Cancer and microenvironment : the functional interplay between intra- and extracellular nucleotide metabolisms / Cancer et microenvironnement : dialogue fonctionnel entre les métabolismes nucléotidiques intracellulaire et extracellulaire

Les nucléotides jouent un rôle majeur dans une pléiade de processus biologiques comme la composition des acides nucléiques, la signalisation, ou la régulation de la balance énergétique. Les nucléotides extracellulaires exercent également des fonctions biologiques. Par conséquent, des dérégulations des pools de nucléotides impactent l’homéostasie de multiples façons, par exemple en promouvant l’instabilité génétique ou un environnement immunosuppresseur. Or, ces paramètres font partie des « Hallmarks du Cancer » décrits par Hanahan et Weinberg. Ces observations confirment l’éventualité d’un rôle clé des nucléotides dans le cadre du cancer.cN-II et CD73 sont des 5’-nucléotidases impliquées respectivement dans les métabolismes nucléotidiques intra- et extracellulaire. Elles sont de nouvelles cibles thérapeutiques en oncologie. Cependant, leurs rôles dans la biologie de la cellule cancéreuse, ou le possible impact de leur utilisation en tant que cible thérapeutique sur le comportement des cellules tumorales sont peu connus. Considérant l’implication de ces enzymes dans les métabolismes nucléotidiques, nous avons enquêté sur les modifications de l’agressivité de la cellule cancéreuse ou sur sa capacité à interagir avec son microenvironnement, dans le cas d’une invalidation ou une diminution d’expression de cN-II et/ou CD73. cN-II semble donc impliquée dans l’adaptabilité métabolique et la combinaison des invalidations de cN-II et CD73 est associée à une modification d’expression d’enzymes du métabolisme du glucose. CD73 peut aussi moduler l’expression de gènes de la migration cellulaire. cN-II est impliquée dans la migration cellulaire, via l’axe COX-2/PGE2, et dans la sensibilité à des agents modulant ce paramètre. Ces caractéristiques sont plus marquées en association avec une invalidation de CD73. Ici, cN-II et CD73 ne semblent pas jouer de rôle dans la prolifération ou le dialogue avec une sous-population de cellule de l’immunité innée / Nucleotides play a major role in nucleic acids constitution and are involved in various cell phenomena. Indeed, intracellular ATP, GTP, AMP, GMP and their cyclic forms are components of cell signaling and define the energetic balance. Extracellularly, they also play multiple roles. Thus, when nucleotide pools are deregulated various processes are impacted. For example, a low availability of nucleotides supports genetic instability and aberrant levels of extracellular adenosine can lead to an immunosuppressive microenvironment. Interestingly, the cited parameters are among the Cancer Hallmarks described by Hanahan and Weinberg. These observations confirm the possibility of a key role of these molecules in this pathology. cN-II and CD73 are 5’-nucleotidases, involved in intra- and extracellular nucleotide metabolism respectively and have been identified as possible targets for new anti-cancer therapies. Nevertheless, very little is known about their biological roles on cancer cells and what parameters of cell biology could be impacted by such strategies. Considering the involvement of these purines in cell metabolism, we wondered what changes a decrease in cN-II and/orCD73 expressions or their silencing could trigger in cancer cells as well as in the interplay with their microenvironment.We studied cancer cell aggressiveness and the interplay with innate immune cells under cN-II and CD73 modulations. We observed that cN-II is involved in metabolic adaptability. The association of cN-II and CD73 invalidations results in glucose-metabolism-related gene modifications. CD73 can regulate migration-related genes expression but does not affect the process. cN-II is also involved in cell migration, via the COX-2/PGE2 axis. Again, these characteristics are accentuated when associated with CD73 deficiency. Here, cN-II and CD73 do not seem to be involved in cancer cell proliferation or in their interplay with a subset of innate immune cells

The goal of this thesis is to evaluate erosion processes and runoff conditions in a part of catchment Zubčický potok containing the land arrangement Věžovatá Pláně. I drew up the area description for this catchment in terms of geomorfological, geological, pedological, climatic and phytogeographical aspect. For the evaluation of erosion processes in the solved area was used the universal soil loss equation by Wischmeier and Smith and in the microcatchment was used the CN method. Further, flow conditions was evaluated using Čerkašin equation. The result of this thesis is evaluation of erosion and runoff conditions, concept of erosion control measures and evaluation their impact on erosion and runoff conditions in the microcatchment.

The diploma thesis focuses on the analysis of rainfall-runoff processes and erosion characteristics of selected catchment in different landscape management. This analysis was solved at the Lukavecký brook catchment, which is located in the Pelhřimov district in Vysočina region. The theoretical part of the thesis describes the rainfall-runoff process and then focuses more on individual processes. It also characterizes water erosion, its consequences, forms and erosion control. At the end of the theoretical part are described methods of calculation of water runoff and water erosion. The practical part describes the catchment of the Lukavecký brook, its characteristics of natural conditions and the threat of water erosion. Then there is a procedure for processing of bases and data in the program ArcGIS. Subsequently, it includes an assessment of the different measures and changes in the type of land to drain water from the catchment and erosion threat.

First estimate the water demand in climatological experimental catchment representative of the Savannah. Defined by the difference in the occurrence of potential evapotranspiration and precipitation minimal likely to occur on a scale ten days. In the estimation of potential evapotranspiration compared to the empirical equations of the methods Blaney-Criddle, Hargreaves, ASCE Penman-Monteith, Penman, Priestley-Taylor method with the standard Penman-Monteith FAO 56. The method presented Blaney-Criddley is recommended when there is limited availability of climatological data. To estimate the probability distribution used the gamma distribution for the time series of precipitation and evapotranspiration and precipitation time series with zero values, we used the incomplete gamma function. The adhesion of the estimated probabilities to the observed data was verified by testing, non-parametric Kolmogorov-Smirnov test, with significance level (α-0, 05) which fitted well the model distributions. The occurrence of precipitation and evapotranspiration decendial likely P (X ≥ x) for different probability levels showed drought periods of ten days between 4:32, with critical values 25-39 mm, on a scale ten days, the months from May to September. In a second stage it was considered the the reservoir of soil water management and proposes a model allowing to calculate the irrigation water demand of crops of beans and corn planting dates in two different in drought and rainy. The bowl was hydrology by the method of the turn number (CN). Simulated the water demand and availability required in ascertaining what is the risk of meeting the irrigated area. The proposed model was effective for water management in the basin throughout the year. It was found that corn and beans require different amounts of water throughout the year, and that optimizing the additional water demand is a management tool able to minimize conflicts over water use. hydrology of the basin presents risks of not having met the demand required in the dry period is necessary to establish infraestrurura water for water storage / O estudo da simulação e da gestão da disponibilidade hídrica da bacia foi abordado de duas formas. Erosion is an issue, which causes huge damages not only on agricultural land. This diploma thesis describes practical evaluation of erosion control measures (usage of leaving crop residue, circuit strips, contour tillage), which are include in Good Agriculture and Environmental Condition standart, which was issued by European union. Efficiency of usage of narrow-row and wide-row crops is also modeled. Each measures were evaluated in two different modeling programmes HydroCad for calibration and ERCN for results comparison. Seven different scripts are used in the thesis. Subcatchment area P6 in catchment area of Kopaninský creek is used as model area. Literature review describes the precipitation and runoff process, issue of erosion, and agriculture land protection against water erosion. Is also describes GAEC standards (today DZES).

Analýza faktorů ovlivňujících přímý odtok z povodí / Analysis of factors affecting the direct runoff

This diploma thesis deals with the analysis of the factors which influence the direct drainage from the drainage area. The thesis specifically deals with the drainage area of the Kopaninský brook. Drainage is always influenced by several factors at the same time. These factors are above all climate, anthropogenic impact, underdrainage, shape and extent of a drainage area, land use, geologic structure, location and altitude of a drainage area. The direct drainage can be separated by different methods (modificated semi-graphical method, GROUND method, method of hydrogram separation, method of digitals filtres) or CN- curves can be used for calculation. This thesis used the method of CN curves to process the results of a given drainage area. In the methodical part of the thesis I reached to the creation of direct drainage maps with the utilisation of the BPEJ maps together with the input data gained by the method of CN curves. These outputs were created at different values of design rains repetition time N= 2, 10, 50 and 100 years. The volume of a direct drainage was calculated by a classic method and by a method using the ArcMap programme. Another part of the thesis was simulation of different area utilisation situations (real condition versus grassing of ploughland) and saturation of drainage area, or more precisely water content in soil (dry soil, middle dry soil, wet soil).

The linear chains were characterized by resonance Raman, thermal analysis, ion bombardment and using high resolution and scanning transmission electron microscopy. The HRTEM images obtained here shown the chain within the innermost MWCNT and in this work we obtain the first image of ``cross-section$"$ described for this system Cn@MWCNT. A Raman experiment was carried out in high pressure conditions (from 0 to 9.54 GPa) using a paraffin oil as pressure transmitting medium.The G band frequency of nanotube increase as pressure increase while the C-C stretching mode of linear carbon chain decreases. Theoretical results based on atomistic simulations allowed an understanding of vibrational behavior of the chains. Furthermore, we conducted works that involved instrumentation in order to set in the Department of Physics of the Federal University of Ceara, the instrumentation needed for the development of a device that enables to perform Raman spectroscopy and AFM measurements. For this purpose is necessary to integrate a scan-head, a controller system that controls the probe scan-head and an optical system that communicates with an APD (avalanche photodiode device), responsible for carrying the images with a spectrometer dedicated for obtaining Raman spectra. The system is functional as regards the ability to perform confocal measurements and ready for setting the near field measurements. / Apresentamos um estudo de cadeias lineares de carbono encapsuladas em nanotubos de carbono de paredes múltiplas (Cn@MWCNT) utilizando microscopia eletrônica e espectroscopia Raman em condições ambientes e extremas. As cadeias lineares foram caracterizadas por espectroscopia Raman ressonante, análise térmica, bombardeamento de íons e com microscopia eletrônica de transmissão. As imagens de alta resolução aqui obtidas mostraram a presença das cadeias dentro do tubo mais interno do MWCNT e neste trabalho obtivemos a primeira imagem de "seção transversal" descrita na literatura para um sistema deste tipo. Um experimento Raman foi conduzido em condições de altas pressões (0 - 9,54 GPa), utilizando um óleo de parafina como meio transmissor de pressão. A frequência da banda G dos nanotubos aumentou com o aumento da pressão enquanto o modo de estiramento C-C das cadeias de carbono lineares diminuiu. Resultados teóricos baseados em simulações atomísticas permitiram uma compreensão do comportamento vibracional da cadeia. Além disso, realizamos trabalhos que envolveram instrumentação afim de instalar no Departamento de Física da Universidade Federal do Ceará toda a instrumentação necessária para o desenvolvimento de um equipamento que permite realizar espectroscopia Raman e medidas de microscopia de força atômica (AFM). Para isto, é necessário integrar uma "scan-head", um sistema controlador que controla a sonda da "scan-head" e um sistema óptico que se comunica com uma APD (dispositivo fotodiodo avalanche) responsável pela aquisição das imagens, com um espectrômetro dedicado à obtenção dos espectros Raman. O sistema ficou funcional no que diz respeito a capacidade de executar medidas confocal e está pronto para a realização de medidas de campo próximo.

Um experimento Raman foi conduzido em condições de altas pressões (0 - 9,54 GPa), utilizando um óleo de parafina como meio transmissor de pressão. A frequência da banda G dos nanotubos aumentou com o aumento da pressão enquanto o modo de estiramento C-C das cadeias de carbono lineares diminuiu. Resultados teóricos baseados em simulações atomísticas permitiram uma compreensão do comportamento vibracional da cadeia. Além disso, realizamos trabalhos que envolveram instrumentação afim de instalar no Departamento de Física da Universidade Federal do Ceará toda a instrumentação necessária para o desenvolvimento de um equipamento que permite realizar espectroscopia Raman e medidas de microscopia de força atômica (AFM). Para isto, é necessário integrar uma "scan-head", um sistema controlador que controla a sonda da "scan-head" e um sistema óptico que se comunica com uma APD (dispositivo fotodiodo avalanche) responsável pela aquisição das imagens, com um espectrômetro dedicado à obtenção dos espectros Raman. O sistema ficou funcional no que diz respeito a capacidade de executar medidas confocal e está pronto para a realização de medidas de campo próximo.
