Aplicação da técnica da dupla camada na soldagem de dutos em operação

A intervenção por soldagem em dutos em operação adquiriu nos últimos anos extrema importância na indústria do petróleo, visto que por meio dela é possível instalar derivações bem como reparar trechos danificados na malha dutoviária sem a interrupção do fluxo dos produtos escoados, o que certamente impactaria em elevados custos nas operações. Os principais limitantes da técnica de soldagem em operação é o risco de perfuração do duto pelo arco durante a soldagem e principalmente, o elevado risco das juntas soldadas sofrerem fissurações induzidas pelo hidrogênio. Estas últimas surgem devido à ocorrência de três fatores em conjunto: alto nível de hidrogênio no processo de soldagem, tensão residual e, microestrutura susceptível. Neste trabalho, são feitas algumas considerações acerca dos métodos atualmente empregados para se contornar os limitantes da técnica da soldagem em operação aplicada pelo processo de eletrodo revestido (ER). Mais especificamente, estudou-se uma metodologia da técnica da dupla camada, amplamente empregado nos procedimentos de soldagem que visam evitar a formação de microestrutura susceptível à trincas por hidrogênio e, avaliação de dois softwares comumente empregados para avaliar o risco de perfuração do duto pelo arco durante a soldagem em operação. Paralelamente investigou-se equações para estimativas de durezas máxima da ZAC publicada por diversos autores. Ficou evidente que a metodologia avaliada para a prevenção da perfuração do duto durante a soldagem em operação é bastante confiável e pode ser aplicada com segurança. Quanto às estimativas de dureza máxima na ZAC, os resultados não foram unânimes dentre as equações dos autores investigados. A metodologia da técnica da dupla camada estudada se mostrou eficiente, porém com algumas limitações. / Welding interventions onto in-service pipe lines have acquired relevant importance in the last few years in petroleum industry, since with this technique it is possible to install derivations as well to repair damaged pipe lines portions without interrupt the flux of its products, which in turn would results in high costs. The in-service welding techniques limitations are mainly the risk of pipe wall burn through by the electric arc and, the hydrogen cold crack joint susceptibility. The last one is due to the action, at the same time, of three mainly factors: high hydrogen levels and, susceptible microstructure. In this work it is made some considerations about methods usually applied to work out the limitations of shielded metal arc welding technique applied to in-service welding. Specifically talking, it was studied a temper bead technique commonly used in procedures which the aim is to avoid the presence of hydrogen cold crack susceptible microstructure and, evaluate two commonly software used to prevent the wall pipe burn-through risk by the arc welding. Additionally, it was investigated some published equations to estimate the maximum HAZ hardness. It was clarified that the evaluated methodology to prevent the pipe wall burn-through risk during the welding process is very confident and safety to be applied in practice situations. The studies conducted about the maximum HAZ hardness showed that just a few of the investigated equations and authors, the results are reliable. The temper bead technique studied as presented as an efficiently method, although some limitations must be taken in account.

Method development for enrichment of autoantibodies from human plasma

Antibodies are naturally occurring in humans, with the function to protect the body from pathogens. Occasionally, antibodies towards the body’s own proteins are produced. These so called autoantibodies are present in healthy individuals but are also highly associated with diseases with autoimmune involvement. Research on autoantibodies in healthy individuals as well as in patients is important to gain knowledge and facilitate prognostics, diagnostics and treatment. However, a method for purification of these antibodies has not previously been described. In the present project, an enrichment procedure of circulating autoantibodies found in human plasma is described. Twenty protein fragments previously known to be highly reactive were attached to magnetic microbeads, enabling autoantibodies from eight human plasma sample pools to be captured. The six antigens with highest shown reactivity were chosen for elution procedure. Using pH alterations and heat treatments, a successful elution and enrichment procedure was developed. With analysis of the eluted autoantibodies, it can be established that the enrichment was successful on multiple sample pools. In the scaled-up procedure, autoantibodies could be enriched in all positive antigen-sample combinations. Concentration measurements indicated amounts of up to 0.23 mg antibodies per ml eluate. This implies sufficient concentrations for further applications of the enriched autoantibodies. / Antikroppar förekommer naturligt i människor, med syftet att skydda kroppen från patogen. I vissa fall skapas av misstag antikroppar som angriper kroppens egna proteiner. Dessa autoantikroppar förekommer hos alla människor, såväl friska som sjuka, men de är också starkt förknippade med autoimmuna sjukdomar. Kunskapen om autoantikroppar hos friska personer och hos patienter är idag begränsad, men fortsatt forskning inom området förväntas i framtiden underlätta prognostik, diagnostik och behandling. Hittills har ingen metod för anrikning av autoantikroppar ur blodplasma beskrivits. I detta projekt beskrivs en anrikningsmetod för autoantikroppar ur blodplasma från människa. Tjugo tidigare kända högreaktiva proteinfragment fästes på magnetiska mikrokulor. Dessa antigen-täckta mikrokulor användes för att fånga in autoantikroppar från åtta plasmaprover. De sex proteinfragment som hade högst reaktivitet i dessa prover valdes ut för elueringsförsök. Eluering genomfördes under basiska följt av sura förhållanden, tillsammans med värmebehandling. Denna elueringsmetod fungerade för anrikning av några autoantikroppar från flera av plasmaproverna. I ett utökat experiment kunde autoantikroppar anrikas ur alla kombinationer av antigen och plasmaprov som förväntades ge signal. Koncentrationen av autoantikroppar i eluaten uppskattades till högst 0.23 mg/ml. Denna koncentration är tillräcklig för flera vanliga metoder där antikroppar används.

Magnetic DNA detection sensor for point-of-care diagnostics

This thesis focuses on inductive base sensor design at MHz range frequency. The background theory, design, experiments and results for a new magnetic particles sensor is presented. A new magnetic sensor based on a planar coil was investigated for DNA pathogen detection. Change in inductance of the planar coil due to the presence of magnetic particles with varying mass was measured. The experimental set-up consisted of different sized planar coil with associated electronics for inductance measurements. The best sensor performance was accomplished using two different inductors while oscillating at frequencies 2.4MHz using 9.5μH inductor and 7.2MHz with 85μH inductor. The sensor has very large signal to noise ratio (580×103), while the average amount of frequency drift was 0.58. This sensor was tested with various types of magnetic particles. In addition, iron-oxide nanoparticles were synthesized through water in oil microemulsion method and with an average size of 25nm. The best sensitivity achieved for detection of 50μg iron-oxide particles was with the bead size of 10nm. 81Hz frequency shift was attained in regard to that amount of particles. This research shows that increasing the resonance frequency to 7.2MHz can cause the larger output signal difference (frequency shift) in the presence of magnetic particles; however, the sensor stability is the most important factor for determining the detection resolution and sensitivity. The sensitivity is better if the sensor can detect smaller amount of magnetic sample. The results of this research demonstrate that while the sample consists of smaller size particles, the sensor can detect the lower amount of sample. This is due to the heating effect of nanoparticles. On the other hand the sample distance from the sensor has a major impact on the sensitivity too; the shorter the distance, the higher the sensitivity. This technique can potentially be extended to detect several different types of bacterial pathogens and can be modified for multiplex quantitative detection. This sensing technique will be incorporated into a handheld, disposable microfluidic chip for point-of-care diagnostics for sexually transmitted diseases.

Particle interactions in a magnetophoretic system

The continuous flow separation of magnetic particles from a mixture of particles could improve the performance of magnetic bead based assays but the formation of agglomerates limit the separation efficiency. Bead agglomerates are formed as a result of magnetic binding forces while the hydrodynamic fluid environment strongly influences their movement. The ability to predict the interaction between nearby beads will help to determine a threshold separation distance which will be recommended for use when obtaining measurement within a magnetic bead assay for a specified time interval. The introductory part of this thesis explored the development of a two dimensional numerical model in Matlab which predicts the trajectory pattern as well as magnetic induced velocities between a pair of super-paramagnetic beads suspended in water within a uniform field. The movement of a bead pair interacting due to both magnetic and hydrodynamic forces within a magnetophoretic system was recorded using an optical system; the beads' movements were compared with the simulated trajectories and gave a good agreement. The model was used to predict the shortest agglomeration time for a given separation distance which is of practical benefit to users of bead based assays. The concluding part of this thesis expanded the simulation into a three dimensional model to predict the interactions among three super-paramagnetic beads within a magnetophoretic system. In order to determine the height of the magnetic beads, a Huygens-Fresnel model was implemented in Matlab which was compared with off-focused diffracted images of the beads viewed under an optical system. A good comparison was obtained by comparing the simulated three-dimensional trajectories with experimental data.

Development of Sandwich Assays for Potential Protein Biomarkers in Neurodegenerative Diseases

As the aging population is increasing worldwide, so is the prevalence of neurodegenerativediseases such as Alzheimer’s disease (AD), Parkinson’s disease (PD), frontotemporal dementia(FTD) and amyotrophic lateral sclerosis (ALS). Reliable biomarkers able to aid the diagnosis anddifferentiation of these diseases are needed in order to start the right treatment as early as possible.Due to its representative state of the central nervous system, cerebrospinal fluid (CSF) is afavorable sample material for biomarker discovery within neurodegenerative diseases. Alteredprotein levels of this body fluid might serve as a biomarker, but further validation of earlierfindings is needed. The aim of this project was to validate earlier studies suggesting potentialprotein biomarkers in CSF. From a list of 80 potential biomarkers in the CSF of patient samples,eight were chosen to be included in this validation effort. By utilizing a suspension bead array ina sandwich assay setup, 21 antibodies were tested in an initial screening. Antibody pairs that couldmeasure the protein levels in a dilution dependent manner was further optimized before individualpatient samples were analyzed. Sandwich assays targeting the three proteins Amphiphysin(AMPH), Chitotriosidase-1 (CHIT1) and Beta-synuclein (SNCB) were successfully developed andcorrelated to earlier generated data using a suspension bead array with a single binder setup.Therefore, the earlier findings of elevated levels of AMPH and SNCB in AD patients and CHIT1in ALS patients were successfully validated. / Prevalensen av neurodegenerativa sjukdomar såsom Alzheimers sjukdom (AD), Parkinsonssjukdom (PD), frontallobsdemens (FTD) och amyotrofisk lateralskleros (ALS) ökar i takt med denåldrande populationen. Ryggmärgsvätska, enkroppsvätska tillhörande det centrala nervsystemet, kan ge en inblick i det centrala nervsystemetstillstånd. Förändrade proteinnivåer i denna kroppsvätska skulle därför kunna fungera sombiomarkörer. Målet i detta projekt var att validera tidigare föreslagna proteinbiomarkörer iryggmärgsvätska. Utifrån en lista av 80 tidigare analyserade proteiner i ryggmärgsvätska hospatienter, inkluderades åtta proteiner i detta valideringsförsök. En antikroppsbaserad så kalladsandwich assay användes i en suspension bead array för att testa 21 stycken antikroppar i ett initialtscreeningsförsök. Antikroppspar som kunde mäta proteinnivåer på ett spädningsberoende vis i detinitiala screeningsförsöket optimerades vidare innan den utvecklade sandwich assayn användes föratt analysera proteinnivåer i individuella prover. Sandwich assays gentemot Amphiphysin(AMPH), Chitotriosidase-1 (CHIT1) och Beta-synuclein (SNCB) kunde bli framtagna ochkorrelerade gentemot tidigare genererat data från en single binder assay på ett framgångsrikt sätt.Projektet kunde därmed validera tidigare fynd som indikerat förhöjda nivåer av AMPH och SNCBi AD patienter, samt förhöjda nivåer av CHIT1 i ALS patienter.

Real-time estimation of MIG welding weld bead width using an IR camera

Current manufacturing process controls are principally based only on statistical performance. The next evolution is to make physics based models combined with the state of the art sensors and actuators to control the manufacturing processes. In this paper, metal inert gas welding is used as an example of how the first steps in developing a reliable estimation technique to implement a physics based controller. The weld bead geometry will be the main focus because it is crucial to creating a quality weld. This paper uses an IR camera to generate and evaluate multiple weld bead width estimation techniques and characterizes their corresponding standard deviations. Also a Gaussian Mixture Model (GMM) is used to fit the temperature linescan data to fit an analytical function to the numerical data. The GMM is then used to estimate the weld bead width. Finally, the optimal linescan location is calculated to produce the best possible weld bead estimation. The result is that only one of the estimation techniques actually follows a step input and vi the optimal linescan location is 4 mm from the back of the arc. Furthermore, the GMM provides an excellent fit to the temperature linescan, but does not increase the accuracy of the estimate. / text
