光儲存媒體經營策略之個案研究

林昺宏 Unknown Date (has links)

Regulation of S6K1 Protein Kinae Activation by its C-Terminal Autoinhibitory Domain

Ragan, Timothy James 20 June 2008 (has links)
Signal transduction kinases lie at the heart of the cell's ability to respond to environmental cues. These kinases are typically controlled by post-translational modification, most commonly by phosphorylation. S6K1alphaII is a member of the AGC subfamily of serine-threonine protein kinases, whereby catalytic activation requires phosphorylation of critical residues in the conserved T-loop (T229) and hydrophobic motif (T389) regions of its catalytic kinase domain. In addition to its kinase domain, S6K1 contains a C-terminal autoinhibitory domain (AID, residues 399-502), which inhibits T-loop and hydrophobic motif phosphorylation. Autoinhibition is relieved upon multi-site Ser-Thr phosphorylation of the AID by MAP kinase(s). We developed an optimized PCR-based gene synthesis method, which I utilized to build expression constructs for the AID alone as well as the kinase domain and full length S6K1alphaII. A fully activated form of S6K1alphaII was purified from Sf9 cells by co-expression with PDK1, and was used for in vitro analysis of the signaling pathway. AID was successfully purified in a soluble form from E. coli despite the fact that PONDR analysis predicted a highly disordered structure. Aberrant mobilities in both SDS-PAGE and size-exclusion chromatography, as well as low chemical shift dispersion in 1H-15N HSQC spectra and far UV CD data showing a lack of secondary structure, confirmed that purified recombinant AID is largely unfolded. Despite this, addition of purified AID effectively inhibited PDK1-catalyzed T-loop phosphorylation of a catalytic kinase domain construct of S6K1 and inhibition was decreased when the tetraphospho-mimic mutant AID(D2ED) was used. These studies, along with the reagents produced by them, will allow for further exploration of the emerging field of disordered regulatory domains.

Studies on the 10KD heat shock protein expression regulation and clerodane diterpenoid cytotoxicity in colorectal cancer

Chen, Wei-cheng 19 August 2010 (has links)
A new compound, clerodane diterpenoid, CD, was isolated from the bark of Polyalthia longifolia var. pendula. . In previously study, CD was proved to induce cell apoptosis. We analysis differentially expressed proteins of colorectal cancer cells under drug CD treatment by 2-D electrophoresis and find drug response proteins, e.g. Hsp10, Profilin-1, Peroxindoxin-1. The expression change of protein had been further confirmed by RT-PCR and western blotting. It is interesting to reveal the role of these proteins in the colorectal cancinogenesis and anti-tumor drug response. In most kind of cancers, Hsp10 is often overexpress as indicated in many reports. Under CD treatment, Hsp10 is down-regulation in our experiment. The major aim of this study is to investigate the effect of Hsp10 on colorectal cancer cell lines SW480, and also investigate the relation of Hsp10 to drug treatment. Here,we overexpress Hsp10 in SW480. MTT assay, and flow cytometric analysis are utilized to investigate the effect of Hsp10 and CD cytotoxicity when overrxpress Hsp10 in SW480.

Vliv rozšíření internetu na pokles prodejů hudebních nosičů CD / Effect of the Spread of the Internet Use on the Sales of Audio CDs

Trš, Lukáš January 2010 (has links)
This thesis is concerned with the effect of widespread internet use on the sales of audio CDs. It analyses trends in the American and Czech markets from the end of the 1990's to today. Its analysis is based on the relationship between musical genres and their respective target audiences with the hypothesis being that genres with listeners who have adopted the internet more rapidly are more affected than other genres. The thesis proves that the decrease in music sales in the last ten years, the bulk of which have been CDs, has been caused by the graudual establishment of the internet with its probable illegal downloading of recordings and the change in format to MP3 from CD.

Effect of MP3 Use on CD Consumption Behavior

Deng, Yan-Ru 23 June 2000 (has links)
Today, more and more Websites provide digital music with MP3 formats on the Internet. MP3 is a format that can compress the digital files. Without this format the music program would be overlarge, and the small transmission capacity of the World Wide Web would be insufficient to support such a device. With this technological improvement, the consumers will be benefited from the conveniency of MP3, while the records companies would be threatened by the challenge confronted with the music industry. On the one hand, MP3 may reduce the sales of CDs in the market. On the other hand, it may help bring about various advantages for the record companies in terms of decreasing the marketing cost. This thesis aims to discuss the effect of the use of MP3 on CD consumption behavior by surveying the users of the MP3 Websites. The research is based on a well-designed questionnaire on the Internet. Through the promotion in thirteen days, it is received 1532 reply of the questionnaire, and there are 1293 entities considered to be valid samples for the study. The result shows that the correlation between the MP3 use and the CD consumption behavior is positive. That is to say, the users who spend longer time in MP3 Websites will buy more CDs. The more the use frequency of MP3 Websites is, the more the frequency of CD consumption. By comparing those who bought CDs with those who did not during the past twelve months, we find that the former spend longer time in MP3 Websites. To analyze the consumer's recognition about the influence of MP3 use on CD consumption behavior, our study shows that the consumers think the MP3 Websites use will not effect the CD consumption behavior. It further demonstrates an increasing tendency that the more CDs the consumers purchase, the less influence they think of the MP3 use. Therefore, according to the research, the conclusion of this thesis stands to argue that the use of MP3 Websites will not reduce the CD sales, but rather the records companies may take great advantages of it by promoting the CD market through the performance of MP3 in the Internet.

Removable media software engineering

Zabava, Bruce A. 22 February 2011 (has links)
This report focuses on software solutions that are fully contained on CD, DVD, or USB storage devices. When the removable media is selected as the boot device on a computer system, it will launch its own operating system and then execute the actions it was written to perform. Organizations often have many types of removable media solutions that are engineered to perform various tasks; however, this report will focus on only two types of removable media projects that are often needed by original equipment manufacturers (OEMs): the end user reinstallation media (EURM) and the field service reinstallation media (FSRM). In this document we explain the purpose of EURM and FSRM solutions, the engineering methods used to create them, what engineering measurements can be used to achieve project goals, what to consider when implementing process improvements, and finally how to analyze the results of said process improvements. / text
