposures

Pharmaceuticals and personal care products are a growing concern in the aquatic environment. Compounds from the class of non-steroidal anti-inflammatory drugs are commonly detected in surface waters and have the potential to negatively affect aquatic organisms. The purpose of this experiment was to determine the acute and chronic effects of ibuprofen on rainbow trout (Oncorhynchus mykiss). Cyclooxygenase (COX) activity, vitellogenin (VTG) concentration and ethoxyresorufin-O-deethylase (EROD) activity were evaluated following waterborne ibuprofen exposure of trout to 1 and 10 mg/L in the acute exposure and 1, 32 and 1000 μg/L in the chronic exposure, along with an experimental control, E2 control of 1000 μg/L and an E2-ibuprofen mixed treatment. Ibuprofen did not inhibit COX enzyme activity in either gill or kidney tissue. To evaluate the estrogenic effects of ibuprofen, VTG concentrations were measured; by the end of the 56 day chronic exposure VTG concentrations significantly increased in all of the ibuprofen treatments relative to the controls. EROD activity may have been inhibited by ibuprofen but definitive conclusions could not be made. These findings indicate that more research needs to be done studying ibuprofen in aquatic systems. / UOIT
