Interferon γ is an important T-cell helper type 1 (Th1) cytokine involves in antimicrobial immunity. It is a part of the inflammatory immune response in the site of infection. However, for its proper function, the regulation of immunity is necessary to avoid injury of the tissue caused by long-term inflammation. While interferon γ triggers expression of proinflammatory genes, it also regulates genes which inactivate immune response. The B7-H1 molecule belongs among these inhibitory regulators. Furthermore, antitumour effect of interferon γ is well-known as well. After extensive experiments, interferon γ was tested as an immunotherapeutic drug against melanomas in clinical trials. However, the trials had to be terminated prematurely because of unsuccessful results. It started to be clear that interferon γ could have also a protumour effect. Interferon γ upregulates the expression of B7-H1 molecule which aids tumour in escape from immunity. The B7-H1 molecule possesses a binding site for interferon regulatory factor 1 (IRF-1) in its promoter region. This IRF-1 is induced by interferon γ - JAK/STAT signalling pathway. Multiscale analysis of emulsions and suspensions with surface effects

The better understanding of the behavior of emulsions and suspensions is important in many applications. In general, emulsions allow the delivery of insoluble agents to be uniformly distributed in a more efficient way. At the same time suspensions of rigid particles are used as “smart materialsâ€� as their properties can be changed by the interaction with a magnetic or electric field. In the first part of the talk we consider a periodic emulsion formed by two Newtonian fluids in which one fluid is dispersed under the form of droplets of arbitrary shape, in the presence of surface tension. We assume the droplets have fixed centers of mass and are only allowed to rotate. We are interested in the time-dependent, dilute case when the characteristic size of the droplets aε, of arbitrary shape, is much smaller than the period length ε. We obtain a Brinkman type of fluid flow for the critical size aε = O(ε3) as a replacement of the Stokes flow of the emulsion. Additionally, using Mosco convergence and semigroup theory we extend the convergence to the parabolic case. For the case when the droplets convect with the flow, it can be shown again using Mosco-convergence that, as the size of the droplets converges to zero faster than the distance between the droplets, the emulsion behaves in the limit like the continuous phase and no “strangeâ€� term appears. Moreover, we determine the rate of convergence of the velocity field for the emulsion to that of the velocity for the one fluid problem in both the H1 and L2 norms. Additionally, a second order approximation is determined in terms of the bulk and surface polarization tensors for the cases of uniform and non-uniform surface tension. The second part of the talk is devoted to the study of MR fluids. We consider a suspension of rigid magnetizable particles in a non-conducting, viscous fluid with an applied external magnetic field. Thus, we use the quasi-static Maxwell equations coupled with the Stokes equations to capture the magnetorheological effect. We upscale using two scale asymptotic expansions to obtain the effective equations consisting of a coupled nonlinear system in a connected phase domain as well as the new constitutive laws. The proposed model generalizes the model of Rosenweig by coupling the velocity of the fluid and the magnetic field intensity. Using the finite element method we compute the effective coefficients for the MR fluid. We analyze the resulting MR model for Poiseuille and Couette flows and compare with experimental data for validation.

Trata-se de biopolímero aniônico que possui propriedades como solubilidade em água, atoxicidade, além de ser comestível e biodegradável. Devido a estas propriedades, possui muitas aplicações ambientais, dentre elas, o uso como incremento ao processo de floculação, no tratamento de água. O ácido γ-poliglutâmico pode ser produzido, através da fermentação da soja utilizando-se da bactéria Bacillus subtilis encontrada em alimentos como o natto, consumido como iguaria no Japão. Grande parte das aplicações de biopolímeros está diretamente envolvida com seu potencial de resposta às alterações do meio em que se encontram dissolvidos. Uma abordagem voltada ao entendimento das alterações estruturais, conformações e associações de cadeias poliméricas apresenta alto interesse científico, assim como aplicações práticas. O objetivo deste trabalho volta-se pela extração e quantificação do ácido γ-poliglutâmico de produtos comerciais, natto e PGα21Ca, para estudos como agente floculante. Observou-se diferença expressiva, na produção de ácido γ-poliglutâmico, em marcas distintas de natto. A metodologia de extração γ-PGA que representou maior eficiência foi através do uso de metanol refrigerado como agente de precipitação, onde em apenas uma extração obteve-se o biopolímero com atividade floculante, quando comparado à extração com o álcool etílico e acetona. Quanto ao método de extração de γ-PGA do PGα21Ca, a extração por álcool etílico apresentou-se eficiente na separação do ácido γ-poliglutâmico, sendo observado apenas pequeno residual de cálcio. / The γ-polyglutamic acid (γ-PGA) is a homopolyamide composed of D- and L- units of glutamic acid. It is anionic biopolymer that has properties such as water solubility, no toxicity, in addition to being edible and biodegradable. Due to these properties, it has many environmental applications, among them, the use as an increment to the flocculation process, in the treatment of water. γ-polyglutamic acid can be produced by fermenting soybeans using Bacillus subtilis bacteria found in foods such as natto, consumed as a delicacy in Japan. Most applications of biopolymers are directly involved with their potential to respond to changes from the environment in which they are dissolved. An approach aimed at understanding the structural changes, conformations and associations of polymer chains presents a high scientific interest, as well as practical applications. The objective of this work is the extraction and quantification of γ-polyglutamic acid from commercial products, natto and PGα21Ca, for studies as a flocculating agent. There was an expressive difference in the production of γ-polyglutamic acid in different natto brands. The methodology of γ-PGA extraction that represented greater efficiency was through the use of cooled methanol as a precipitation agent, where in only one extraction was obtained the biopolymer with flocculating activity, when compared to the extraction with ethyl alcohol and acetone. As for the γ-PGA extraction method of PGα21Ca, the ethyl alcohol extraction was efficient in the separation of the γ-polyglutamic acid, with only small residual of calcium being observed.

RELATIONSHIPS BETWEEN ANIMAL TEMPERAMENT AND SYSTEMIC IMMUNE RESPONSES IN BEEF CATTLE EXPOSED TO CONDITIONS ASSOCIATED WITH CONVENTIONAL MANAGEMENT

Measures of temperament have been shown to influence physiological responses. Exit velocity (EV) has been identified as an objective, robust measure of temperament that can be used to predict subsequent performance of cattle. Additionally, previous studies from our lab indicate this measure of temperament may be related to production of interferon-γ (IFN-γ), a cytokine associated with cell-mediated immunity (CMI). Whereas research has investigated effects of EV upon immune responses, the overall goal of these studies was to examine this relationship under a variety of scenarios including human handling, transportation, and exposure to endophyte-infected tall fescue (E+) for determination of its ability to influence CMI in cattle. In each of 5 experiments, calves were classified as either high or low EV animals, based upon measurements obtained prior to initiation of experimental periods. The hypothesis for these studies was that calves with high exit velocities would have lower systemic immune responses to applied treatments. Two experiments were designed to examine the relationship between exit velocity and lymphocyte IFN-γ production during and following a period of exposure to E+ seed and increased temperature humidity index conditions. Preliminary measures of this cytokine indicated a positive relationship with EV. During application of heat and E+ treatment application, no differences in IFN-γ production were detected between EV or endophyte treatment groups. However, in both experiments, after temperatures were returned to thermoneutral and E+ heifers were placed on the endophyte-free treatment, the positive relationship between exit velocity and total lymphocyte production of IFN-γ observed in baseline samples was reestablished. Similarly, during an experiment examining IFN-γ production by lymphocytes in steers during the 4 weeks following a 10h, 805 km transport study, average lymphocyte production of IFN-γ was higher and lymphocyte proportions producing IFN-γ lower in low EV steers, but total lymphocyte production of this cytokine did not differ between exit velocity treatments. In a grazing and finishing study, cattle were placed on E+ or novel endophyte pastures, with balanced representation of low and high EV treatments within each pasture. During the subsequent finishing period, blood samples for lymphocyte IFN-γ production were collected from a single high EV calf from each pasture group. Neither endophyte nor exit velocity was detected to be related with lymphocyte production of IFN-γ. In an experiment examining changes in cytokine gene expression changes during acclimation to human handling, IFN-γ, Il-6, IL-10, and IL-12 were observed to increase linearly over the experimental period in all calves, irrespective of exit velocity designation. In the same experiment, whole period pro-inflammatory tumor necrosis factor-α expression was higher for high EV calves, but interferon-γ (IFN-γ) was lower in this same treatment group. These studies, cumulatively, indicate EV may be related to systemic production of IFN-γ, but abrupt changes to an animal’s environment may serve to mask this relationship.

Design, Synthesis, Applications of Polymers and Dendrimers

WHO has reported that antibiotic resistance is the third major cause of human death all over the globe. Recent study, has focused on the development of new antibacterial resistance drugs. Herein, we tried to synthesis a series of polymers that can mimic the HDPs. HDPs can target the bacterial cell membrane and they have less chances to develop bacterial resistance. We synthesized the amphiphilic polycarbonates that are highly selective to Gram-positive bacteria, including multidrug resistant pathogens. The membrane disruption activity of these polymers was proved by fluorescence and TEM studies and the drug resistance study showed that the polymers don’t develop bacterial resistance. In order to further design the molecules that can target a broad spectrum of bacteria, we have designed a series of lipidated dendrimers that can target the Gram-positive and Gram-negative bacteria. These dendrimers mimic the HDPs and target the bacterial cell membrane. Dendrimers are reported to inhibit the formation of bacterial biofilm which makes them promising for their future development of antibiotic agents. Apart from the synthesis of polymers and dendrimers as antibacterial agents, we have designed a series of small molecular antibacterial agents that are based on the acylated reduced amide scaffold and small dimeric cyclic guanidine derivatives. These molecules display good potency against a panel of multidrug-resistant Gram-positive and Gram-negative bacterial strains. Meanwhile, they also effectively inhibit the biofilm formation. Mechanistic studies suggest that these compounds kill bacteria by compromising bacterial membranes, a mechanism analogous to that of host-defense peptides (HDPs). Lastly, we also demonstrate that these molecules have excellent in vivo activity against MRSA in a rat model. This class of compounds could lead to an appealing class of antibiotic agents combating drug-resistant bacterial strains.

Structure-based Design, Synthesis and Applications of a New Class of Peptidomimetics: <em>'Y</em>-AA Peptides and Their Derivatives

Peptidomimetics can mimic hierarchical structures of peptides and proteins. Thus, they are extensively studied for therapeutic applications. To break the limitation of backbones and frameworks and expand the peptidomimetics family, a new class of peptidomimetics - “γ-AApeptides” was developed. Design of γ-AApeptides is based on the chiral peptide nucleic acids (PNAs) backbone. The World Health Organization estimates that one -third of all deaths in the world are on account of infectious diseases. AMPs are important because of their high activity against broad spectrum microbes, less susceptible to grow resistance and selectivity in binding to bacterial cells over human cells. γ-AApeptides as a new class of peptidomimetics have increased stability and enhanced chemical diversity. We have developed polymyxin mimic cyclic peptides, small linear molecules and hydantoin derivatives as potent antibiotic agents with γ-AApeptides. They have good bioactivity and selectivity. Combinatorial library is key technology for accelerating the discovery of novel therapeutic agents. One-bead-two-compound γ-AApeptides-based library was developed and screened against SMYD2 protein which is essential for tumor growing.

Sonochemical and impregnated Co-W/γ-Al2O3 catalysts : performances and kinetic studies on hydrotreatment of light gas oil

γ-Al2O3 supported Co-W based catalysts with varying Co (1 - 3 wt %) and W (7 - 13 wt %) loadings were prepared using impregnation and sonochemical methods. All prepared catalysts were characterized with elemental analysis, BET analysis, X-ray diffraction (XRD), NH3 temperature programmed desorption (TPD), temperature programmed reduction (TPR) and thermogravimetry analysis (TGA). <p>The performances of all the synthesized catalysts were tested at a pressure of 8.9 MPa, LHSV of 2 h-1 and temperatures of 340, 350 and 360 °C in a laboratory trickle bed microreactor for hydrodesulphurization (HDS) and hydrodenitrogenation (HDN) of light gas oil (LGO) derived from Athabasca bitumen. The performance tests with impregnated catalysts indicated a maximum in activity for HDS and HDN reactions (sulfur and nitrogen conversions at 93.0 and 57.1 % at 360 °C) for Co(3 wt %)-W(10 wt %)/γ-Al2O3 whereas the performance tests with sonochemically prepared catalysts showed a maximum in activity (sulfur and nitrogen conversions at 87.9 and 42.5 % at 360 °C) for Co(3 wt %)-W(11.5 wt %)/ γ-Al2O3. These two catalysts were selected for detail performance, optimization and kinetic studies. The effects of reaction temperature (340 - 380 °C), pressure (7.6 - 10.3 MPa), liquid hourly space velocity (1.5 - 2.0 h-1) and hydrogen gas/gas oil ratio (400 - 800 mL/mL) were examined on HDS and HDN of LGO with these catalysts. The reaction kinetics for HDS was best fitted with a Power Law model whereas same for HDN was found to be best represented by a Langmuir-Hinshelwood model with a reasonable accuracy (0.90 <R2 <0.95). The activation energy for HDS of LGO were 14 and 12 kJ/mol for selected impregnated and sonochemically prepared catalysts whereas the same for HDN were 9 and 14 kJ/mol for these catalysts, respectively. Calculation showed that the fitted HDS rate expressions were apparent and HDN rate expressions were intrinsic under existing reaction conditions. It also showed that the pore diffusion resistances for both HDS and HDN increased with an increase in reaction temperature from 340 to 380 °C.

Observations of nearby Galaxy Clusters with the Fermi Large Area Telescope : Towards the first Gamma Rays from Clusters

Galaxy clusters are the most massive bound systems known in the Universe and are believed to have formed through large scale structure formation. They host relativistic cosmic-ray (CR) populations and are gravitationally bound by large amounts of Dark Matter (DM), both providing conditions in which high-energy gamma rays may be produced either via CR interactions with the intracluster medium or through the annihilation or decay of DM particles. Prior to the launch of the Fermi satellite, predictions were optimistic that these sources would be established as γ-ray-bright objects by observations through its prime instrument, the Large Area Telescope (LAT). Yet, despite numerous efforts, even a single firm cluster detection is still pending. This thesis presents a number of studies based on data taken by the LAT over its now seven year mission aiming to discover these γ rays. Using a joint likelihood technique, we study the γ-ray spectra of a sample of nearby clusters searching for a CR-induced signal due to hadronic interactions in the intracluster medium. While we find excesses in some individual targets, we attribute none to the cluster. Hence, we constrain the maximum injection efficiency of hadrons being accelerated in structure formation shocks and the fraction of CR-to-thermal pressure. We also perform a refined search targeting the Coma cluster specifically due to its large variety of existing observations in other wavebands. In the latter case we find weak indications of an excess which however falls below the detection threshold. Because the cluster emission we consider is inherently extended, we need to take into account the imperfect modeling of the foreground emission, which may be particularly difficult such as is the case with the Virgo cluster. Here, we assess the systematics associated with the foreground uncertainties and derive limits based on an improved background model of the region. For the first time we derive limits on the γ-ray flux from CR and DM-interactions in which we take into account the dynamical state of the system. For DM we also include the contribution from substructure. The DM domain is further explored by searching for line-like features as they arise from the annihilation of DM into two photons in a large sample of clusters, including Virgo and Coma. Finding no evidence for γ-ray lines, we derive limits on the DM annihilation cross section that are roughly a factor 10 (100) above that derived from observations of the galactic center assuming an optimistic (conservative) scenario regarding the boost due to DM substructure. / <p>At the time of the doctoral defense, the following papers were unpublished and had a status as follows: Paper 3: Submitted. Verlauf der zellulären Immunantwort bei Lebendnierenempfängern - Messung von IFN-γ und IL-17 im Elispot-Assay

Die Nierentransplantation ermöglicht Patienten die Wiederherstellung der Nierenfunktion. Aufgrund der begrenzten Verfügbarkeit an Organen nimmt dabei die Zahl der Transplantationen von einem lebenden Spender stetig zu. Zudem ermöglichen die präzisen und genauen Vorbereitungen und Abläufe bei Lebendnierenspenden eine bessere 5-Jahres-Überlebensrate als bei Kadaverspenden. Die genetische Verschiedenheit zwischen Spender und Empfänger bedingt jedoch eine lebenslange immunsuppressive Therapie, um Abstoßungsreaktionen und damit das Scheitern einer Organtransplantation zu verhindern. An den Universitätskliniken Leipzig und Halle/Saale besteht diese Therapie aus einer Dreifachkombination von Tacrolimus, Mycophenolat-Mofetil und Prednisolon, wobei mögliche Nebenwirkungen wie opportunistische Infektionen, kardiovaskuläre und metabolische Erkrankungen sowie Tumore in Kauf genommen werden. Zudem besteht für den immunsupprimmierten Organismus die ständige Gefahr einer Abstoßungsreaktion. Diese Aspekte führen bei den Empfängern zu einer massiven Einschränkung der Gesundheit und Lebensqualität. Inwieweit die ausgeprägte Immunsuppression notwendig ist, bleibt unklar und muss individuell festgelegt werden. Bisher existiert kein geeignetes Verfahren für ein Immunmonitoring, weshalb in vielen Fällen eine umfangreiche und überdosierte Immunsuppression in Kauf genommen wird. Im Rahmen dieser Arbeit wurde ein geeignetes Testverfahren, der Elispot-Assay, für die Expression der beiden proinflammatorischen Zytokine IFN-γ und IL-17 erstellt. Dafür wurden die PBMC der Spender und Empfänger aus Vollblut separiert, um sie anschließend sowohl separat als auch in einer Lymphozytenmischreaktion zu untersuchen. Die Darstellung von IL-17 konnte nur aufgrund einer zusätzlichen Stimulation mit OKT3 gelingen, während der IFN-γ-Elispot sowohl im Leerwert als auch unter Stimulation mit IL-2 zu ausreichenden Spotanzahlen führte. Die Spotanzahlen der Spender-PBMC wurden mit Hilfe von γ-Strahlung signifikant reduziert (IFN-γ: p=0,047 | IFN-γ + IL-2: p=0,007 | IL-17: p = 0,001), um in den Lymphozytenmischreaktionen die alleinige Zytokinausschüttung der Empfänger-PBMC messen zu können. Die Spender- PBMC fungierten dabei nur als Antigene. Insgesamt konnten zwischen 2009 und 2012 zwölf von siebzehn Patientenpaaren in die Studie eingeschlossen werden. Die Spotanzahlen der Paare wurden dabei sowohl im IFN-γ- als auch im IL-17-Elispot-Assay zu vier unterschiedlichen Zeitpunkten gemessen (vor Transplantation | 21±3 d postoperativ | 28±3 d postoperativ | 75±15 d postoperativ). In den meisten Fällen zeigte sich vor Transplantation eine erhöhte Spotanzahl im Vergleich zu den drei postoperativen Werten. Zudem stiegen die Spotanzahlen sowohl für IFN-γ als auch für IL-17 nach niedrigen Messergebnissen kurz nach der Transplantation im postoperativen Verlauf wieder an und erreichten in einigen Fällen die Spotanzahl der präoperativen Ausgangswerte. Ein signifikanter Unterschied konnte aufgrund der geringen Fallzahl nicht erreicht werden. Die kurzfristige Reduktion der Spotanzahlen postoperativ ist dabei aller Wahrscheinlichkeit nach auf die hohen Dosen an immunsuppressiven Medikamenten zurückzuführen. Insgesamt zeigten die Verläufe der IFN-γ- und der IL-17- Elispot-Assays ähnliche Verläufe. Daraus lässt sich schlussfolgern, dass der IL-17-Elispot- Assay in Bezug auf mögliche Abstoßungsreaktionen eine ähnliche Aussagekraft besitzen könnte wie der bereits vielfach untersuchte IFN-γ-Elispot-Assay. Weiterhin wurden die Messergebnisse mit der Serumkreatininmolarität verglichen. Diese zeigte präoperativ höhere Molaritäten als postoperativ, wobei die postoperativen Molaritäten im Verlauf, im Gegensatz zu den Elispot-Messungen, abnahmen, was das Einsetzen der Nierenfunktion widerspiegelt. Unter den zwölf Patientenpaaren gab es keine einzige nachgewiesene akute Abstoßungsreaktion, der Verlauf der Serumkreatininmolaritäten war bei allen zwölf Empfängern vergleichbar. Demzufolge konnten die Werte der Elispot-Assays nicht herangezogen werden, um an ihnen eine Abstoßungsreaktion der transplantierten Nieren erkennen zu können. Das präoperative Abschätzen einer möglichen Abstoßungsreaktion anhand der Elispot-Assays konnte aufgrund fehlender Abstoßungsreaktionen ebenfalls nicht untersucht werden. Zusätzlich wurde bei den Patienten eine HLA-Typisierung vorgenommen, wobei der Bereich von optimalen bis maximal ungünstigen Konstellationen reichten (HLA-Mismatch: 0-0-0 bis 2-2-2). Auch hier konnten die Ergebnisse nicht mit möglichen Abstoßungsreaktionen verglichen werden. In der vorliegenden Arbeit wurden zahlreiche Varianten untersucht, die das Abschätzen einer Immunreaktion nach Nierentransplantation (Immunmonitoring) ermöglichen könnten. Aufgrund fehlender Abstoßungsreaktionen bei den Empfängern konnte das Testverfahren nicht an den klinischen Verläufen validiert werden. Mit dem in dieser Arbeit entwickelten Messverfahren kann jedoch eine neue und größer angelegte Studie erfolgen, die in Zukunft ein Immunmonitoring bei Patienten nach Nierentransplantation ermöglicht. / Introduction Since the first kidney transplantation in the 1950ies, kidney transplantation is still being challenged by graft dysfunction and complete graft failure. Permanent immunsuppressive treatment is mandatory to avoid an unfavourable outcome. The treatment with Prednisolone, Tacrolimus and Mycophenolat-Mofetil may cause toxic side effects resulting in Diabetes mellitus, hypertension, infections and cancer. In the present study we tried to demonstrate that the amount of spots in the Enzyme linked immunospot assay (Elispot-Assay) of IFN-γ and IL-17 correlates with the probability of graft dysfuction and complete graft failure. We also compared the results to clinical parameters. Methods Between the years 2009 and 2012, twelve pairs of related living kidney transplantations were included in this study. From each pair blood samples were taken at four time points (before transplantation, and at 21±3, 28±3 and 75±15 days after kidney transplantation, respectively). After establishing the technique of IFN-γ- and IL-17-Elispot-Assays, we separated the periphale blood mononuclear cells (PBMC) and performed follow up examinations at the four time points mentioned above. The PBMC of each donor and each recipient were examined separatly, and in addition together in a lymphocyte mixed reaction. We stimulated the PBMC of the IFN-γ-Elispot with Interleukin-2 (IL-2) and the PBMC of the IL-17-Elispot with OKT3 to get significant characteristics. PBMC of the donors were irradiated with 30 Gy before mixing them with the PBMC of the recipients. We also took the HLA-matches and serum creatinine molarity to compare important clinical parameters with the results of the Elispot-Assays. Results Sufficient spots were measured using the unstimulated and stimulated IFN-γ-Elispot and the stimulated IL-17-Elispot. Radiation was significant at all three tests (IFN-γ: p=0,047 | IFN-γ + IL-2: p=0,007 | IL-17: p = 0,001). All twelve recipients showed a high number of spots before transplantation in both types of Elispot-Assays and most of them an increasing number of spots after a minimal turning point three weeks after transplantation. Due to the small number of cases, no significant results could be obtained at follow up. Non recipient developed a graft rejection as proven by biopsy or graft failure. The molarity of serum creatinine was permanently reduced whereas it was high before transplantation. Because of the abscence of any rejection episodes, HLA matches could not be compared. Discussion Due to the absence of rejection episodes or graft failure, no prediction for rejection by the IFN-γ- and IL-17-Elispot was possible. The low number of cases of living related kidney transplantation demonstrated the challange of the investigation of living related kidney transplantation. Although we could prove a significant effect of the irradiation of PBMC, there was no significant result in the follow up investigations. A higher number of cases are needed in future investigations. The established method of the IFN-γ- and IL-17-Elispot can be used in a future study with an extended number of cases and a longer follow up of time.
