(2) Métodos: Foram utilizados ratos divididos em três grupos (n=8), submetidos à eutanásia aos 14 dias e 28 dias pós-cirúrgicos. Em cada animal foi realizado um defeito ósseo de 5mm na calvária, sendo o Grupo Coágulo (GC) com o defeito preenchido somente com coágulo; Grupo Osso Autógeno (GA), preenchido com osso autógeno e Grupo Bone Ceramic (GBC), com osso aloplástico (Straumann®). Foi avaliada a área de tecido ósseo presente na região central dos defeitos ósseos. Para a comparação entre os valores obtidos, foi realizado o teste ANOVA e como pós-teste, Tukey. Foi adotado como nível de significância p<0,05. Foi realizada análise imunoistoquímica, com os anticorpos primários contra OC, RUNX 2, TRAP e VEGF, para a avaliação dos diferentes estágios do processo de reparo ósseo. Como análise adicional, foram obtidads imagens 3D das cálvarias em microtomografia. (3) Resultados: O local do defeito ósseo originalmente tratado com BC foi totalmente preenchido por um conglomerado de biomateriais e osso neoformado, o qual foi estatisticamente superior ao GC, em ambos os períodos analisados (p<0,05). Remanescentes de BC eram visíveis e estavam em íntimo contato com o tecido ósseo. Além disso, no grupo GA as observações foram semelhantes, porém foi notado fechamento total do defeito aos 28 dias e houve maior área de neoformação óssea em todos os períodos analisados, comparando-se aos demais biomateriais (p<0,05). Defeitos que foram preenchidos com CO apresentaram apenas um tecido conjuntivo delgado fechando o defeito... / (1)Objectives: To evaluate by histometric and immunohistochemical analysis, the osteoconductive potential of hydroxyapatite granules and beta-tricalcium phosphate (Bone Ceramic - Straumann ®) in the repair of critical bone defects in rat calvaria process. (2)Methods: The rats were divided into three groups (n = 8) and submitted to euthanasia at 14 days and 28 days post-surgery. In each animal a 5 mm calvaria bone defect was performed. In Blood Clot Group (CG) the defect was filled only with clot; in the Autogenous Bone Group (AG) the defect was filled with autogenous bone and in Bone Ceramic Group (BC), with alloplastic bone (Straumann ®).The bone tissue area was evaluated in the central region of bone defects. The results were submitted to ANOVA and Tukey’s test at 5% level of significance. For the evaluation of the different stages of the bone healing process, immunohistochemical analysis was performed with primary antibodies against OC, RUNX 2, TRAP and VEGF. As an additional analysis, calvaria’s 3D images were obtained in microtomography. (3)Results: The bone defect treated with BC was filled with a conglomerate of biomaterials and bone formation, which was statistically higher than CG in both periods analyzed. Remnants of BC were visible and were in intimate contact with bone tissue. Furthermore, the AG group observations were similar, but it was observed a complete defect closure after 28 days and there was a greater area of bone formation in all periods analyzed, comparing to other biomaterials (p <0.05). The defects filled with CO showed only a slender conjunctive tissue closing the bone defect. The TRAP protein was lightly immunomarked, while immunostaining of VEGF was moderate. No caso de materiais ortopédicos, estes devem apresentar resistência mecânica adequada ao seu emprego. Peças fabricadas de metais celulares de titânio é uma alternativa para redução do módulo de elasticidade, aproximando sua rigidez à do tecido ósseo, pois fornece uma combinação de baixa densidade e grande capacidade de absorção de energia. Neste trabalho foram confeccionadas estruturas celulares da liga Ti-6Al-4V, processadas por sinterização a laser direta em metais (DMLS), que é uma técnica de manufatura aditiva que reproduz protótipos virtuais 3 D camada-a-camada de forma a constituir o objeto desejado, que faz uso da energia do laser para sintetizar partículas de pó da liga Ti-6A1-4V. Para construção dos modelos virtuais utilizou-se o software SolidWorks 3D CAD Design, com um grupo de amostras com poros de geometria quadrada e um outro grupo com geometria esférica, tamanho de poros distintos de 500, 650 e 750 um e espessura de paredes celulares sempre a metade do tamanho dos poros, para obtenção de porosidades próximas a 60%. As amostras foram submetidas a tratamento térmico a 800ºC e resfriamento lento, sendo caracterizadas por microscopia eletrônica de varredura (MEV), tomografia computadorizada, dureza, densidade relativa e módulo de elasticidade (rigidez). Tanto o pó da liga quanto as amostras sinterizadas apresentam estrutura martensítica. As paredes celulares apresentam elevada rugosidade à presença de partículas do pó não sinterizadas. As imagensde MEV apresentam divergências entre tamanho de poros e espessura de parede celular. As imagens de tomografia computadorizada mostram microtrincas nas paredes celulares. Os valores de porosidade experimental diferem os valores teóricos, em média, 2% e valores de... / Abstract: The growing demand for biomaterials has required of science great advances in the development of techniques that generate better quality of life for people in need of implants. In the case celular metals of titanium are an alternative for reducing the modulus of elasticity, approaching its rigidity to the bone tissue, as it provides a combination of low density, high rigidity and high energy absorption capacity. In this work made celular made cellular structures of Ti-6Al-4V alloy, processed by direct metal laser sitering (DMLS), which is an additive manufacturing techique that reproduces layer-to-layer 3D virtual prototypes in order to form the desired object, which makes use of laser energy to sintering powder particles of Ti-6Al-4V alloy. For construction of virtual models it was used the SolidWorks 3D CAD Desing software, with a group of sampls with square geometry pores, with size of 500, 650 and 750 um, and thickness cell walls always half the size of the pores, for obtaining porosity close to 60%. The samples were submited to heat treatment at 800ºC and slow cooling, being characterized by scanning electron microscopy (SEM), computed tomography, relative density, hardness and modulus of elasticity (stiffness). Both the alloy powder and sintered samples presented high roughness due to the presence of non-sintered powder particles. The SEM images presented differences between pore size and cell wall thickness. The images of computed tomography show micro failure on the cell walls. The experimental porosity values differ from the theoretical values, on average, 27% and relative density values above 0.30 were obtained for all samples. The hardness values for all cellular metals are superior to commercial alloy. The values of modulus of elasticity (siffness) suffered significant reductions (when compared with commercial alloy), with values varying between 9 and 22 GPa.
