Papain: a Novel Urine Adulterant.

The estimated number of employees in the United Stated screened annually for illicit drugs is approximately 20 million, with marijuana being the most frequently abused drug. Urine adulterants provide an opportunity for illicit drug users to obtain a false negative result on commonly used primary drug screening methods such as the Fluorescence Polarized Immunoassay (FPIA) technique. Typical chemical adulterants such as nitrites are easily detected or render the urine specimen invalid as defined in the proposed federal guidelines for specimen validity testing based on creatinine, specific gravity and pH. Papain is a cysteine protease with intrinsic ester hydrolysis capability. The primary metabolite of the psychoactive chemical in marijuana, 11-norcarboxy-delta-9-tetrahydrocannibinol (THC-COOH), was assayed by FPIA in concentrations ranging from 25 to 500 ng/mL, at pH values ranging from 4.5 to 8, over the course of 3 days with papain concentrations ranging from 0 to 10 mg/mL. FPIA analysis of other frequently abused drugs: amphetamines, barbiturates, benzodiazepines, cocaine, opiates, and phencyclidine, along with gas chromatography/mass spectrometry (GC/MS) of THC-COOH and high pressure liquid chromatography/ultraviolet detection (HPLC/UV) of nordiazepam was performed in order to determine if the mechanism of urine adulteration by papain was analyte specific. Control and adulterated urine specimens (n=30) were assayed for creatinine, specific gravity and pH to determine if papain rendered the specimens invalid based on the proposed federal guidelines. There was a direct pH, temperature, and time dependent correlate between the increase in papain concentration and the decrease in THC-COOH concentration from the untreated control groups (p<0.01). The average 72 hour THC-COOH concentration decrease at pH 6.2 with a papain concentration of 10 mg/mL was 50%. Papain did not significantly decrease the concentration of the other drugs analyzed with the exception of nordiazepam. GC/MS of THC-COOH and HPLC/UV of nordiazepam revealed a 66% and 24% decrease in concentration of the respective analyte with 10 mg/mL papain after 24 hours at room temperature (~23 °C). No adulterated specimens were rendered invalid based on the SAMHSA guidelines. Immediate FPIA analysis is suggested to minimize the interfering effects of papain with regards to primary drug screening.

Performance and ageing quantification of electrochemical energy storage elements for aeronautical usage / Evaluation des performances et du vieillissement des éléments de stockage d’énergie électrochimiques pour l’usage aéronautique

Dans un contexte de progression du stockage d’énergie sous forme électrochimique dans les transports, notamment dans l’aéronautique, les problématiques de performance, de fiabilité, de sureté de fonctionnement et de durée de vie du stockeur sont essentielles pour utilisateurs. Cette thèse se focalise ces voltes pour l’avion plus électrique. Les technologies étudiées correspondent à des éléments commerciaux de dernière génération de type Lithium-ion (NMC/graphite+SiO, NCA/graphite, LFP/graphite, NMC/LTO), Lithium-Soufre (Li-S), supercondensateur et hybride (LiC). Une première partie de ce manuscrit s’attache à la quantification des performances des différents éléments dans l’environnement aéronautique [-20°C, 55°C] et pour l’usage aéronautique. Un modèle comportemental de type électro-thermique est développé et validé. La seconde partie est consacrée à la quantification du vieillissement des différents éléments. Les résultats de vieillissement calendaire et en cyclage actif sont présentés ainsi que ceux des tests abusifs. Une méthode d’estimation de l’état de santé (SOH) des éléments basés sur l’analyse de la capacité incrémentale (ICA) est proposée. Enfin, l’évaluation de la robustesse des éléments de stockage lors de tests de vieillissement accéléré avec un profil spécifique à l’usage aéronautique est proposé. Les modèles de vieillissement et la méthode d'estimation de SOH proposés précédemment sont utilisés ici pour évaluer l'impact de la température sur la vitesse de dégradation et pour estimer le SOH des cellules vieillies à l’aide de ce profil aéronautique. / In the context of progress in the electrochemical energy storage systems in the transport field, especially in the aeronautics, the issues of performance, reliability, safety and robustness of these elements are essential for users. This thesis is focused on these issues for the more electric aircraft. The technologies studied correspond to the latest generation commercial elements of Lithium-ion batteries (NMC/ graphite + SiO, NCA/graphite, LFP/graphite, NMC/LTO), Lithium-Sulfur (Li-S), Supercapacitor and Lithium-ion capacitors. The first part of this manuscript is dedicated to the performance quantification of the different electrochemical energy storage elements in aeronautical environment [-20°C, 55°C] and usage. An efficient and accurate electro-thermal model is developed and validated. The second part is devoted to the calendar and power cycling ageings as well as to the presentation of abuse testing results. A State Of Health (SOH) estimation based on incremental capacity analysis method is proposed. Finally, the robustness of the storage elements during accelerated ageing tests with a specific profile for the aeronautical usage is evaluated. The ageing models and SOH estimation methods proposed in the previous sections are used here to evaluate the impact of temperature on the degradation rate and to estimate the SOH of the cells with this aeronautical profile.

Qualitat en l’anàlisi de drogues d’abús en cabell i en fluid oral

The use of hair and oral fluid for drugs of abuse testing has increased over the last years. For this reason, the assurance that results provided using these matrices are reliable and error-free is needed. The objective of this thesis is to develop tools to assess the quality of results provided by laboratories analysing drugs of abuse in hair and in oral fluid and to evaluate the effect on the quality of results of different actions carried out. For this reason, intercomparison exercises have been organized and some studies have been performed to develop appropriate quality control materials. Regarding the analysis of drugs of abuse in hair, nine different intercomparison exercises have been organized. The evaluation of qualitative and quantitative results reported by laboratories together with the study of the methodology used, has led to know the quality of the results, to identify the sources of error and to know the corrective actions that should be developed. Concerning the quality control material, these exercises have enabled to know the influence on the results of the type of hair used to perform the analysis. Concerning the analysis of drugs of abuse in oral fluid, a method has been developed to identify and quantify 6-monoacetyl morphine, morphine, codeine, cocaine and benzoylecgonine in oral fluid samples. On the other hand, stability studies of the main drugs of abuse in oral fluid have been done to establish the optimal preparation, transport and storage conditions, and finally, two intercomparison exercises have been conducted to know the performance of analytical laboratories when analysing drugs of abuse in oral fluid and to know the stability of some drugs of abuse in two commercial collection devices. / La creixent utilització del cabell i del fluid oral per a l’anàlisi de drogues d’abús, ha portat a la necessitat d’assegurar que els resultats obtinguts utilitzant aquestes matrius són fiables i lliures d’error. L’objectiu d’aquesta tesi és desenvolupar eines que permetin avaluar la qualitat dels resultats dels laboratoris que analitzen drogues d’abús en cabell i en fluid oral i avaluar l’efecte de diferents accions sobre la qualitat final dels resultats. Per això s’han organitzat exercicis interlaboratori i s’han realitzat estudis per desenvolupar material d’assaig adequat. Pel que fa a l’anàlisi de drogues d’abús en cabell, s’han realitzat nou exercicis interlaboratori. A través de l’avaluació dels resultats qualitatius i quantitatius informats pels laboratoris i mitjançant l’estudi de la metodologia emprada, ha estat possible conèixer la qualitat dels resultats obtinguts, detectar les fonts d’error i conèixer quines mesures correctives caldria desenvolupar. Pel que fa al material d’assaig, aquests exercicis han permès conèixer la influència que té, en els resultats, el tipus de cabell del qual es parteix per realitzar l’anàlisi. Pel que fa a l’anàlisi de drogues d’abús en fluid oral, s’ha desenvolupat i validat un mètode analític que ha permès identificar i quantificar 6-monoacetil morfina, morfina, codeïna, cocaïna i benzoïlecgonina en mostres de fluid oral. Per altra banda, s’han realitzat estudis d’estabilitat de les principals drogues en fluid oral que han permès establir les condicions òptimes de preparació, transport i conservació del material d’assaig i, per últim, s’han realitzat dos exercicis interlaboratori que han permès conèixer la qualitat dels resultats analítics obtinguts pels laboratoris i també l’estabilitat de les drogues en dos dispositius de recollida comercials.
