Porovnání koncentrace tuhých znečišťujících látek uvnitř stáje v rozdílných technologiích velkochovů drůbeže

Intensive poultry farming releases contaminated gases, odors, microorganisms and large concentrations of dust particles. Dust particles have a negative effect on the health of humans and animals. Dangerous particles with a diameter of less than or equal to 4 m, which can travel to the gas exchange area of the human respiratory system, pose a great health risk. The aim of this thesis is to measure PM10 dust particles in intensive poultry farms depending on the age of the chickens. Another goal is to compare the results between farms and compare them with the values shown in Table 5.15 on page 157 of document Final TWG meeting for review of the IRPP (Integrated Pollution Prevention and Control) BREF (Reference Document on Best Available Techniques). The measurements were carried out at the farm Alas a.s. in Hartmanice and at the farm Farma u lesa a.s. in Sudoměřice u Bechyně.

Měření emisí zápachu a tuhých znečišťujících látek z intenzivního chovu drůbeže

As agricultural production grows, the focus is on improving air quality, which affects not only residents around farms, but also climate change on a global scale. This thesis deals with the measurement of particulate pollutants and odor emissions in intensive poultry farming. The measurements were carried out at the Farma U Lesa in Sudoměřice u Bechyně in the spring and autumn of 2018. The production specific emission of odorous substances into the air was evaluated using dynamic olfactometry according to ČSN EN 13 725, which ranged from 0.01 to 0 according to the fattening cycle phase, 3 OUE. Ks-1. s-1. Compared to the BREF reference document, which lists emission odor values from 0.032 to 0.7 OUE. Ks-1. s-1 the measured values on the farm Mr. Šonky came out very low. Dust particles of PM10 fraction were measured according to the ČSN EN 12 341 methodology for the measurement of dust particles and the production specific emission of PM10 particles was found, which during the whole fattening cycle was measured to a maximum of 0.0025 kg. Ks-1.year-1. The measurement results were compared with the BREF reference document, where the maximum permissible production of solid pollutants of this fraction from 0.004 to 0.025 kg. Ks-1.year-1. In comparing the measured dust emission values with the BREF, the production of harmful emissions was very low.

Charakterizace virových nanočástic odvozených od myšího papilomaviru / Characterization of viral nanoparticles derived from mouse papillomavirus

The L1 and L2 capsid proteins of papillomaviruses are characterized by the ability to self- assemble into viral capsids, which can be divided into pseudovirions (PsVs) and virus-like particles (VLPs) by inner content. In addition to the fact that such particles can serve as "nano-containers" for diagnostic and therapeutic agents, it has also been shown that papillomaviruses, whether wild, PsVs or VLPs have a higher affinity for tumor tissue than non-tumor tissue. This thesis deals with relatively newly discovered (2011) mouse papillomavirus (MusPV) and nanoparticles derived from this virus. This papillomavirus has been chosen for its positives, including easy preparation of VLPs and PsVs, as well as an available model organism for possible testing. Furthermore, MusPV has the potential for use in gene therapy and cancer diagnosis, because there is no immune response in the human population. The aim of this diploma thesis is to prepare an expression system for the production of PsVs and VLPs. In additional it will also look at the quality and quantity of PsVs and VLPs, characterization of these particles and verification of existing postulates regarding higher affinity of papillomaviruses for tumor cells. Analýza emisí tuhých znečišťujících látek v chovu drůbeže / Analysis of emissions of particulate matter in poultry farming

This thesis was focused on monitoring production of dust particles in poultry farming. This measurement was made with the Dusttrak II 8530 which monitored the environment in the stable every 3 seconds for 24 hours. For monitoring were selected companies in the South Bohemian Region, specifically MTD Ústrašice and Farma u lesa. Results of Dusttrak measurements showed that none of the stables exceeded the permissible limits of dust particles. Additionally microscopic analysis of the samples of dust particles revealed that the main component of source of dust was feed mixture.

The objective of this diploma thesis is to assess the factors affecting the concentration of dust particles during forage pressing. For this purpose, five presses for square bales and five presses for round bales were selected. They were harvesting different kinds of straw. Thanks to measurement using an aerosol monitor, the concentration of dust particles during the harvest was detected. The measured values were evaluated and the amount of concentration of the dust particles between the square and round bale presses was compared. There were also set the methodological steps of the measurement procedure.

Student: Josef Mašek Title of Thesis: Particulate medicated systems This diploma thesis was focused on experimetal study of influence of technological parameters on the characteristics of nanoparticles made of terpolymer of aliphatic hydroxy acids with tripentaerythritol. In the theoretical part of this thesis nanoparticles as a system for targeted drug distribution were described. Methods of preparation of these nanoparticles, microencapsulation techniques, targeting, basic parameters of the nanoparticles, their size and zeta potential were mentioned in the theoretical part. The experiments were focused on influence of formulation factors, as detergent, concentration of the drug and polymer device on polydispersity of the nanoparticles, their zeta potential and encapsulation effectivity. The influence of detergent formulation on formation and parameters of the nanoparticles was proved. Methylsulfonylmethan was proved for the next experimental procedure as a suitable non-toxic solvent in compositions of biodegradable polymers.

In past years, nanoparticles have been studied as possible platform to be used in biomedicine. In order to establish the application potential of nanoparticles, its impact to biological systems have to be determined. Herein, several silicon-based nanoparticles of different origins were studied in respect of their influence on metabolic activity of human cells, namely osteoblast cell line SAOS-2 and monocytic cell line THP-1. The obtained results proposed that the impact of nanoparticles on cells is highly dependent on cultivation conditions in which nanoparticles are administered to cells. Furthermore, microscopy experiments were implemented in order to localize the particles within cells, where conventional microscopy limitations are evident. This paper examines the influence of air pollution on humans. Suggesting the possible consequences of each action of air pollutants on human health but also the possible economic impacts of air pollution. It primarily exploers the effects of concentration of suspended particulate matter (PM10) on the incidence of chronic obstructive pulmonary disease. The response variable in the regression model serves to determine the effect of PM10 on the incidence of chronic obstructive pulmonary disease was a percentage share of patients with chronic obstructive pulmonary disease among the clients of the General Health Insurance Company in individual regions of the Czech Republic. Explanatory variables were the mean annual concentration of PM10 and gross added value per capita. The model suggests that there is a statistically significant positive correlation between the incidence of chronic obstructive pulmonary disease in the Czech Republic and PM10 concentrations.

The aim of this thesis is to perform comparative analysis of the state policies of the Czech Republic and Poland aimed at reducing dust emissions from industrial and power generation sources of air pollution in the Moravia-Silesia and in the Silesian Voivodeship. The analysis is meant to facilitate improving the regulation requirements of these sources in both districts. The extent of regulation of the sources of air pollution in the Silesia Voivodeship influences the amount of dust emissions produced by such sources and, due the influence of the existence of cross-border transfer of emissions, this influences the concentration of particulate matter in the atmosphere in the Moravian-Silesian Region. The thesis arrives at the conclusion that the industrial and power generation sources of air pollution in the Czech Republic in the Moravian-Silesian Region faces stricter regulation requirements in comparison with similar sources in Poland in the former Silesian Voivodeship. Stricter conditions of regulation for Czech sources are the reason for the lower level of emissions and the resulting higher control costs such sources. The thesis concerns comparative analysis of the control costs a sample group of metallurgic and power generation plants in the Moravian-Silesian Region and in the Silesian Voivodeship.

Zařízení pro magnetické míchání suspenze s feromagnetickými částicemi / Device for magnetic stirring of suspension with ferromagnetic particles

The aim of this work deals with the design and manufacture of a device for mixing of magnetic beads in solution by magnetic field. It is based on my previous bachelor thesis, where the device was created for one microtube (one sample). In this following work, the three improved variants for mixing and separation of 96 samples at one time were designed. The first variant of device, derived from functional prototype from bachelor thesis, was manufactured. First, the magnetic circuit and power converter were designed. Subsequently, the device was manufactured just for one microtube due to functionality testing. The third variant was realized only experimentally, the construction was designed in CAD program, and individual parts were created using 3D printer. As an alternative solution, which should be the compromise among electromagnetic and mechanical stirring, additional prototype was manufactured using 3D printer and CNC instrument. This prototype utilizes neodymium magnets and a stepper motor. The capability of magnetic beads mixing in solution was verified.
