No. of bitstreams: 1 Dissertação -Pricilla Louise Leite da Silva.pdf: 1906908 bytes, checksum: 31683da2a9bf5ed9615a30941ba001dd (MD5) Previous issue date: 2014-03-26 / Não Informada / The present dissertation describes the physical-chemical, chemical and biological characteristics of latex utilis Couma (Apocynaceae), fruit species, genuinely Brazilian, endemic of Amazon region, popularly known as rowan, whose latex is used in food. After extraction, the latex is separated in three phases: rubber particles (F1), serum (F2) and lutoids (F3). Morphological characterization of latex, previously heated at 45°C was carried out by Microscopy of Atomic Force and was observed the formation of films and roughness from raw latex, anticoagulant serum, diluted, centrifuged and filtered serum. Presence of proteins was not detected in Maldi-TOF spectrometer analysis. However, filtered and lyophilized serum extract, by chromatographic fractioning and further analysis by NMR of isolated compound, allowed identifying the occurrence of α-amyrin acetate. The antimicrobial activity of F2 serum was evaluated using microorganisms: Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Salmonella typhimurium , Aspergillus Niger and Candida albicans. An inhibition of growth for Staphylococcus aureus and Candida albicans in the final concentration of 50 μl/ml of F2 was observed. / A presente dissertação descreve as características físico-químicas, químicas e atividades biológicas do látex de Couma utilis (Apocynaceae), espécie frutífera, genuinamente brasileira, endêmica da Região Amazônica, conhecida popularmente como sorveira, cujo látex é utilizado em alimentação. O látex, após a extração, separa-se em três fases: partículas de borracha (F1), soro (F2) e lutóides (F3). A caracterização morfológica do látex, previamente aquecido a 45°C, foi realizada por Microscopia de Força Atômica e observou-se a formação de filmes e rugosidade a partir do látex bruto, soro com anticoagulante, soro diluído, filtrado e centrifugado. Não se detectou a presença de proteínas em análise no Espectrômetro de Massas Maldi-TOF. Entretanto, o extrato filtrado e liofilizado do soro, através de fracionamento cromatográfico e posterior análise por RMN do composto isolado, permitiu identificar a ocorrência de acetato de α-amirina. A atividade antimicrobiana do soro (F2) foi avaliada empregando os micro-organismos: Pseudomonas aeruginosa, Staphylococcus aureus, Bacillus subtilis, Escherichia coli, Salmonella typhimurium, Aspergillus niger e Candida albicans. Observou-se a inibição de crescimento para Staphylococcus aureus e Candida albicans na concentração final de 50 μL/mL de F2.
