Izolace DNA z vybraných zeleninových výrobků (paprika) / DNA isolation from selected vegetable products (paprika)

Gőghová, Sabína January 2018 (has links)
The diploma thesis deals with micromethod of DNA isolation from ten differently processing food products containing pepper (Capsicum annum). PCR ready DNA was isolated by magnetic particles PGMA functionalized by carboxyl groups from homogenates prepared in lysis buffer with CTAB. Quantity and quality of DNA was estimated using spectrophotometric measurements and verified using PCR methods with primers specific for plant rDNA. Quality of isolated DNA varied depending on processing technology. DNA isolated from smoked grinded peppers and from heat treated food products was degraded and amplified with primers F_26S and R_26S (PCR product 220 bp) in contrary to the primers F_18S and R_5.8S (PCR product 700 bp). DNA isolated from the other food products was amplified with primers F_18S and R_5.8S (PCR product 700 bp). PCR product from one grinded pepper (Žitavská paprika) was cloned and sequenced.

Sitek, Tomáš January 2018 (has links)
This diploma thesis deals with issues of fine and ultrafine particles formation during thermal decomposition and combustion of biomass. An introduction is devoted to a utilization of biomass as fuel. A following part describes the gas products emerging generally during combustion process. A current emission and air pollution situation in the Czech Republic as well as the health impacts of solid pollutants on human are also mentioned. Within an experimental measurement three basic factors that have impact on fine particles formation during combustion of beech wood (i. e. oxygen content in combustion atmosphere, type of the sample and size of the sample) were thoroughly analyzed. Attention was paid to a progress of thermal decomposition and combustion of sample with respect to the particles formation. Finally, a measurement of fine particles amount was accomplished in a real boiler successively with four different biomass pellet types.

Environmentální a zdravotní zátěž z dopravy a možnosti jejich snižování v konkrétních podmínkách / The Environmental and Health Burden of Transport and the Possibility of its Reduction in Particular Conditions

Dvořáková, Dita January 2018 (has links)
Air quality is affected by many factors, one of the most important is transport, especially road transport. In this thesis, the transport intensity is compared with number/concentrations of particulate matter in the air. The evaluation is carried out for days, weeks and months, analyses are focused mainly on Brno. In addition, the particulate matter concentrations are analysed depending on meteorological conditions (temperature). The conclusion is focused on the possible measures which can reduce the impact of transport on air pollution.

In Vitro Study of the Effect of Particle Characteristics and Flow Rate on Regional Deposition in Human Airways / In Vitro Study of the Effect of Particle Characteristics and Flow Rate on Regional Deposition in Human Airways

Bělka, Miloslav January 2018 (has links)

Optimalizace metody pro hodnocení špinivosti ETICS / Optimization of method for evaluation ETICS dirtiness

Gabriel, Jindřich January 2021 (has links)
This diploma thesis deals with the issue of adhesion of pollution to the final surface treatments of the ETICS system. The theoretical part deals with important factors and processes that affect the degree of adhesion of pollution. The practical part of the diploma thesis follows the diploma thesis of Ing. Jakub Krejčík. Optimization of test methods for simulation of artificial pollution on tested samples was proposed. These samples were then evaluated in the laboratory and also compared with plasters that were exposed to real pollution from the natural environment.

PCR identifikace nepatogenních bakterií izolovaných ze sýrů / PCR identification of nonpathogenic bacteria strains in cheeses

Jurečková, Nela January 2010 (has links)
Different species of genus Bifidobacterium are part of human and animal intestinal flora. These bacteria have benefit effects and therefore they are used in foods and pharmaceutical products as probiotics. Cheese is now suitable as a probiotic matrix except yoghurts and fermentated milks. This diploma thesis was focused on optimalization of DNA isolation from bacteria of genus Bifidobacterium. Magnetic microparticles (P(HEMA-co¬-GMA)) were used for DNA isolation in presence of 8% polyethyleneglycol PEG 6000 and 5 M sodium chloride. Phenol extraction weas also used as an isolation method. Isolated DNA was used for amplification in domain, genus and species specific PCRs. Optimized method was tested for detection of bacteria of genus Bifidobacterium in experimentaly prepared probiotic cheeses. These cheeses contained potential probiotic bacteria from Laktoflóra collection. Bacteria were identified into species using species specific PCR. Species Bifidobacterium animalis was identified in all samples of probiotic cheeses.
