Altered bone metabolism in the protein deprived rat

In contrast to the vast amount of active research and available knowledge on many aspects of protein-calorie malnutrition, surprisingly little attention has been focused on the skeletal effects of this syndrome. As early as the 1920's it was realised that the skeletal metabolism in protein-calorie malnutrition was deranged. Notwithstanding this, there is still to date inadequate understanding of the mechanisms by which these skeletal changes are brought about. This is partly due to the fact that the research workers were "victims of their times" in that the techniques utilised gave limited information and it has only been in the past decade or two that more sophisticated techniques have become available. (Evidence for this will be borne out in the following chapter where the literature is reviewed). It was with this in mind that the work presented in this thesis was undertaken. Part I of the thesis consists of a review of the literature, followed by a dissertation on the normal physiological concepts of calcium metabolism as well as a description of the calcium pools used in kinetic models. Part II includes all the chapters on the various experiments performed. Included in each chapter are the methods used, results obtained as well as a discourse relevant to that particular study, with special reference to similar experiments performed by other authors. Part III encompasses the integrated summary of the results and a discussion theorising the mechanisms involved in those changes. Finally, an appendix which deals with the statistical methods used for analyses, as well as details of those techniques which were used, but were established by other authors.

The condition was apparently known even in Hunter's day and most of its characteristic features were described before the end of the nineteenth century. Nevertheless, the evolution of these curious lesions, their unusual distribution and the associated abnormalities of growth are still incompletely defined and largely unexplained.

Sever's disease, or Calcaneal Apophysitis, is a common condition affecting children. It has been examined at a review level at best since first being described in 1907. The aim of this study was to compare the characteristics of a group of children with Sever's disease with a non-symptomatic group of children of similar age. This prospective study had two main objectives. Firstly, to examine issues which had been anecdotally raised in the literature over the years; secondly, to determine the quality of life effects on children with Sever's disease. / A literature review of Sever's disease articles was undertaken to identify what was known, and what was surmised about the disease. The literature was also reviewed to allow selection of the correct tools to determine if there were any differences between the two groups of children. Issues such as reliability and validity as well as suitability of questionnaires were examined and justified from the literature. / Several areas arose which the literature did not adequately cover, including the reliability of foot measures, validity of a new measure - the Foot Posture Index, and a reliable and valid way to measure the dorsiflexion of the foot on the leg. Experiment 1 established that the intra rater reliability of the measurements to be used in Experiment 4 was good, but that the inter rater reliability was poor to moderate. Experiment 2 investigated the criterion validity of four components of the foot Posture Index, a new measurement that was to be used in Experiment 4, concluding that gross movements could be shown, but not subtle foot position changes. Experiment 3 developed equipment for the measurement of ankle joint dorsiflexion, another parameter to be measured in Experiment 4, and established that the intra and inter rater reliability of this apparatus was excellent. / Experiment 4 involved the gathering of data relating to Body Mass Index, activity, foot posture, traditional foot measures, ankle joint dorsiflexion and quality of life from the two aforementioned groups. Binary logistic regression was used to determine the odds ratios of various factors for the Sever's disease and non-Sever's disease group. The only significant differences between the groups were in quality of life scales and in forefoot to rearfoot determination. Bone Marrow and Bone as a Source for Postmortem RNA

No / The susceptibility of RNA to enzymatic degradation has been considered as a tool to estimate time-since-death in forensic samples, and it has previously been demonstrated that the choice of tissue is an important factor. In this study we have extracted RNA from decaying bone and bone marrow under the hypothesis that the delayed onset of putrefaction may render them a useful source in this context. In a preliminary study, total RNA was extracted from bone and bone marrow that had been sampled from six skeletally mature rabbits at time points between zero and 31 days after death. The levels of three specific RNA transcripts could be quantified using real-time polymerase chain reaction. Bioanalyzer results show rRNA bands in bone marrow samples up to 21 days postmortem. We hereby propose bone marrow as a potential source for postmortem RNA in forensic studies.
