Crystals of the title compound are monoclinic with a= 8.314 (5), b= 22.723 (9), c= 12.589 (6) A, /3= 123.0 (2t, Z=4, space group P2dc. The structure was determined by Patterson and Fourier methods and refined by full-matrix least squares to a final R of 0.030 for 2043 independent reflexions. The U atom has pentagonal bipyramidal coordination and the N-methylacetylacetoneamine is bonded to U via O. There are two intramolecular N-H. . .0 hydrogen bonds which govern the geometry of the adduct molecule.

Synthesis and characterization of new adsorbents for CO2 capturingPiet, Marvin January 2014 (has links)
Philosophiae Doctor - PhD / Carbon dioxide emissions have become a major concern as they are one of the contributing factors to the “green-house” effect. Recently, much effort has been put into separating carbon dioxide (CO2) from flue gases linked to the combustion processes at fixed point sources. The development of solid sorbents for adsorption based on CO2 capture has attracted much attention. Ordered Mesoporous Silica (OMS) materials have recently attracted much attention as solid adsorbents for capturing CO2. OMS have been investigated for this purpose owing to their high pore volume, large surface area and ease of functionalizationIn this work we report on the synthesis of OMS viz. MCM-41 and SBA-15 along with amorphous silica as adsorbents for CO2 capture. MCM-41 was prepared with surfactants having different alkyl chain lengths (C14TABr, C16TABr and C18TABr) where TABr is trimethylammonium bromide. SBA-15 was prepared using a Triblock copolymer as a structure directing agent for the organization of polymerizing silica species. Commercial amorphous silica gel was used for comparative purposes. Initial characterization OMS with powder X-Ray diffraction (XRD) and small angle diffraction (SAXS) yielded diffraction patterns which may be associated with well-ordered structures of hexagonal mesoporous material. Ease of preparation for MCM-41 materials allowed for convenient scale- up, obtaining highly ordered mesoporous silica MCM-41 at room temperature. SBA-15 was also found to be scaled up with considerable ease through increasing the volume of the autoclave during hydrothermal treatment. Structural, morphological and textural properties of the adsorbents were characterized by N2 physisorption measurements, Transmission Electron Microscopy (TEM), Scanning Electron Microscopy (SEM) and Thermogravimetric Analysis (TGA). TEM confirmed the hexagonal structure; SEM showed that C14MCM-41 had spheroidal particle morphology whereas SBA-15 displayed rod-like structures. High surface areas of up to 1302, 1186, 1211 and 1024 m2/g for C14MCM-41, C16MCM-41, C18MCM-41 and SBA-15, respectively were obtained. The pore size of MCM-41 materials was increased from 2.6 nm for C14MCM-41 to 4.4 nm in diameter for C18MCM-41 using surfactants with different alkyl chain lengths. CO2 adsorption characteristics of OMS were studied using CO2-temperature programmed desorption (TPD). The results showed that C14MCM-41, C16MCM- 41, C18MCM-41 and SBA-15 desorbed 0.19, 0.16, 0.11 and 0.26 mmol/g respectively. The synthesized OMS were then further modified by grafting various amine moieties on their surfaces in order to increase their CO2 adsorptive capabilities. 3-(Aminopropyl)triethoxysilane (APS), N-[3-(trimethoxysilyl)propyl]ethylene- diamine (TPED), 3-chloropropyl)-trimethoxysilane (CPS), ethyl 2-bromopropanoate, tris(2-aminoethyl)amine (TREN) and guanidine. Several characterization techniques such as XRD, SAXS, HRTEM, HRSEM, TGA, Fourier Transform Infra-Red (FT-IR) spectroscopy, CO2 isotherms and CO2-TPD were used to analyze amine grafted solid sorbents for CO2 capture. The results revealed that the structural integrity of the amine modified sorbents was not compromised during the grafting process. The structural properties of the supports, such as surface area and pore size, nature of amine and the number of amine groups, affected the loading and CO2 adsorption capacity of chemically grafted sorbents. APS grafted amorphous silica gel adsorbed 0.67 mmol/g CO2, which proved to be the highest compared to C18MCM-41 and SBA-15 grafted with APS and TPED. C18MCM-41, SBA-15 and amorphous silica gel were also grafted with TREN and adsorbed 0.42, 0.51 and 0.27 mmol/g of CO2, respectively. A systematic study of guanidine grafted on C14MCM-41, C16MCM-41 and C18MCM-41 was investigated, for the first time, to the best of our knowledge. Structural properties like variation in pore size, proved to enhance the adsorption capacity of the adsorbent, coupled with the guanidine molecules grafted on MCM-41 materials. C18MCM-41-guanidine showed the highest CO2 uptake of the guanidine grafted MCM-41 materials, adsorbing 0.30 mmol/g. A novel synthetic route to TPED-TREN grafted C18MCM-41 and SBA-15, using ethyl 2-bromopropanoate as a linker, was investigated as a potential adsorbent for CO2 capture, for the first time. C18MCM-41-TPED-TREN and SBA-15-TPED-TREN CO2 adsorption capacity were found to be 0.14 and 0.14 mmol/g, respectively.

Amine Transaminases in Multi-Step One-Pot ReactionsAnderson, Mattias January 2017 (has links)
Amine transaminases are enzymes that catalyze the mild and selective formation of primary amines, which are useful building blocks for biologically active compounds and natural products. In order to make the production of these kinds of compounds more efficient from both a practical and an environmental point of view, amine transaminases were incorporated into multi-step one-pot reactions. With this kind of methodology there is no need for isolation of intermediates, and thus unnecessary work-up steps can be omitted and formation of waste is prevented. Amine transaminases were successfully combined with other enzymes for multi-step synthesis of valuable products: With ketoreductases all four diastereomers of a 1,3-amino alcohol could be obtained, and the use of a lipase allowed for the synthesis of natural products in the form of capsaicinoids. Amine transaminases were also successfully combined with metal catalysts based on palladium or copper. This methodology allowed for the amination of alcohols and the synthesis of chiral amines such as the pharmaceutical compound Rivastigmine. These examples show that the use of amine transaminases in multi-step one-pot reactions is possible, and hopefully this concept can be further developed and applied to make industrial processes more sustainable and efficient in the future. / <p>QC 20170113</p>

L'entreprise Juxta produit des résines pour des revêtements transparents de fortes épaisseurs en polyuréthane. Ces revêtements sont fabriqués à partir de deux produits dangereux : les isocyanates et un catalyseur à base de mercure. Pour protéger ses employés et consommateurs, l'entreprise Juxta a lancé un projet de substitution des isocyanates dans les polyuréthanes. Pour ce faire, trois nouvelles voies de synthèse ont été envisagées :La voie cyclocarbonate/amine est la plus novatrice. Dans un premier temps, la réactivité de cyclocarbonates et d'amines modèles a été étudiée ainsi que l'effet de différents catalyseurs. Les meilleurs catalyseurs sont de la famille des thiourées, urées et guanidines. Puis une voie d'accès à des cyclocarbonates incolores a été mise au point. Enfin, une étude a permis de mettre en évidence que les liaisons hydrogènes étaient responsables du faible avancement de la réaction lors de la synthèse de polyhydroxyuréthanes linéaires.La voie époxy/amine a permis de développer une gamme de matériaux transparents avec des Tg allant de 18 à 47°C. Une étude IR a été réalisée, permettant de mieux appréhender le phénomène de carbonatation de l'amine, récurent lors de la synthèse de résines époxys à température ambiante. L'utilisation de prépolymères aminotéléchéliques permet de contourner ce phénomène.La voie acrylate/amine a permis de développer un matériau transparent de basse Tg, -12°C. L'étude de la réactivité des acrylates avec les amines a été réalisée par DSC et RMN. The thioureas, ureas and guanidines proved to be very efficient catalyst families for the cyclocarbonate aminolysis. Then a synthesis was developed to obtain transparent cyclocarbonates. To conclude, a study conducted on linear polyurethanes demonstrated that hydrogen bondings are responsible for the low conversions observed with this reaction.Transparent materials with Tg ranging from 18°C to 47°C were synthesized using the epoxy/amine pathway. An IR study was performed to better understand the amine carbonation phenomenon. It is a frequent phenomenon when the epoxy resins are synthesized at room temperature. Aminotelechelic prepolymers can be used to circumvent it.A low Tg (-12°C) transparent material was synthesized using acrylate/amine synthesis. The couple reactivity was studied by DSC and NMR. PART I Animals need to constantly evaluate their external environment in order to survive. In some cases the internal state of the animal changes to cope with it’s surrounding. In our study we wanted to investigate the role of amines in modulating internal states of Drosophila. We have designed a behavioral paradigm where the flies are fixed in space but can walk on a small styrofoam ball suspended by a gentle stream of air. The walking activity of flies was used as behavioral readout. PART I Animals need to constantly evaluate their external environment in order to survive. In some cases the internal state of the animal changes to cope with it’s surrounding. In our study we wanted to investigate the role of amines in modulating internal states of Drosophila. We have designed a behavioral paradigm where the flies are fixed in space but can walk on a small styrofoam ball suspended by a gentle stream of air. The walking activity of flies was used as behavioral readout. An operant training paradigm was established by coupling one of the walking directions to incidence of heat punishment. We observed that animals quickly realized the contingency of punishment with walking direction and avoided walking in the punished direction in the presence of punishment, but did not continue walking in the unpunished direction in the absence of the punishment. This would indicate that the flies do not form a memory for the punished direction or rapidly erase it under new conditions. On having established the paradigm with heat punishment we have attempted to activate selected subsets of neuronal populations of Drosophila while they were walking on the ball. The selective activation of neurons was achieved by expressing the light-activated ion channel channelrhodopsin-2 (ChR2) using the Gal4-UAS system and coupling the unidirectional walking of the animals on the ball with the incidence of blue light required to activate the channels and depolarize the neurons. The feasibility of this approach was tested by light-activating sugar sensitive gustatory receptor neurons expressing ChR2, we found that when the light was actuated the flies preferred to turn in one direction the optically “rewarded” direction. Next we similarly activated different subsets of aminergic neurons. We observed that in our setup animals avoided to turn in the direction which was coupled to activation of dopaminergic neurons indicating that release of dopamine is disliked by the animals. This is in accordance with associative learning experiments where dopamine is believed to underlie the formation of an association between a neutral conditioned stimulus with the aversive unconditioned stimulus. However, when we activated tyraminergic/octopaminergic neurons we did not observe any directional preference. The activation of dopaminergic and tyraminergic/octopaminergic neurons led to arousal of the animals indicating that we were indeed successful in activating those neurons. Also, the activation of serotonergic neurons did not have any effect on directional preference of the animals. With this newly established paradigm it will be interesting to find out if in insects like in mammals a reward mediating system exists and to test subsets of aminergic or peptidergic neurons that could possibly be involved in a reward signaling system which has not been detected in our study. Also, it would be interesting to localize neuropile regions that would be involved in mediating choice behavior in our paradigm. PART II In collaboration with S. Kneitz (IZKF Wuerzburg) and T. Nuwal we performed genome-wide expression analysis of two pre-synaptic mutants - Synapsin (Syn97) and Synapse associated protein of 47 kDa (Sap47156). The rationale behind these experiments was to identify genes that were up- or down-regulated due to these mutations. The microarray experiments provided us with several candidate genes some of which we have verified by qPCR. From our qPCR analysis we can conclude that out of the verified genes only Cirl transcripts seem to be reproducibly down regulated in Synapsin mutants. The Cirl gene codes for a calcium independent receptor for latrotoxin. Further qPCR experiments need to be performed to verify other candidate genes. The molecular interactions between CIRL and SYN or their genes should now be investigated in detail. / Teil I Lebewesen müssen beständig ihre äußere Umgebung auswerten, um überleben zu können. Manchmal ändern sich innere Zustände der Tiere, damit sie sich der Außenwelt anpassen. In unseren Untersuchungen wollten wir die Rolle von Aminen untersuchen, die für die Modulation von inneren Zuständen bei Drosophila notwendig sind. Wir haben ein Verhaltensparadigma entwickelt, bei dem die Fliege räumlich fixiert ist, aber auf einem Styroporball laufen kann, der auf einem Luftpolster schwebt. Die Laufaktivität der Fliege wird durch die Ballbewegungen anzeigt. Mit diesem Versuchsaufbau wurde ein operantes Lernparadigma etabliert, bei dem eine Laufrichtung mit Bestrafung durch Hitze gekoppelt wird. Wir konnten feststellen, dass die Tiere schnell den Zusammenhang zwischen dem Auftreten der Bestrafung und der Laufrichtung realisieren und die bestrafte Seite vermeiden. Wurde die eine Laufrichtung nicht mehr bestraft, so vermieden die Fliegen sie nicht mehr. Dies zeigt dass die Fliegen kein Gedächtnis für die bestrafte Richtung ausbilden oder es bei veränderten Bedingungen rasch löschen . Nachdem sich der Versuchsaufbau mit Hitzebestrafung bewährt hatte, wurde versucht, bestimmte Sub-population von Neuronen von Drosophila zu aktivieren, während die Fliege auf dem Ball läuft. Die selektive Aktivierung von Neuronen wurde durch die Expression des lichtaktivierten Jonenkanals Channelrhodopsin-2 (ChR-2) mit Hilfe des Gal4-UAS-System und Beleuchtung der Fliege mit Blaulicht erreicht, das die Kanäle aktiviert. Nun erfolgte eine Kopplung einer Laufrichtung auf dem Ball mit dem Auftreten von blauem Licht. Die Durchführbarkeit derartiger Experimente wurde durch die Aktivierung von zuckersensitiven gustatorischen Rezeptorneuronen getestet. Die Ergebnisse zeigten, dass die Tiere bevorzugt die Richtung wählen, welche die Zuckerrezeptorneurone aktiviert. Anschließend aktivierten wir verschiedene Untergruppen von Neuronen des aminergen Systems. In diesem Versuchsaufbau beobachteten wir, dass die Tiere die Laufrichtung vermieden, die gekoppelt war mit der Aktivierung dopaminerger Neurone. Diese Ergebnisse stehen in Übereinstimmung mit Versuchen zum assoziativen Lernen, bei dem Dopamin wahrscheinlich notwendig ist für die Assoziation zwischen dem neutralen Konditionierungsstimulus und dem aversiven unkonditionierten Stimulus. Wenn wir jedoch die tyraminergen/oktopaminergen Neurone aktivierten, konnte keine gerichtete Präferenz nachgewiesen werden. Die Aktivierung dopaminerger und tyraminger/oktopaminerger Neurone führte jedoch zur Aktivitätssteigerung der Tiere, wodurch die erfolgreiche der Aktivierung der Neurone belegt wurde. Die Aktivierung serotonerger Neurone führte ebenfalls zu keinem Effekt in der Präferenz der Tiere. In zukünftigen Experimenten mit dem neu entwickelten Paradigma wäre es interessant, herauszufinden, ob in Insekten auch ein belohnungsabhängiges System existiert, vergleichbar dem von Säugern. So wäre die Identifizierung von Subpopulationen aminerger oder peptiderger Neurone des Belohnungssystems bei Insekten wichtig, da dies nicht in unseren Experimenten entdeckt wurde. Eine weitere interessante Fragestellung wäre, welche Gehirnstruktur die Richtungswahl auf dem Ball vermittelt. Teil II In Zussamenarbeit mit S. Kneitz (IZKF, Würzburg) und T. Nuwal wurde in der vorliegenden Arbeit die genomweite Genexpression einer Synapsin-Mutante (Syn97) und einer Mutante für das Synapsen-assoziierte-Protein von 47kDa (Sap47156) untersucht. Bei beiden Proteinen handelt es sich um präsynaptische Proteine von Drosophila. Ziel dieses Experiments war es, Gene zu identifizieren die aufgrund dieser Mutationen hoch bzw. herunterreguliert vorliegen. Durch das Microarray-Experiment wurden mehrere Kandidatengene entdeckt, wovon einige dieser Gene per qPCR-Versuchen verifiziert wurden. Aufgrund der qPCR-Analysen lässt sich schlussfolgern, dass nur das Cirl-Transkript in den Synapsin-Mutanten reproduzierbar herunterreguliert vorliegt. Das Cirl gene kodiert für einen Calcium independent receptor for Latrotoxin Weitere qPCR-Experimente sind notwendig, um die anderen Kandidatengene zu bestätigen.
