Potencial acaricida, anti-helmíntico e estudos fitoquímicos de extrativos vegetais obtidos de Achyrocline satureioides (Lam.) D.C. (Asteraceae) /

Apesar disso, o setor enfrenta perdas consideráveis devido à ocorrência de infestações massivas de ectoparasitas como os carrapatos da espécie Rhipicephalus (Boophilus) microplus. Ainda dentro do agronegócio, outra produção que se encontra em amplo crescimento e demonstrando grande potencial de ganho de espaço no mercado internacional é a ovinocultura, com rebanho estimado em 14 milhões de animais, colocando o Brasil em 18º lugar no ranking mundial de exportações. Porém, existem grandes perdas econômicas devido à existência de infestações com endoparasitas, como o nematódeo gastrintestinail da espécie Haemonchus contortus. Em paralelo à preocupação com os prejuízos financeiros no agronegócio, outra espécie de carrapato, Rhipicephalus sanguineus, desperta interesse dos pesquisadores por ser o vetor de microrganismos causadores de importantes doenças em cães nas regiões tropicais e subtropicais. Devido ao mau uso, acaricidas e nematicidas sintéticos tem ocasionado o fenômeno de resistência, com isso a busca por novas substâncias a partir de espécies vegetais configura-se numa alternativa muito promissora para o controle desses parasitas. Nevertheless, the sector is facing considerable losses due to the occurrence of massive infestations of ectoparasites such as tick species of Rhipicephalus (Boophilus) microplus. In the agribusiness, another production that is in ample growth and showing great potential for space gain in the international market is the sheep industry, with herd estimated at 14 million animals, placing Brazil in 18th position in the world exports ranking. However, there are great economic losses due to the infestations of endoparasites, such as the gastrointestinal nematodes species of Haemonchus contortus. In parallel to the concern about the financial losses in agribusiness, other tick species, Rhipicephalus sanguineus, arouses interest of researchers for being the vector of microorganisms that causes major diseases to dogs in tropical and subtropical regions. Due to misuse of synthetic antiparasitic, which causes the phenomenon of resistance to them, the search for new substances from plant species sets up a very promising alternative for the biological control. Carrapatos duros (Acari : Ixodidae) associados a hospedeiros domésticos em diferentes regiões da Colômbia e sua interação com Rickettsia spp. /

No entanto, na Colômbia o registro e distribuição das espécies da família Ixodidae é escasso e pouco se sabe sobre a interação destes ectoparasitas com as diferentes espécies de Rickettsia na região, deixando uma lacuna em estudos e em conhecimento ixodológico e zoonótico no país. Desta forma, o presente trabalho buscou: a) realizar o levantamento e identificação de espécies de carrapatos duros associados a hospedeiros domésticos em regiões da Colômbia, utilizando caracteres morfológicos externos e marcadores moleculares; b) aferir a existência de caracteres morfohistológicos do sistema reprodutor masculino de carrapatos do gênero Amblyomma e c) verificar a ocorrência de bactérias Rickettsia spp. em carrapatos duros em regiões da Colômbia. Durante o período de agosto de 2014 e maio de 2016, foram coletados 1.745 carrapatos diretamente de hospedeiros domésticos e em ativa alimentação, em 17 municípios de 10 departamentos da Colômbia. Os indivíduos coletados foram identificados com base em sua morfologia externa e preparados para aplicação de técnicas de microscopia, bem como técnicas de análise molecular dos genes ITS2, citocromo oxidase I (COI) e 16S rDNA. However, in Colombia, species registry and distribution for the family Ixodidae is scarce, and little is known of the interaction of these ectoparasites with the different Rickettsia species in the region, leading to gaps regarding the ixodological and zoonotic study and knowledge in the country. In this context, this study aimed to: a) establish and identification hard-bodied ticks associated with domestic hosts in various regions of Colombia, using external morphological characters and molecular markers; b) assess the existence of morphohistological characters of the male reproductive system of Amblyomma ticks; and c) verify the occurrence of Rickettsia spp. in hard-bodied ticks in various regions of Colombia. During August of 2014 and May of 2016, 1745 ticks were collected actively feeding on domestic hosts in 17 municipalities of 10 departments of Colombia. Frente a isso, há crescente incentivo na busca por alternativas de controle do ectoparasito, como o desenvolvimento de vacinas. O presente estudo teve como objetivo avaliar a utilização de uma proteína recombinante da aquaporina do Rhipicephalus (Boophilus) microplus como antígeno em uma vacina contra R. sanguineus, em cães domésticos. A investigação foi realizada em cooperação científica com o United States Department of Agriculture/Agricultural Research Service (USDA/ARS). Os efeitos da vacina foram avaliados nos diferentes estádios do ectoparasito em cães previamente vacinados com o antígeno + adjuvante (Montanide) (G1) e em animais de um grupo controle inoculados apenas como adjuvante (G2). Foram avaliados: i. potencial biótico (parâmetros biológicos) dos carrapatos; ii. Therefore, there is an increasing incentive in the search for alternatives to the parasite's control, such as the development of vaccines. This study aimed to evaluate the use of a recombinant aquaporin R.(Boophilus) microplus protein as antigen in a vaccine against R. sanguineus in domestic dogs. The research was conducted on scientific cooperation with the United States Department of Agriculture / Agricultural Research Service (USDA / ARS). The vaccine's effects were evaluated at different stages of the ectoparasite in dogs previously vaccinated with antigen + adjuvant (Montanide) (G1) and in animals of a control group inoculated only with adjuvant (G2). It was evaluated: i. biotic potential (biological parameters) of ticks; ii. serum antibodies (IgG) titers of the dogs after immunization, by indirect enzymelinked immunosorbent assay (ELISA); iii. Histopathological evaluation of the tick's attachment site at various times post-fixation on the host's skin, including the inflammatory cell counts migrated into the inflammatory site; iv. Detecção molecular de Ehrlichia canis em cães e em órgãos de seus respectivos carrapatos /

Esta bactéria multiplica-se nas células epiteliais do intestino, nos hemócitos e nas células das glândulas salivares do carrapato Riphicephalus sanguineus, onde ocorre a transmissão transestadial e provavelmente não ocorre transmissão transovariana. O objetivo do presente estudo foi detectar molecularmente Ehrlichia canis em amostras de linfonodos e medulas ósseas de cães e nos intestinos, ovários e glândulas salivares de seus respectivos carrapatos R. sanguineus. Assim, 720 artrópodes fêmeas (dez de cada animal) foram removidos dos 72 cães examinados. Em seguida, foram efetuadas punções aspirativas de linfonodos e medulas ósseas. Após a dissecação dos carrapatos, seus órgãos (intestinos, ovários e glândulas salivares), bem como as amostras puncionadas de linfonodos e medulas ósseas dos cães foram testadas por meio da Reação em Cadeia da Polimerase (nested-PCR) para amplificação de um fragmento do gene 16S do ácido ribonucleico ribossomal (rRNA) de E. canis e testados também por meio da PCR em Tempo Real Quantitativa (qPCR) para E. canis, baseada em um fragmento do gene dsb. A detecção por meio da nested-PCR foi de 80,5% nos linfonodos e 44,4% nas medulas ósseas, havendo diferença significativa (p<0,05). This bacteria multiplies in the epithelial cells of the intestine, in the hemocytes and in the salivary glands of the tick Riphicephalus sanguineus, where there is a transestadial transmission and probably does not occur a transovarial transmission. The objective of our study was to molecularly detect Ehrlichia canis in samples of lymph nodes and bone marrow of dogs and in the intestines, ovaries and salivary glands of their respective ticks R. sanguineus. Therefore, 720 female arthropods (ten of each animal) were removed from the 72 evaluated dogs. Next, aspirative punches were made of lymph nodes e bone marrow. After the dissection of the ticks, their organs (intestines, ovaries and salivary glands), like the punches samples of lymph nodes and bone marrow of the dogs were tested by Polymerase Chain Reaction (nested-PCR) for amplification of the gene 16S rRNA of E. canis and also tested by the Quantitative Real Time PCR (qPCR) to E. canis, based on a fragment of the gene dsb. The detection of the refer pathogen by the nested-PCR was 80,5% in lymph nodes and 44,4% in bone marrows, showing a significant difference (p<0,05). ).
