Utilização do extrato de levedo de cerveja como suplemento de meio à base de glicerol residual para produção de bioinseticida por Bacillus thuringiensis / Utilization of brewer\'s yeast extract as a supplement to medium based on residual glycerol for bioinsecticide production by Bacillus thuringiensis

A utilização de bioinseticida à base de Bacillus thuringiensis tem se mostrado eficaz no combate às larvas de insetos veiculadores de doenças como a dengue, elefantíase e malária. O bioinseticida apresenta alta especificidade, ausência de resistência nos insetos alvos e baixo impacto ambiental comparado aos inseticidas químicos. Porém, a difusão do uso de bioinseticida apresenta alguns desafios como o custo da produção e da formulação. O levedo excedente da indústria cervejeira pode tornar a produção do bioinseticida economicamente viável devido ao seu baixo custo, assim como o glicerol, resíduo da produção de biodiesel. A escolha adequada dos componentes do meio de cultivo é essencial para o sucesso de um produto e deve ser realizada buscando-se obter uma alta produtividade com o menor custo. Além disso, no caso do bioinseticida, os componentes do meio exercem uma grande influência no crescimento e na síntese de toxinas pela bactéria. No presente trabalho, propôs-se formular um meio de cultivo para Bacillus thuringiensis var. israelensis utilizando levedo de cerveja e glicerol proveniente da fabricação de biodiesel, visando obter um meio fermentado com elevada toxicidade e de menor custo, comparado aos obtidos atualmente. Inicialmente foi definido um procedimento de preparo do extrato de levedo de cerveja. Constatou-se que o melhor método consiste da esterilização da suspensão de levedo de cerveja (121 ºC por 20 minutos) seguida de filtração qualitativa. Foi definida também a faixa de concentração de extrato de levedo de cerveja que seria utilizada no estudo de composição do meio (10 g/L a 30 g/L). Os ensaios foram realizados em frascos Erlenmeyer de 1000 mL em incubadora de movimento recíproco. Os componentes do meio de cultivo (glicerol residual, extrato de levedo de cerveja, KNO3, CaCl2.2H2O, MgSO4.7H2O, MnSO4, K2HPO4 e KH2PO4) foram avaliados segundo um planejamento experimental e, nas condições estudadas, não exerceram influência significativa sobre a produção de toxinas. Definiu-se então a composição do meio de fermentação em termos de fonte de carbono e nitrogênio. O glicerol e o extrato de levedo de cerveja não exerceram influência significativa sobre o consumo de substrato pela bactéria. Quanto à concentração celular máxima, o aumento da concentração de glicerol apresentou efeito negativo, enquanto o aumento da concentração de extrato de levedo de cerveja apresentou efeito positivo. Embora as concentrações das fontes de carbono e nitrogênio não tenham sido significativas para a atividade larvicida, a interação entre os fatores foi significativa em seu nível negativo. O melhor resultado quanto à atividade larvicida do meio foi apresentado pelo meio composto por 10 g/L de glicerol residual, 30 g/L de extrato de levedo de cerveja, 5 g/L de KNO3, 0,12 g/L de CaCl2.2H2O, 1,5 g/L de MgSO4.7H2O, 0,09 g/L de MnSO4, 1,5 g/L de K2HPO4 e 1,5 g/L de KH2PO4. Essa mesma composição de meio de cultivo foi testada em um ensaio em biorreator, obtendo-se um meio fermentado mais eficiente que o ensaio realizado em incubadora, e com CL50 de 2,43 µL. / The use of bioinsecticide based on Bacillus thuringiensis has been shown to be effective against larvae of insects able to spread diseases such as dengue, malaria and elephantiasis. The bioinsecticide has high specificity, absence of resistance in target insects and low environmental impact compared to chemical insecticides. However, there are some challenges in the way of widespread use of bioinsecticide, such as the costs of production and formulation. The surplus yeast from the brewing industry can make the production of bioinsecticide economically feasible due to its low cost, as well as the residual glycerol from the biodiesel production. The proper choice of the components of the culture medium is essential for the success of a product and must be carried out in order to obtain a high productivity at the lowest cost. Furthermore, in the case of bioinsecticide, medium components have a major influence on the growth and synthesis of toxins by the bacteria. In the present study it was proposed the formulation of a fermentation medium for Bacillus thuringiensis var. israelensis using brewer\'s yeast and glycerol from the biodiesel manufacturing, seeking to obtain a fermented broth with high toxicity and lower cost compared to those currently obtained. Initially it was defined a procedure to prepare the extract of brewer\'s yeast. It was found that the best method is the sterilization of the brewer\'s yeast suspension (121 °C for 20 minutes) followed by qualitative filtration. The range of brewer\'s yeast extract concentration to be used in the study of the medium composition was also defined (10 g/L to 30 g/L). Assays were performed in 1000 mL Erlenmeyer flasks in a reciprocal motion shaker. The components of the medium (glycerol waste, brewer\'s yeast extract, KNO3, CaCl2.2H2O, MgSO4.7H2O, MnSO4, K2HPO4 e KH2PO4) were evaluated according to an experimental design and, considering the studied conditions, they did not have significant influence on the toxin production. Thus, the composition of the fermentation medium was defined in terms of nitrogen and carbon source. Glycerol and brewer\'s yeast extract did not have any significant influence on the substrate consumption by the bacteria. For the maximum cell concentration, the increase of glycerol concentration showed a negative effect, while increasing the concentration of brewer\'s yeast extract showed a positive effect. Although the concentrations of carbon and nitrogen sources were not significant for the larvicidal activity, the interaction between such factors was significant in its negative level. The best result of larvicidal activity was shown by the following medium: 10 g/L of residual glycerol, 30 g/L of brewer\'s yeast extract, 5 g/L of KNO3, 0.12 g/L of CaCl2.2H2O, 1.5 g/L of MgSO4.7H2O, 0.09 g/L of MnSO4, 1.5 g/L of K2HPO4 and 1.5 g/L of KH2PO4. This same composition of the culture medium was tested in a bench fermenter, resulting in a more efficient fermented broth than the one obtained in shaker flask, and with LC50 of 2.43 µL.

Bacillus thuringiensis é uma bactéria do solo, gram-positiva, que se destaca pela capacidade de sintetizar um cristal protéico, adjacente ao esporo, responsável pela ação tóxica contra diversas espécies de dípteros, como a larva do mosquito Aedes aegypti, o qual é o principal vetor da dengue. A dengue hoje é uma das doenças de maior preocupação para as agências de saúde brasileiras, uma vez que o Brasil possui temperatura e condições socio-econômicas que favorecem proliferação do mosquito transmissor. O Brasil se destaca por desenvolver combustíveis alternativos aos combustíveis de origem petrolífera, como etanol e biodiesel. Porém, durante a reação de transesterificação de óleos/gorduras para a produção de biodiesel é gerado também glicerina e, por não haver uma demanda deste sub-produto no mercado mundial, seu destino se tornou um problema e um desafio para as indústrias de biodiesel. Logo, desenvolver um bioinseticida para o combate da dengue empregando a glicerina como fonte de carbono é uma forma de encontrar novas aplicações para a glicerina e contribuir para o controle de uma doença de impacto nacional. Estudos realizados comprovaram que o ácido acético é um composto produzido e consumido pelas células de Bacillus thuringiensis durante seu crescimento. Além disso, apresenta papel fundamental para a produção de um composto chamado poli-?-hidroxibutirato (PHB), que é fundamental no fornecimento de energia para a síntese da toxina. No presente trabalho estudou-se o efeito do ácido acético sobre o crescimento de Bacillus thuringiensis e sobre a atividade larvicida do meio fermentado contra larvas de Aedes aegypti. Foram avaliadas as concentrações de 1 g/L, 3 g/L e 5 g/L de ácido acético nas seguintes fases: (i) início da fermentação, (ii) ponto de valor mínimo de pH e (iii) ponto de desaceleração do aumento do pH. Os ensaios foram realizados em frascos Erlenmeyer de 1000 mL em incubadora de movimento recíproco. Os resultados obtidos demonstraram que a adição de ácido acético no início e no tempo igual a 10 horas de fermentação favorece o crescimento celular e a produção de toxinas. O resultado mais expressivo de produção de toxinas foi obtido com adição de 5 g/L de ácido no início da fermentação. Neste ensaio o resultado de porcentagem de morte de larvas foi 8 superior ao resultado encontrado no ensaio sem adição de ácido (ensaio controle). Já no ensaio com adição de ácido com 20 horas de fermentação foi observada a maior concentração celular máxima, porém, obteve-se uma menor atividade larvicida do meio fermentado, comparado com o ensaio controle. / Bacillus thuringiensis is a gram-positive soil bacterium that is distinguished by the ability of synthesizing a protein crystal adjacent to the spore. This protein is responsible for the toxic action against several species of Diptera, as the larvae of the Aedes aegypti mosquito, which is the main vector of dengue. As Brazil possesses temperature and socioeconomic conditions that favors proliferation of the dengue transmitting mosquito, it is one of the major concerns to health agencies in Brazil. Brazil stands out for developing alternative fuel sources to petroleum fuels such as ethanol and biodiesel. However, during the transesterification reaction of oils/fats for the production of biodiesel glycerin is also generated. As there is no demand for this sub-product in the global market, its fate has become a problem and a challenge for the biodiesel industry. Therefore the development of an insecticide for fighting dengue using glycerol as the carbon source is a way to find new applications for glycerin and to contribute to the control of a disease with nationwide impact, considered a public health problem. Studies have shown that acetic acid is a compound produced and consumed by Bacillus thuringiensis\' cells during their growth. Moreover, it presents a major role in the production of a compound called poly-?-hydroxybutyrate (PHB), which is critical for providing energy for the toxin synthesis. In the present study, it was studied the effect of acetic acid on the growth of Bacillus thuringiensis and on the larvicidal activity of the fermented broth on the larvae of Aedes aegypti. The concentrations tested were 1 g/L, 3 g/L and 5 g/L of acetic acid, added in the following stages: (i) beginning of the fermentation, (ii) time of minimum pH value and (iii) time of deceleration of the increasing pH value. The assays were performed in 1000 mL Erlenmeyer flasks in a reciprocating motion shaker. The results showed that the addition of acetic acid at the beginning and at 10 hours of fermentation improves cell growth and toxin production. The most significant result regarding toxin production was achieved with the addition of 5 g/L of acid at the beginning of the fermentation. In this assay the net percentage of killing of the larvae was eight times higher than the results found in the test without the addition of acid (control test). For the test in which acetic acid was added at 20 hours of fermentation it was observed a higher cellular concentration, however, there was a lower larvicidal activity of the fermented broth, compared to the control test.

Os plantios de eucalipto sofrem com severos ataques de insetos-praga, cujos danos comprometem seriamente a produtividade. O controle biológico é a principal estratégia adotada pelas empresas do setor florestal, envolvendo sistemas de monitoramento das populações de insetos-praga. Um dos mais importantes grupos de desfolhadores é o dos lepidópteros desfolhadores. Espécies nativas brasileiras se adaptaram às plantações de eucalipto ao longo dos anos e a mais notável é a lagarta parda do eucalipto, Thyrinteina arnobia (Stoll, 1782) (Lepidoptera: Geometridae). Com ampla distribuição no território brasileiro, esta espécie se caracteriza pelos surtos nos períodos secos do ano, quando encontra condições favoráveis para expressar seu elevado potencial reprodutivo, causando severas desfolhas em grandes extensões. Medidas de controle são frequentemente necessárias, sendo realizadas pulverizações aéreas de bioinseticidas a base de Bacillus thuringiensis (Berliner, 1915) e liberações de inimigos naturais. Entretanto, epizootias virais podem ser observadas como reguladores naturais de tais populações. Eucalyptus plantations suffer severe attacks from insect-pests, whose damages compromise seriously the productivity. Biological control is the main strategy adopted by the forest companies, involving pest population monitoring systems. One of the most important defoliators group is the lepidopterous defoliators. Native Brazilian species had adapted to the eucalyptus plantations over the years and the most remarkable one is the eucalyptus brown looper, Thyrinteina arnobia (Stoll, 1782) (Lepidoptera: Geometridae). Spread all over Brazil's territory, this species is characterized by the outbreaks during the dry periods of the year, when it has good conditions to express its high reproductive potential, causing severe defoliation in large extensions. Control measures are frequently necessary, thus aerial spraying with Bacillus thuringiensis (Berliner, 1915) based bioinseticides and releases of natural enemies are carried out. However, sometimes viral epizootics can be observed acting as natural regulators for such populations. The interactions between B. thuringiensis and native entomopathogenic viruses are unknown to T.arnobia in eucalyptus plantations. In this context, the present work had as objectives: (I) the toxicity analysis of six different Cry1 proteins of B. thuringiensis on populations of T. arnobia and their effect on the emergence of latent viruses in these populations; (II) complete genome sequencing, phylogenetic analysis and ultrastructural characterization of a cypovirus found in T. arnobia caterpillars. O delineamento utilizado foi de blocos casualisados em esquema fatorial 2x3, sendo 2 manejos de inseticidas (com e sem inseticidas para controle de lepidopteros) e 3 modos de utilização de cultivares: área 100% transgênica (Nuopal), área 100% não transgênica (Deltaopal) e área com interior transgênico (80% Nuopal) e bordadura não transgênico (20% Deltaopal); com 5 repetições. O manejo das pragas foi realizado conforme os níves de controle preconizados no MIP. Realizaram-se avaliações semanais da infestação de pragas, observando-se em 15 plantas por parcela os seguintes parâmetros: número de lagartas de Alabama argillacea, Heliothis virescens e Spodoptera frugiperda; número de adultos de Bemisia tabaci biótipo B; e número de adultos e ninfas de Aphis gossypii. Avaliou-se 100 botões florais por parcela anotando-se o número de estruturas reprodutivas atacadas por Anthonomus grandis. Realizou-se a medição da produção e a analise econômica dos tratamentos. Pela análise dos resultados concluiu-se que: a ocorrência de A. argillacea e H. virescens foi menor nos tratamentos com a cultivar transgênica. Não ocorreram diferenças entre a cultivar transgênica e convencional quanto à ocorrência de S. frugiperda, A. gossypii, B. tabaci biótipo B e A. grandis. Estatisticamente não ocorreram diferenças na produtividade... / The cotton crop presents a complex of pests that can limit the yield. A new alternative in integrated pest management is the use of genetically modified plants resistant to caterpillars. Aiming at to study the occurrence of pests and costs yield on cotton genetically modified, with the introduction of the bacterium Bacillus thuringiensis, comparatively to the conventional cotton in the Cerrado region: two experiments were carried out at field conditions, in the seasons 2006/2007 and 2007/2008 in Chapadão do Sul, State of Mato Grosso do Sul, Brazil, with design blocks, in the 2x3 factorial system, with 2 applications of insecticides (with and without insecticides for lepidoptera control) and 3 modes of use of cultivars: 100% area-transgenic (Bt- Nuopal), area 100% non-transgenic (Deltaopal) and transgenic area with 80% (Bt-Nuopal) and border non-transgenic (20%-Deltaopal), with 5 replicates. The pests management was carried out according to the recommended levels of control in IPM. Weekly assessment were made to verify of pest infestation, observing 15 plants per plot the following parameters: number of larvae of Alabama argillacea, Heliothis virescens and Spodoptera frugiperda, number of adults of Bemisia tabaci biotype B, and number of adults and nymphs of Aphis gossypii. It was evaluated 100 flower buds per plot recording the number of structures affected by Anthonomus grandis. Was made the measurement of production and economic analysis of treatments. The occurrence of Alabama argillacea and Heliothis virescens was more reduced in treatments with the transgenic cultivar than conventional cultivar. There were no differences in infestations of S. frugiperda, A. gossypii, B. tabaci biotype B and A. grandis. There were no differences in cotton production among cultivars transgenic and conventional. The cultivar conventional obtained the highest profitability, in the two seasons evaluated

Obtenção de bioinseticida a partir de Bacillus thuringiensis var. israelensis e aplicação de bioensaios em culicídeos /

Orientador: Iracema de Oliveira Moraes / Coorientador: Vanildo Luiz Del Bianchi / Resumo: A bactéria Bacillus thuringiensis var. israelensis (Bti) gera certas toxinas com ação inseticida que podem ser usadas no controle de doenças transmissíveis através de culicídeos, especialmente o Aedes aegypti, vetor da dengue. Este bioinseticida tem sido produzido através de fermentação submersa e, no Brasil, esta produção tem sido realizada através de pequenos centros de investigação e, mais recentemente, por uma pequena empresa. Para a implementação de um programa de controle de vetores viável através de bioinseticidas, alguns estudos sobre meio de cultura são essenciais para unir eficiência e baixos custos. Assim, tem-se utilizado resíduos ou subprodutos agroindustriais como fontes de nutrientes em meio de cultura. Neste estudo, alguns resíduos e subprodutos tais como água de maceração de milho ("milhocina"), um subproduto do processamento industrial do milho, "manipueira", um subproduto do processamento da farinha de mandioca e sangue bovino oriundo de abatedouros foram avaliados com o intuito de se verificar a eficiência em processo fermentativo para obtenção de bioinseticidas. O crescimento celular foi avaliado por densidade óptica a 620 nm, a produção de esporo por plaqueamento em ágar nutriente e a CL 50 (Concentração letal média), por bioensaios contra larvas de 4º instar de Ae. aegypti. Foi verificado, em estudos com "manipueira", que quando este subproduto é utilizado em baixa concentração, há o mesmo ou um melhor efeito do que quando se utiliza em concentração alta. A esterilização parece ser necessária ao processo fermentativo, pois quando se comparam ambos os meios de "manipueira", um submetido à esterilização e outro sem este processo, o primeiro produziu mais esporos e uma melhor eficiência em bioensaios. (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: The bacterium Bacillus thuringiensis var. israelensis (Bti) generates certain toxins with pesticid action, which can be used on the control of transmissible diseases by culicides, specially Aedes aegypti, the dengue's vector. This biopesticide has been produced by submerged fermentation and, in Brazil, this production has been made by very little research centers and, more recently, by a unique small enterprise. For the implementation of a viable vectors control program through bioinsecticides, some studies about culture media are essential in order to join efficiency and low costs. So, agroindustrial wastes or by-products have been utilized as nutrient source for the culture media production. In this study, some wastes and by-products like corn steep liquor ("milhocina"), a corn industrial processing byproduct, "manipueira", a by-product from cassava flour processing and cattle blood proceeding from slaughterhouses were utilized as substrate in order to verify the efficiency on fermentative process to obtain bioinsecticides. Cellular growth was evaluated by optical density at 620 nm, spore production by pour plate count and LC50 by bioassays against 4th instar larvae. It was verified, in studies with "manipueira", that a low concentration of this byproduct has the same or better effect compared with high concentrations. The sterilization was a requisite to the fermentative process because when compared both sterilized and no sterilized "manipueira" media, the first yielded more spores and better efficiency in bioassays. Likewise, when cattle blood is added to "manipueira" media, there is an increase on the spores number and effectiveness against Ae. aegypti larvae. It was compared a traditional nitrogen source, peptone, with "manipueira", with and without salts and glucose, tested separately and joined. Pela análise dos resultados concluiu-se que: a ocorrência de A. argillacea e H. virescens foi menor nos tratamentos com a cultivar transgênica. Não ocorreram diferenças entre a cultivar transgênica e convencional quanto à ocorrência de S. frugiperda, A. gossypii, B. tabaci biótipo B e A. grandis. A new alternative in integrated pest management is the use of genetically modified plants resistant to caterpillars. Aiming at to study the occurrence of pests and costs yield on cotton genetically modified, with the introduction of the bacterium Bacillus thuringiensis, comparatively to the conventional cotton in the Cerrado region: two experiments were carried out at field conditions, in the seasons 2006/2007 and 2007/2008 in Chapadão do Sul, State of Mato Grosso do Sul, Brazil, with design blocks, in the 2x3 factorial system, with 2 applications of insecticides (with and without insecticides for lepidoptera control) and 3 modes of use of cultivars: 100% area-transgenic (Bt- Nuopal), area 100% non-transgenic (Deltaopal) and transgenic area with 80% (Bt-Nuopal) and border non-transgenic (20%-Deltaopal), with 5 replicates. The pests management was carried out according to the recommended levels of control in IPM. Weekly assessment were made to verify of pest infestation, observing 15 plants per plot the following parameters: number of larvae of Alabama argillacea, Heliothis virescens and Spodoptera frugiperda, number of adults of Bemisia tabaci biotype B, and number of adults and nymphs of Aphis gossypii. It was evaluated 100 flower buds per plot recording the number of structures affected by Anthonomus grandis. Was made the measurement of production and economic analysis of treatments. The occurrence of Alabama argillacea and Heliothis virescens was more reduced in treatments with the transgenic cultivar than conventional cultivar. There were no differences in infestations of S. frugiperda, A. gossypii, B. tabaci biotype B and A. grandis. Obtenção de bioinseticida de Bacillus thuringiensis a partir de resíduo da fabricação de biodiesel / Obtainment of bioinsecticides from Bacillus thuringiensis by the waste of biodiesel production

A crescente demanda por combustíveis provenientes de fontes renováveis, em função de questões ambientais e possível escassez dos derivados de petróleo, juntamente com a obrigatoriedade de sua adição no óleo diesel empregado atualmente, levou a uma \"explosão\" do número de empresas que produzem biodiesel. Este produto é obtido pela transesterificação de gorduras animais e óleos vegetais na presença de catalisador, processo que resulta um único sub-produto, a glicerina, a qual teve um aumento de produção na medida do aumento de produção do biodiesel. Uma forma de contribuir para diminuir os problemas advindos do acúmulo da glicerina é utilizá-la como base para formular meios de fermentação e obter produtos de interesse sócio-econômico. Um exemplo é o bioinseticida produzido pela bactéria Bacillus thuringiensis para o controle do mosquito transmissor do vírus da dengue. Este produto apresenta várias vantagens quando comparado aos inseticidas químicos, pois impede que os insetos adquiram resistência, não polui e é seletivo, não oferecendo riscos de intoxicação para os insetos benéficos e animais vertebrados. Cabe salientar que as toxinas produzidas por esta bactéria são letais também para larvas dos mosquitos transmissores da malária e da elefantíase. Assim, no presente trabalho propõe-se obter um bioinseticida com princípio ativo produzido por Bacillus thuringiensis var. israelensis, empregando o resíduo da fabricação de biodiesel a partir de óleo vegetal. Para tanto, o trabalho compreendeu três fases: 1. Definição de um tratamento para o resíduo que proporcionasse o crescimento da bactéria e a produção de toxina. 2- Estudo do processo fermentativo com o objetivo aumentar a atividade larvicida do meio fermentado em frascos (definição da composição do meio e de condições de fermentação) e 3. Estudo da transferância de oxigênio em diferentes fases do desenvolvimento da bactéria, em biorreator. O estudo em frasco foi realizado em Erlenmeyer de 1.000 mL, provido ou não de chicanas, com 200 mL de meio. O estudo da transferência de oxigênio foi realizado em fermentador Bioflo III, (New Brunswick Sci. Co.) de 1,25 L. Com os resultados obtidos, foi possível concluir que o melhor tratamento para o resíduo em estudo, consistiu do abaixamento do pH até 4 com ácido fosfórico, seguido de decantação por 24 horas. Com este tratamento foi possível obter uma fonte de carbono mais viável enocomicamente e, pelos estudos de otimização de processo, foi possível determinar a melhor composição de meio de cultura (10 g/L de glicerina tratada, 6 g/L de extrato de levedura, 3 g/L de (NH4)2SO4, 0,12 g/L de CaCl2.2H2O, 1,5 g/L de MgSO4.72H2O, 0,09 g/L de MnSO4, 1,5 g/L de K2HPO4 e 1,5 g/L de KH2PO4) e a melhor condição de aeração (manter a concentração de oxigênio dissolvido em 45% da saturação durante todas as fases do desenvolvimento da bactéria). Desta forma, foi possível produzir um meio fermentado 583 vezes mais potente que o ensaio controle empregando glicerol PA. / The increasing demand for fuels originated from renewable sources, due to environmental issues and possible scarce of petroleum products, together with the mandatory of its inclusion in diesel oil currently used, generated a significant increase in the number of biodiesel producting companies. This product is obtained through transesterification reaction of animal fat and vegetable oils in the presence of a catalyzer, process that results in a single by-product, the glycerin, which had an increase of its production as biodiesel production increases. A way to contribute for the reduction of problems generated by the excess of glycerin is to use it in the composition of fermentation medium and obtain products that are demanded by society. An example is the bioinsecticides produced by the bacteria named Bacillus thuringiensis, used in the controlling of mosquito vectors of the dengue viruses. This product presents several advantages when compared to chemical insecticides because it prevents insects resistance, it does not pollute the environment and it is selective, which means it does not create risks of intoxication for other insects and vertebrate animals. It is important to mention that toxins produced by these bacteria are lethal as well to the larvae of malaria and elephantiasis mosquito vectors. Thus, in this work it is intended to obtain a bioinsecticide containing active ingredient from Bacillus thuringiensis var. israelensis by using the biodiesel production waste from vegetable oils. In order to achieve that, this work consisted of three steps: 1. Definition of a treatment for the waste of biodiesel to provide the growth of Bacillus thuringiensis and its toxin production; 2. Study of the fermentative process to increase larvicidal activity of the fermentation broth in flasks (definition of the medium composition and the fermentative conditions) and 3. Study of the oxygen transference in bioreactor in diferent development phases of the bacteria. The study in flask was made in a 1000 mL Erlenmeyer flask, with and without baffles, and 200 mL of medium. The study of oxygen transference was realized in a 1,25 L bench fermentor (New Brunswick Sci. Co./Bioflo III). With the results obtained it was possible to conclude that the best treatment to the waste of biodiesel consisted of decreasing pH until 4 with phophoric acid, followed by 24 hours of decantation. With this treatment it was possible to obtain a promising carbon source more and, by the process optimization studies, it was possible to determine the best composition of culture medium (10 g/L of treated glycerin; 6 g/L of yeast extract; 3 g/L of (NH4)2SO4; 0,12 g/L of CaCl2.2H2O; 1,5 g/L of MgSO4.7H2O; 0,09 g/L of MnSO4; 1,5 g/L of K2HPO4 and 1,5 g/L of KH2PO4) and the best aeration condition (mantaining the oxygen concentration at 45% of saturation during all phases of bacterial development). So, it was possible to obtain a fermented broth 583 times more potent than the control test using pure glycerol.

The development of new strategies for the control of Aedes aegypti is of utmost importance to minimize or eliminate the cases of the disease. An alternative for the control of this vector is the use of biological control agents such as Bacillus thuringiensis, which is toxic to this insect order of epidemiological importance. In this study, we sought to investigate the biological control of A. aegypti, with the use of B. thuringiensis isolated from Cerrado Maranhense. Selective bioassays were conducted to determine the entomopathogenic activity against the larvae of A. aegypti, quantitative bioassays in order to determine the toxicity of isolates, by means of the estimation of Median Lethal Concentration (LC50) and PCR tests (Polymerase Chain Reaction) to verify the presence of mosquitocidal genes (cry and cyt). From 45 soil samples collected in 17 counties in the State, it was obtained 1225 bacterial colonies, with 383 (31.26%) identified as B. thuringiensis. Of the 383 isolates tested, 12 (3.13%) showed larvicidal activity greater than 50% (BtMA 25, BtMA 64, BtMA 104, BtMA 131, BtMA 194, BtMA 251, BtMA 401, BtMA 410, BtMA 413, BtMA 450, BtMA 451 and BtMA 560), which were selected for subsequent realization of the quantitative bioassay and molecular characterization. The quantitative bioassays allowed to observe that the lower LC50 values were obtained with the isolate 401 (4.0 x 104 spore-crystals/mL) and with the standard bacteria Bti (0.32 x 107 spore-crystals/mL). The others tested isolates showed low larvicidal activity. The PCR analysis showed that from eight tested genes: cry4Aa, cry4Ba, cry10Aa, cry11Aa, cry11Ba, cyt1Aa, cyt1Ab and cyt2Aa, five of them: cry11Aa, cry11Ba, cyt1Aa, cyt1Ab and cyt2Aa were detected in ten isolates, being absent in only two of 12 isolates tested. These data demonstrate that the isolates of B. thuringiensis from Cerrado Maranhense showed high pathogenicity and potential for biological control of A. aegypti. / Atualmente a dengue é um grave problema de saúde pública mundial, no Brasil verifica-se aumento do número de casos a cada ano e a circulação de vários sorotipos virais. O desenvolvimento de novas estratégias para o controle do Aedes aegypti é de extrema importância, para minimizar ou erradicar os casos da doença. Uma alternativa para o controle deste vetor é a utilização de agentes de controle biológico, como Bacillus thuringiensis, que apresenta toxicidade a essa ordem de inseto de importância epidemiológica. Neste estudo, buscou-se investigar o controle biológico de A. aegypti, com o uso de B. thuringiensis isolados do Cerrado Maranhense. Foram realizados bioensaios seletivos para determinar a atividade entomopatogênica contra as larvas de A. aegypti, bioensaios quantitativos com a finalidade de determinar a toxicidade dos isolados, por meio da estimativa da Concentração Letal Mediana (CL50) e testes de PCR (Reação em Cadeia da Polimerase) para verificar a presença de genes mosquitocida (cry e cyt). A partir de 45 amostras de solo coletadas em 17 municípios do estado, obteve-se 1225 colônias bacterianas, sendo 383 (31,26%) identificadas como B. thuringiensis. Dos 383 isolados testados, 12 (3,13%) mostraram atividade larvicida superior a 50% (BtMA 25, BtMA 64, BtMA 104, BtMA 131, BtMA 194, BtMA 251, BtMA 401, BtMA 410, BtMA 413, BtMA 450, BtMA 451 e BtMA 560), aos quais foram selecionados para posterior realização do bioensaio quantitativo e caracterização molecular. Os bioensaios quantitativos permitiram observar que os menores valores de CL50 foram obtidos com o isolado 401 (4,0 x 104 esporos-cristais/mL) e com a bactéria padrão Bti (0,32 x 107 esporos-cristais/mL). Os demais isolados testados mostraram baixa atividade larvicida. Os testes de PCR mostraram que dos oito genes testados: cry4Aa, cry4Ba, cry10Aa, cry11Aa, cry11Ba, cyt1Aa, cyt1Ab e cyt2Aa, cinco destes: cry11Aa, cry11Ba, cyt1Aa, cyt1Ab e cyt2Aa foram detectados em dez isolados, estando ausentes em apenas dois dos 12 isolados testados. Esses dados demonstram que os isolados de B. thuringiensis do Cerrado Maranhense apresentaram alta patogenicidade e potencial para o controle biológico de A. aegypti.

Effets des bioinsecticides à base de Bacillus thuringiensis sur la physiologie intestinale de la Drosophile / Effects of Bacillus thuringiensis bioinsecticides on the Drosophila gut physiology

Le tube digestif est la première barrière contre les agresseurs présents dans la nourriture (virus, bactéries, produits chimiques, pesticides etc...). Il doit donc maintenir au mieux son intégrité structurale et fonctionnelle tout au long de la vie de l'individu. Bien que l'impact délétère d'une intoxication aiguë puisse être surmonté par la capacité de défense et de régénération de la muqueuse digestive, une agression prolongée ou répétée peut compromettre l'équilibre physiologique (l'homéostasie) du tube digestif. Parmi les agresseurs pouvant être ingérés avec la nourriture, on trouve la bactérie Bacillus thuringiensis (Bt) et représente 70% des ventes de bioinsecticides. Bt est une bactérie Gram+ sporulante qui produit, pendant la sporulation, des toxines nommées Cry. Parmi les différentes souches de Bt, certaines ont été sélectionnées pour la spécificité d'action de leurs toxines Cry contre des nuisibles et sont commercialisées sous forme de sporanges. Certaines de ces souches sont utilisées en agriculture biologique en France et l'accroissement de leur utilisation fait qu'elles sont de plus en plus présentes dans la nourriture, source de contamination potentielle pour l'homme et l’environnement. La question qui se pose maintenant est de savoir si un tel accroissement de l’utilisation de Bt peut avoir des impacts sur des espèces non cibles. Mon projet de thèse a consisté en l’étude des conséquences de l'ingestion de Bt (sous forme végétative ou de sporanges) sur la physiologie intestinale de la drosophile (animal non sensible à Bt en termes de toxicité aigüe / The digestive tract is continuously subjected to multiple aggressions through virus, bacteria, toxins and chemicals mixed in the feed. Therefore the gut lining has established a mechanism of replenishment in order to maintain the physiological function of the organ called the gut homeostasis. Although the deleterious impact of acute poisoning can be overcome by the defense capacity and regeneration of the gut mucosa, prolonged or repeated intoxication can impair its homeostasis. Among the aggressors hidden in the feed, there is the bacterium Bacillus thuringiensis (Bt). Bt is worldwide used as bioinsecticide. Indeed the multitude of Bt strains produces a broad range of crystalline toxins, named Cry toxins, which certain have been selected in organic farming owing to their lethal properties against specific pests. Because of incentive programs for sustainable development, the use of Bt bioinsecticides as an alternative to chemical pesticides will further increase in the next decades. Although the specificity of the acute toxicity of Cry toxins has been proved since many years, data are scarce on adverse effects that could result from chronic exposure. The question now is how far non-target organisms will be potentially impacted by the resulting augmentation of the Bt bacterium and its Cry toxins in the environment. To answer this challenge, I used Drosophila (a non-target organism) to study the impacts of Bt bioinsecticides on the gut physiology because 1/ the digestive tract is the main entrance for feed contaminated by Bt bioinsecticides and 2/ Bt and its toxins are known to impair the gut epithelium of sensitive pests

Caracterización molecular de cepas de Bacillus Thuringiensis con propiedades entomocidas, para vectores transmisores de enfermedades metaxénicas (dengue)

Bacillus thuringiensis (B. thuringiensis) es una bacteria Gram positiva que forma un cristal paraesporal con actividad insecticida contra insectos de diferentesórdenes. Los bioinsecticidas fabricados con las esporas y cristales de B. thuringiensis son una alternativa frente a los pesticidas químicos sintéticos para el control de mosquitos que son vectores de enfermedades metaxénicas, como por ejemplo, malaria y dengue. Se caracterizaron 53 cepas nativas de B. thuringiensis, procedentes de suelos peruanos, de los departamentos de Junín, Lima (Huaral), Ica, Cuzco, Tacna, Arequipa, Chiclayo y Cajamarca; dos de ellas codificadas como Ica10 y Caj1 por su procedencia, presentaron actividad entomocida contra larvas de Aedes aegypti. Se observan cepas que presentan bandas (toxinas) con pesos moleculares en el órden de los 36 kDa y mayores a 116 kDa; otras cepas presentan bandas a 97 y 75 kDa, como las de, Cajamarca, Arequipa y Chiclayo, que serían entomocidas para dípteros, algunas cepas presentaron bandas de 130 kDa que corresponderían al gen cry1, tóxico para lepidópteros. El gen cry2 fue el más frecuente (92%), seguido de cry1 (45%) y cry4 (42%), lo cual significaría que las cepas tendrían principalmente, actividad entomocida contra dípteros. El secuenciamiento confirmó la presencia de genes cry en 62 de 67 cepas analizadas. Se determinó la cepa B. thuringiensis Cuz5 como variedad cry2Ab. El método del PCR fue efectivo para caracterizar rápidamente las cepas en base al contenido de genes que amplifican con secuencias de oligonucleótidos conocidos. El método REP-PCR mostró patrones de bandas diferentes en las cepas aisladas que podría significar variabilidad genética entre las cepas estudiadas, el dendograma permitió agruparlas en tres clusters y subclusters que podrían deberse a las variedades del gen cry. Bioinsecticides manufactured with B. thuringiensis spores and crystals are a useful alternative to synthetic chemical pesticide in agriculture and for mosquito control. B. thuringiensis has also been used successfully to suppress the population levels of medically important dipteran pests. Examples include the mosquito vectors of malaria, virus diseases including dengue. It was characterized 53 strains peruanas of Bacillus thuringiensis with entomocidal properties from soil Peruvian’s farms of the deparments of Junín, Lima (Huaral), Ica, Cuzco, Tacna, Arequipa, Chiclayo y Cajamarca, two: Ica10 y Caj1 with activity against Aedes aegypti. There are strains with toxins betwen 36 y 116kDa. Strains with bands to 97 and 70KDa, as those of Arequipa, Cajamarca and Chiclayo, which would be entomocidal with band of 130 kDa that correspond to cry gen, toxic to Lepidoptera and Diptera. Cry2 gene was the most abundant (92%), followed by cry1 (45%) and cry 4 (42%), which means that the strains would have activity against Dipterans. The sequencing confirms the content of cry genes in most of the analyzed strains. The strain Cuz5 as variety cries 2Ab. The PCR method is effective to rapidly characterize the strains based on the content of genes that amplify with known sequences of oligonucleótidos and REP-PCR shows patterns of different bands in the isolated strains that could mean genetic variability among the studied strains, the dendogram allows to group them into three clusters and subclusters, that might be due to the varieties of the gene cry.
