Os indivíduos alocados em um dos tanques eram 8 alimentados com uma mistura de alta proteína (MAP; δ13C = -22,62‰), os alocados em 9 um segundo tanque eram alimentados com uma mistura de baixa proteína (MBP; δ13C = 10 -14,34‰) e os animais dos outros tanques tinham acesso livre a ambas, quatro vezes ao 11 dia. Foram realizadas coletas de músculo, fígado e sangue a cada cinco dias por 86 dias, 12 exceto para o grupo alimentado com MBP (alimentado por 120 dias). As amostras 13 foram analisadas em espectrômetro de massa e as proporções consumidas das misturas 14 foram estimadas a partir do enriquecimento isotópico do carbono (δ13C). O 15 balanceamento realizado indicou redução no consumo relativo de proteína. Contudo, 16 esta relação não apresentou padrão definido em relação ao peso dos indivíduos, ou seja: 17 o balanceamento de proteína e energia para tilápia do Nilo está relacionado 18 principalmente à idade dos animais e não é claro se este processo está relacionado 19 também ao seu peso. As estimativas de consumo de um grupo animais desta espécie por 20 meio dos isótopos estáveis de carbono são semelhantes aos obtidos por outras técnicas, 21 portanto este método permite observação da diversidade deste comportamento no 22 autobalanceamento entre os indivíduos que compõe tal grupo. / This study aimed to assess the Nile tilapia ability to balance its own diet, 6 when two ingredient mixes were offered, using carbon stable isotopes. The physical and geochemical characteristics of diamonds from the Artemisia Kimberlite (Northern Slave Craton, Nunavut, Canada) and the micro-/macro-diamond relationship

This thesis presents the results of a study on diamonds from the Artemisia kimberlite, Nunavut, Canada. This study integrates the isotopic, chemical and physical characteristics of the diamonds in an attempt to provide insight into the nature of the micro-/macro-diamond genetic relationship, and its possible implication pertaining to the use of size frequency distributions in modern kimberlite exploration programs. Despite geochemical commonalities of the Artemisia micro- and macro-diamonds, distinct signatures are observed, particularly in regards to enriched and depleted carbon isotopic compositions, the abundance of nitrogen and hydrogen impurities and different resorption histories. This implies that, despite an observed lognormal size frequency distribution for Artemisia diamonds, micro- and macro-diamonds do not represent a single population at this locality.

Regulation of stomatal development by environmental conditions and physiological processes in the leaf

Stomatal development and its regulation by environmental conditions (light, CO2 concentration) and physiological processes in the leaf of higher plants were investigated. The study was based on the assumptions that stomatal development should be regulated by signals coming from both external environment and leaf interior, and that the signal should be transduced from cotyledons to leaves. Transgenerational effect in stomatal development was also studied. Molecular and physiological approaches were applied to reveal the relationship between leaf environment, stomatal development, stomatal function and leaf physiology.

La composition isotopique (¹³C/¹²C) du CO₂ respiré par les feuilles et les racines dans l'obscurité et celle des substrats respiratoires putatifs, y compris les sucres solubles et les acides organiques (malate et citrate), l'activité de la PEPc ainsi que les échanges gazeux des feuilles ont été déterminés sur le haricot (Phaseolus vulgaris L.) et l’épinards (Spinacia oleracea L.) cultivés dans du sable avec différents rapports NH₄⁺: NO₃⁻ dans N fourni. Le CO₂ respiré par les feuilles était enrichi en ¹³C sous NO₃⁻ s’appauvrissant progressivement avec l’augmentation de la fraction de NH₄⁺ dans le N fourni, tandis que la composition isotopique du CO₂ issu de la respiration des racines restait inchangée quel que soit le rapport NH₄⁺: NO₃⁻. Nous avons suggéré qu'une plus grande quantité de pools enrichis en ¹³C fixés par la PEPc via la voie anaplérotique contribuait à la respiration foliaire sous NO₃⁻. Cependant, un effet similaire dans les racines attendu sous NH₄⁺ a été masqué en raison d'une refixation (par la PEPc) du CO₂ respiré (appauvrie en ¹³C). De manière inattendue, les modifications de la composition isotopique du C des métabolites individuels, leurs quantités, ainsi que l'activité de l’enzyme PEPc, présentaient des profils différents entre les deux espèces étudiées. Des expériences de double marquage (¹³C et ¹⁵N) sont nécessaires pour mieux comprendre l’impact de la plasticité métabolique du TCA sur l’écart isotopique entre le malate et le citrate et sur la composition isotopique du CO₂ respiré chez différentes espèces sous différentes formes d’azote. / C-isotope composition of leaf- and root-respired CO₂ in the dark and that of putative respiratory substrates including soluble sugars and organic acids (malate and citrate), PEPc activity, as well as leaf gas exchanges were determined on bean (Phaseolus vulgaris L.) and spinach (Spinacia oleracea L.) plants grown in sand with varying ratios of NH₄⁺: NO₃⁻ in supplied N. Leaf-respired CO₂ was ¹³C enriched under NO₃⁻ nutrition and became progressively ¹³C depleted with increasing amount of NH₄⁺ in supplied N, while C-isotope composition of root-respired CO₂ remained unchanged across N-type gradient. We suggested that a higher amount of ¹³C enriched C-pools fixed by PEPc through anaplerotic pathway contributed to respired CO₂ in leaves under NO₃⁻ nutrition. However, a similar effect in roots expected under NH4+ nutrition was masked because of a rather ¹³C depleted C source (respired CO₂) refixation by PEPc. Unexpectedly, the changes in C-isotope composition of individual metabolites and their amounts as well as PEPc activity exhibited different patterns between the two species. Double labelling experiments (¹³C and ¹⁵N) are needed for better understanding the impact of metabolic plasticity of TCA on isotopic gap between malate and citrate and on C-isotope composition of respired CO₂ in different species under varying N-type nutrition.

Tracking climate signals in tropical trees : new insights from Indonesian stable isotope records

The tropical warm pool waters surrounding Indonesia are one of the equatorial heat and moisture sources that are considered as a driving force of the global climate system. The climate in Indonesia is dominated by the equatorial monsoon system, and has been linked to El Niño-Southern Oscillation (ENSO) events, which often result in severe droughts or floods over Indonesia with profound societal and economic impacts on the populations living in the world's fourth most populated country. The latest IPCC report states that ENSO will remain the dominant mode in the tropical Pacific with global effects in the 21st century and ENSO-related precipitation extremes will intensify. However, no common agreement exists among climate simulation models for projected change in ENSO and the Australian-Indonesian Monsoon. Exploring high-resolution palaeoclimate archives, like tree rings or varved lake sediments, provide insights into the natural climate variability of the past, and thus helps improving and validating simulations of future climate changes. Centennial tree-ring stable isotope records | Within this doctoral thesis the main goal was to explore the potential of tropical tree rings to record climate signals and to use them as palaeoclimate proxies. In detail, stable carbon (δ13C) and oxygen (δ18O) isotopes were extracted from teak trees in order to establish the first well-replicated centennial (AD 1900-2007) stable isotope records for Java, Indonesia. Furthermore, different climatic variables were tested whether they show significant correlation with tree-ring proxies (ring-width, δ13C, δ18O). Moreover, highly resolved intra-annual oxygen isotope data were established to assess the transfer of the seasonal precipitation signal into the tree rings. Finally, the established oxygen isotope record was used to reveal possible correlations with ENSO events. Methodological achievements | A second goal of this thesis was to assess the applicability of novel techniques which facilitate and optimize high-resolution and high-throughput stable isotope analysis of tree rings. Two different UV-laser-based microscopic dissection systems were evaluated as a novel sampling tool for high-resolution stable isotope analysis. Furthermore, an improved procedure of tree-ring dissection from thin cellulose laths for stable isotope analysis was designed. The most important findings of this thesis are: I) The herein presented novel sampling techniques improve stable isotope analyses for tree-ring studies in terms of precision, efficiency and quality. The UV-laser-based microdissection serve as a valuable tool for sampling plant tissue at ultrahigh-resolution and for unprecedented precision. II) A guideline for a modified method of cellulose extraction from wholewood cross-sections and subsequent tree-ring dissection was established. The novel technique optimizes the stable isotope analysis process in two ways: faster and high-throughput cellulose extraction and precise tree-ring separation at annual to high-resolution scale. III) The centennial tree-ring stable isotope records reveal significant correlation with regional precipitation. High-resolution stable oxygen values, furthermore, allow distinguishing between dry and rainy season rainfall. IV) The δ18O record reveals significant correlation with different ENSO flavors and demonstrates the importance of considering ENSO flavors when interpreting palaeoclimatic data in the tropics. The findings of my dissertation show that seasonally resolved δ18O records from Indonesian teak trees are a valuable proxy for multi-centennial reconstructions of regional precipitation variability (monsoon signals) and large-scale ocean-atmosphere phenomena (ENSO) for the Indo-Pacific region. Furthermore, the novel methodological achievements offer many unexplored avenues for multidisciplinary research in high-resolution palaeoclimatology. / Die tropischen Gewässer um Indonesien sind eine der äquatorialen Wärme- und Feuchtigkeitsquellen, die als treibende Kraft des globalen Klimasystems betrachtet werden können. Das Klima in Indonesien ist geprägt durch das Australisch-Indonesische Monsunsystem. Weiterhin besteht eine Verknüpfung mit El Niño-Southern Oszillation (ENSO) Ereignissen, die oft zu schweren Dürren oder Überschwemmungen in der Region mit tiefgreifenden gesellschaftlichen und wirtschaftlichen Folgen führen. Der neueste IPCC-Bericht legt dar, dass ENSO auch in den nächsten 100 Jahren das vorherrschende Klimaphänomen im tropischen Pazifik bleiben wird. Ferner wird davon ausgegangen, dass sich die ENSO-bezogenen Niederschlagsextrema intensivieren werden. Wenig Übereinstimmung herrscht jedoch bislang zwischen den Klimasimulationsmodellen in Bezug auf die voraussichtlichen Veränderungen von ENSO und dem Australisch-Indonesischen Monsunsystem. Hochaufgelöste Paläoklima-Archive, wie z.B. Jahrringe oder warvierte Seesedimente, geben Auskunft über die natürliche Klimavariabilität der Vergangenheit und können somit dazu beitragen, die Computersimulationen der künftigen Klimaentwicklung zu verbessern und zu validieren. Hundertjährige stabile Jahrring-Isotopenchronologien | Das Hauptziel dieser Doktorarbeit war es, dass Potenzial von tropischen Jahrringen zur Aufzeichnung von Klimasignalen herauszustellen und deren Evaluierung als Paläoklimaproxys. Im Detail wurden stabile Kohlenstoff- (δ13C) und Sauerstoff- (δ18O) Isotopenverhältnisse in Teakbäumen analysiert, und die ersten gut replizierten hundertjährigen (AD 1900-2007) stabilen Isotopenchronologien aus Java (Indonesien) erstellt. Dabei wurden verschiedene klimatische Einflussgrößen getestet, ob diese signifikante Korrelationen mit den Jahrringparametern aufzeigen. Weiterhin wurden hochaufgelöste intra-annuelle Sauerstoffisotopenzeitreihen erstellt, um den Transfer des saisonalen Niederschlagssignals in den jeweiligen Jahrring zu bemessen. Die ermittelte Sauerstoff-Isotopenchronologie wurde anschließend auf mögliche ENSO Signale hin untersucht. Methodische Errungenschaften | Ein zweites Ziel dieser Arbeit war es neue Verfahren zur Analyse stabiler Isotope in Baumjahrringen zu entwickeln und zu optimieren. Zwei verschiedene UV-Lasermikrodissektions-Systeme wurden getestet als neues präzises Präparationswerkzeug für stabile Isotopenstudien. Darüber hinaus wurde eine verbesserte Methode für die Probenaufbereitung stabiler Isotopenmessungen anhand von Zellulose-Dünnschnitten entwickelt. Die wichtigsten Ergebnisse dieser Doktorarbeit sind: I) Die hier vorgestellten neuartigen Techniken zu Probenvorbereitung verbessern die Analyse stabiler Isotope für Jahrringstudien in Hinsicht auf Präzision, Effizienz und Qualität. Es wurde gezeigt, dass die UV-Lasermikrodissektion eine wertvolle Technik ist, um die Beprobung von Pflanzengewebe in höchster Auflösung und beispielloser Präzision durchzuführen. II) Es ist gelungen, einen Leitfaden für ein modifiziertes Verfahren der Zelluloseextraktion an Gesamtholz-Dünnschnitten und der anschließenden Jahrringaufbereitung zu erstellen. Diese neuartige Methode optimiert die Analyse stabiler Isotopenzeitreihen in zweierlei Hinsicht: schnellere und effiziente Zelluloseextraktion und präzise Trennung der Jahrringsequenzen in inter-annueller bis intra-annuelle Auflösung. III) Die hundertjährigen stabilen Jahrring-Isotopenchronologien weisen signifikante Korrelationen mit dem regionalen Niederschlag auf. In den hochaufgelösten stabilen Sauerstoffisotopenwerten spiegelt sich deutlich das Niederschlagssignal der Trocken- und der Regenzeit wieder. IV) Die stabile Sauerstoffisotopenzeitreihe zeigt signifikante Korrelationen mit verschiedenen ENSO Phasen. Dies betont, dass die verschiedenen ENSO Phasen bei der Interpretation von tropischen Paläodaten zu berücksichtigen sind. Die Ergebnisse der Dissertation zeigen, dass saisonal aufgelöste stabile Sauerstoffisotopenchronologien von indonesischen Teakbäumen ein geeigneter Proxy für mehrhundertjährige Rekonstruktionen der regionalen Niederschlagsvariabilität (Monsun-Signale) und großräumiger Ozean-Atmosphären-Systeme (ENSO) für den Indopazifik ist. Darüber hinaus bieten die neuartigen methodischen Errungenschaften viele neue Ansätze für multidisziplinäre hochaufgelöste Studien in der paläoklimatologischen Forschung.

Quellen von DOC im hydrologischen Einzugsgebiet der Wilzsch (Westerzgebirge) – eine Multitraceranalyse

Im Fokus der Untersuchungen steht das hydrologische Einzugsgebiet der oberen Wilzsch im Westerzgebirge. Veränderte Stoffausträge des bewaldeten und zum Teil vermoorten hydrologischen Einzugsgebietes führen zu erhöhten Stoffkonzentrationen im Hauptzufluss der Trinkwassertalsperre Weiterswiese (Carlsfeld). Seitens der Trinkwasseraufbereitung stellen insbesondere die gestiegenen Gehalte an gelösten organischen Kohlenstoffen (DOC) Mehraufwendungen dar. Im Zuge der Wiedervernässung der Moorflächen kommt es zu relevanten bodenchemischen Veränderungen, die im Abfluss messbar sind. Eine räumliche Betrachtung der Teileinzugsgebiete des hydrologischen Einzugsgebietes der oberen Wilzsch ermöglicht, das Abflussgebiet in verschiedene Herkunftsräume zu gliedern. Dieser raumbezogene Ansatz leistet einen wesentlichen Beitrag in der Herkunftsanalytik von Wässern, deren Fließwege durch Standorte unterschiedlicher Nutzung geprägt sind. Die Arbeit bedient sich hydrochemischen Analysen sowie multitraceranalytischen Verfahren. Die Analyse der stabilen Umweltisotope ²H und 18O am Wasser, 13C am DIC und DOC, 34S und 18O am Sulfat sowie 15N und 18O am Nitrat hilft bei der Identifizierung von Stoffquellen und Transformationsprozessen. Die Arbeit verfolgt das Ziel, den Beitrag der verschiedenen Stoffsysteme am gesamten Stoffaustrag des Vorfluters aufzuzeigen sowie die atmosphärische Grundlast an den Messstandorten nachzuweisen. Dazu wurden von Mai 2009 bis August 2010 fünf Beprobungskampagnen angelegt. Das Messdesign erstreckte sich auf 19 Standorte in ausgewählten Teileinzugsgebieten, die maßgeblichen Einfluss auf die Vorflut haben. Parallel dazu erfolgte die Untersuchung des Niederschlags an der Station Talsperre Weiterswiese. Erfasst wurden die Inhalte von Wässern verschiedener Moorgebiete, Moorwasserpegel unterschiedlich degradierter Moorbereiche, Gräben und Bodendrainagen im Fichtenforst, der Vorfluter Wilzsch und Kranizsch und des Grundwassers im Fichtenforst. Es kann festgehalten werden, dass je nach Lage bzw. Entfernung der Ursprungsgebiete der Anteil des DOC - Exports unterschiedlich stark ist. So sind die DOC - Austräge saisonal sehr verschieden. Im Ergebnis ist der hydrochemische Beitrag - im Besonderen DOC - der unterschiedlichen Stoffsysteme des hydrologischen Einzugsgebietes letztlich am finalen Pegel der Wilzsch messbar. Die vom Basisabfluss gesteuerten silikatischen Standorte im Fichtenforst leisten einen wesentlichen Anteil zum hydrochemischen Milieu der Vorfluter. Die im Untersuchungsgebiet eingeleiteten Renaturierungsmaßnahmen beeinflussen die Trinkwasserqualität in der Talsperre Carlsfeld nicht. Eine Abgrenzung der atmosphärischen Grundlast ist ebenfalls möglich, atmogen eingetragene Stoffe können an den Standorten ausgewiesen werden.

Niterói, RJ / O sistema de circulação oceânica desempenha um importante papel na regulação e manutenção do clima no planeta. Uma vez que o Atlântico Equatorial representa o ponto de conexão entre o Atlântico Sul e Norte, estudos paleoceanográficos nesta área podem contribuir para o entendimento dos processos e fatores envolvidos nas variações climáticas ao longo do tempo geológico, principalmente durante o Quaternário. Dentro desse contexto, o presente estudo visou descrever elementos representativos da paleoprodutividade do Atlântico Equatorial Oeste no Pleistoceno tardio, por meio da análise de dados sedimentológicos e geoquímicos do testemunho sedimentar MD09-3243CQ coletado no talude da margem continental do nordeste do Brasil. A partir desse testemunho, foi recuperado o período entre 33.700 e 10.150 anos cal A.P., o qual foi dividido em seis fases principais, de acordo com a análise de agrupamento das frações granulométricas representativas do testemunho. As fases apresentaram características distintas em função de mudanças das condições paleoambientais, paleoceanográficas e da paleoprodutividade. O cenário do último glacial (Fase V), caracterizado pela alta produtividade carbonática e baixa produtividade orgânica (COT = 0,40 ± 0,12%; NT = 0,05 ± 0,01%; derivados de clorofila = 0,06 ± 0,04 SPDU; teor de carbonato = 51,04 ± 6,39%), foi gradualmente substituído por ambiente com certa influência terrígena em virtude do clima mais seco e maior erosão, como sugerido pela razão C/N e δ13C, que proporcionou o aumento da produtividade fitoplanctônica, e, assim, das concentrações de COT (0,90 ± 0,25%), NT (0,09 ±0,02%) e derivados de clorofila (0,12 ± 0,07 SPDU) durante o Último Máximo Glacial (Fase IV). No H1, por outro lado, foi observada uma redução abrupta das concentrações destas variáveis e da razão C/N, concomitante com o aumento do teor de carbonato, δ13C e δ15N, indicando uma produtividade estritamente marinha, com maior remineralização da matéria orgânica. Essas alterações na paleoprodutividade estão relacionadas ao enfraquecimento da AMOC e aumento da temperatura no Atlântico Equatorial durante o H1, que resultaram em um ambiente mais oligotrófico e com maior precipitação de carbonatos. O YD (Fase II) apresentou uma tendência oposta ao H1 e semelhante ao LGM. O tardiglacial (final da Fase II e Fase I), por sua vez, apresentou uma redução do COT, NT e derivados de clorofila de 1,21 ± 0,24%, 0,11 ± 0,02% e 0,27 ± 0,08 SPDU para 0,83 ± 0,06%, 0,08 ± 0,01% e 0,15 ± 0,05 SPDU, respectivamente, acompanhado de uma ligeira diminuição do C/N (de 10,98 ± 1,05 para 10,47 ± 0,59) e valores menos negativos de δ13C (-21,92 ± 1,26‰ para -20,31 ± 0,25‰). De uma forma geral, a relação entre a razão C/N e δ13C sugeriu a presença de matéria orgânica predominantemente marinha ao longo do testemunho, com alguns períodos com tendências terrígenas. A taxa de sedimentação foi relativamente baixa 0,013 ± 0,051 cm.ano-1, apresentando maiores valores nos primeiros 100 cm. Os resultados mostram que a paleoprodutividade nessa região foi influenciada pelas alternâncias de inputs terrestres e produção de carbonatos ao longo do Pleistoceno tardio / The ocean circulation system plays an important role in climate on Earth. Once the Equatorial Atlantic is the connection point between the South and North Atlantic, paleoceanographic studies in this area can contribute to the understanding of the processes and factors involved in climate variations over geological time, especially during the Quaternary. Within this context, The present study aimed to describe representative elements of the western equatorial Atlantic paleoproductivity during the Last Glacial through the sedimentological and geochemical analysis of sedimentary MD09-3243CQ core collected in the slope of the continental margin of northeastern Brazil. It was recovered the period between 33,700 and 10,150 years B.P., which was divided into six main phases, according to the cluster analysis of representative size fractions. Stages had different characteristics due to changes of paleoenvironmental conditions, paleoceanographic and paleoproductivity. The Last Glacial scenario (Phase V), characterized by high productivity and low carbonate marine primary productivity (TOC = 0.40 ± 0.12%; TN = 0.05 ± 0.01%, derived from chlorophyll = 0.06 0.04 ± SPDU; carbonate = 51.04 ± 6.39%) was gradually replaced by environmental influenced by terrigenous material because the drier climate and greater erosion, as suggested by the C/N ratio and δ13C that provided increased phytoplankton productivity, and thus the concentrations of TOC (0.90 ± 0.25%), TN (0.09 ± 0.02%) and derivatives of chlorophyll (0.12 ± 0.07 SPDU) during the Last Glacial Maximum (Phase IV). During H1, on the other hand, an abrupt reduction of the concentrations of these variables and the C/N ratio, concomitant with the increase in carbonate content, δ13C and δ15N was observed, indicating a strictly marine productivity with increased remineralization of organic matter. These changes in paleoproductivity are related to the weakening of AMOC and increase of temperature in the Equatorial Atlantic during H1, which resulted in a more oligotrophic environment and greater precipitation carbonates. The YD (Phase II) had an opposite trend to H1 and was similar to LGM. The Late Glacial (end of Phase II and Phase I), in turn, showed a reduction in TOC, TN and derivatives of chlorophyll 1.21 ± 0.24%, 0.11 ± 0.02%, and 0.27 ± 0.08 SPDU, to 0.83 ± 0.06%, 0.08 ± 0.01% and 0.15 ± 0.05 SPDU, respectively, accompanied by a slight decrease in the C/N (10.98 ± 1.05 to 10.47 ± 0.59) and less negative values of δ13C (-21.92 ± 1.26 ‰ to -20.31 ± 0.25 ‰). In general, the relation between the C/N ratio and δ13C suggested organic matter predominantly marine with some periods with terrigenous trends. The sedimentation rate was relatively low, 0.013 ± 0.051 cm.year-1, with higher values in the first 100 cm.
