Many studies show the result these chemicals in the reproductive system of animals and even same at man, can cause infertility problem, which can originate, among other factors, the low sperm quality and quantity. This is study aimed at evaluating human sperm quality parameters and the chemical evaluation of the core of the sperm trough Raman Scaterring Spectroscopy, after in vitro exposure to different concentrations of 2,4-dichlorophenoxyacetic acid (2,4-D) and atrazine. The choice of the use of the these herbicides was because they are widely use throughout the world, especially in Brazil, highlighted the west Paulista region, used at cane sugar plantation, significant culture of the region. In this context, sperm samples were selected according to the normal testing parameters that evaluate sperm quality, such as liquefaction, viscosity, appearance, volume, pH, sperm, vitality, morphology and motility. Semen samples of 10 healthy donor were exposed to study herbicides varying the concentration and time. After the different exposures (0.05, 0.5, 1.0, 5.0 and 125 &#956;g/mL, 30 and 60 min), the analysis of motility and vitality were again evaluated. The Raman Scaterring spectra were obtained from samples control sperm and after exposure to the herbicide 2,4-D e atrazine at concentration of 125 &#956;g/mL and 60 min incubation. The progressive motility and total motility decreased from the concentration 0,5 &#956;g/mL at times 30 and 60 minutes while the percentage of sperm properties increased. The progressive motility and total motility decreased from the concentration 0.5 mg / mL at times 30 and 60 minutes while the percentage of sperm immotile increased. The vitality (viable sperm) decreased significantly (p <0.05) from the exposure concentration 0.5 &#956;g/mL to 2,4-D and from 1.0 &#956;g/mL atrazine, at times 30 and 60 minutes, showing that there is a functional change of trend of human sperm after exposure to these herbicides, and this change is intensified with the exposure time factor and concentration of herbicides. Through the Raman Scaterring technique allowed us to characterize normal human sperm and exposed to herbicides and verify the change in the molecular structure in the exposed sperm, evidenced by changes in relative intensities of the bands 785, 1095, 1255, 1336, 1374, 1486 and 1575 cm-1 treated with 2,4-D and a decrease at 1095 after exposure to atrazine. These results indicate the modification of nitrogenous bases and the phosphate group that make up DNA present in the nucleus of the sperm. / A utilização desenfreada de praguicidas, além de contribuir com o agravamento da poluição ambiental, também está influenciando de forma negativa a saúde dos seres vivos, inclusive a saúde das pessoas. Muitos estudos mostram a interferência desses produtos químicos no sistema reprodutor de animais e até mesmo do homem, podendo acarretar problemas de infertilidade, que podem ser originados, dentre vários fatores, pela baixa quantidade e qualidade espermática. Este estudo visa á avaliação de parâmetros de qualidade espermática humana e a avaliação química do núcleo do espermatozoide, por meio de Espectroscopia de Espalhamento Raman, após a exposição in vitro a diferentes concentrações de ácido 2,4-diclorofenoxiacético (2,4-D) e atrazina. Estes herbicidas são amplamente utilizados em todo o mundo, principalmente no Brasil, em destaque a região do Oeste Paulista, por serem empregados na cana-de-açúcar, cultura significativa da região. Neste contexto, foram selecionadas amostras de espermatozoides de acordo com os parâmetros normais de testes que avaliam a qualidade espermática, como: liquefação, viscosidade, aparência, volume, pH, concentração de espermatozoides, vitalidade, morfologia e motilidade. Amostras de sêmen de 10 doadores saudáveis foram expostas aos herbicidas de estudo, variando o tempo e a concentração. Após as diferentes exposições (0,05; 0,5; 1,0; 5,0 e 125 &#956;g/mL, 30 e 60 min), as análises de motilidade e vitalidade foram novamente avaliadas. Os espectros de Espalhamento Raman foram obtidos das amostras de espermatozoide controle e após a exposição aos herbicidas 2,4-D e atrazina na concentração de 125 µg/mL em 60 min de incubação. A motilidade progressiva e motilidade total diminuíram a partir da concentração 0,5 &#956;g/mL nos tempos 30 e 60 min enquanto que a porcentagem de espermatozoides imóveis aumentou. A vitalidade (espermatozoides viáveis) diminuiu significativamente (p<0,05) a partir da exposição da concentração 0,5 &#956;g/mL para o 2,4-D e a partir de 1,0 &#956;g/mL para a atrazina, nos tempos 30 e 60min, mostrando-se que há uma tendência de alteração funcional do espermatozoide humano após a exposição a estes herbicidas, e esta alteração é intensificada com o fator de tempo de exposição e concentração dos herbicidas testados. Por meio da técnica de Espalhamento Raman foi possível caracterizar o espermatozoide humano normal e os expostos aos herbicidas e verificar a alteração na estrutura molecular nos espermatozoides expostos, evidenciadas pelas alterações nas intensidades relativas das bandas 785, 1095, 1255, 1336, 1374, 1486 e 1575 cm-1 do tratado com 2,4-D, e uma diminuição na banda de 1095 após a exposição à atrazina. Estes resultados indicam a alteração das bases nitrogenadas e do grupo fosfato que compõem o DNA presente no núcleo do espermatozoide.

Presenza di erbicidi triazinici e loro metaboliti nelle acque di falda di aree maidicole italiane / PRESENCE OF TRIAZINE HERBICIDES AND THEIR METABOLITES IN GROUNDWATER OF ITALIAN MAIZE-GROWING AREAS

La presenza di pesticidi nelle acque di falda è emersa come un importante problema ambientale negli ultimi decenni. In base a “Rapporto nazionale pesticidi nelle acque - Dati 2015-2016” (ISPRA, 2018), in Italia, gli erbicidi triazinici e i loro metaboliti sono tra le sostanze identificate con più frequenza nelle acque, sebbene l’atrazina sia stata bandita nel 1992 e l’utilizzo di terbutilazina sia sottoposto a severe restrizioni. Lo scopo di questo progetto è valutare e quantificare la presenza di atrazina, terbutilazina e dei loro metaboliti nelle acque di falda in aree maidicole italiane. I risultati dimostrano come le molecole oggetto di studio siano state frequentemente individuate nei campioni di acqua di falda, anche se a concentrazioni inferiori al limite stabilito dalla Commissione Europea. Considerando che l’utilizzo di questi erbicidi è proibito o fortemente regolamentato, è interessante valutare il loro comportamento in una scala temporale pluriennale al fine di mettere in evidenza la contaminazione delle acque di falda dovute all’uso diffuso e alla persistenza ambientale di tali molecole. / The presence of pesticides in groundwater has emerged as an important environmental issue in the last decades. According to “Rapporto nazionale pesticidi nelle acque - Dati 2015-2016” (ISPRA, 2018), in Italy triazine herbicides and their metabolites are among the substances most frequently detected in water. While terbuthylazine is still used under restriction, atrazine was banned in 1992. The aim of research project is to investigate the presence of atrazine, terbuthylazine and their metabolites in groundwater in Italian maize-growing areas in order to supply data and information on the groundwater quality. The results show that atrazine, terbuthylazine and their metabolites were often detected in groundwater, even if their concentrations were below the limit established by European Commission. Considering these molecules are banned or heavily regulated, it is interesting to evaluate their behavior on a multiannual time scale in order to highlight the groundwater contamination due to the widespread use and to the environmental persistence of these substances.

Relations entre les coefficients d'adsorption de l'atrazine et les caractéristiques physico-chimiques et colorimétriques de la phase organique de différents sols

Ce projet d'étude, qui a pour but de développer un indice simple à déterminer fournissant des informations sur la rétention de l'atrazine dans les sols, a mis en relation les différents coefficients d'adsorption existants (Kd, Kf, Koc) et deux indices colorimétriques. La méthode utilisée pour déterminer les indices colorimétriques est celle au pyrophosphate de sodium de Kaila. Cette dernière est facile, rapide et accessible à tous les laboratoires d'analyses. Les principales conclusions de l'étude sont les suivantes : 1. L'indice colorimétrique au pyrophosphate de sodium de Kaila est applicable aux sols minéraux. Les lectures obtenues en absorbance sont valables et reproductibles pour les 11 sols minéraux de l'étude. 2. La méthode colorimétrique au pyrophosphate de sodium de Kaila est une méthode simple, rapide et relativement peu coûteuse à effectuer. Les modifications apportées à la méthode n'entraine pas de changements majeurs au niveau des coûts et de la faisabilité. 3. L'indice colorimétrique de Kaila standard ne semble pas être relié avec les paramètres de la rétention de l'atrazine tels que 1/Kd et Kdoc. 4. L'indice colorimétrique de Kaila en version modifiée semble être relié avec les paramètres de la rétention de l'atrazine tels que 1/Kd et Kdoc. 5. L'indice colorimétrique de Kaila en version modifiée semble être inversement proportionnel à la rétention de l'atrazine. 6. Les indices colorimétriques de Kaila (standard ou modifié) semblent être des outils qui peuvent nous apporter des informations sur la rétention de l'atrazine dans les sols minéraux ainsi que dans les sols organiques.

Characterization of Acetolactate Synthase-Inhibiting Herbicide-Resistant Smooth Pigweed and Corn Weed Management Programs Utilizing Mesotrione in Combinations with Other Herbicides

Repeated use of acetolactate synthase (ALS)-inhibiting herbicides in recent years has resulted in the selection of 89 weed species resistant to these herbicides. One management strategy that can eliminate or slow the development of resistance is applying mixtures of herbicides with different modes of action. This research involved the characterization of ALS-inhibiting herbicide-resistant smooth pigweed (<i>Amaranthus hybridus</i> L.), as well as investigations on weed management programs in corn (<i>Zea mays</i> L.) utilizing mesotrione, a triketone, in mixtures with other herbicides. ALS-inhibiting herbicide-resistant smooth pigweed biotypes were collected from fields in Virginia, Delaware, Maryland, and Pennsylvania to evaluate response to ALS-inhibiting herbicides and to determine the molecular mechanisms of resistance. Sequencing of the ALS genes from these biotypes revealed two amino acid substitutions known to confer resistance, Ala<sub>122</sub> to Thr and Ser<sub>653</sub> to Asn, and one that has not been previously reported in plants, Asp<sub>376</sub> to Glu. The smooth pigweed biotype with an Asp<sub>376</sub> substitution displayed resistance to four classes of ALS-inhibiting herbicides that included imidazolinone (IMI), sulfonylurea (SU), pyrimidinylthiobenzoate (PTB), and triazolopyrimidine sulfonanilide (TP) chemistries. Transformation of this smooth pigweed ALS gene into <i>Arabidopsis thaliana</i> confirmed that the Asp<sub>376</sub> substitution is responsible for the resistance. Other biotypes that had a substitution at Ala<sub>122</sub> exhibited resistance to an IMI herbicide, little to no resistance to SU herbicides, and increased sensitivity to a PTB and a TP herbicide, whereas, biotypes that had a substitution at Ser<sub>653</sub> exhibited high-level resistance to an IMI herbicide and lower resistance to PTB and SU herbicides. Experiments were also conducted to investigate the effectiveness of mesotrione in preemergence (PRE) and postemergence (POST) corn weed management programs in Virginia. Mesotrione applied PRE in mixtures with <i>S</i>-metolachlor and atrazine controlled common lambsquarters (<i>Chenopodium album</i> L.), smooth pigweed, common ragweed (<i>Ambrosia artemisiifolia</i> L.), and morningglory (<i>Ipomoea</i> spp.) species when a timely rainfall followed application. POST applications of mesotrione controlled common lambsquarters and smooth pigweed, but common ragweed and morningglory species were not always controlled. Common ragweed and morningglory species were controlled by mesotrione in a mixture with atrazine POST. Large crabgrass [<i>Digitaria sanguinalis</i> (L.) Scop.] and giant foxtail (<i>Setaria faberi</i> Herrm.) control was generally better when the ALS-inhibiting herbicides nicosulfuron plus rimsulfuron or rimsulfuron plus thifensulfuron plus atrazine were applied in a mixture with mesotrione. Mixtures of mesotrione with other POST herbicides in a total POST program produced corn yields comparable to standard PRE followed by POST weed management programs. / Ph. D.

A atrazina (ATZ) é um herbicida utilizado em larga escala no Brasil de forma intensiva em culturas de cana-de-açúcar, milho e sorgo. A ATZ possui caráter persistente e recalcitrante, de forma que os tratamentos convencionais de efluentes não são capazes de removê-lo. Nesse contexto, os processos oxidativos avançados, como a fotocatálise heterogênea, têm se mostrado eficientes para remoção deste poluente. Este trabalho tem por objetivo avaliar o desempenho dos fotocatalisadores TiO2 P25 e C-TiO2 Kronos vlp 7000 modificado com carbono para a degradação do pesticida atrazina em solução aquosa, utilizando um reator fotoquímico com coletor parabólico composto irradiado por um simulador solar. Os catalisadores TiO2 P25 e C-TiO2 Kronos foram caracterizados por meio de difração de raios-X, espectroscopia de infravermelho, área superficial específica BET e análise de reflectância difusa. As concentrações de TiO2 P25 e C-TiO2 Kronos empregadas foram 500 e 100 mg L&-1, respectivamente. Os resultados obtidos mostraram significativa remoção de ATZ ao final de 120 minutos (100; 98; 98; 93; 86% para concentrações iniciais de ATZ de 1; 5; 10; 20 e 30 mg L-1, respectivamente) com emprego do catalisador TiO2 P25. No entanto, as análises de carbono orgânico total (COT) indicaram que não houve mineralização significativa da ATZ, com valores inferiores a 20% com o uso do TiO2 P25. Para o catalisador C-TiO2 Kronos não se obteve remoção importante da ATZ ao final de 120 minutos de experimento, quando comparado com o TiO2 P25. Neste caso, o catalisador C-TiO2 Kronos permitiu obter remoções do pesticida de 37; 35; 38; 39 e 45%, aproximadamente, para as concentrações iniciais de ATZ de 1; 5; 10; 20 e 30 mg L-1, respectivamente. Apesar disso, o catalisador C-TiO2 Kronos apresentou melhor desempenho em mineralizar a ATZ a CO2 e H2O em relação ao proporcionado pelo TiO2 P25, com aproximadamente 38% de mineralização. O modelo de Langmuir-Hinshelwood (LH) constitui uma aproximação adequada para a cinética de degradação da atrazina para ambos os catalisadores estudados, com valores de Kr e Kads iguais a 1,54 mg L-1 min-1 e 15,47 L mg-1, respectivamente, para o catalisador TiO2 P25 e de 1,34 mg L-1 min-1 e 132,95 L mg-1, respectivamente para o catalisador C-TiO2 Kronos vlp 7000. / Atrazine (ATZ) is a widely used herbicide in Brazil, where it is intensively applied in sugarcane, corn and sorghum cultivations. However, conventional wastewater treatments are not able to remove ATZ due to its persistent and recalcitrant properties. Advanced oxidation processes, such as heterogeneous photocatalysis, have been proved effective for the removal of this pollutant. This study aims to evaluate the performance of TiO2 P25 and carbon-modified TiO2 Kronos vlp 7000 photocatalysts in the degradation of ATZ in aqueous systems using a tubular photochemical reactor equipped with a compound parabolic collector (CPC) irradiated by simulated solar light. Catalysts TiO2 P25 and C-TiO2 Kronos were characterized by X-ray diffraction, infrared spectroscopy, BET analysis and diffuse reflectance analysis. The concentrations of TiO2 P25 and C-TiO2 Kronos were 500 and 100 mg L-1, respectively. For TiO2 P25, the results showed significant ATZ removals after 120 minutes of irradiation (100; 98; 98; 93; 86% for initial ATZ concentrations of 1; 5; 10; 20, and 30 mg L-1, respectively). In contrast, the total organic carbon (TOC) analyses indicated that no significant ATZ mineralization occurred, with values lower than 20% for TiO2 P25. In comparison with TiO2 P25, C-TiO2 was not able to completely remove ATZ at the end of 120 minutes of irradition. In this case, pesticide removals of about 37; 35; 38; 39 and 45% were obtained for initial ATZ concentrations of 1; 5; 10; 20, and 30 mg L-1, respectively. Nevertheless, C-TiO2 Kronos showed better performance for ATZ mineralization to CO2 and H2O in comparison with TiO2 P25, with approximately 38% total carbon organic removal. The Langmuir-Hinshelwood (LH) model was shown to be an appropriate approximation for the degradation kinetics of atrazine for both catalysts, with values of Kr and Kads equal to 1.54 mg L-1 min-1 and 15.47 mg L-1, respectively, for the P25 TiO2 and 1.34 mg L-1 min-1 and 132.95 mg L-1, respectively, for C-TiO2 Kronos vlp 7000.

Development of Photocatalysts Supported on Graphitic Carbon Nitride for the Degradation of Organic Water Pollutants

Graphitic carbon nitride (g-C3N4) heterojunction composites with the semiconducting metal oxides, CeO2, ZnO and TiO2 are prepared in situ by co-calcination of the precursor materials or by a solvothermal method. The structural, morphological and the optical properties of the prepared materials are studied using various microscopy and spectroscopy techniques. The synthesized composite materials, CeO2/g-C3N4, ZnO/g-C3N4 and TiO2/g-C3N4 are more efficient in the photocatalytic degradation of the water pollutants indigo carmine (IC) and atrazine than the pure metal oxide, g-C3N4, or their physical mixtures. The CeO2/g-C3N4 and ZnO/g-C3N4 composites also exhibit improved degradation efficiencies of atrazine as compared to the individual metal oxide or g-C3N4 materials. The improved photocatalytic activity of the composites are attributed to the effective electron-hole charge separation within composite heterojunction, resulting from the well matched energy levels of the metal oxide and g-C3N4. This strategy could be helpful for the synthesis of other metal oxide and g-C3N4 composites for photocatalytic applications.

Biodégradation des herbicides en sols tempérés - Contrôle des communautés bactériennes dégradantes par la bioturbation du sol

L'intensification de l'agriculture s'est accompagnée d'une utilisation importante de pesticides qui a généré une pollution généralisée des sols et des eaux, problème environnemental majeur et actuel. Sous la pression de sélection liée à l'usage régulier de pesticides (molécules xénobiotiques) des bactéries du sol se sont adaptées à ces molécules et ont acquis la capacité de les utiliser comme source nutritive et donc de les dégrader. La biodégradation constitue un service écologique majeur fournit par le sol, puisqu'elle est à la base des capacités épuratrices du sol et au-delà, de la résilience des écosystèmes. Le sol étant également un grand réservoir de biodiversité, ces bactéries dégradantes sont sous contrôle de différentes interactions biotiques et notamment celles impliquant les lombriciens, qualifiés d'organismes ingénieurs des sols de par leur action de bioturbation. Grâce à un développement méthodologique important et novateur (RT-qPCR, SIP-ARN), nous avons étudié l'impact de la bioturbation du sol par la macrofaune lombricienne sur les communautés bactériennes du sol intervenant dans la biodégradation de molécules xénobiotiques. L'atrazine a été utilisée comme molécule modèle à double titre : d'un point de vu fondamental, son utilisation pendant plus de 50 ans en France a permis aux bactéries du sol de s'adapter et au titre de l'actualité, bien qu'elle soit interdite en France depuis 2003, il s'agit toujours du principal polluant retrouvé dans les eaux souterraines et de surface. Nous avons analysé par une double approche quantitative et qualitative l'impact de la bioturbation du sol par les lombriciens sur l'abondance, l'activité et la diversité des bactéries indigènes du sol et sur celles dégradant l'atrazine. Nous avons mis en évidence que : (i) la digestion du sol par les lombriciens stimule l'activité d'une partie des bactéries du sol mais qu'une autre fraction ne résiste pas au passage dans le tube digestif des vers, (ii) la bioturbation du sol par les lombriciens génère des ‘hot spot' pour l'activité de dégradation de l'atrazine. Ainsi dans les parois de galeries les bactéries dégradantes sont sélectionnées, la dissipation de l'atrazine est rapide et les premiers acteurs du processus de dégradation ont été identifiés, (iii) la dégradation accélérée de l'atrazine dépend d'espèces bactériennes clés interagissant au sein de consortia dégradants, ainsi la diversité des bactéries dégradantes n'est pas corrélée à la fonction de dégradation. L'ensemble des résultats obtenus nous montre également que l'impact de la bioturbation par la macrofaune lombricienne sur l'activité de dégradation dépend des propriétés physico-chimiques et biologiques initiales du sol. L'ensemble de ces connaissances présente un intérêt dans un contexte de bioremédiation in situ des sols pollués puisque les lombriciens constituent une grande part de la macrofaune dans nos sols tempérés et qu'ils modifient significativement les bactéries dégradant l'atrazine au sein des microsites de sols qu'ils génèrent.

Regulation of elements of the thyroid hormone and corticosteroid systems by stress, hormone treatment, and atrazine during ontogeny of red drum (Sciaenops ocellatus)

Survival of teleost larvae requires growth and development which depend upon endocrine processes. In this dissertation I have examined the ontogeny of elements of the thyroid hormone (TH) and corticosteroid (CS) systems in red drum (Sciaenops ocellatus). Basal cortisol production was first detected 3 days post-hatch (DPH) and a cortisol stress response was present from 6 DPH forward. Changes in steroidogenic enzyme mRNA (CYP11B and CYP21) levels did not correlate with these events. The time necessary to reach peak cortisol levels as well as return to basal levels declined as larvae developed. A second set of studies examined ontogenetic patterns in levels of mRNAs encoding thyroid (soTR[alpha] and soTR[beta]) and corticosteroid (soGR) hormone receptors and assessed the regulation of these mRNAs by exogenous triiodothyronine (T3) and cortisol. soTR[alpha], soTR[beta] and soGR were expressed in all stages of red drum examined. soTR[alpha] levels increased during the time when surging TH levels have been reported. soTR[beta] levels did not differ significantly during development. soGR levels were strongly correlated with those of soTR[alpha]. T3 up-regulated soTR[alpha] and soTR[beta] levels in 7 DPH, but not older larvae. Cross-regulation of receptor mRNAs by exogenous treatment with T3 or cortisol was not observed. Finally, I assessed the influence of a common herbicide, atrazine, on receptor mRNA, TH levels and growth of red drum. In two experiments, red drum exposed to environmentally relevant levels of atrazine did not alter hormone receptor mRNA levels, or TH content. However, atrazine did depress growth in some instances. In conclusion, the expression patterns of hormone receptor mRNA in embryos suggests receptor proteins could be activated by maternal hormones prior to the onset of endogenous hormone production. A correlation between soTR[alpha] and soGR mRNA levels suggests coordinated function of TH and CS systems, although regulatory interactions between these systems were not evident under the conditions in this study. Sendo os pesticidas constituídos por moléculas biologicamente ativas e de difícil remoção durante o processo convencional de tratamento de água, o uso de filtros lentos de areia seguidos por filtros de carvão ativado biologicamente pode representar um sistema eficiente no tratamento de água. Considerando estes aspectos, o presente estudo teve como objetivo avaliar a eficiência da filtração lenta e da filtração utilizando carvão ativado biológico (CAB) na remoção do herbicida atrazina em escala de bancada e a composição fenotípica e genotípica dos microrganismos associados na degradação do herbicida. A remoção de atrazina atingiu uma remoção próxima de 100% nos efluentes dos filtros de areia e carvão. Foi verificada a presença de microrganismos (Bacillus e Salirhabdus) em ambos conjunto de filtros, podendo estes estarem ligados à degradação do herbicida da água. Os resultados deste estudo demonstraram eficiência na remoção de atrazina em níveis resguardados pela legislação sanitária, possibilitando o uso deste sistema na remoção do herbicida avaliado, assegurando a distribuição e consumo humano desta água. Além de avaliar a viabilidade do uso de filtros biológicos de carvão antecedidos por filtros lentos de areia na remoção de pesticidas, o presente trabalho poderá contribuir para realização de novas pesquisas... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The increased contamination of watercourses by pesticides along with better knowledge of the harmful properties of these pollutants have increased interest in the removal of these compounds during treatment of drinking water. Since pesticides consisting of biologically active molecules and hard to remove during conventional water treatment, use of slow sand filters followed by biologically activated carbon filters may represent an efficient system for water treatment. Considering these aspects, this study aimed to evaluate the efficiency of slow sand filtration and biological filtration using activated carbon (BAC) in the removal of the herbicide atrazine in bench scale and phenotypic and genotypic composition of the microorganisms involved in degradation of the herbicide. The removal of atrazine reached a level close to 100% in effluent from sand and carbon filters compounds. We observed the presence of microorganisms (Bacillus and Salirhabdus) in both set of filters and they can be linked to degradation of the herbicide from water. The results showed removal efficiency of atrazine at levels controlled by the health legislation, enabling the use of this system in the removal of the herbicide evaluated, ensuring the distribution and consumption of water. Besides evaluating the feasibility of using biological filters charcoal preceded by slow sand filters for removal of pesticides, this study may contribute to new research on the technologies used in the treatment of drinking water contaminated by recalcitrant compounds. / Mestre

Hodnocení subchronického působení atrazinu na raka (Cherax destructor)

The evaluation of the sub-chronic exposure to atrazine on crayfish The aim of this study is to evaluate the sub-chronic effect of atrazin on a behaviour, oxidative stress, antioxidant enzyme aktivities and biochemical profile of haemolymph in. These complex data should help to appraise the impact of this substance in the environment. The total test duration was 28 days and was divided into two periods. The first 14 days the crayfish were exposed to two concentrations of atrazine: 6.86 micrograms per liter (ATRenv = environmental concentration in the water in the Czech Republic) and 1.21 milligrams per liter (ATR10% = is coincident to 10% LC50). After the atrazine treatments the depuration 2 weeks phases in water without any chemicals followed. The results indicate that sub-chronic effect of atrazine influenced neither the behaviour of the crayfish nor the level of oxidative stress (measured by TBARS), whereas the changes of superoxiddismutase (SOD) were observed in all tissues (muscles, gills and hepatopancreas). The changes of enzyme activity were observed in catalase (CAT; hepatopancreas and the muscle tissue), glutathione S-transferase (hepatopancreas and the gills tissue), glutathione reductase (GR; the hepatopancreas tissue) and reduced glutathione (the muscle tissue). The influence of ATRenv on the biochemical profile of haemolymph at the following parameters was estimated only for lactate and alkaline, however phosphatase changes made by ATR10% were significant for glucose, ammonia, lactate and alkaline phosphatase measurements. The sub-chronical effect changed the activity of all antioxidant enzymes in hepatopancreas, muscles and the gills tissue of the observed crayfish. The presented results in this study are giving compact information of impact of atrazine on the crayfish and the whole water environment. The suggestion of using the crayfish for tests of toxicity looks like an ideal supplement for triazine herbicide estimations.
