Por esse motivo o grupo demonstra ser um objeto de estudo de grande importância para a ecologia comparativa. Este estudo avalia os atributos ecológicos de vários táxons associada à filogenia reconstruir a história evolutiva do grupo, identificando caracteres ancestrais. Foram coletados dados morfológicos, merísticos, alimentares e reprodutivos das 13 espécies (Calamodontophis paucidens; Gomesophis brasiliensis; Pseudotomodon trigonatus; Ptychophis flavovirgatus; Tachymenis peruviana; Thamnodynastes hypoconia; T. lanei; T. nattereri; T. pallidus; T. rutilus; T. strigatus e Tomodon dorsatus). Para tanto foram obtidos dados da literatura e de coleções herpetológicas brasileiras e do exterior. Por meio da otimização de caracteres foi possível reconhecer a evolução de padrões morfológicos, merísticos, bem como definir a dieta da maioria das espécies estudadas / Abstract: The Tachymenini tribe has great diversity on the morphology, habitat use and feeding habits. For this reason, the group proves to be a very important object of study for comparative ecology. This study evaluates the ecological attributes of various taxa and associate then to phylogeny to the reconstruct of the evolutionary history, identifying ancestral characters of the tribe. Morphological, meristic, feeding and reproductive data were colected for 13 species (Calamodontophis paucidens; Gomesophis brasiliensis; Pseudotomodon trigonatus; Ptychophis flavovirgatus; Tachymenis peruviana; Thamnodynastes hypoconia; T. lanei, T. nattereri, T. pallidus, T. rutilus, T. strigatus and Tomodon dorsatus). Therefore, we obtained data from the literature and many herpetological collections from South America. Comparação da capacidade neutralizante dos antisoros botrópicos comercial e monoespecífico frente a peçonha de B. Erythromelas

O antiveneno botrópico produzido no Brasil inclui apenas cinco espécies desse gênero, dentre as quais não se encontra a espécie Bothrops erythromelas (jararaca-da-seca), serpente botrópica endêmica da região Nordeste. Primeiramente foram determinadas as dose mínimas que constituíram o desafio de neutralização para os antivenenos em cada atividade. Em seguida a neutralização foi calculada e expressa como DE50, dose que inibe 50% da atividade testada. As Doses Mínimas encontradas foram: 22,119 μg/camundongo (hemorrágica), 78,38 μg/camundongo (necrosante), 21,37 μg/ml (Coagulante) e 0,05 mg (Fosfolipásica). A DL50 foi 5,11 mg/kg. A eletroforese em gel de poliacrilamida, sob condições redutoras, da peçonha de B. erythromelas, corada por Comassie blue mostrou sete bandas confirmadas pelo imunoblotting quando revelado com os antivenenos comercial e monoespecífico diluídos 1/16.000. Foi demonstrado também, através da técnica de imunoprecipitação, que o antiveneno monoespecífico apresenta maior capacidade de formar imunocomplexos com os determinantes antigênicos presentes na peçonha de B. erythromelas, enquanto que a capacidade de complexação do antiveneno comercial não foi suficiente para precipitar os antígenos dessa peçonha. Nos experimentos de neutralização, foi observada uma eficácia cerca de 2x maior do antiveneno monoespecífico em relação ao comercial, em todas as atividades testadas. As Doses Efetivas 50% para o antiveneno comercial e monoespecífico, respectivamente, foram: 49,21 μl/mg peçonha e 26,95 μl/mg peçonha (hemorragia); 9,50 μl/mg e 7,34 μl/mg (necrosante); 85,2 μl/mg e 51,2 μl/mg (letal); 12,61 μl/mg e 6,35 μl/mg (coagulante) e, 285,38 μl/mg e 166 μl/mg (fosfolipásica). Neste trabalho, foi demonstrado que o antiveneno botrópico monoespecífico foi mais efetivo que o botrópico comercial na neutralização das atividades testadas (letal, hemorrágica, necrosante, coagulante e fosfolipásica), na capacidade de formar imunocomplexos in vitro com a peçonha de Bothrops erythromelas e no reconhecimento das proteínas separadas eletroforeticamente

Cloning and functional expression of Taiwan cobra chymotrypsin inhibitor

Previous studies showed that dendrotoxins and B chain of b-Bungarotoxin shared sequence and structural homology with Kunitz-type protease inhibitors. In the present study, the cDNA of Kunitz¡Vtype protease inhibitor was successfully amplified from Taiwan cobra venom gland total RNAs using the primers designed from the B chain of b-Bungarotoxin. The deduced amino acid sequence of the cDNA exhibited the structural character of chymotrypsin inhibitor, and the mature protein contained 57 amino acids with six Cys residues. The chymotrypsin inhibitor was subcloned into pET29a(+) and transformed into BL21(DE3) E.coli strain. The expressed protein was isolated from inclusion bodies of E.coli and subjected to refolding into its folded structure. The inhibitor potency of the recombinant protein on chymotrypsin activity had a Ki value of 461.3 mM. However, removal of its N-terminal fused peptide with thrombin further increased the Ki value to 31.7 mM. Removal of the N-terminal residues further reduced its inhibitory potency, and the inhibitory activity completely lost after deleting three residues at the N-terminus of mature protein. This reflects that the N-terminal region of protease inhibitor should be associated with its activity. The genomic DNA encoding the precursor of the inhibitor was also amplified using PCR. The genetic structure composed of three exons and three introns, which shared the same organization with the b-Bungarotoxin B chain gene. Moreover, the two genes showed a high degree of sequence identity up to 83%. This observation emphasizes the idea that the B chain of b-Bungarotoxin and protease inhibitor are evolutionarily related.

A language and toolkit for the specification, execution and monitoring of dependable distributed applications

This thesis addresses the problem of specifying the composition of distributed applications out of existing applications, possibly legacy ones. With the automation of business processes on the increase, more and more applications of this kind are being constructed. The resulting applications can be quite complex, usually long-lived and are executed in a heterogeneous environment. In a distributed environment, long-lived activities need support for fault tolerance and dynamic reconfiguration. Indeed, it is likely that the environment where they are run will change (nodes may fail, services may be moved elsewhere or withdrawn) during their execution and the specification will have to be modified. There is also a need for modularity, scalability and openness. However, most of the existing systems only consider part of these requirements. A new area of research, called workflow management has been trying to address these issues. This work first looks at what needs to be addressed to support the specification and execution of these new applications in a heterogeneous, distributed environment. A co- ordination language (scripting language) is developed that fulfils the requirements of specifying the composition and inter-dependencies of distributed applications with the properties of dynamic reconfiguration, fault tolerance, modularity, scalability and openness. The architecture of the overall workflow system and its implementation are then presented. The system has been implemented as a set of CORBA services and the execution environment is built using a transactional workflow management system. Next, the thesis describes the design of a toolkit to specify, execute and monitor distributed applications. The design of the co-ordination language and the toolkit represents the main contribution of the thesis.

Caracterização morfológica e molecular de Strongyloides ophidiae (nematoda, strongyloididae) parasitas de serpentes /

A primeira foi descrita em serpente Mastigodryas bifossatus e a segunda em lagartixa Hemidactylus mabouya. Na descrição de S. ophidiae apenas a fêmea partenogenética foi caracterizada em 1929 e, desde então, nenhum outro relato sobre esta espécie foi publicado. Estudos moleculares foram realizados com o objetivo de contribuir para a caracterização de Strongyloides spp. foram realizados, porém estes não incluíram S. ophidae. No presente estudo foi realizada a caracterização morfológica e molecular de S. ophidiae coletados em serpentes da região de Botucatu, Estado de São Paulo, Brasil. De um total de 125 animais, apenas quatro encontraram-se parasitados por S. ophidiae, porém fêmeas partenogenéticas foram recuperadas de apenas um animal. Coproculturas foram realizadas para obtenção das larvas L1, L2 e L3, bem como machos e fêmeas de vida livre, os quais foram morfologicamente analisados. Os produtos da PCR apresentaram 350 pb e as seqüências de nucleotídeos (número de acesso Genbank EU287935) isoladas deste nematódeo de Oxyrhopus guibei apresentaram 98% de identidade com Strongyloides procyonis (número de acesso AB272234.1 e AB205054.1) e 97% de identidade com Strongyloides cebus, Strongyloides stercoralis, Strongyloides sp. "ex snake" e Strongyloides fuelleborni (Genbank AB277236.1, AF279916.2, AJ417031.1 e AJ407030.1, respectivamente). A análise filogenética das espécies da família Strongyloididae e Rhabdiasidae demonstrou que, embora apresente 97% de similaridade com outras espécies, S. ophidiae encontra-se isolado em um ramo da árvore filogenética, caracterizando uma identidade diferente em relação a outras espécies analisadas. O presente estudo caracterizou morfológica e molecularmente S. ophidiae, parasita de serpentes da região de Botucatu, São Paulo, Brasil. / Abstract: In Brazil the genus Strongyloides includes two reptile parasite: S. ophidiae and S. cruzi. The former was described in Mastigodryas bifossatus snake and the other in Hemidactylus mabouya gecko. In the description of S. ophidiae just the partenogenetic females was characterized in 1929 and, ever since, no other report about this species was published. Molecular studies for the characterization of Strongyloides spp. were reported, however these did not include S. ophidae. In the present study the morphological and molecular characterization of S. ophidiae obtained from snakes in Botucatu region, São Paulo State, Brazil was accomplished. Among the 125 studied animals only four snakes were found infected by S. ophidiae, however partenogenetic females were recovered in only one animal. Fecal exams had been performed for obtaining L1, L2 and L3 larvae, as well as free-living males and females, which were morphologically analyzed. PCR products amplified from S. ophidiae partenogenetic female samples presented about 350 bp. Nucleotide sequences (Genbank accession numbers EU287935) isolates from these nematodes from Oxyrhopus guibei presented 98% identity with Strongyloides procyonis (Genbank acession number AB272234.1 and AB205054.1) and 97% identity with Strongyloides cebus, Strongyloides stercoralis, Strongyloides sp. ex snake and Strongyloides fuelleborni (Genbank AB277236.1, AF279916.2, AJ417031.1 and AJ407030.1, respectively). The analysis of the phylogenetic relationship among the species of Strongyloididae and Rhabdiasidae demonstrated that, although there are similarity above 97% with the other species, S. ophidiae was allocated in an isolated branch of the phylogenetic tree, characterizing a different identity in relation to other species analyzed. As proteases purificadas de veneno são geralmente dassificadas por sua estrutura em: serinoproteases e metaloproteases, e apresentam potente efeito na ativação ou inibição da coagulação sangüínea, agindo na hemostasia, trombose, fibrinólise e agregação plaquetária. A presente dissertação teve por objetivo purificar e caracterizar a serinoprotease Uruprot do veneno de Bothrops altematus, uma serpente da família Viperidae. Uruprot foi purificada em dois passos cromatográficos: Sephadex G75 e DEAE 5PW, em sistema HPLC. A homogeneidade de Uruprot foi monitorada em gel SDS-PAGE e em coluna de fase reversa C18. O gel de eletroforese em condições desnaturantes evidenciou uma banda com massa molecular aparente de 32 kDa na presença de 0,1 M de DTT, sugerindo que esta é formada por uma única cadeia polipeptídica. A eletroforese de duas dimensões de Uruprot demonstrou ser seu ponto isoelétrico em torno de 4,6, confirmando massa molecular observada em SDS-PAGE. A análise da composição de aminoácidos de Uruprot apresentou alta concentração dos aminoácidos glicina, treonina, fenilalanina, isoleucina, serina e glutamina e baixos níveis de aminoácidos básicos como histidina e arginina, ratificando o resultado da 2D caracterizando um pl ácido. Uruprot teve reação positiva quando submetida à coloração para carboidrato, sugerindo a existência de carboidratos ligados covalentemente à sua estrutura. O estudo da homologia seqüencial da região N-terminal da seqüência consenso (WGGDECNINEHR-LVAI) indicou alto grau de homologia com as serinoproteinases: TVSPA (94%), KLE 11 (94%) e Ancrod (92%) entre outras. Uruprot não induziu a agregação em plaquetas lavadas, mas inibiu a indução da aglutinação de eritrócitos na presença de Ristocetina na concentração mínima de 1 O ~g/mL. O conjunto de resultados obtidos com Uruprot monstram fortes indícios de que essa pode desempenhar a função de ativadora do plasminogênio, entretanto o papel da atividade "in vivo" das serinoproteases de venenos em envenenamentos patológicos é pouco conhecido. Estudos adicionais são necessários para a obtenção de maiores informações sobre esse grupo de enzimas / Abstract: Snake venoms contain a high variety of toxic compounds and enzymes. Venom purified proteases are structurally dassified as serine proteases and metalloproteases, and showa potent effect on blood coagulation, acting on hemostasis, thrombosis, fibrinolysis and platelet aggregation. The aim of this work was the purification and characterization of a serine protease from the venom of Bothrops altematus snake, called Uruprot. Uruprot was purified in a two step chromatography: Sephadex G-75 and DEAE 5PW both in HPLC system. Its homogenity was monitored in SDS-PAGE and reversedphase C18 chromatography. In denaturing conditions, 0,1 M DTT, a band of 32 kDa apparent molecular mass was evidenced, suggesting that the protein consists in one polypeptidic chain. The two dimensional electrophoresis of Uruprot showed its isoelectric point at about 4,6, confirming the apparent molecular mass observed in SDS-PAGE. Uruprot's amino acid analysis composition showed a high concentration of glycine, threonine, phenylalanine, isoleucine, serine and glutamine and low levels of basic amino aCids, as histidine and arginine, ratifying the result obtained from bidimensional electrophoresis wich showed an acid pl. Uruprot showed a positive reaction when submitted to carbohydrate coloration indicating the presence of carbohydrate covalentely bound to its structure. The N-terminal sequence homology study showed a consensus sequence (WGGDECNINEHR-LVAI) and indicated a high homology with serine proteinase TSV-PA (94%), KLE 11 (94%), Ancrod (92%), and others. Uruprot did not induce platelet aggregation but inhibits erytrocytes agglutination in the presence of ristocetin, in a minimum concentration of 10llg/mL. Ali the results obtained indicates that Uruprot is a serine protease plasminogen activator, however, the role of "in vivo" activity of venom serine proteases in envenomation remains unclear.
