Preeclampsia and selected immunogenetic factors Our objective was to evaluate plasma levels of the eight most common antiphospholipid antibodies (antiphosphatidylserine, antiphosphatidylethanolamine, antiphosphatidylinositol, antiphosphatidylglycerol, antiphosphatidic acid, antiannexin V, anticardiolipin and anti 2-glycoprotein I antibodies) by ELISA method and selected inherited thrombophilia (F V- Leiden mutation, FII mutation G20210A, C677T and A1298C variants of the gene for methylene tetrahydrofolate reductase-MTHFR) by DNA analysis of peripheral blood lymphocytes using the real-time PCR in fifty-five women with preeclampsia in the period immediately before urgent termination of pregnancy. Fifty-five healthy women without preeclampsia was considered as a controll group. Entered data were examined using a non-parametric Wilcoxon's test, univariate analysis were perfomed using the Fisher's exact test and statistical dependence between variables was assessed using Spearman's rank correlation coefficient. We demonstrated that women with preeclampsia had significantly higher levels of anticardiolipin antibodies in the isotope IgG (p <0.01) and IgM (p <0.01), elevated levels of antiphosphatidylserine antibodies in the isotope IgG(p <0.01) and antiethanolamine antibodies in the isotope IgM (p <0.01) when...

Are gingivitis, periodontitis and peri- implantitis associated with autoantibodies- A litterature review

Introduction: Periodontal disease is one of the most common inflammatory diseases in the world. A possible autoimmune aspect behind the local tissue destruction in periodontal disease, as a result of the invasion of oral pathogens over time has been reported in previous studies, but the correlation is yet unclear. Purpose: The aim of this literature review was to shed light on the topic if autoantibodies and autoimmune reactions are associated with gingivitis, periodontitis or peri-implantitis and the progression of these inflammatory diseases. Material and methods: A search in the Pubmed database was done resulting in 138 hits. To follow a systematic approach for selecting the studies to include, we used predefined inclusion and exclusion criteria Results: 26 articles studying a broad variety of different autoantibodies was included for this literature review. A vast majority of the included studies were of case-control design and, because of the broad variety and different variables and data, we decided that a meta-analysis could not be performed. Conclusion: Many studies where results could be compared due to similar comparisons, regarding the incidence of periodontal disease and the prevalence of certain autoantibodies, showed opposite results which makes it hard to reach a conclusion. The main part of the included studies were of small size and therefore more comparable studies are needed to clarify the possible association between periodontal disease and an autoimmune reaction mediated by autoantibodies.

Immunosignature of Alzheimer's Disease

abstract: The goal of this thesis is to test whether Alzheimer's disease (AD) is associated with distinctive humoral immune changes that can be detected in plasma and tracked across time. This is relevant because AD is the principal cause of dementia, and yet, no specific diagnostic tests are universally employed in clinical practice to predict, diagnose or monitor disease progression. In particular, I describe herein a proteomic platform developed at the Center for Innovations in Medicine (CIM) consisting of a slide with 10.000 random-sequence peptides printed on its surface, which is used as the solid phase of an immunoassay where antibodies of interest are allowed to react and subsequently detected with a labeled secondary antibody. The pattern of antibody binding to the microarray is unique for each individual animal or person. This thesis will evaluate the versatility of the microarray platform and how it can be used to detect and characterize the binding patterns of antibodies relevant to the pathophysiology of AD as well as the plasma samples of animal models of AD and elderly humans with or without dementia. My specific aims were to evaluate the emergence and stability of immunosignature in mice with cerebral amyloidosis, and characterize the immunosignature of humans with AD. Plasma samples from APPswe/PSEN1-dE9 transgenic mice were evaluated longitudinally from 2 to 15 months of age to compare the evolving immunosignature with non-transgenic control mice. Immunological variation across different time-points was assessed, with particular emphasis on time of emergence of a characteristic pattern. In addition, plasma samples from AD patients and age-matched individuals without dementia were assayed on the peptide microarray and binding patterns were compared. Ocorrência de anticorpos par o vírus da artrite equina em cavalos criados nas mesorregiões macro metropolitana paulista e Campinas /

Resumo: O presente estudo investigou a ocorrência de anticorpos contra o vírus da arterite eqüina, utilizando a técnica de soroneutralização viral em 1.400 eqüinos criados nas Mesorregiões Macro Metropolitana Paulista e Campinas, pertencentes a 42 municípios do estado de São Paulo (SP), Brasil entre os meses de janeiro de 2007 a dezembro de 2008. Do total das amostras, 80 (5,71%) apresentaram anticorpos para o vírus (títulos entre 4 e 4096). Dentre os 42 municípios amostrados, 15 (35,7%) apresentaram pelo menos um animal sororeagente. Foram analisadas 238 propriedades das quais 41 apresentaram ao menos um animal sororeagente. A ocorrência de animais reagentes foi maior em cavalos destinados ao esporte, particularmente das raças de Salto e Quarto de Milha e foi semelhante entre machos e fêmeas. A ocorrência também foi maior em animais acima de 24 meses de idade. Os resultados obtidos sugerem a circulação do vírus nos criatórios amostrados e alertam para o impacto econômico- sanitário da doença para a eqüideocultura do estado de São Paulo / Abstract: This study investigate the occurence of arteritis virus antibodies using virus neutralization teste in 1,400 equines from Campinas and Macro Metropolitan Paulista Mesoregions which belong to the 42 cities located on State of Sao Paulo, Brazil between january of 2007 until december of 2008. All the samples, 80 (5.71%) showed antibodies to equine arteritis virus (titers ranging from 4 to 4096). Among the 42 cities, 15 (35.7%) presented at least one positive animal to equine arteritis virus. Were studied 238 farms, and from these 41 showed at least one soropositive animal. The prevalence was higher in sport horses like Jumping Horses, Quarter Horses and was similar between females and males. The seropositive ocurrence was higher in animals above 24 months of age. Komplement-vážící protilátky u pacientů po transplantaci orgánů a jejich klinický význam / Complement-binding antibodies in patients after organ transplantation and their clinical relevance

The diagnosis of antibody-mediated rejection after liver transplantation is complicated, due to the fact that the clinical and pathological signs of this life- threatening complication are often overlapping with non-immunological symptoms, like biliary obstruction, ischemia, thrombosis and others. Furthermore, the transplanted liver is to a great extent resistant to this type of rejection. Like in the transplanted kidney and heart, the main pathological factors of graft injury are antibodies directed to the mismatched HLA antigens of the organ donor, i.e. donor- specific antibodies. Besides analysis of HLA specificity of antibodies, research lately has been directed to define whether these antibodies are complement-binding or not. Literature data on this are however up till now limited. Therefore, the aim of this diploma thesis was to study the clinical relevance of complement-binding antibodies against HLA antigens in patients after liver transplantation. Our preliminary results suggest that there might be a correlation between the presence of complement-binding antibodies and the development of antibody-mediated rejection. This finding may play a role for improvement of the prognosis of patients after liver transplantation. Key words HLA, antibodies, C4d, complement, liver transplantation
