It is widely distributed in tropical and subtropical regions, including Brazil. Some studies show that the latex of P. rubra has gastric antiulcer effect,antioxidant and vasodilator activity. The aim of this study was to evaluate the effect of isolated proteins from Plumeria rubra latex (PrLP) in arthritis induced by zymosan (Zy). METHODOLOGY: This study was approved by the Ethics Committee on Animal Research of the UFC-CEPA (82/14). Arthritis was induced in female Swiss mice (n = 6) by injection of 0.1 mg Zy intra-articular. After 1 hour of Zy injecton, the animals were treated with saline or PrLP (12.5, 25, 50 and 100 mg/kg i.v.); the dexamethasone (2 mg/kg i.v.) was administered thirty minutes before Zy treatment. After six hours of arthritis induction, the animals were euthanized, the joint washing carried and collected for leukocyte count, determination of the myeloperoxidase (MPO) activity and inflammatory mediators (NO, TNF-α, IL-1β, IL-6 and IL-10). The edema was evaluated by measuring the diameter articular and the vascular permeability through extravasation evans blue. In addition, immunohistochemistry was performed for cyclooxygenase 2 (COX-2), NOSi, NfkB e TNF- α in the tissue.The mechanical hypernociception was induced by intra-articular injecton of Zy or intra-plantar injection of carrageenan or prostaglandin E2.The effect antinociceptive of PrLP was evaluated in the absence and in the presence of Naloxone (1 mg / kg; i.v.) and L-arginine (1g / kg; i.p.), which were administered 30 minutes before of the administration of PrLP. The statistical analysis were performed using the ANOVA/ Newman Keuls or Kruskal Wallis/Dunn post hoc test. Values of p <0.05 was considered statistically significant. RESULTS PrLP at doses of 25, 50 and 100 mg /kg iv reduced the leukocyte count (899.8 x 106 / ml ± 148; 1225 x 106 / ml ± 89.59 and 890 x 106 / ml ± 165.7, respectively), the neutrophil count (860.3 ± 97.57; 1108 ± 127.6 and 908.7 ± 159.9, respectively) and the MPO activity (12.1 ± 1.3;14.3 ± 0.6 and 8.9 ± 3.5 UMPO / ml, respectively). PrLP (25 mg/kg) modulated the concentrations of the inflammatory mediators such as NO, IL-1β, TNF-α and IL-6 (71.84 ± 9.1 mM;, 4569 ± 712.5;, 1409 ± 221.6 and 847.3 ± 260.4 pg/ml respectively) when compared to the saline group. PrLP (25 mg/kg). Reduced the edema, which significantly reduced joint diameter in the third (0.89 ± 0.001 mm) and in the sixth hour (0.99 ± 0.02 mm) and also reduced the extravasation of evans blue to the joint (40.46 ± 3.6 μg/g). In the tissue histopathological study, PrLP (25 mg / kg) significantly reduced the inflammatory infiltrate (with reduction in the neutrophil count), edema, synovial membrane involvement and haemorrhage. PrLP (25 mg / kg) was also able to reduces the immunohistochemistry labeling for COX-2, NOSi, TNF-α and NFkB. In mechanical hypernociception, PrLP (25 mg / kg) significantly reduced both the third (0.86 ± 1.3) and the sixth hour (1.08 ± 2.3), and this effect was reversed by the administration of L-arginine (1g / kg, ip) and naloxone (1mg / kg, iv). In addition, PrLP (25 mg / kg) significantly reduced carrageenin-induced plantar hypernociception in the third (1.74 ± 0.4) and fifth hour (1.5 ± 0.4) and prostaglandin-induced hypernociception in the third hour (1.4 ± 0.4). CONCLUSION: PrLP has antinociceptive and anti-inflammatory activity in the model of arthritis induced by Zy. The effect seems to be related to the decreased migration of neutrophils and the release of inflammatory mediators, reducing damage to the synovial membrane and hypernociception. / Plumeria rubra L. é uma planta laticífera que pertence à família Apocynaceae. Conhecida popularmente como jasmim, jasmim-manga, frangipani ou árvore-pagode, é originária da América Central e encontra-se amplamente distribuída pelas regiões de clima tropical do mundo, inclusive no Brasil. Estudos com a fração do látex de Plumeria rubra demonstraram apresentar efeito antiulcera gástrica, antioxidante e atividade vasodilatadora. O presente trabalho teve como objetivo avaliar o efeito da fração proteica isolada do látex de Plumeria rubra (PrLP) na artrite induzida por zymosan (Zy) em camundongos. METODOLOGIA: Esse estudo foi aprovado na comissão de ética em pesquisa animal da Universidade Federal do Ceará (n°82/2014). A artrite foi induzida em camundongos fêmeas swiss (n=6) através da administração de 0,1 mg Zymosan (Zy) i.a. Os animais foram tratados com salina ou dexametasona (2 mg/kg e.v.) meia hora antes ou com PrLP (12,5; 25, 50 e 100mg/kg e.v.) 1 hora após o Zy. Seis horas após a indução os animais foram eutanasiados e foi feita a coleta do lavado articular para contagem total e diferencial de leucócitos, atividade da mieloperoxidase (MPO) e concentração de mediadores inflamatórios (NO, TNF-α, IL-1β, IL-6 e IL-10). O edema foi avaliado pela medição do diâmetro transversal da articulação e pela avaliação da permeabilidade vascular com azul de evans. Adicionalmente foi avaliado o efeito de PrLP sobre processo inflamatório na membrana sinovial e sobre a expressão de NOSi, COX, NFkB e TNF- α no tecido. A hipernocicepção mecânica foi estudada utilizando uma versão eletrônica (VonFrey) e foi investigada tanto na hipernocicepção articular induzida por zymosan quanto na plantar induzida por carragenina e prostaglandina. O mecanismo do efeito antinociceptivo de PrLP foi investigado fazendo uma modulação com Naloxona (1 mg/kg; e.v.) ou L-arginina (1g/kg; i.p.) 30 minutos antes da administração de PrLP. Para as análises estatísticas foi utilizado o teste ANOVA/Newman Keuls ou Kruskal Wallis/Dunns; p <0,05 foi considerado significante. RESULTADOS: O tratamento com PrLP nas doses de 25, 50 e 100 mg/kg e.v. reduziu de forma significativa a contagem de leucócitos (899,8 x 106/ml ± 148; 1225 x 106/ml ± 89,59; 890 x 106/ml ± 165,7, respectivamente), a contagem de neutrófilos (860,3±97,57; 1108±127,6 e 908,7±159,9, respectivamente) e a concentração de MPO (12,1±1,3; 14,3±0,6; 8,9±3,5 UMPO/ml respectivamente). PrLP na dose de 25mg/kg reduziu as concentrações de mediadores inflamatórios tais como NO, IL-1β, TNF-α e IL-6 (71,84±9,1 mM; 4569±712,5; 1409±221,6 e 847,3±260,4 pg/ml respectivamente) quando comparados ao grupo controle. Também foi observado uma redução do edema pela administração de PrLP 25 mg/kg, esse tratamento reduziu de forma significativa o diâmetro articular na terceira (0,89 ± 0,001 mm) e na sexta hora (0,99±0,02 mm) e também reduziu o extravasamento de azul de evans para a articulação (40,46±3,6 µg/g). No ensaio histopatológico do tecido observou-se redução significativa do infiltrado inflamatório (com redução na contagem de neutrófilos), edema, comprometimento da membrana sinovial e hemorragia nos animais que receberam PrLP (25 mg/kg) e na imunohistoquímica houve redução significativa na expressão de COX-2, NOSi, TNF-α e NFkB. Na hipernocicepção mecânica articular o tratamento com PrLP (25 mg/kg) reduziu de forma significativa tanto na terceira (0,86 ± 1,3) como na sexta hora (1,08±2,3) e esse efeito foi revertido pela administração prévia de L-arginina (1g/kg, i.p.) e de naloxona (1mg/kg; e.v.). Adicionalmente, PrLP (25 mg/kg) reduziu de forma significativa a hipernocicepção plantar induzida por carragenina na terceira (1,74±0,4) e na quinta hora (1,5±0,4) e a hipernocicepção induzida por prostaglandina na terceira hora (1,4±0,4). CONCLUSÃO: PrLP apresenta atividade antinociceptiva e anti-inflamatória no modelo de artrite induzida por zymosan. O efeito parece estar relacionado com a diminuição da migração de neutrófilos e da liberação de mediadores inflamatórios, reduzindo o dano na membrana sinovial e a hipernocicepção.

The HdLP (0.5, 5.0 and 50.0 mg/Kg) was administered IV 30 minutes before absolute ethanol (0.2 mL/animal, oral dose) and its gastroprotective effect was observed only at the 5.0 mg/Kg dose, which significantly (p<0.05) reduced the gastric lesions induced by ethanol by 83%, besides also significantly reducing the histopathological lesion scores. The gastroprotective mechanism of the HdLP was investigated at the 5.0 mg/Kg dose. The HdLP gastroprotective effect was inhibited in animals pretreated with L-NAME (20 mg/Kg, s.c.), a nonspecific NOS inhibitor, with glibenclamide (5 mg/Kg, IP), an ATP-sensitive potassium channel (KATP) blocking drug, or with ODQ (10 mg/Kg, IP), a guanylate cyclase selective inhibitor, and the lesion areas in these groups increased significantly (p<0.05) by 63%, 290% and 249%, respectively, compared with the group pretreated only with the fraction. The HdLP also induced an increase of NO in the gastric mucosa lesioned by ethanol. On the other hand, pretreating the animals with indomethacin (10 mg/Kg, oral dose), a nonspecific COX inhibitor, or with capsazepine (5 mg/Kg, IP), a TRPV1 receptor antagonist drug, did not inhibit the HdLP gastric protection in the experimental model. In addition, the pretreatment with HdLP significantly (p<0.05) re-established the NP-SH (glutathione) gastric levels which had been depleted by the ethanol. Therefore, the results show, through different mechanisms, that the HdLP exert a gastroprotective function, with the involvement of NO, cGMP, activation of the KATP channels and antioxidant action / HdLP, uma fração protéica isolada do látex de Himatanthus drasticus, popularmente conhecida como Janaguba, foi avaliada no modelo de lesões gástricas induzidas por etanol em camundongos. O mecanismo gastroprotetor da HdLP foi investigado na dose de 5,0 mg/Kg. Em animais pré-tratados com L-NAME (20 mg/Kg, s.c.), um inibidor inespecífico da NOS, com glibenclamida (5 mg/Kg, i.p.), droga bloqueadora de canais de potássio dependentes de ATP, ou com ODQ (10 mg/Kg, i.p.), um inibidor seletivo da guanilato ciclase, o efeito gastroprotetor da HdLP foi inibido e as áreas lesionadas nestes grupos aumentaram significativamente (p<0,05) em 63%, 290% e 249%, respectivamente, em relação ao grupo pré-tratado somente com a fração. HdLP induziu também aumento de NO na mucosa gástrica com lesões por etanol. Por outro lado, o pré-tratamento dos animais com indometacina (10 mg/Kg, v.o), inibidor inespecífico da COX ou com capsazepina (5 mg/Kg, i.p.), droga antagonista dos receptores TRPV1, não inibiu a proteção gástrica da HdLP no modelo estudado. Em acréscimo, o pré-tratamento com HdLP restabeleceu de forma significativa (p<0,05) os níveis gástricos de NP-SH (glutationa) os quais foram depletados pelo etanol. Deste modo, o conjunto de resultados demonstra uma função gastroprotetora para HdLP, através de diferentes mecanismos com envolvimento do NO, GMPc, ativação dos canais de KATP e ação antioxidante

Apocynacea do Parque Estadual do Ibitipoca, Minas Gerais, Brasil / Apocynaceae of Ibitipoca State Park, Minas Gerais, Brazil

Este trabalho apresenta o levantamento das Apocynaceae do Parque Estadual do Ibitipoca. O Parque está situado na Cadeia da Mantiqueira, entre os paralelos 21°40’15" a 21°43’30" S e 43°52’35" a 43°54’15" W. Abriga diversas formações vegetais como cerrados de altitude, florestas montanas e principalmente campos rupestres. A família Apocynaceae, incluindo Asclepiadaceae, é cosmopolita, com ca. de 415 gêneros e 4550 espécies, das quais ca. 870 ocorrem no Brasil, em diferentes hábitats. No Parque ocorrem 10 gêneros e 28 espécies: Aspidosperma olivaceum, A. spruceanum, Blepharodon ampliflorum, Condylocarpon isthmicum, Ditassa acerosa, D. bicolor, D. conceptionis, D. cordata, D. laevis, D. linearis, D. mucronata, D. tomentosa, Forsteronia australis, F. velloziana, Jobinia lindbergii, Mandevilla atroviolacea, M. illustris, M. pohliana, M. sellowii, M. tenuifolia, Oxypetalum appendiculatum, O. insigne, O. lanatum, O. minarum, O. patulum, O. strictum, Peplonia organensis e Tassadia subulata. São apresentados chaves de identificação para os gêneros e espécies, descrições, ilustrações e comentários sobre distribuição e fenologia. / The Apocynaceae species inventory of the Ibitipoca State Park is presented. The Park is located in Mantiqueira Range, between 21°40’15" - 21°43’30" S and 43°52’35" - 43°54’15" W. Within the Park there are several types of vegetation, such as altitude savannas, montane forests and mostly campos rupestres. Relaxamento do corpo cavernoso de coelho induzido pela fraÃÃo alcaloidal F3-5 de Aspidosperma ulei Markgr / Relaxation on of rabbit corpus cavernosum induced by the alkaloidal fraction F3-S from - Aspdidosperma ulei Markgr

CoordenaÃÃo de AperfeiÃoamento de NÃvel Superior / Erectile dysfunction is a worldwide public health issue and, in spite of advances brougth by the utilization of type-5 phosphodiesterase inhibitors, there is still much interest in new treatment alternatives, especially when derived from natural products. Some investigators observed the pro-erectile effects of an alkaloidal-rich fraction from Aspidosperma ulei Markgr., named F3-5. In this study, it was evaluated the degree of relaxation induced by F3-5 on rabbit corpus cavernosum, as well as possible Pharmacological mechanisms involved, in vitro. Several experimental assays were performed, utilizing the methods of cascade tissue superfusion and isolated tissue bath. WÃth strips ot corpus cavernosum pre-contracted with phenilephrine (5ÂM), dose-response curves of relaxation for F3-5 papaverine and DMSO were produced. were There were also assays fo, evaluation of the effects of L-NAME, 7-NI, 000, propranolol, atropine, sildenafil and F3-5 on the relaxations mediated by sodium litroprusside, acetylcholine, isoproterenol, sildenafil and F3-S. With depolarized preparations of corpus cavernosum, in a calcium-free, potassium-rich (60 mM) medium, the effects of F3-S and DMSO on the contractions induced by the progressive increase in Ca2+ concentration (1 - 300 mM) were observed. F3-5 was capable of inducing complete relaxation of rabbit corpus cavernosum with magnitude similar to that of papaverine for all doses tested, except 3 mg, when Ãt presented a significantly higher relaxation. Superfusions of L-NAME, 7-NI and ODQ did not significantly inhibit the relaxatÃons provoked by F3-5, suggesting that it acts independently of the nitrergià pathway. Propranolol and atropine also did not significantly interfere with relaxations mediated by F3-5 indicating that B-adrenergic or rnuscarinic receptors also might not be involved. F3-5 did not significantly amplify the relaxations promoted by sodium nitroprusside or acetylcholine, as opposed to sildenafil, suggesting that it does not act through inhibition of type-5 ohosphodiesterase. Finally, in cavernous tissue pre-incubated with F3-5, the minimum dose of Ca2+ necessary for muscular contraction was thirty times superior to that utilizedon the tissue without previous treatment, or treated with OMSO. This inhibition of the contractions of corpus cavernosum mediated by Ca2+ suggests, therefore, that F3-S can act through blockade of voltage-dependent calcium channels. / A disfunÃÃo erÃtil à um problema mundial de saÃde pÃblica e, apesar dos avanÃos trazidos pela utilizaÃÃo dos inibidores da fosfodiesterase tipo-5, ainda hà muito interesse em novas alternativas de tratamento, especialmente quando derivadas de produtos naturais. Alguns pesquisadores observaram os efeitos prÃ-erÃteis de uma fraÃÃo rica em alcalÃides de Aspidosperma ulei Markgr., denominada F3-5. Neste estudo, foi avaliado o grau de relaxamento induzido por F3-5 no corpo cavernoso de coelho, bem como possÃveis mecanismos farmacolÃgicos envolvidos, in vitro. Foram realizados diversos ensaios experimentais, utilizando-se os mÃtodos de superfusÃo de tecido em cascata e de banho isolado de tecido. Com tiras de corpo cavernoso prÃ-contraÃdas com fenilefrina (5 &#956;M), foram produzidas curvas de dose-resposta de relaxamento para F3-5, papaverina e DMSO. TambÃm foram realizados ensaios para avaliaÃÃo dos efeitos de L-NAME, 7-NI, ODQ, propranolol, atropina, sildenafil e F3-5 sobre os relaxamentos mediados por nitroprussiato de sÃdio, acetilcolina, isoproterenol, sildenafil e F3-5. Com preparaÃÃes de corpo cavernoso despolarizadas, em um meio livre de cÃlcio e rico em potÃssio (60 mM), os efeitos de F3-5 e DMSO sobre as contraÃÃes induzidas pelo aumento progressivo na concentraÃÃo de Ca2+ (1 â 300 mM) foram observados. F3-5 foi capaz de induzir o relaxamento completo do corpo cavernoso de coelho, com magnitude similar à da papaverina para todas as doses testadas, exceto 3 mg, quando apresentou relaxamento significantemente maior. As superfusÃes de L-NAME, 7-NI e ODQ nÃo inibiram significantemente os relaxamentos provocados por F3-5, sugerindo que este age independentemente da via nitrÃrgica. Propranolol e atropina tambÃm nÃo interferiram significantemente com os relaxamentos mediados por F3-5, indicando que receptores &#946;-adrenÃrgicos ou muscarÃnicos tambÃm nÃo devem estar envolvidos. F3-5 nÃo amplificou significantemente os relaxamentos promovidos por nitroprussiato de sÃdio ou acetilcolina, ao contrÃrio de sildenafil, sugerindo que nÃo age por inibiÃÃo da fosfodiesterase tipo-5. Finalmente, em tecido cavernoso prÃ-incubado com F3-5, a dose mÃnima de Ca2+ necessÃria para contraÃÃo muscular foi trinta vezes superior Ãquela utilizada em tecido sem tratamento prÃvio ou tratado com DMSO. Esta inibiÃÃo das contraÃÃes de corpo cavernoso mediadas pelo Ca2+ sugere, portanto, que F3-5 pode atuar atravÃs do bloqueio de canais de cÃlcio voltagem-dependente.

O Parque está situado na Cadeia da Mantiqueira, entre os paralelos 21°40’15” a 21°43’30” S e 43°52’35” a 43°54’15” W. Abriga diversas formações vegetais como cerrados de altitude, florestas montanas e principalmente campos rupestres. A família Apocynaceae, incluindo Asclepiadaceae, é cosmopolita, com ca. de 415 gêneros e 4550 espécies, das quais ca. 870 ocorrem no Brasil, em diferentes hábitats. No Parque ocorrem 10 gêneros e 28 espécies: Aspidosperma olivaceum, A. spruceanum, Blepharodon ampliflorum, Condylocarpon isthmicum, Ditassa acerosa, D. bicolor, D. conceptionis, D. cordata, D. laevis, D. linearis, D. mucronata, D. tomentosa, Forsteronia australis, F. velloziana, Jobinia lindbergii, Mandevilla atroviolacea, M. illustris, M. pohliana, M. sellowii, M. tenuifolia, Oxypetalum appendiculatum, O. insigne, O. lanatum, O. minarum, O. patulum, O. strictum, Peplonia organensis e Tassadia subulata. São apresentados chaves de identificação para os gêneros e espécies, descrições, ilustrações e comentários sobre distribuição e fenologia. / The Apocynaceae species inventory of the Ibitipoca State Park is presented. The Park is located in Mantiqueira Range, between 21°40’15” - 21°43’30” S and 43°52’35” - 43°54’15” W. Within the Park there are several types of vegetation, such as altitude savannas, montane forests and mostly campos rupestres. Em um ambiente caracterizado por um regime de chuvas irregulares, como o semiárido brasileiro, ela pode atuar como um fator determinante do conjunto de espécies capazes de se estabelecer em diferentes estágios do processo de regeneração natural. Apesar de ser reconhecida como um componente importante do sistema de defesa das folhas contra a perda de água para o ambiente, ainda não foi esclarecido de que maneira a cera epicuticular interfere no metabolismo foliar. Nesse trabalho, investigamos como a cera epicuticular contribui para a redução da perda de água para o ambiente e o seu impacto nas trocas gasosas em diferentes estádios do processo de sucessão ecológica. Para tanto, utilizamos indivíduos de Aspidosperma pyrifolium (Mart.) de mesma idade, localizados em três áreas em diferentes estádios do processo de sucessão: inicial, intermediário e tardio. Em cada indivíduo foram avaliadas folhas intactas e folhas que sofreram remoção mecânica da cera epicuticular das faces abaxial e adaxial, utilizando uma solução de goma arábica (1,5 g. mL⁻¹). Avaliamos as trocas gasosas, o status hídrico e o teor de ceras epicuticulares, fenóis e pigmentos fotossintéticos. As medições foram realizadas no mês de Maio de dois anos consecutivos, com baixa (2013) e alta (2014) pluviosidade. Os dados foram submetidos à análise de variância (ANOVA) dois fatores, seguida do teste de Student Newman Keul’s a 5% de probabilidade. Os resultados mostraram que a cera contribui fortemente para a manutenção de altas taxas fotossintéticas nas folhas, que são capazes de manter seus estômatos abertos por mais tempo e com maior eficiência no uso da água mesmo no ano mais seco. Em 2013, o menor potencial hídrico e as maiores taxas de fotossíntese foram encontrados nos indivíduos do estádio inicial, mas em 2014 as maiores taxas fotossintéticas foram encontradas nos estádios intermediário e tardio. O teor de clorofila a, carotenoides e fenóis foi mais elevado no ano de 2014 e semelhante entre os diferentes estádios sucessionais. No ano de 2013, o teor de cera das folhas foi maior do estádio inicial ao tardio, mas no ano de 2014 não apresentou diferença entre os estádios sucessionais, sendo sempre igual ou inferior ao ano anterior. Nossos resultados indicam que as ceras epicuticulares ajudam a proteger as folhas contra a dessecação, pois promovem uma barreira física à perda de água, permitindo, assim, que elas mantenham seus estômatos abertos e consigam fixar mais CO₂. A cera epicuticular exerce um papel importante nas folhas de A. pyrifolium, contribuindo fortemente para a manutenção de altas taxas de trocas gasosas com maior eficiência no uso da água, independentemente do estádio sucessional. / The epicuticular wax has been named as an important physical adaptation to a great range of environmental factors as drought, high irradiance and herbivory. In a typically irregular rainfall environment, as the Brazilian semiarid, it can act as a determining factor of the set of species capable of stablishing at the different stages of the natural regeneration process. Despite being known as an important element of the leaves defense system against environmental water loss, it has not been clarified how the epicuticular wax intervenes on the leaf metabolism. On the present work we investigated how the epicuticular wax contributes to the reduction of environmental water loss and its impact on gas exchanges on different stages of ecological succession. In order to do so we used same aged Aspidosperma pyrifolium (Mart.) subjects located on three areas at the different stages of the succession process: early, mid and late. In each subject we evaluated intact leaves and leaves that went through a mechanical removal of their adaxial and abaxial epicuticular waxes, using a Arabic gum solution (1,5 g. mL⁻¹). We evaluated gas exchanges, water status and the epicuticular wax, phenols and photosynthetic pigments contents. The measurements were performed on May from two consecutive years with low (2013) and high (2014) rainfall. Data was submitted to a factorial nested variance analysis (ANOVA) followed by a de Student Newman Keul’s test at 5% probability. The results show that the wax strongly improves the maintenance of high photosynthetic rates on the leaves which are capable of keeping their stomata open longer and with higher water use efficiency even on the dryer year. In 2013 the lowest water potential and photosynthetic rates were found in the early stage individuals, but in 2014 the higher photosynthetic rates were found on the mid and late successional stages. The leaves chlorophyll a, carotenoids and phenols contents were higher on 2014 and similar between different successional stages. In 2013 the leaves wax content was higher from the early to the late stage, but in 2014 it was equal between successional stages, and lower than the previous year. Our results indicate that the epicuticular wax helps protect the leaves against desiccation by promoting a physical barrier against water loss, thereafter allowing them to keep their stomata open and fixate more CO₂. The epicuticular wax exerts an important role on A. pyrifolium’s leaves, strongly contributing to the conservation of high gas exchange rates and water use efficiency, regardless of the successional stage.

It is widely distributed in tropical and subtropical regions, including Brazil. This plant is commonly used for the treatment of syphilis, fever, and as a purgative. Some studies show that the latex of P. rubra has antioxidant and vasodilator activity. The aim of this study was to evaluate the gastroprotective effect of laticifers proteins of Plumeria rubra (PrLP) in ethanol-induced gastric damage, to investigate the possible involvement of TRPV1 receptors, NOcGMPKATP pathway and glutathione, and possible toxic effects. Animal handling and experimental protocols were registered on the Institutional Ethics Committee under number 057/2010. Swiss mice (n=8), fasting for 16 hours, were treated intravenously (i.v.) with PrLP doses of 0.05, 0.5, 5 and 50 mg/kg. After 30 min the animals received 0.2 ml of absolute ethanol per oral (p.o.). After 60 min of ethanol administration, the animals were sacrificed, their stomachs removed and analyzed to determine lesion index. To investigate the involvement of mediators in PrLP effect, animals received indomethacin (10 mg/kg, p.o.), L-NAME (20 mg/kg, i.p.), ODQ (10 mg/kg, i.p.), glibenclamide (5 mg/kg, i.p.) and capsazepine (5 mg/kg, i.p.) prior to treatment with PrLP (0.5 mg/kg i.v.). Misoprostol (50 µg/kg; p.o.), L-arginine (600 mg/kg; i.p.), diazoxide (3 mg/kg; i.p.) and capsaicin (0.3 mg/kg; p.o.) were used as standard-drug. To evaluate the effect of PrLP on the levels of NO3-/NO2, the dosage was performed in the homogenate of the stomach, but to investigate a possible antioxidant effect, it was carried out the measurement of the GSH levels in normal and injured stomachs. For sub-chronic toxicity animals were treated for 7 days with 50 mg/kg i.v., followed by evaluation of various parameters, such as: body weight, complete blood count, biochemical (urea, ALT, AST) and wet weight of vital organs (heart, spleen, liver and kidney). PrLP at doses of 0.5, 5 and 50 mg/kg was able to inhibit the gastric lesions by 81.9, 72.8 and 68%, respectively, retrieving the GSH levels in the mucosa by 105% compared with the ethanol group and PrLP did not alter the GSH levels in animals that were not ethanol-lesioned stomachs. Additionally, PrLP also increased in 26% NO3-/NO2- levels that were reduced by ethanol administration. Indometacin, L-NAME, ODQ, glibenclamide and capsazepine were able to reverse the PrLP protective effect, demonstrating the involvement of prostaglandins, NO, GMPc, potassium channels ATP-dependent and TRPV1 receptors in its mechanism of action. Furthermore, the treatment for 7 days with PrLP did not change any parameter evaluated showing safety in their use. These results indicate that PrLP have gastroprotective pharmacology activity on the gastric mucosa which seems to be mediated in part by modulation of prostaglandin, NO/cGMP/KATP pathway and TRPV1 receptors, which play a fundamental role in maintaining blood flow and gastric mucosa defense. PrLP acts avoiding depletion of GSH levels ethanol-induced. Since this is important for the maintenance of mucosal antioxidant defenses. Moreover, PrLP did not shown acute toxicity in animals. / A Plumeria rubra L. é uma planta laticífera pertencente à família Apocynaceae, popularmente conhecida como Jasmim. Esta amplamente distribuída em regiões tropicais e subtropicais, incluindo o Brasil. Essa planta é utilizada popularmente para o tratamento de sífilis, febre e como purgativo. Alguns estudos mostram que o látex de P. rubra possui atividade antioxidante e vasodilatadora. O objetivo desse trabalho foi avaliar o efeito gastroprotetor das proteínas laticíferas da Plumeria rubra (PrLP) em modelo de úlcera gástrica induzida por etanol, investigar o possível envolvimento dos receptores TRPV1, da via NOGMPcKATP e da glutationa, e possíveis efeitos tóxicos. A manipulação dos animais e os protocolos experimentais foram registrados no Comitê de Ética Institucional sob o número 057/2010. Camundongos Swiss (n = 8), em jejum de 16h, foram tratados por via intravenosa com PrLP nas doses de 0,05; 0,5; 5 e 50 mg/kg. Após 30 min da administração de PrLP os animais receberam 0,2 ml de etanol absoluto v.o. Decorridos 60 min dessa administração, os animais foram sacrificados, os estômagos removidos e analisados para determinação do índice de lesão. Para investigar o envolvimento de mediadores no efeito de PrLP, os animais receberam indometacina (10 mg/kg; v.o.), L-NAME (20 mg/kg; i.p.), ODQ (10 mg/kg; i.p.), glibenclamida (5 mg/kg; i.p.) e capsazepina (5 mg/kg; i.p) antes do tratamento com PrLP (0,5 mg/kg; i.v.). Misoprostol (50 µg/kg v.o), L-arginina (600 mg/kg; i.p.), diazóxido (3mg/kg; i.p.) e capsaicina (0,3 mg/kg; v.o.) foram utilizados como droga-padrão. PrLP nas doses de 0,5; 5 e 50 mg/kg foi capaz de inibir lesão gástrica em 81,9; 72,8 e 68%, respectivamente, recuperou os níveis de GSH na mucosa em 105% quando comparados com o grupo etanol e não alterou os níveis de GSH em animais que não tiveram seus estômagos lesionados pelo etanol. Adicionalmente, PrLP também aumentou em 26% os níveis de NO3-/NO2- que foram reduzidos pela administração de etanol. Indometacina, L-NAME, ODQ, Glibenclamida e capsazepina foram capazes de reverter o efeito de PrLP, demonstrando o envolvimento das Prostaglandinas, NO, GMPc, canais de KATP e dos receptores TRPV1 em seu mecanismo de ação. Além disso, o tratamento por 7 dias com PrLP não alterou nenhum parâmetro avaliado, mostrando segurança no seu uso. Estes resultados indicam que PrLP possui atividade farmacológica gastroprotetora sobre a mucosa do estômago ao qual parece ser mediada em parte pela modulação de prostaglandina, pela via NO/GMPc/KATP e pelos receptores TRPV1, ao qual possuem papel fundamental na manutenção do fluxo sanguíneo e na defesa da mucosa gástrica. PrLP atua evitando a depleção dos níveis de GSH induzidas pelo etanol. Sendo isto importante para a manutenção das defesas antioxidantes da mucosa. Além do mais, PrLP não apresenta toxicidade aguda nos animais.
