Biodegradace polychlorovaných bifenylů pomocí ligninolytických hub a jejich enzymů / Biodegradation of polychlorinated biphenyls by white - rot fungi and their enzymes

Polychlorinated biphenyls (PCB) represent relevant persistent organopollutants of the environment and the estimated amount of PCB released into the environment is 750000 metric tons. White-rot fungi have been studied for long time due to their degradative potential toward various aromatic pollutants and it is known that these fungi are able to decompose PCB in vivo. Biodegradation of PCB by the fungus Pleurotus ostreatus was studied in the frame of this work. A high degradative efficiency of P. ostreatus was observed in the first set of experiments, even in the presence of relative high amount of added PCB. Fungus was able to transform 780±50 µg out of the intial amount 1000 µg in 20 ml of a cultivation media within 42 days. A decrease in toxicity was recorded during the degradation that suggests the suitability of this organism for a practical use in decontamination. In vitro experiments with purified laccase induced with Cu2+ from this fungus did not prove any participation of the enzyme in the first step of PCB transformation. The enzyme did not show an ability to degrade PCB even after purification from cultivation media containing PCB. Bioremediation of polychlorinated biphenyls (PCBs)-contaminated soil by phytoremediation with Chromolaena odorata(L) R.M. King and Robinson

The ability of Chromolaena odorata propagated by stem cuttings and grown for six weeks in the greenhouse to thrive in soil containing different concentrations of PCB congeners found in Aroclor and transformer oil, and to possibly remediate such soil was studied under greenhouse conditions. Chromolaena odorata plants were transplanted into soil containing 100, 200, and 500 ppm of Aroclor and transformer oil (T/O) in 1L pots. The experiments were watered daily at 70% moisture field capacity. Parameters such as mature leaves per plant, shoot length, leaf colour as well as the root length at harvest were measured. C. odorata growth was negatively affected by T/O in terms of shoot length and leaf numbers, but no growth inhibition was shown by Aroclor. At the end of six weeks of growth, Plants size was increased by 1.4 and 0.46%, but decreased at -1.0% in T/O, while increases of 45.9, 39.4 and 40.0% were observed in Aroclor treatments. Mean total PCB recoveries were 6.40, 11.7, and 55.8μg in plants tissues at Aroclor treated samples resulting in a percentage reduction of PCB from the soil to 2.10, 1.50, and 1.10 at 100, 200, and 500mg/kg Aroclor treatments respectively. There was no PCB recovery from plants in transformer oil treatments as a result of its inhibition to growth. Root uptake was found to be the probable means of remediation of PCB-contaminated soil by C. odorata, this was perhaps aided by microbes. This study has provided evidence on the ability of C. odorata to remediate PCB contaminated soil. The effect of protozoan grazers on the cycling of polychlorinated biphenyls (PCBs) in marine systems

In marine systems, the microbial loop influences organic carbon dynamics by recycling a significant fraction of dissolved and particulate organic matter. The goal of this thesis was to understand how these recycling processes affect chlorobiphenyl (CB) cycling in marine systems by monitoring CB dynamics among organic carbon pools represented by dissolved organic matter, bacterial prey and phagotrophic protozoan grazers. Initially, I studied the extent to which a protozoan grazer (Uronema sp.-10[micro]m ciliate) equilibrated with aqueous PCBs within 2-3 hours. Initial calculations predicted rapid equilibration via passive diffusion. Experimentally, no difference in equilibration time was noted between grazing and non-grazing protozoa, indicating that diffusion was the primary uptake pathway for these organisms. The results were extended to determine the transition size of an organism where the rates of diffusive and ingested uptake are equivalent (100-500[micro]m). Disassociation rate constants were estimated for complexes of CB congeners and dissolved organic carbon (DOC). CB-DOC complexes enhanced the diffusive uptake rate constant for Tenax resin and, by inference, protozoan grazers. In the second phase of this work, concentrations of surfactants, organic carbon and cells were monitored over time in protozoan cultures. The effects of bacterial growth substrate and protozoan species were examined. Surfactants increased during protozoan exponential growth while total DOC concentrations decreased. Production of / (cont.) surface-active material in ciliate cultures was significantly higher than in flagellate cultures, and all protozoan cultures were higher than the bacterial control. Common headspace vessels were then used to compare and contrast the affinity of protozoan and bacterial culture filtrates (<0.2[micro]m) for PCBs relative to a seawater control. Affinities were normalized to bulk DOC and surfactant concentrations to determine underlying relationships among these parameters. Values of equilibrium partition coefficients (K[oc]) ranged from 10⁴·⁶ in Vineyard Sound seawater to 10⁵·⁴ and 10⁵·⁵ in protist cultures, indicating that "grazer-enhanced" DOM was a better sorbent for PCBs than DOM in bacterial controls and Vineyard Sound seawater. / by Elizabeth Belle Kujawinski. / Ph.D.

Byla provedena studie bioakumulace POP v potravním řetězci a historických trendů hladin POP v sedimentech Brněnské přehrady. Současné úrovně kontaminantů byly stanoveny v jelci tloušti z řeky Svratky a rovněž byla posouzena hygienická kvalita ryb z rekreačního rybolovu. Současné úrovně POP v terestrickém ekosystému byly posouzeny pomocí analýz jehličí jako přirozeného indikátoru znečištění. Studium jedinců tří druhů dravých ptáků z ČR bylo zaměřeno na zhodnocení kontaminace těchto vrcholných predátorů POP. S pomocí analýz vybraných druhů ryb ze Záhlinických rybníků byl rovněž studován proces bioakumulace. Posouzení parametrů dvou moderních extrakčních technik a jejich využitelnosti pro izolaci POP z různých matric bylo námětem poslední části dizertace.

La neurotoxicité développementale associée au méthylmercure, au plomb et aux biphényles polychlorés : l’attention et le traitement visuel à l’étude

L’influence des contaminants environnementaux (CE) a été largement étudié en contexte d’exposition aigüe et d’intoxication. Qu’en est-il d’une exposition chronique à plus faibles doses sur le développement cognitif et sensoriel des enfants? Les études longitudinales de la présente thèse ont été réalisées au Nunavik. Bien que géographiquement éloignée des centres industriels, la communauté inuite est exposée aux CE via leur transport par les courants atmosphériques et océaniques ainsi que par leur bioaccumulation dans le poisson et les mammifères marins. Puisque l’alimentation autochtone traditionnelle, notamment la chair de béluga, est encore fréquemment consommée par la communauté, une proportion substantielle d’enfants inuits est exposée in utero aux CE. Ceux-ci sont également continuellement exposés à ces agents neurotoxiques durant leur développement postnatal. Or, la variation considérable dans l’adoption de l’alimentation traditionnelle au sein de la communauté représente une opportunité pour étudier la relation entre les niveaux d’exposition aux CE et le développement cognitif et sensoriel des enfants. Bien que certains déficits aient déjà été mis en lien avec l’exposition chronique aux CE dans la littérature, la présente thèse s’intéressa à cette relation plus spécifiquement chez les enfants inuits vivant dans le Grand Nord et plus exposés aux CE en raison de leur alimentation. Par ailleurs, les protocoles qui ont été développés pour cette thèse permettront d’évaluer des aspects qui ont été peu étudiés en lien avec les CE, soit l’attention visuospatiale et le traitement visuel cérébral précoce. Dans le premier volet de cette thèse, la relation entre trois CE et l’attention visuospatiale a été étudiée à l’aide d’une version adaptée de la tâche de Posner (M.I. Posner et al., 1980). Cette tâche psychophysique a été administrée à des enfants inuits (âge moyen = 11.2 ans) dont les niveaux d’exposition au mercure (Hg), au plomb (Pb) et aux biphényles polychlorés (BPCs) ont été documentés durant les périodes pré et postnatale. Les expositions in utero au Pb et aux BPCs ont été significativement associées à de l’impulsivité et de l’inattention, respectivement, alors que l’exposition postnatale au Pb a été associée à des temps de réaction plus longs. Bien qu’aucune relation spécifique avec l’attention visuospatiale n’ait été trouvée, les résultats de cette étude suggèrent que l’exposition aux CE est associée à une diminution des capacités attentionnelles générales chez les enfants résidant au Nunavik. Dans le second volet, le traitement cérébral précoce de l’information visuelle a été examiné à l’aide de potentiels évoqués visuels auprès d’enfants de la même communauté (âge moyen = 10.9 ans). La concentration de Hg dans le sang de cordon ombilical mesurée à la naissance a été associée à une réduction de l’amplitude et à une augmentation de la latence de la composante N75. L’exposition prénatale au Pb a quant à elle été associée à un délai dans la latence de la composante N150. Les résultats obtenus suggèrent ainsi que l’exposition aux métaux lourds, en particulier durant la période gestationnelle, serait associée à des altérations dans le développement visuel. Les résultats présentés dans cette thèse soutiennent l’hypothèse selon laquelle le cerveau en développement est vulnérable lors d’une exposition chronique aux CE, et cela même à des niveaux de concentration inférieurs aux limites recommandées par les organismes de santé publique. Les résultats permettent également d’apporter un éclairage nouveau sur les déficits cognitifs et sensoriels associés aux CE. / The deleterious impact of environmental contaminants (EC) has been extensively studied in acute exposure and poisoning events. What about the chronic exposure to lower doses on cognitive and sensory development of children? The longitudinal studies of this thesis were conducted in Nunavik. Although geographically distant from industrial centers, the Inuit community is exposed to EC via their transport by atmospheric and oceanic currents and their bioaccumulation in fish and sea mammals. Since traditional native foods, especially beluga meat, are still frequently consumed by this community, a substantial proportion of Inuit infants are exposed in utero to EC. Inuit children are also continuously exposed to these neurotoxic agents during postnatal development. The considerable variation in consumption of traditional food within this community provides an opportunity to investigate the relation between degree of exposure to EC and the effects on cognitive and sensory development of children. Although some deficits have already been related with chronic exposure to environmental contaminants in the literature, this thesis is interested more specifically at these relations in Inuit children living up North. Furthermore, the protocols that were developed for this thesis will evaluate new aspects related to EC, which are the visuospatial attention and the early brain visual processing. The first part of this thesis was designed to assess the effect of three EC on visuospatial attention with a modified Posner paradigm (M.I. Posner, et al., 1980). This psychophysical task was administered to Inuit children (mean age = 11.2 years) for whom the levels of exposure to mercury (Hg), lead (Pb) and polychlorinated biphenyls (PCBs) were documented for pre and postnatal periods. In utero exposures to Pb and PCBs were significantly associated with greater impulsivity and inattention, respectively, while current exposure to Pb was significantly associated with longer reaction times. Although no specific effect has been found on visuospatial attention, the results of this study suggest that exposure to EC is associated with decreased in general attentional abilities in children living in Nunavik. In the second part, early brain processing of visual information was assessed using visual evoked potentials with children from the same community (mean age = 10.9 years). Cord blood Hg level measured at birth was associated with a reduction of the amplitude and an increase of the latency of the N75 component. Prenatal exposure to Pb was associated with a delay of the N150 latency. These results suggest that heavy metal exposure, in particular during the gestational period, is associated with alterations in visual development. The results presented in this thesis support the hypothesis that the developing brain is vulnerable during a chronic exposure to EC, even at concentration levels below the limits recommended by public health agencies. The results also shed new light on cognitive and sensory deficits associated with EC.

The remediation of persistent chlorinated aromatic compounds has become a priority of great relevance due to the teratogenic, carcinogenic and endocrine-disrupting properties of these xenobiotics. The use of biological methodologies for the clean-up of contaminated sites, collectively referred to as "bioremediation", has been gaining an increasing interest in recent years because it represents an effective, cost-competitive and environmentally friendly alternative to the physico-chemical and thermal treatments. In this respect, "white rot" fungi, an ecological subgroup of filamentous fungi, display features that make them excellent candidates to design an effective remediation technology ("mycoremediation"). In spite of this, fungi have not been widely exploited for their metabolic capabilities and the mechanism by which they are able to degrade the aforementioned pollutants has not been fully elucidated yet. Within this frame, the present Ph.D thesis was aimed at: i) assessing the efficiency of different mycoremediation strategies for the clean-up of a polychlorinated biphenyl (PCBs)-contaminated soil; ii) understanding the fungal degradation pathways of polychlorinated biphenyls and their major metabolites, namely chlorobenzoic acids (CBAs) and hydroxylated polychlorinated biphenyls (OH-PCBs). i)...

Evaluating organic compound sorption to several materials to assess their potential as amendments to improve in-situ capping of contaminated sediments

Contaminated sediments represent a common environmental problem because they can sequester large quantities of contaminants which can remain long after the source of pollution has been removed. From the sediment these hazardous compounds are released into the sediment porewater where it can partition into organisms in the sediment and bioaccumulate up the food web; leading to an ecological and human health concern. The objective of this work is to investigate an emerging option in contaminated sediment remediation; specifically an option for in-situ treatment known as active capping. Conventional capping uses clean sediment or sands to separate contaminated sediment from overlying water and biota. Active capping is the use of a sorptive amendment to such a cap to improve its effectiveness. This work focuses on granular materials as direct amendments to conventional caps including; granular activated carbon (GAC), iron/palladium amended GAC, alumina pillared clay, rice husk char, and organically modified clays. All materials were investigated in batch sorption tests of benzene, chlorobenzene, and naphthalene in DI water. Additionally porewaters from three sites were extruded and the concentrations of polycyclic aromatic hydrocarbons (PAHs) and polychlorinated biphenyls (PCBs) were measured. At Manistique Harbor and Ottawa River PCBs were identified as the primary contaminant of concern while PAHs were the contaminant of concern at the Grand Calumet River. At these sites a solvent extraction method was used to analyze the sediment concentrations of the contaminants of concern. From the former batch tests activated carbon and a commercially available organoclay were chosen for further investigation. This includes PAHs in batch sorption tests using extruded sediment porewater to investigate matrix effects, and PCB sorption in distilled water. / text

Delivery of hydrophobic substrates to degrading organisms in two-phase partitioning bioreactors

This thesis examined the use of two-phase partitioning bioreactors (TPPBs) for the biodegradation of poorly water-soluble compounds. TPPBs are stirred tank bioreactors composed of a biocatalyst-containing aqueous phase and an immiscible second phase containing large amounts of poorly water-soluble or toxic substrates. Degradation of the bioavailable substrate in the aqueous phase will result in equilibrium-driven partitioning of additional substrate from the immiscible phase into the aqueous phase, theoretically allowing for complete substrate degradation. Fundamental work was undertaken with the PCB-degrading organisms Burkholderia xenovorans LB400 in liquid-liquid and solid-liquid TPPBs. Initially biphenyl was used as the sole carbon source due to its hydrophobic nature and structural similarity to the environmentally relevant PCBs. The critical LogKO/W (octanol/water partitioning coefficient) of the organism was determined to be 5.5 and its growth kinetics on biphenyl were determined in a liquid-liquid TPPB. A polymer selection strategy for solid-liquid TPPBs was developed in the next chapter, and it was shown in the following chapter that biphenyl degradation in solid-liquid TPPBs was mass transfer limited, as described mathematically utilising the previously estimated microbial kinetics. The fundamental knowledge gained in the early chapters was then applied to the degradation of PCBs by the same organism. It was shown that the aqueous phase availability of PCBs is the rate-limiting step in biphasic bioreactors, and not the mass transfer rate. The low specific microbial degradation rates, resulting from substrate-limited growth were addressed with increased biomass concentrations; however, it was also found that an additional carbon source was required to maintain microbial activity over an extended period of time. Pyruvic acid was selected as a carbon source which, once added to actively PCB-degrading cells, maintained the cells’ activity towards PCBs and up to 85 % of 100 mg l-1 was degraded in 15 h. It was shown as the final contribution in this thesis that TPPBs can be combined with a PCB soil extraction step as a potential remediation scheme for PCB contaminated soil. From Mono- to Tetraphosphines – A Contribution to the Development of Improved Palladium Based Catalysts for Suzuki- Miyaura Cross Coupling Reaction

Im ersten Teil der Arbeit wird die Synthese neopentyl- und neosilylsubstituierter Phosphane zur Verwendung als Liganden in katalytisch aktiven Palladiumkomplexen beschrieben. Die Aktivität wurde in der Suzuki-Miyaura Kreuzkupplungsreaktion getestet. Während die neosilylsubstituierten Phosphane 2:1 Addukte (5b und 5d) mit geeigneten Palladiumsalzen bilden, welche moderate Katalyseaktivität zeigen, untergehen die neopentylsubstituierten Komplexe schnelle Cyclometalierungsreaktionen in Gegenwart von Basen und bilden die katalytisch wenig aktiven Palladacyclen (6a, 6e, and 6g). Die deaktivierende Cylometallierung konnte durch Darstellung der Palladiumcomplexe ausgehend von Pd(cod)Cl2 in Abwesenheit von Basen vermieden werden. Die erhaltenen 2:1 Phosphaneaddukte zeigten deutlich verbesserte Aktivität. Daraus wurde geschlossen, dass die Cyclomettalierung als Nebenreaktion eine wichtige Deaktiverungsmöglichkeit darstellt, diese Überlegung veranlasste uns Trialkylphosphane mittlerer Größe, mit Substituenten die nur schwer eine Cyclometallierungen eingehen können zu testen. Die Verwendung der Phosphoniumsalze 4h (R = Cy, R‘ = neopentyl) und 4m (R = iPr, R‘ = CH2Cy) führt zu höheren Aktivitäten in der Suzuki-Miyaura Kreuzkupplung, als bestes Katalysatorsystem hat sich die Kombination aus Pd2(dba)3 oder Pd(OAc)2 und entsprechendem Phosphoniumsalz ergeben. Im zweiten Teil dieser Arbeit werden Synthesen zu neuen biphenylbasierten Diphosphanen (70, 71, 76, and 77) vorgestellt. Die Palladiumkomplexe wurden ebenfalls auf ihre Eignung als Katalysatoren in palladiumkatalysierten Suzuki-Miyaura Kreuzkupplungen getestet und zeigen für diese Klasse von Komplexen gute Aktivität. Das Tetraphosphan 82 wurde für die Synthese des zweikernigen Palladium(II)-komplex 83 eingesetzt. Durch die Koordination des D2h-symmetrischen Tetraphosphanes an die Palladiumatome wird die Symmetrie des Moleküls erniedrigt und folglich erhält man den formal D2-symmetrischen Komplex 83. / In the first part of this thesis, the synthesis and catalytic activity of neopentyl and neosilyl substituted phosphine palladium complexes is described. The complexes have been tested in the Suzuki-Miyaura cross-coupling reaction. Whereas the neosilyl substituted phosphines form 2:1 adducts (5b and 5d) with Palladium salts which showed moderate activity, the neopentyl complexes quickly undergo cyclometallation in presence of bases to form Palladacycles (6a, 6e, and 6g) which showed only moderate catalytic activity. Cyclometallation could be avoided by the preparation starting from Pd(cod)Cl2 in the absence of bases. The obtained 2:1 phosphine adducts showed superior activity. We concluded that cyclometallation process is an important deactivation pathway, this prompted us to test trialkyl phosphine ligands with medium size but substituents not reliable to cyclometallation. We have been pleased to find that 4h (R = Cy, R‘ = neopentyl) and 4m (R = iPr, R‘ = CH2Cy) showed good activity in the Suzuki-Miyaura cross-coupling reaction. The best results have been obtained by in situ preparation of active catalyst from Pd2(dba)3 or Pd(OAc)2 and the appropriate phosphonium salt. In the second part of this thesis, the first synthesis of a new family of biphenyl based bisphosphine ligands (70, 71, 76, and 77) has been reported. Their palladium complexes were successfully tested as catalyst in the Suzuki cross-coupling reaction. Within the class of bisphosphine based palladium complexes they show good activity in Suzuki-Miyaura cross-coupling reaction. Systematically, was expanded our synthesis strategy and we were able to introduce the first synthesis of a highly symmetric 2,2',6,6'-tetraphosphinobiphenyl. Tetraphosphine 82 was used as ligand in a dinuclear palladium(II) complex 83. Upon complexation the D2h symmetric 2,2’,6,6’-tetraphosphine lead to a chiral D2 symmetric complex 83.
