Insulated Metabolic Exchange for Sustainable Agriculture and Biotechnology / Insulerat metaboliskt utbyte för hållbart jordbruk och bioteknik

Mellan 1960 och idag har världspopulationen dubblerats medan matproduktionen tredubblats. Det har möjliggjorts av kemiskt framställda gödsel som introducerades på mitten av 1900-talet. Sedan 1965 har användandet av kvävegödsel ökat sexdubblet. Kvävegödsel ökar lönsamheten och avkastningen men har stora koldioxdutsläpp och orsaker övergödning. Därför finns det en efterfrågan på andra alternativ. Ett alternativ skulle kunna vara en syntetsik föränding av rhizosfären. För att undersöka om det är möjligt kommer ett modellsystem för insulerat metaboliskt utbyte att skapas. Modellsystemet kommer att ha en kvävefixarande bakterie som fixerar kväve från atmosfären och producerar en kvävekälla till en kolfixerande bakterie, som kommer modellera en växt i system. Den kolfixerande bakterien kommer att fixera koldioxid from atmosfären och producera en kolkälla för den kvävefixerande bakterien att använda. I den här studien har olika produktioner av olika kvävekällor i Escherichia coli och Anabaena sp. undersökts. Fyra olika plasmider har skapats för produktionen av alanin, guanin och allantoin. Alla odlingar med plasmiderna växte bättre än kontrollodlingarna, men de har inte blivit analyserade för produktion av metaboliterna på grund av tidsbrist. Tilläggningsvis, två av tre replikat av Anabaena sp. med en plasmid för allantoin produktion i media med urinsyra verkar ha varit stressade för okända anledningar / Between 1960 and today, the world population has doubled while food production has tripled. This has been made through chemical fertilisers being introduced in the middle of the 20th century. From 1965, the usage of nitrogen fertilisers has increased by sixfold. Nitrogen fertilisers improve crop profitability and yield but have large carbon dioxide emissions as well as cause eutrophication. Thus, there is a demand for an alternative. One alternative would be to engineer the rhizosphere. To investigate whether this is possible, a model system for insulated metabolic exchange will be created. The model system will have a nitrogen-fixating bacteria that will fixate atmospheric nitrogen and produce a nitrogen compound to a carbon-fixating bacteria, that will act as a plant in the model system. The carbon-fixating bacteria will fixate carbon dioxide from the atmosphere and produce a carbon source for the nitrogen-fixating bacteria to utilise. In this study, different productions of different nitrogen compounds in Escherichia coli and Anabaena sp. have been investigated. Four different plasmids have been constructed for the production of alanine, guanine and allantoin, respectively. The strains containing the plasmids all grew better than the control cultures, however, they have not been analysed for producing the metabolites due to time constraints.

Increase of urate formation by stimulation of sympathetic hepatic nerves, circulating noradrenaline and glucagon inthe perfused rat liver

In the isolated rat liver perfused in situ stimulation of the nerve bundles around the portal vein and the hepatic artery caused an increase of urate formation that was inhibited by the α1-blocker prazosine and the xanthine oxidase inhibitor allopurinol. Moreover, nerve stimulation increased glucose and lactate output and decreased perfusion flow. Infusion of noradrenaline had similar effects. Compared to nerve stimulation infusion of glucagon led to a less pronounced increase of urate formation and a twice as large increase in glucose output but a decrease in lactate release without affecting the flow rate. Insulin had no effect on any of the parameters studied.

Ureide accumulation in faba bean (Vicia faba, L.)

Faba bean (Vicia faba L.) is a cool season crop that uses symbiotic biological nitrogen fixation to obtain atmospheric nitrogen (N), a limiting macronutrient, for growth and maintenance of the plant. Most cool season legumes like faba bean transport N from the nodules as amides, which are metabolized in destination tissues. Ureide metabolism is a catabolic process that produces N rich compounds from purine rings. Many warm season legumes such as soybean and common bean produce ureides (allantoin and allantoate) in their root nodules and then use these molecules to transport fixed nitrogen from root to shoot. Non-ureide exporting plants such as faba bean also produce ureides in normal purine recycling whereby these compounds may play a role in response to abiotic stress. This research aims to examine possible differences in ureide metabolism across genotypes and to assess the role of ureides in response to water limitation. In field grown faba bean, total ureides were found in highest concentrations in leaf tissue, followed by reproductive parts, stems, and nodules, but were not found to differ significantly among genotypes. Ureide concentrations varied throughout the growing season, decreasing over time as the plants reached physiological maturity. A water limitation experiment of faba bean grown in a controlled environment showed that faba bean accumulated the ureides allantoin and allantoate after six to eight days of water limitation when all data were pooled. However, no consistent trend was observed comparing results by genotype, and inoculated versus non-inoculated plants. Overall, results indicate that faba bean likely does not use ureides to transport symbiotically fixed N and that ureide accumulation in field grown plants is most likely in response to abiotic stress or remobilization of purine N from senescing tissues.

Alguns decênios atrás, o advento da nanotecnologia abriu perspectivas inovadoras permitindo alcançar novos alvos. Na área farmacêutica, as nanopartículas abriram alternativas ineditas de acesso para tratar órgãos e tecidos. Dentre as diversas nanopartículas existentes, este trabalho assentou sobre as nanopartículas lípidicas sólidas porque elas permitem a utilização de substâncias naturais, que representam um interesse cada vez maior devido às suas potencialidades diversificadas e comprovadas. Assim, foram escolhidos o óleo de copaíba e a alantoína devido às suas propriedades farmacológicas. Da mesma maneira que existem vários tipos de nanopartículas, existem várias técnicas para produzi-las. Neste estudo, o método por homogeneização à alta pressão foi selecionado devido às diversas vantagens. Para validar essas opções tecnológicas e caracterizar as nanopartículas, análises morfológicas, físico-químicas e térmicas foram realizadas. O segundo propósito desta pesquisa foi a avaliação do potencial antifúngico das nanopartículas contra fungos leveduriformes e filamentosos multirresistentes, devido às diversas propriedades microbiológicas do óleo de copaíba. Consequentemente, diversos ensaios micológicos foram feitos a fim de determinar onde e como essas nanopartículas agiram sobre esses fungos. Os resultados mostraram que a produção de nanopartículas lípidicas sólidas homogêneas e estáveis físico-quimicamente foi possível. Além disso, essas nanopartículas, compostas por substâncias naturais, demonstraram atividade antifúngica contra fungos multiresistentes, fato que não ocorreu com as matérias-primas isoladas. Consequently, many mycological tests were performed to determine where and how these nanoparticles acted on these fungi.
