Vliv intrauterinní aplikace autoplazmy na plodnost klisen / Effect of intrauterine autoplasma application on mare fertility

The objective of this study was to evaluate the effect of intrauterine plasma infusion on conceiving rate in selected group of mares during the breeding seasons 2008 and 2009. The fresh sperm insemination was followed by the infusion of self-plasma and subsequently the successivity of the insemination was compared with the control group of mares. Next, there were observed following factors which could influence the effectiveness of conceiving: age of the mares, number of insemination doses, size and location of preovulating follicle, month of the year. There was also evaluated the difference between multiparous (with one and more births) and nulliparous mares. The statistical analyses were proceeded by the use of contingency tables and the one-factorial analysis of variance (ANOVA). Next, the regression and correlation analyses were used. All the statistical procedures were performed with the use of statistical software STATGRAPHICS Plus 5.0. The results showed that there were the same results in observing of both number of insemination doses and fertile heats evaluation. There was found no significant effect of plasma infusion on the conceiving rate. In the model with the effects of plasma infusion and the age of the mare there were found statistically significant differences. It means that the effect of plasma infusion on conceiving rate was confirmed in the mares of the age up to 14 years. The result of the following analysis is the non-significant difference in conceiving rate between the groups of multiparous mares with or without plasma infusion.

Estes isômeros podem contaminar não só o meio ambiente como também a população que tenha contato direto ou indireto com os resíduos. Nas décadas de 70 e 80, foi utilizado um produto comercial composto pelo ativo HCH comercializado como \"Hexabel®\" no controle de insetos xilófagos no Acervo Histórico de Joinville em Santa Catarina. A aplicação do inseticida foi realizada pelos próprios funcionários do Arquivo Histórico, e houve intensa manipulação dos documentos tratados durante uma mudança do prédio o presente trabalho teve como objetivo validar uma metodologia de determinação de resíduos do HCH e seus isômeros em soro sangüíneo, e analisar o grau de exposição dos funcionários e ex-funcionários do Arquivo Histórico de Joinville ao produto. Foram realizados exames sorológicos em todos os funcionários e ex - funcionários do local e também na população que sabidamente nunca tiveram contato com o ativo, denominada população controle. As análises foram realizadas após a validação do método multiresíduos que obtém todos os ativos em uma única extração e tem detecção por Cromatografia a gás com detector de captura (GC-ECD) de elétrons. Os resultados comprovaram comprovaram a eficiência do método, através de dados em conformidade com os critérios do ensaio. No. of bitstreams: 0 / O presente trabalho tem como objetivo avaliar se o complexo Európio- Clorotetraciclina (EuCTc) ou o corante Tioflavina T (ThT) podem atuar como biossensores precisos e eficientes de colesterol numa fração específica, através de procedimentos simples. Para isso estudaram-se as propriedades ópticas do complexo EuCTc e ThT na presença da Lipoproteína de Baixa Densidade (LDL) em seu estado nativo e em seu estado oxidado. O primeiro estudo realizado verificou a melhor razão molar entre o Európio e a Clorotetraciclina, em seguida verificou-se a influência da diálise da LDL na emissão do complexo EuCTc, para obtenção de um protocolo para quantificação da concentração da LDL. Foram traçadas as curvas de calibração da emissão do complexo EuCTc com várias concentrações da LDL nativa, LDL oxidada com íons de Cobre e LDL oxidada com íons de Ferro. Em seguida obteve-se o tempo de vida do íon Európio no complexo EuCTc na presença de diferentes concentrações de LDL nativa e LDL oxidada por íons de Cobre. Na segunda parte do trabalho estudou-se a emissão do corante Tioflavina T na presença da LDL nativa e LDL oxidada. Na terceira etapa as propriedades ópticas dos biossensores EuCTc e ThT foram investigadas na presença do plasma sanguíneo e foram comparadas às emissões do complexo EuCTc e o corante ThT com a LDL ultracentrifugada, para verificar a possibilidade de quantificar a LDL diretamente no plasma sanguíneo. Inicialmente foram criados cirurgicamente defeitos de retração Classe I de Miller nos caninos superiores de seis cães de raça indefinida. Após um mês de cronificação, os defeitos bilaterais semelhantes foram aleatoriamente designados a receber os seguintes tratamentos: Lado 1: enxerto de tecido conjuntivo subepitelial associado ao uso de PRP; Lado 2: enxerto de tecido conjuntivo subepitelial. Decorridos 45 dias do tratamento os animais foram sacrificados e foram obtidas as peças para análise histológica dos seguintes parâmetros histométricos: novo cemento formado com fibras inseridas, novo osso, extensão do epitélio sulcular e juncional, área de adaptação conjuntiva e extensão do defeito. Observou-se uma maior extensão linear de novo cemento estatisticamente significante (p⿤ 0,05) nos dentes tratados com o PRP (2,18 ± 0,78 mm) quando comparados aos dentes do lado controle (1,19 ± 0,62 mm). Todos os outros parâmetros não tiveram diferenças estatísticas. As médias obtidas nos lados teste e controle, respectivamente, foram: extensão de epitélio sulcular e juncional, 2,04 ± 0,57mm e 2,49 ± 0,82mm; adaptação conjuntiva 0,29 ± 0,28mm e 0,23 ± 0,18mm; novo osso â¿¿ 0,57 ± 0,95mm e â¿¿ 0,46 ± 1,34mm, e extensão do defeito 4,13 ± 0,83mm e 4,47 ± 0,58mm. Considerando os limites deste estudo, pode-se concluir que a associação do PRP ao enxerto de tecido conjuntivo, no tratamento de defeitos de retração, promoveu maior neoformação cementária quando comparado ao tratamento controle / Abstract: The aim of this study was to histometrically evaluate the healing process of gingival recessions treated by PRP in combination with subepithelial connective tissue graft (SCTG) and to compare it to that obtained with SCTG alone (Control). Six mongrel dogs were used in the experiment. Gingival recessions (5x7mm) were surgically created and exposed to plaque accumulation for 1 month. Contralateral defects were then randomly assigned to test group or control. Dogs were sacrificed 45 days after healing, and the blocks containing the experimental specimens were processed for histological analysis. The histometric parameters evaluated were: length of sulcular and junctional epithelium, connective tissue adaptation, new cementum, new bone and defect extension. A superior length of new cementum, statistically significant, was observed in sites treated with PRP (2.18 ± 0.78mm) in comparison with the control (1.19 ± 0.62mm). No statistical differences in any other parameters evaluated were detected. The extension of the sulcular and junctional epithelium was 2.04 ± 0.57 mm for the PRP group and 2.49 ± 0.82mm for the control. The new connective tissue adjacent to the root without cementum formation was 0.29 ± 0.28 mm and 0.23 ± 0.18 mm for the PRP group and control, respectively. Bone formation was â¿¿ 0.57 ± 0.95 mm for the PRP group and â¿¿ 0.46 ± 1.34mm for the control. These isomers may contaminate both the environment and the people who had direct or indirect contact with the HCH residues. On 70\'s and 80\'s, its was used a commercial product which contained HCH on its formula and was commercialized as Hexabel®. The product was used to control the xylophage insect population on the Historical Archieve of Joinville, at Santa Catarina, Brazil. The own employees did the insecticide application and there was an intensive manipulation of the treated material during a building change. The objective of this work was to validate a methodology to quantify the HCH residues and its isomers on blood plasma and analyze the employee\'s exposure extent to the chemical. It was done serological investigation on employees, former-employees and on non-exposed people, which was classified as the control population. The analyses were done after the methodology validation. The method was capable to obtain all isomers on a unique extraction and the detection and quantification were done by gas chromatography with electron-capture detector (GC-ECD). The results obtained showed that the proposed method is accurate, and that the amount of HCH residues on blood plasma of all individuals analyzed was lower than the quantification limits established to the method, which were 0,04 μg dL-1 for the alpha- and gamma- HCH (lindana) and 0,08 μg dL-1 for the beta- and delta- HCH.

Sur un procédé hautes pressions de sécurisation du plasma sanguin humain / High pressure processing for assuring the safety of human blood plasma

La mise en œuvre des hautes pressions permet d'inactiver des micro-organismes contaminants (levures, bactéries, spores, virus…) au sein de divers milieux. Cette technologie a tout d'abord été utilisée puis développée à l'échelle industrielle pour décontaminer des produits alimentaires. Du fait de la faible énergie générée par les hautes pressions, seules les liaisons faibles et correspondant à une valeur de ?V négative sont altérées, ce qui, en outre, ouvrait la voie à de nombreuses applications biologiques. L'objectif de cette thèse était d'inactiver l'ensemble des micro-organismes pathogènes au sein d'un produit sanguin : le plasma. L’enjeu principal était d'une part de sauvegarder la fonctionnalité des principales protéines contenues dans le plasma (facteurs de coagulation notamment) et d'autre part d'inactiver les micro-organismes contaminants. Afin d’atteindre cet équilibre, plusieurs paramètres ont été utilisés : - la valeur de la pression (de 100 à 300 MPa), - la durée du traitement, - les températures négatives… Plusieurs résultats originaux seront présentés, notamment en ce qui concerne la différence de sensibilité aux hautes pressions des protéines d'une part et l'inactivation de micro-organismes d'autre part. / High hydrostatic pressure (HHP) has shown its capacity to inactivate contaminating micro-organisms (yeasts, bacteria, spores, viruses…) present in various media. This technology has been first used and developed to an industrial scale for the food industry. Because of the low energy raised using high pressure, only weak interactions characterized by a negative ?V will be altered. This statement opens the way to numerous applications in the biology field. The objective of this PhD work was to inactivate all the pathogens that may contaminate blood products, and blood plasma in particular. The main challenge was to inactivate the contaminating micro-organisms while maintaining the activity of the plasma proteins (such as blood coagulation factors). To achieve this balance, several parameters were used: - the pressure value (from 100 to 300 MPa), - the treatment duration, - negative temperatures… Some original results will be presented, concerning differences in the high pressure sensitivity of proteins and the inactivation of pathogens.

The effect of a balanced amino acid infusion on hepatic regeneration in rats

The efficacy of a balanced amino acid infusion on recovery from temporary hepatic insufficiency and its effect on plasma ammonia levels in partially hepatectomized rats were studied. Polyethylene implant buttons were developed to secure animals to the infusion apparatus. Rats under went jugular vein cannulation, and 70% partial hepatectomy. Test solutions were continuously infused (30 ml/d). Plasma samples were taken through the jugular vein cannula and analyzed for bilirubin concentration, ammonia concentration, LDH activity, and GPT activity. LDH and bilirubin results were inconclusive. Results from GPT activity assays suggest that both Freamine III and the balanced formula provided injured hepatocytes with better nutrition than saline infusions and ad libitum food consumption. Ammonia assay results suggest that the balanced formula was more effective than Freamine in maintaining plasma ammonia levels in the normal range after partial hepatectomy.

A MULTI-DIMENSIONAL METHOD FOR THE ANALYSIS OF HUMAN BLOOD PLASMA METABOLOME

<p>The comprehensive analysis of human blood plasma metabolome has been completed using derivatization gas chromatography-mass spectrometry (GC-MS) analysis, liquid chromatography-mass spectrometry (LC-MS) analysis and a comprehensive LC-GC-MS analysis approach wherein LC fractions were collected, derivatized and analyzed using GC-MS. In all cases blood plasma samples were deproteinized using solvent precipitation prior to chromatography and MS analysis.</p> <p>In GC-MS analyses, the progress of all derivatization reactions was monitored by adding 9-anthracenemethanol and 1,3-diphenylacetone to all reaction mixtures; their conversions to 9-anthracenemethanol trimethylsilyl ether and the oxime derivative of 1,3-diphenylacetone were used as measures of the completion of these derivatization reactions. Any reactions with completions less than 99% were repeated.</p> <p>GC-MS analysis of blood plasma samples detected 100 peaks; 44 were positively identified by comparing retention indices and mass spectra with those of authentic standards. LC-MS analyses were conducted on a HILIC column (aminopropyl phase) with MS detection in both negative ion and positive ion modes and resulted in the identification of 97 peaks; 47 were observed in the positive ion mode, 58 in the negative ion mode with 8 peaks observed in both modes.</p> <p>The multi-dimensional LC-GC approach was not designed as a routine analytical method; rather the purpose of this approach was to see how many compounds could be observed in the sample and to obtain better quality mass spectra and retention index values. The LC separation afforded 16 fractions which upon derivatization GC-MS analysis gave an additional 176 peaks from a total of 276 peaks. The MS data from these additional spectra can be used to develop selected ion monitoring GC-MS or tandem mass spectrometry analytical methods. This thesis has demonstrated the power of off-line comprehensive methods to identify compounds that neither the GC nor the LC methods detected.</p> / Master of Science (MSc)
