THE ROLE OF THE L-TYPE CALCIUM CHANNEL AND ITS CARBOXYL-TERMINUS

In the heart, the primary role of the L-type calcium channel (LTCC) CaV1.2 is to conduct calcium into cardiomyocytes and initiate contraction. However, part of the CaV1.2 channel itself, the cleaved carboxyl-terminus (CCt) can also localize to the nucleus and regulate gene transcription. Therefore, the goal of this dissertation project was to determine the role and regulation of CCt in the embryonic and adult heart. The global hypothesis of my dissertation project is that CCt localizes to the nucleus in embryonic and adult cardiomyocytes via a calcium-mediated mechanism and regulates transcription. A model of pharmacological LTCC block-induced perturbation of murine embryonic heart development was first utilized to study the role of CCt. Pharmacological block at embryonic day 10 perturbed cardiogenesis and increased CaV1.2 expression. This result was not mimicked by removal of extracellular calcium or inhibition of calcium release from the sarcoplasmic reticulum. Co-currently, pharmacological block decreased CCt nuclear localization in embryonic cardiomyocytes. At the transcriptional level, CCt suppressed the CaV1.2 promoter. This indicated that the observed upregulation of CaV1.2 induced by pharmacological block may be caused by nuclear localization of the transcriptional repressor, CCt. Therefore, the conclusion was made that pharmacological LTCC block perturbed embryonic cardiogenesis by decreasing nuclear localization of the transcription factor CCt; implying a role for CCt in embryonic heart development. Next, CCt regulation was studied in the adult heart. Similar to the embryonic heart, pharmacological LTCC block decreased nuclear localization of CCt. Inhibition of the calcium activated phosphatase calcineurin also decreased CCt nuclear localization. To determine a role for CCt in the adult heart, CCt nuclear localization was measured in response to hypertrophic stimuli. Serum-induced cardiomyocyte hypertrophy significantly increased nuclear localization of CCt. In conclusion, this dissertation supports the hypothesis that CCt localizes to the nucleus in embryonic and adult cardiomyocytes, and that this regulation is mediated by calcium entry into the cardiomyocyte. Furthermore, data from this dissertation suggests that CCt nuclear localization may play an important role in embryonic heart development and adult cardiac hypertrophy.

Signal Detection for Overloaded Receivers

In this work wireless communication systems with multiple co-channel signals present at the receiver are considered. One of the major challenges in the development of such systems is the computational complexity required for the detection of the transmitted signals. This thesis addresses this problem and develops reduced complexity algorithms for the detection of multiple co-channel signals in receivers with multiple antennas. The signals are transmitted from either a single user employing multiple transmit antennas, from multiple users or in the most general case by a mixture of the two. The receiver is assumed to be overloaded in that the number of transmitted signals exceeds the number of receive antennas. Joint Maximum Likelihood (JML) is the optimum detection algorithm which has exponential complexity in the number of signals. As a result, detection of the signals of interest at the receiver is challenging and infeasible in most practical systems. The thesis presents a framework for the detection of multiple co-channel signals in overloaded receivers. It proposes receiver structures and two list-based signal detection algorithms that allow for complexity reduction compared to the optimum detector while being able to maintain near optimum performance. Complexity savings are achieved by first employing a linear preprocessor at the receiver to reduce the effect of Co-Channel Interference (CCI) and second, by using a detection algorithm that searches only over a subspace of the transmitted symbols. Both algorithms use iterative processing to extract ordered lists of the most likely transmit symbols. Soft information can be obtained from the detector output list and can then be used by error control decoders. The first algorithm named Parallel Detection with Interference Estimation (PD-IE) considers the Additive White Gaussian Noise (AWGN) channel. It relies on a spatially reduced search over subsets of the transmitted symbols in combination with CCI estimation. Computational complexity under overload is lower than that of JML. Performance results show that PD-IE achieves near optimum performance in receivers with Uniform Circular Array (UCA) and Uniform Linear Array (ULA) antenna geometries. The second algorithm is referred to as List Group Search (LGS) detection. It is applied to overloaded receivers that operate in frequency-flat multipath fading channels. The List Group Search (LGS) detection algorithm forms multiple groups of the transmitted symbols over which an exhaustive search is performed. Simulation results show that LGS detection provides good complexity-performance tradeoffs under overload. A union bound for group-wise and list-based group-wise symbol detectors is also derived. It provides an approximation to the error performance of such detectors without the need for simulation. Moreover, the bound can be used to determine some detection parameters and tradeoffs. Results show that the bound is tight in the high Signal to Interference and Noise Ratio (SINR) region.

A Channel Approach to Fashion

Background – Previous research has evaluated different distribution systems and chan-nels that companies operate through. However, research primarily has focused on the richness of online channels rather than the value of physical stores. The following report will examine the reasons and implications underlying the choice of channel strategy. Purpose – The purpose of this thesis is to examine the reason why companies within the fashion and apparel industry, operating in Sweden, choose to operate through a certain market channel strategy. The thesis aims to evaluate four types of market channel strat-egies with the help of case companies. Method – A qualitative approach to the research has been chosen which consists of a case study including four companies representing different channel strategies. Data col-lection for the analysis where conducted through interviews with persons having vital positions within one of the four companies. Results and Discussion – The case companies ASOS, Bubbleroom, Cubus and Dressmann each represent one of the chosen market channels. The interviews presented in the results section implied that the companies chosen channel was more appropriate for the market they operated in as well as the target group they were aiming to reach. Conclusion – Even though research states that today’s society is moving away from the physical store market channel, this study proved that one single strategy is not appropri-ate for all businesses. A company will have to research their target market before ap-propriately selecting a channel strategy.

Interference Management in MIMO Wireless Networks

The scarce and overpopulated radio spectrum is going to present a major barrier to the growth and development of future wireless networks. As such, spectrum sharing seems to be inevitable to accommodate the exploding demand for high data rate applications. A major challenge to realizing the potential advantages of spectrum sharing is interference management. This thesis deals with interference management techniques in noncooperative networks. In specific, interference alignment is used as a powerful technique for interference management. We use the degrees of freedom (DoF) as the figure of merit to evaluate the performance improvement due to the interference management schemes. This dissertation is organized in two parts. In the first part, we consider the K-user multiple input multiple output (MIMO) Gaussian interference channel (IC) with M antennas at each transmitter and N antennas at each receiver. This channel models the interaction between K transmitter-receiver pairs sharing the same spectrum for data communication. It is assumed that the channel coefficients are constant and are available at all nodes prior to data transmission. A new cooperative upper-bound on the DoF of this channel is developed which outperforms the known bounds. Also, a new achievable transmission scheme is provided based on the idea of interference alignment. It is shown that the achievable DoF meets the upper-bound when the number of users is greater than a certain threshold, and thus it reveals the channel DoF. In the second part, we consider communication over MIMO interference and X channels in a fast fading environment. It is assumed that the transmitters obtain the channel state information (CSI) after a finite delay which is greater than the coherence time of the channel. In other words, the CSI at the transmitters becomes outdated prior to being exploited for the current transmission. New transmission schemes are proposed which exploit the knowledge of the past CSI at the transmitters to retrospectively align interference in the subsequent channel uses. The proposed transmission schemes offer DoF gain compared to having no CSI at transmitters. The achievable DoF results are the best known results for these channels. Simple cooperative upper-bounds are developed to prove the tightness of our achievable results for some network configurations.

Ένα από τα βασικά μειονεκτήματα που παρουσιάζει ο σχεδιασμός και η υλοποίηση LDPC αποκωδικοποιητών είναι η μεγάλη πολυπλοκότητα που παρουσιάζεται σε επίπεδο υλικού εξαιτίας της εσωτερικής διασύνδεσης των μονάδων επεξεργασίας δεδομένων.H αρχιτεκτονική που επιτυγχάνει το μέγιστο επίπεδο παραλληλότητας και κατά συνέπεια είναι πολύ αποδοτική όσον αφορά την ταχύτητα αποκωδικοποίησης, δεν χρησιμοποιείται συχνά εξαιτίας της πολυπλοκότητας του υλικού λόγω των πολλαπλών κυκλωμάτων διασύνδεσης που απαιτεί. Στην παρούσα διπλωματική εργασία προτείνεται μια νέα αρχιτεκτονική για το δίκτυο διασύνδεσης ενώ παράλληλα έχει υλοποιηθεί και ένας αλγόριθμος για την αποδοτική τοποθέτηση των επεξεργαστικών μονάδων σε αυτό το δίκτυο. Επίσης έχει μελετηθεί και η επίδραση μειωμένης μετάδοσης πληροφορίας σε κάθε επανάληψη του αλγορίθμου αποκωδικοποίησης.Το περιβάλλον που χρησιμοποιήθηκε για την εξομοίωση και την παραγωγή των αποτελεσμάτων είναι η πλατφόρμα της Matlab. Η προτεινόμενη αρχιτεκτονική υλοποιήθηκε και εξομοιώθηκε σε κώδικες LDPC που αποτελούν μέρος του προτύπου DVB - S2 (Digital Video Broadcasting).Το συγκεκριμένο πρότυπο, εκτός των άλλων, καθορίζει και τις προδιαγραφές των κωδίκων LDPC που χρησιμοποιούνται κατά την κωδικοποίηση και αποκωδικοποίηση δεδομένων σε συστήματα ψηφιακής δορυφορικής μετάδοσης. Τα αποτελέσματα των εξομοιώσεων σχετίζονται με την πολυπλοκότητα της προτεινόμενης αρχιτεκτονικής σε υλικό αλλά και της απόδοσης (ταχύτητα αποκωδικοποίησης) και συγκρίνονται με την βασική πλήρως παράλληλη αρχιτεκτονική. / One of the main disadvantages of the design and implementation of LDPC decoders is the great complexity presented at the hardware level because of the internal interconnection of processing units. The fully parallel architecture that achieves the maximum level of parallelism and hence is very efficient in terms of speed decoding is not used often because of the hardware complexity due to the multiple interface circuits required. This MSc thesis proposes a new architecture for the network interface and also introduces an algorithm for the efficient placement of the processing units in this network. In addition to that, a modified version of the decoding algorithm has been implemented. The relative advantage of this algorithm is that in each iteration only a percentage of the processing units exchange information with each other. That approach further reduces the hardware complexity and power usage. The environment used to simulate and produce the results is Matlab. The proposed architecture is implemented and simulated in LDPC codes that are part of the standard DVB - S2 (Digital Video Broadcasting). This standard, among other things, determines the specifications of the LDPC codes used in the channel encoding and decoding process in digital satellite transmission systems. The results of the simulations related to the complexity of the proposed architecture in hardware and performance (decoding speed) are compared with the fully parallel architecture.

Τεχνικές συμπιεσμένης καταγραφής για εκτίμηση και ισοστάθμιση αραιών καναλιών

Κανάλια με αραιή κρουστική απόκριση εμφανίζονται πάρα πολύ συχνά σε εφαρμογές ασύρματων κυρίως τηλεπικοινωνιακών συστημάτων. Παραδείγματα τέτοιων εφαρμογών είναι η εκπομπή HDTV (HighDefinitionΤelevision) ή εκπομπή μέσω υποθαλλάσιων ακουστικών καναλιών. Σε όλες αυτές τις εφαρμογές η μορφή του καναλιού διαμορφώνεται από το φαινόμενο της πολυδιόδευσης. Συνεπώς ο δέκτης λαμβάνει έναν περιορισμένο αριθμό από διαφορετικές εκδοχές του εκπεμπόμενου σήματος καθεμία με διαφορετική εξασθένιση και καθυστέρηση. Ως εκ τούτου η συνάρτηση της κρουστικής απόκρισης ενός τέτοιου καναλιού αποτελείται από ελάχιστα μη μηδενικά στοιχεία σε συγκριση με το μήκος της, καθένα από τα οποία αντιστοιχεί σε ένα από τα μονοπάτια πολυδιόδευσης. Για την ισοστάθμιση αυτών των καναλιών έχουν προταθεί διάφορες τεχνικές, πολλές από τις οποίες εκμεταλλεύονται την ιδιαίτερη αυτή μορφή της κρουστικής απόκρισης. Πολλοί από τους προτεινόμενους ισοσταθμιστές καναλιών απαιτούν την παρεμβολή ακολουθίων εκμάθησης ανάμεσα στην ακολουθία δεδομένων, οι οποίες είναι εκ των προτέρων γνωστές στον δέκτη. Χρησιμοποιούνται δε προκειμένου ο αλγόριθμος εκτίμησης του καναλιού να συγκλίνει όσο το δυνατόν ταχύτερα στην επιθυμητή τιμή. Μειονέκτημα αυτών των μεθόδων είναι η επιβάρυνση του ωφέλιμου εύρους ζώνης που συνεπάγεται. Ωστόσο η εκ των προτέρων γνώση της αραιής μορφής της κρουστικής απόκρισης εχει δώσει αφορμή για την σχεδίαση ισοσταθμιστών με περιορισμένο μήκος αλλά εξίσου καλή απόδοση. Οι συμβατικές τεχνικές εκτίμησης καναλιών, όπως η Least Square μέθοδος, δεν εκμεταλλεύονται αυτή την γνώση. Οι πρόσφατες δε εξελίξεις στην ανακατασκευή αραιών σημάτων μέσω τεχνικών συμπιεσμένης καταγραφής (compressed sensing) έχουν οδηγήσει στην μελέτη της εφαρμογής τέτοιων τεχνικών στο πρόβλημα της εκτίμησης καναλιού. Η μέθοδος της συμπιεσμένης καταγραφής στηρίζεται στη δυνατότητα ανακατασκευής αραιών σημάτων από πλήθος δειγμάτων αισθητά κατώτερο από αυτό που προβλέπει το θεωρητικό όριο του Nyquist. Έχει αποδειχθεί ότι η ανακατασκευή αυτή είναι δυνατή όταν το σήμα ή έστω κάποιος μετασχηματισμός του περιέχει λίγα μη μηδενικά στοιχεία σε σχέση με το μήκος του. Οι εφαρμογές αυτών των τεχνικών εκτείνονται και σε άλλα πεδία όπως η επεξεργασία εικόνας, η μαγνητική τομογραφία, η ανάλυση γεωφυσικών δεδομένων, η επεξεργασία εικόνας radar, η αστρονομία κ.α. Στα πλαίσια αυτής της εργασίας παρουσιάζονταιοι βασικές αρχές που διέπουν την ανακατασκευή αραιών σημάτων μέσω της επίλυσης υποορισμένων συστημάτων γραμμικών εξισώσεων. Παράλληλα παρουσιάζονται οι κυριότεροι αλγόριθμοι που έχουν προταθεί για την υλοποίηση της και εξετάζονται ως προς την απόδοση και την υπολογιστική πολυπλοκότητα τους. Εν συνεχεία εξετάζεται η εφαρμογή αυτών των αλγορίθμων στο πρόβλημα της εκτίμησης αραιών καναλιών. Προτείνονται δε ισοσταθμιστές αραιών καναλιών βασισμένοι σε εκτιμητές απόκρισης που χρησιμοποιούν τεχνικές συμπιεσμένης καταγραφής. / Channels with sparse impulse response are very common in wireless telecommunications systems applications. Example of such channel is HDTV channel where multipath distribution of the transmitted signal results in a sparse form of the channel impulse response. Several different versions of the same signal are received, each one with its own gain and delay. As a result, channel impulse response has a few non zero taps compared to its length, its one corresponding to a different distribution path. Several techniques for estimating and equalizing such channels have been proposed, most of them taking advantage of this sparse form of the impulse response. The transmission of a training sequence known to the receiver is required for this purpose. It is used so that the channel estimation algorithm at the receiver converges faster. The disadvantage of the use of a training sequence is the fact that the useful bandwidth is reduced. However the a priori knowledge of the sparse form of the training sequence has led to the design of equalizers that require short training sequences but have satisfactory performance. Channel estimation techniques based on least square method do not take advantage of this idea. On the other hand recent progress on sparse signal reconstruction using compressed sensing techniques has led scientists to research the potential use of such algorithms in channel estimation. Compressed sensing is based on the idea of reconstructing a sparse signal using less samples that those predicted by Nyquist theorem. It has been proved that such a reconstruction is feasible if the reconstructed signal is sparse enough. In this dissertation several sparse signal reconstruction algorithms are presented and their performance and complexity are evaluated. Then the application of these algorithms on channel estimation equalization problem is analyzed.

Characterisation of novel cardiac and skeletal ion channels on intracellular Ca2+ stores

Excitation-contraction (EC) coupling is the process by which Ca<sup>2+</sup> is released from the sarcoplasmic reticulum (SR) and is fundamental to cardiac and skeletal muscle function. The SR contains many uncharacterised ion channels and proteins which may influence EC coupling and in this thesis I have investigated the biophysical properties of some of these channels. I have demonstrated that the single-channel gating and conducting properties of SR K<sup>+</sup> channels from various mammalian species (rabbit, sheep and mouse) are very similar. I investigated the actions of possible physiological regulators of these channels and demonstrated that luminal Ca<sup>2+</sup> and Mg<sup>2+</sup> can block K<sup>+</sup> flux in a voltage-dependent manner, while luminal Ca<sup>2+</sup>, Ni<sup>2+</sup>, and alkaline pH can reduce Po by additional mechanisms. I also characterised the single-channel properties of the various SR anion channels that are observed after incorporating mammalian SR vesicles into artificial membranes. The trimeric intracellular cation channels (TRIC-A and TRIC-B) and Mitsugumin 23 (MG23) are suggested to be SR cation channels. I have therefore utilised Tric-a KO and Mg23 KO mice to study SR membranes devoid of TRIC-A and MG23. Additionally, I have begun to investigate the single-channel properties of purified c. elegans TRIC-B1 and human TRIC-A. I found that SR K<sup>+</sup> channel function was altered in SR from Tric-a KO or Mg23 KO tissue, however the underlying mechanisms for the observed changes appear to be complex. My initial studies of the purified TRIC-A and TRIC-B proteins show that they are permeable to K<sup>+</sup>, Ca<sup>2+</sup>, choline, and Cl<sup>-</sup>, properties which deviate from those of SR K<sup>+</sup> channels from rabbit, mouse and sheep. This may reflect species differences or alterations to protein function caused during the purification process or that SR K<sup>+</sup> channels remain an unidentified class of ion channel.
