Flipping Biological Switches: Solving for Optimal Control: A Dissertation

Switches play an important regulatory role at all levels of biology, from molecular switches triggering signaling cascades to cellular switches regulating cell maturation and apoptosis. Medical therapies are often designed to toggle a system from one state to another, achieving a specified health outcome. For instance, small doses of subpathologic viruses activate the immune system’s production of antibodies. Electrical stimulation revert cardiac arrhythmias back to normal sinus rhythm. In all of these examples, a major challenge is finding the optimal stimulus waveform necessary to cause the switch to flip. This thesis develops, validates, and applies a novel model-independent stochastic algorithm, the Extrema Distortion Algorithm (EDA), towards finding the optimal stimulus. We validate the EDA’s performance for the Hodgkin-Huxley model (an empirically validated ionic model of neuronal excitability), the FitzHugh-Nagumo model (an abstract model applied to a wide range of biological systems that that exhibit an oscillatory state and a quiescent state), and the genetic toggle switch (a model of bistable gene expression). We show that the EDA is able to not only find the optimal solution, but also in some cases excel beyond the traditional analytic approaches. Finally, we have computed novel optimal stimulus waveforms for aborting epileptic seizures using the EDA in cellular and network models of epilepsy. This work represents a first step in developing a new class of adaptive algorithms and devices that flip biological switches, revealing basic mechanistic insights and therapeutic applications for a broad range of disorders.

Motivations for Indoor Tanning: Theoretical Models

This chapter reviews the literature applying health behavior theories to indoor tanning. Few studies have tried to fit full versions of health behavior models to indoor tanning. Theoretical models from the family of theories referred to as the reasoned action approach (e.g., theory of planned behavior, behavioral alternative model, prototype willingness model, etc.) have been most commonly used to study indoor tanning. Results indicate that these models fit indoor tanning data moderately to extremely well. Two lesser known models, problem behavior theory and the terror management health model, have also demonstrated a reasonable fit. Two other common models, the health belief model and social cognitive theory, have never been fully tested with indoor tanning. However, key constructs from these models (e.g., perceived susceptibility and threat, modeling) have been used to understand indoor tanning. Empirical research conducted represents a solid start toward developing strong, comprehensive models of indoor tanning that can guide intervention efforts. This initial work needs to be expanded by conducting longitudinal studies and by including a broader age range in studies because the majority of existing work has focused on young adults. Incorporating findings related to tanning dependency, peer group affiliation, media influences and other constructs into these foundational models will also improve our understanding and ability to develop efficacious interventions to reduce engagement in this health risk behavior.

Kartografické kulturní dědictví / Cartographic cultural heritage

Disertace je zaměřena na kartografické kulturní dědictví. Zkoumá postupy a stav jeho zpracování i způsoby a nástroje pro zpřístupnění v paměťových institucích. Byly vytčeny dva hlavní cíle práce. Nejprve zpracovat teoretický model online zpřístupnění kartografického kulturního dědictví a ověřit jej na vzorku mapových sbírek. Dále na základě teoretického modelu vytvořit online nástroj pro zlepšení služeb mapových sbírek. K dosažení cíle byly použity kvalitativní výzkumné metody. Konkrétně se jednalo o dotazníkové šetření českých a zahraničních mapových sbírek a také o případové studie českých projektů zaměřených na zpracování a zpřístupnění kartografických památek. K naplnění cílů práce byly podle knihovnického referenčního modelu IFLA navrženy dílčí modely uživatelských úloh, jejichž cílem bylo najít, určit, vybrat, získat a prozkoumat dílo. Na jejich základě byla doporučena kritéria pro hodnocení uživatelských úloh. K ověření funkčnosti modelu došlo testováním 20 digitalizovaných mapových sbírek. Byl vytvořen webový nástroj Databáze digitalizovaných mapových sbírek pro zlepšení služeb mapových sbírek.
