A idade das fêmeas colonizadoras de Hypothenemus hampei (Ferrari) durante o abandonamento do hospedeiro, assim como sua resposta aos voláteis de flores de café, Coffea arabica L. var. Tupi, foram investigadas neste trabalho. Um dispositivo experimental que simulasse as condições de dentro do fruto de café e permitisse a observação da saída dos insetos foi utilizado para a determinação da idade das fêmeas colonizadoras. Em média, os besouros apresentaram 15 dias de idade no momento em que abandonaram o dispositivo experimental. Nesta idade as fêmeas estavam acasaladas, possuiam o tegumento totalmente melanizado, foram capazes de voar e produzir ovos viáveis. Os voláteis de flores de café, foram coletados por aeração, analisados por GC-EAD e GC-MS, e testados em bioensaios olfatométricos. As fêmeas colonizadoras de H. hampei foram atraídas pelos voláteis de flores de café. Dos 50 compostos encontrados nos extratos naturais, sete foram eletrofisiologicamente ativos aos insetos. Destes sete, somente metil salicilato, neral e geranial puderam ser identificados, e a mistura de seus padrões sintéticos foi a mais atrativa nos bioensaios olfatométricos. Tomados juntos, estes resultados trazem novas informações sobre a bioecologia e ecologia química da broca-do-café, as quais poderão ser usadas futuramente em pesquisas de base e naquelas focadas no desenvolvimento de novas estratégias de manejo para H. hampei em cafezais. / The age of Hypothenemus hampei (Ferrari) females during the host abandonment, as well as their response to coffee flower volatiles were determinated under laboratory conditions. A experimental device that simulated the conditions inside a coffee berry, and that allowed the observation the insects coming out was used to determinate the age of colonizing females. On average, the beetles were 15 d-old at the moment they abandoned the experimental device. In this age, the females were mated, had a fully melanized tegument, were able to fly and to produce viable eggs. The volatiles of coffee, Coffea arabica L. var. Tupi, flowers were colected by aeration, analyzed in GC-EAD and GC-MS, and tested in olfactometer bioassays. The colonizing H. hampei females were attracted by the coffee flower volatiles. Of 50 compounds found in the natural extracts, seven were electrophysiologically actives to the insects. Of these seven, only methyl salicylate, neral and geranial could be identified, and the blend with their syntetic standards was the most atractive in the olfactometer bioassays. Taken together, these results bring new information on the bioecology and chemical ecology of the coffee berry borer, which may be used in the future by basic researchs or by those focused on the development of new strategies for the management of H. hampei in coffee farms.

Caracterização patogênica e molecular de isolados de Beauveria sp e Matarhizium sp de diferentes regiões do Brasil para o contole de Diatraea saccharalis Fabricius (Leídoptera: Crambidae) /

O controle biológico com Cotesia flavipes, em função da expansão do plantio, não será suficiente para o controle em áreas de alta infestação. O clima quente e úmido das regiões Noroeste de São Paulo e Centro-Oeste potencializam a utilização dos fungos Beauveria bassiana e Metarhizium anisopliae para o controle desta praga. O presente trabalho teve como objetivo estudar as variações dentre as populações de B. bassiana e M. anisopliae provenientes de diferentes regiões do Brasil e hospedeiros associadas ao controle da Diatraea saccharalis e identificar microrganismos com potencial de uso como agentes para o controle desta praga. Neste sentido, foram realizados isolamentos de fungos de amostras de solos de diferentes culturas e regiões do Brasil e hospedeiros. Destas amostras, após análises morfológicas, foram obtidos 55 isolados de B. bassiana e 14 isolados de M. anisopliae. Isolados provenientes de Institutos de Pesquisa e Empresas privadas foram fornecidos para serem utilizados como referencias, totalizando 73 isolados de B. bassiana e 68 isolados de M. anisopliae. Os isolados foram inicialmente caracterizados quanto ao potencial de patogenicidade, sendo avaliados quanto a capacidade de infectividade sobre todas as fases de desenvolvimento da D. saccharalis. Os resultados do presente estudo demonstraram que ovos, lagartas, crisálidas e adultos de D. saccharalis são suscetíveis à infecção por diversos isolados de M. anisopliae e B. bassiana. Foi possível ainda caracterizar a patogenidade de diversos isolados de M. anisopliae e B. bassiana, comprovando que estes entomopatogênicos apresentam potencial de controle sobre todas as fases de desenvolvimento deste inseto-praga. Biological control with Cotesia flavipes, due to the expansion of the plantation will not be enough to control in areas of high infestation. The hot and humid climate of Northwest region of São Paulo and the Midwest potentiate the use of Beauveria bassiana and Metarhizium anisopliae to control this pest. The present work aimed to study the variations among populations of B. bassiana and M. anisopliae from different regions of Brazil and hosts associated with the control of Diatraea saccharalis and identify microorganisms with potential use as agents to control this pest. In this sense, fungal isolations were made of soil samples from different cultures and regions of Brazil and hosts. From these samples, after morphological analysis, we obtained 55 isolates of B. bassiana and 14 isolates of M. anisopliae. Isolates from research institutes and private companies were provided for use as references, totaling 73 isolates of B. bassiana and 68 isolates of M. anisopliae. The isolates were initially characterized for their pathogenic potential and they were evaluated for their ability to infectivity on all phases of development of D. saccharalis. The results of this study show that eggs, larvae, pupae and adults of D. saccharalis are susceptible to infection by several isolates of M. anisopliae and B. bassiana. It was also possible to characterize the pathogenicity of different isolates of M. anisopliae and B. bassiana, confirming that these entomopathogenics have potential control over all stages of development of this pest insect. Este trabalho teve como objetivo avaliar o efeito da umidade em quatro tipos de solos sobre a emergência de adultos e duração da fase pupal das moscas-das-frutas, Ceratitis capitata (Wiedemann, 1824) e Anastrepha fraterculus (Wiedemann, 1830) e do bicho-furão dos citros, Gymnandrosoma aurantianum Lima, 1927 e na emergência do parasitóide de moscas-das-frutas, Diachasmimorpha longicaudata (Ashmed, 1905), utilizando-se diferentes potenciais mátricos de água no solo e com uma metodologia própria e desenvolvida no presente trabalho. As profundidades de pupação de G. aurantianum e A. fraterculus foram estudadas em tratamento seco e úmido (5%), com a finalidade de se determinar a profundidade adequada para cada espécie utilizada nos experimentos de avaliação da influência da umidade em quatro tipos de solo. G. aurantianum pupou na mesma profundidade em substrato úmido ou seco. A pupação de A. fraterculus é mais superficial em condições secas, com 100% de pupação entre 0 e 1,5 cm. Para C. capitata, a profundidade utilizada no experimento de influência da umidade e do tipo de solo foi a de 3,0 cm. A duração da fase pupal desta espécie foi influenciada diferentemente para machos e fêmeas. Para fêmeas, apenas o tipo de solo influenciou a duração da fase pupal, que foi maior no solo Argiloso. Nos machos, a duração da fase de pupa foi influenciada pela umidade e pelo tipo de solo, isoladamente, ocorrendo redução da fase pupal de machos de C. capitata no solo Muito Argiloso e Argilo arenoso, com maior duração no solo Argiloso. À medida que o potencial mátrico diminuiu, a duração da fase pupal de machos de C. capitata aumentou, independentemente do tipo de solo. A emergência de C. capitata foi influenciada pela umidade, independentemente do tipo de solo, sendo maior em solos mais secos. Para A. fraterculus, a interação da umidade e do tipo do solo influenciou a duração da fase pupal para machos e fêmeas de forma semelhante. As menores durações da fase pupal, para machos e fêmeas de A. fraterculus, foram proporcionadas nos solos com maiores teores de areia (Franco arenoso e Argilo arenoso) e no solo com alto teor de argila (Muito argiloso), neste caso, na maior umidade do solo. Porém, verificou-se, para fêmeas de A. fraterculus, redução da fase pupal à medida que a umidade dos solos Franco arenoso e Argilo arenoso diminuiu; para machos, esta redução foi observada em todas as umidades estudadas nestes solos. A emergência de A. fraterculus foi influenciada pela interação da umidade e do tipo do solo, sendo maior nos solos que apresentaram maior retenção de água nos diferentes potenciais mátricos aplicados aos solos Argilo arenoso e Muito argiloso. A emergência de D. longicaudata foi influenciada pelos fatores tipo de solo e umidade, isoladamente, sendo o número de adultos emergidos de D. longicaudata maior no solo Franco arenoso e menor no Muito argiloso; sempre, o número de adultos emergidos foi maior em altas umidades. A duração da fase de pupa de machos e fêmeas de G. aurantianum foi influenciada pela interação da umidade e do tipo de solo, porém de forma irregular e que não permitiu a construção de um modelo lógico que explique a relação entre maiores ou menores durações com teores de umidade e tipos de solo. A emergência de G. aurantianum foi influenciada pela interação umidade e tipo de solo, sendo maior em solos com maiores teores de areia. O presente trabalho poderá elucidar como alguns fatores do solo afetam o desenvolvimento destas pragas e do inimigo natural, auxiliando na adoção de novas estratégias de manejo, além de fornecer subsídios para a amostragem e para o entendimento da ocorrência destas pragas em ambientes agrícolas. / The goal of this work was to evaluate moisture effect on four soil types in the emergence of adults and duration of the pupal stage of fruit flies, Ceratitis capitata (Wiedemann, 1824) and Anastrepha fraterculus (Wiedemann, 1830) and citrus fruit borer, Gymnandrosoma aurantianum Lima, 1927, and in the emergence of the fruit fly parasitoid, Diachasmimorpha longicaudata (Ashmed, 1905), using different soil water matric potential and a particular methodology, developed in this work. The pupation depths of G. aurantianum and A. fraterculus were studied through a dry and moist (5%) treatment, in order to determine the adequate depth for each species for further use in the investigation of the influence of moisture on four soil types. G. aurantianum pupated at the same depth either under moist or dry medium. A. fraterculus pupation was more superficial under dry conditions, with 100% pupation between 0 and 1.5 cm. For C. capitata, one used a 3.0 cm depth to verify the influence of moisture and soil type. The pupal stage duration of the species was affected differently for males and females. For females, only the soil type influenced the pupal stage duration, which was higher under Clay soil (42,8% clay). In males, the pupal stage duration was influenced by moisture and soil type separately, and the pupal stage of C. capitata was low in Clay (68% clay) and Sandy Clay soil, with higher duration in Clay soil (42,8% clay). As the matric potential decreased, the duration of the pupal stage of C. capitata males increased, regardless of the soil type. The emergence of C. capitata was influenced by the moisture, regardless of the soil type, and was higher in drier soils. For A. fraterculus, the moisture-soil type interaction influenced the pupal stage duration for both males and females similarly. The lower pupal stage durations for A. fraterculus males and females were provided by higher sand content soils (Sandy Loam and Sandy Clay soils) and in high clay content soil (Clay soil, 68% clay), in the higher soil moisture. For A. fraterculus females, however, the pupal stage decreased as the moisture of Sandy Loam and Sandy Clay soils decreased; for the males, such reduction was observed in all moisture degrees studied on these soils. The emergence of A. fraterculus was influenced by the moisture-soil type interaction, higher in higher water retention soils at different matric potentials applied to Sandy Clay and Clay (68% clay) soils. The emergence of D. longicaudata was influenced by the soil type and moisture factors separately, and the number of adults emerging from D. longicaudata was higher in Sandy Loam and lower in Clay (68% clay) soil; the number of emerged adults was always higher under high moisture. The duration of the pupal stage of G. aurantianum males and females was influenced by the moisturesoil type interaction; however, it was irregular and failed to allow the construction of a logical model to explain the relationship between higher or lower durations with moisture contents and soil types. The emergence of G. aurantianum was influenced by the moisture-soil type interaction, higher at higher sand content soils. This work may clarify how some soil factors affect the development of these pests and relating natural enemies, a path to new management strategies, providing subsidies for the sampling and understanding of the occurrence of these pests in agricultural environments.

Hemocytes of the fifth instar European borer, Ostrinia nubilalis Hubner (Lepidoptera: Pyralidae) were examined in stained and unstained preparations. Prohemocytes, plasmatocytes, granular hemocyter, and oenocytoids, and spherule cells were found in this stage, The prohemocytes are characterized by a scant intensely basophilic cytoplasm. The highly pleomorphic plasiratocytes are characterized by a punctate nucleus and production of cytoplasmic extensions in vitro. Granular hemocytes are characterized by a small nucleus, accumulation of lipid droplets, and the production of extremely fine cytoplasmic extensions in living preparations. The oenocytoid is characterized by a small eccentric nucleus in a large amount of homogeneous basophilic cytoplasm. The spherule cell is characterized by large spherules within the cytoplasm which may tend to mask the nucleus.Treatment of larvae with per os doses of Bacillus thuringiensis did not affect the differential hemocyte count. Temperatures Experienced by Emerald Ash Borer and Other Wood-boring Beetles in the Under-bark Microclimate

Most studies of under-bark microclimate have been restricted to observations of a few coniferous trees in wooded southern latitudes. This limitation is worrying because of emerging wood-boring pests that specialize on deciduous trees in Canada, such as emerald ash borer (Agrilus planipennis). Using a large data set that includes 60 ash trees spread across both urban and woodlot sites in 6 different Ontario locations, I found that the under-bark microclimate of deciduous trees can provide wood-boring beetles with an environment in which temperatures which differ from air temperature. On average, daily minimum under-bark temperatures are significantly warmer than air temperatures in the winter months. At temperatures low enough to cause substantial cold-temperature mortality to emerald ash borer, the difference between under-bark and air temperature can be large. In addition, I observed that the difference between daily minimum under-bark and air temperature can vary, and consequently that assumptions of a constant level of difference between the two are not valid. In the spring season, I found that daily under-bark temperature maxima on the south side of the tree are significantly warmer than air temperature maxima. This difference lead to faster predicted development times for beetles in the southern under-bark microclimate of urban trees as compared to predictions based on air temperature, suggesting that city trees may impact overall population dynamics. While it is clear that under-bark temperatures differ from air temperatures, and are important to predicting possible range and population growth of wood-boring insects, large scale measurements of microclimate conditions are not feasible. I tested the ability of a simple Newtonian cooling model to predict under-bark temperature extremes using weather station data. While the model did not predict daily under-bark temperature maxima accurately, predictions of minima were quite accurate (1.31˚C average root mean squared error), especially when compared to the errors from assuming under-bark temperature is the same as air temperature (3.20˚C average root mean squared error). I recommend use of the Newtonian cooling model to predict under-bark temperature minima of deciduous and coniferous trees.

Tri-trophic Analyses of Rice, the Sugarcane Borer, and Putative Biological Control Agents

A three-year field experiment was conducted to evaluate the tolerance and compensatory response of rice (Oryza sativa L.) to injury caused by the sugarcane borer, Diatraea saccharalis (F.). Two mechanisms of within-plant tolerance/compensation were observed. Stem injured plants produced ca. 0.69 more tillers than uninjured plants, while tillers with leaf and leaf sheath injury produced larger panicles, up to 39.5% and 21.0% heavier than uninjured tillers, when injury occurred at 3rd tiller stage and at panicle differentiation, respectively. A 2-year field cage experiment was conducted to determine the biological control potential of Cotesia flavipes (Cameron) against the sugarcane borer on rice. The effective search rate was 49 cm2 ground area (2.2 tillers) parasitoid-1 day-1. A cohortbased age-structured model was developed to simulate the population dynamics and economic value of the sugarcane borer and C. flavipes in rice, as affected by overwintering larval density, timing and rate of parasitoid aerial release, and year-to-year climate (temperature and rainfall). The results suggest C. flavipes was most effective when released during the 1st sugarcane borer generation. The maximum simulated economic benefit ($112.05 ha-1) was ca. 7.8% of that provided by insecticide-based control. The inability of C. flavipes to provide economic control in temperate-subtropical areas is due to its high rearing cost, a low effective search rate, a low maximum rate of parasitism per female, and both spatial and temporal asynchrony of parasitoid emergence with the larval hosts. The biocontrol capability of Trichogramma galloi Zucchi was also simulated. The maximum economic benefit provided by T. galloi was $1128.75 ha-1, ca. 79.0% of that provided by insecticide-based control. Theoretical analyses were conducted to estimate the effectiveness of augmentative releases, using data from previous studies of parasitism of lepidopteran pests by hymenopteran parasitoids with host and parasitoid density as factors. The maximum daily parasitism per female was highest for parasitoids that attack exposed larvae, followed by parasitoids that attack eggs, semi-exposed larvae, and concealed larvae. Simulation analyses were conducted to estimate the population dynamics and economic value of D. saccharalis, herein used as a model host, and each of 5 parasitoid categories (solitary parasitoids that attack eggs, exposed larvae, and semi-exposed larvae, gregarious parasitoids that attack eggs and concealed larvae) in rice, as affected by overwintering larval density, timing of parasitoid release, and year-to-year climate. Among the 13,500 simulations that were conducted, 480 (3.6%) provided a greater economic value than insecticide-based control. All 480 simulations were obtained using solitary parasitoids that attack exposed or semi-exposed larvae. Solitary egg parasitoids provided an average of 42.2% of the economic value provided by insecticide-based control when released 30 days after planting. Gregarious parasitoids that attack eggs or concealed larvae provided almost no positive economic benefit. For parasitoid species that do not overwinter successfully in areas where they are released or that exhibit temporal or spatial asynchrony with their host early in the spring, our results suggest augmentative biological control is only effective for solitary parasitoids that attack either exposed or semi-exposed larvae, with current rearing, shipping and release costs.

A moagem atualmente está em torno de 640 milhões de toneladas, numa área de aproximadamente 10 milhões de hectares por todo o Brasil. A produção cada vez maior nos canaviais, em parte, é devido ao melhoramento genético, resultando em cultivares capazes de se desenvolverem em áreas de expansão, proporcionando ganhos importantes para diferentes regiões. Mas apesar da diversidade de cultivares existentes no mercado, dos diferentes programas de melhoramento, são escassas algumas informações, quanto à suscetibilidade/tolerância de pragas, predisposição ao florescimento, variáveis tecnológicas, entre outros, nos diversos cultivares atualmente utilizadas no Brasil, ressaltando a importância do estudo do comportamento dos diferentes cultivares. Para o sucesso na utilização de plantas geneticamente modificadas visando o controle deste inseto, faz-se necessário conhecer sua dispersão. Estudos da dinâmica de vôo também podem ajudar na compreensão do comportamento do inseto, e de suas interações ecológicas. O estudo da dispersão de D. saccharalis foi possível através da marcação, liberação e captura dos machos adultos. A marcação foi realizada por meio da incorporação de corante lipossolúvel à dieta das larvas. As armadilhas de feromônio foram dispostas nos talhões de cana-de-açúcar nos sentidos norte, sul, leste e oeste, circundando o ponto de liberação. Os resultados obtidos mostram que mais de 45 % dos machos marcados capturados permaneceram nas áreas mais próximas ao ponto de liberação. Fatores climáticos como a temperatura, umidade relativa, velocidade do vento e precipitação não interferem decisivamente na captura dos machos liberados e dos machos do campo. A direção do vento exerce um papel fundamental na dispersão de D. saccharalis, uma vez que a maioria dos insetos voou a favor do vento. O modelo mais adequado para explicar a relação entre a distância de vôo e o número de indivíduos capturados é dado por y = a2/(1+(2a1,8bx))2,6. Com isso, para o manejo de resistência de D. saccharalis em cultivos de cana-de-açúcar geneticamente modificada expressando proteínas de Bacillus thuringiensis Berliner, deve se manter no máximo 800 m de distância entre as áreas de refúgio / Abstract: The sugarcane borer, Diatraea saccharalis (Fabr.), is one of the most important sugarcane pests in Brazil. For a successful utilization of genetically modified crops aiming at the control of this insect, it is necessary to know its dispersal. Studies of flight dynamics can also help understand the insect's behavior and its ecological interactions. The study of D. saccharalis dispersal was possible through labeling, releasing and capturing adult males. Labeling was performed by incorporating oil soluble dyes into the larval diet. The pheromone traps were placed in a sugarcane field following the cardinal directions (North, South, East, West) around the release site. The results showed that more than 45 % of the labeled males were captured closer to the release site. Climatic factors such as temperature, relative humidity, wind speed and rain do not affect decisively in the release or wild males capture. The wind direction is important in D. saccharalis dispersal, once the majority of insects flew towards the wind. The most adequate model to explain the relationship between flight distance and number of captured insects is y = a2/(1+(2a1,8bx))2,6. A moagem atualmente está em torno de 640 milhões de toneladas, numa área de aproximadamente 10 milhões de hectares por todo o Brasil. A produção cada vez maior nos canaviais, em parte, é devido ao melhoramento genético, resultando em cultivares capazes de se desenvolverem em áreas de expansão, proporcionando ganhos importantes para diferentes regiões. Mas apesar da diversidade de cultivares existentes no mercado, dos diferentes programas de melhoramento, são escassas algumas informações, quanto à suscetibilidade/tolerância de pragas, predisposição ao florescimento, variáveis tecnológicas, entre outros, nos diversos cultivares atualmente utilizadas no Brasil, ressaltando a importância do estudo do comportamento dos diferentes cultivares.
