Effect of Density Gradient Centrifugation on Quality and Recovery Rate of Equine Sperm

Density gradient centrifugation of sperm is a common assisted-reproduction procedure in humans used to improve semen quality. The technique allows sperm separation based on their isopycnic points. Sperm with morphologic abnormalities are often more buoyant, leading to their retention above centrifuged density gradients, with structurally normal sperm passing through the gradient. Three experiments were conducted to evaluate the effects of tube size, sperm number following centrifugation, and density gradient volume (height) on stallion sperm quality and recovery rate in sperm pellets following centrifugation. In all three experiments, equine semen was initially centrifuged to increase sperm concentration. In Experiment 1, one-mL aliquots were layered over EquiPure? Bottom Layer (1-Layer) or over-tiered EquiPure? Top and Bottom Layers (2-Layer). For Experiment 2, one-mL aliquots were layered over three different heights of EquiPure? Bottom Layer in 15-mL or 50-mL conical-bottom tubes. For Experiment 3, four different aliquots containing a sperm load of 1-4x were layered over a constant volume of EquiPure? Bottom Layer in 15-mL or 50-mL conical bottom tubes. The tubes were then centrifuged. Resulting sperm pellets were evaluated for morphologic quality, DNA integrity, motility and recovery rate. Sperm-EquiPure? centrifugation yielded improvements in motility, morphology and DNA integrity parameters (P<0.05), as compared to controls. The 1-Layer method resulted in a higher recovery rate than the 2-Layer method (P<0.05). Sperm processed in the 15-mL tubes yielded higher velocity and higher recovery rates than sperm processed in the 50-mL tubes (P<0.05). Within tube type, gradient volume did not impact parameters of semen quality or recovery rate. An increase in sperm number for density gradient centrifugation resulted in a decreased recovery rate (P<0.05) when 15-mL tubes were used.

Density gradient films, lanthanide electrochemistry, and magnetic field effects on hydrogen evolution, oxygen reduction, and lanthanide electrochemistry

Electroanalytical techniques are used to investigate mass transport through density gradient films; lanthanide triflate reduction and oxidation in a Nafion/acetonitrile matrix; and magnetic field effects on hydrogen evolution reaction (HER), oxygen reduction reaction (ORR), and lanthanide electrochemistry. Graded density films are more dense at the electrode surface and become less dense out into solution due to a brush polymer structure. Fick's second law expands to account for a diffusion coefficient that varies with distance x normal to the electrode surface. Confocal microscopy, cyclic voltammetry, and computer simulations are used to investigate density graded Ficoll® films. Mass transport approaches steady state (scan rate independence) at slow scan rates where the diffusion length samples the entire film. The use of Ficoll to template an ion exchange polymer is explored by casting Nafion® Ficoll composites. Lanthanide electrochemistry is enabled in acetonitrile at a Nafion modified platinum electrode in the presence of triflate ligands. Formal potentials are shifted into the voltage window of acetonitrile accessible due to triflate complexation. The Nafion further solubilizes the compounds. The mechanism (ECEC) is studied with cyclic voltammetry and x-ray photoelectron spectroscopy. Magnetic field effects on electrochemical systems have been of interest to researchers for the past 65 years. Mass transport effects, such as magnetohydrodynamics and magnetic field gradient effects have been reported, but the Leddy group focuses on electron transfer effects. Electrode surfaces are modified with composite films of magnetic microparticles suspended in ion exchange polymer Nafion. Effects are verified to be electron transfer related and due to the magnetization of chemically inert microparticles. The magnets catalyze the rates of important electron transfer reactions such as hydrogen evolution and oxygen reduction. Magnetic field effects on HER at various noncatalytic metal electrodes are explored with linear scan voltammetry. There is a correlation between the magnetic susceptibility of the electrode metals and the HER exchange currents (reaction rates). Exchange currents are 103× larger for a paramagnetic metal electrode than a diamagnetic one with the same work function. The overpotential at diamagnetic electrodes is decreased by modification with a Nafion + magnetic microparticle composite film. A decrease in overpotential of ∼70 % for all electrodes except platinum is observed. The overpotential decrease correlates with the magnetic susceptibility of the particles. Magnets can enhance differences between lanthanide cyclic voltammograms by shifting current densities at a given potential and enhancing current based on the number of 4f electrons and magnetic moment of each lanthanide ion. Magnetic field effects on ORR in acetonitrile are investigated with cyclic voltammetry. In aprotic solvents, ORR proceeds by a one electron transfer reaction to paramagnetic O2.–. Enhanced reversibility and electron transfer kinetics are observed as well as a decrease in overpotential of ∼100 mV. Magnetic field effects on ORR in a lanthanide triflate solution are also examined. Electron transfer kinetics and reversibility are further enhanced in the presence of lanthanide triflate.

Experimental characterization of drift-wave turbulence in the sheared, cylindrical slab

Plasma turbulence on a uniform density gradient with unfavorable magnetic curvature is investigated extensively in the Helimak device. The turbulence is strong with density and electrostatic potential fluctuation levels in excess of 40%. Measurements of the dispersion relation, k[subscript z], and k[subscript parallel lines] identify the the fluctuations with drift-waves, which propagate in the poloidal direction at the diamagnetic drift velocity and have a small, but nite parallel wavenumber. A non-zero phase shift between the density and potential fluctuations gives rise to turbulent cross-eld particle transport, which is measured using spectral techniques. In addition, the electrostatic drift-wave fluctuations have a small magnetic component that is driven by the turbulent parallel current [scientific symbols]. An examination of nonlinear processes associated with the plasma turbulence uncovers high levels of intermittency near the plasma edge and long-time persistence of the density fluctuations on the order of the parallel confinement time. An analysis of the bispectrum conrms the existence local and nonlocal three-wave interactions between unstable drift-waves although the turbulent saturation of the density fluctuation spectrum is likely due nonlinear processes acting in the time domain. / text

Para cumprir esta tarefa é utilizada, principalmente, a retificação com rebolo de diamante sob baixas taxas de remoção de material. O torneamento com ferramenta de diamante pode ser aplicado para a maioria das cerâmicas, mas esbarram em condições limites de remoção de material, como profundidade de corte nanamétricas, que se ultrapassadas mudam o regime de remoção de dúctil para frágil. Uma alternativa é a usinagem da peça no estado verde, a qual apresenta fácil usinagem com a diminuição da introdução de defeitos prejudiciais à resistência mecânica. Após a sinterização invariavelmente ocorrem distorções devido à distribuição heterogênea de gradientes de densidade que se localizam nas porções mais externas do compactado. Na tentativa de remover estes gradientes, o presente trabalho analisou a influência da remoção de sobrematerial no estado verde na distorção da peça após a sinterização. Corpos-de-prova de alumina, com 99,8% de pureza, foram torneados com sobremateriais de 1,0, 2,0 3,0, 4,0 e 5,0 mm por uma ferramenta de metal duro sob condições constantes de usinagem. Além da distorção, outras variáveis de saída foram analisadas, com desgaste de ferramenta, força de corte (Fc) e rugosidade média aritmética (Ra) do compactado em verde e sinterizado. Os resultados apontam uma redução da distorção da peça sinterizada de até 81,4% com o maior valor de remoção de sobrematerial em verde e a influência do desgaste da ferramenta na rugosidade da peça em verde e na correspondente peça sinterizada / Advanced ceramics machining is generally done with the objective of correcting distortions in pieces after sintering. In order to comply with this task grinding with a diamond grinding wheel is mainly used with low material removal rates. Turning can be done diamong tool to most of the ceramics, but is runs into very narrow material removal limit conditions, as nanometric cutting depths, which if exceeded change the removal system from ductile to brittle. An alternative is machining the piece on the green state, which presents easy machining without the introduction of harmful defects to mechanical resitance. However, after sintering there are invariably distortions caused by the heterogeneous distribution of density gradients that are located in the most outlying portions of the compacted. In an attempt to remove these gradients, this study examined the influence of different allowance values removed and the corresponding distortion after sintering. Alumina speciments are used for this procedure, with 99,8% purity, from which allowances of 1.0,2.0,3.0,4.0, 5.0 mm are removed by turning with a cermented carbine tool under constant machining conditions. Besides distortion, other output variables were analyzed, such as tool wear, cutting force and surface roughness of the green ceramics and the sintered. The results presents a reduction of the distortion of the sintered piece of 81,4% with the greater allowance removal values and the influence of tool wear on the green machining surface roughness and on the corresponding sintered piece

Modelling CO2-Brine Interfacial Tension using Density Gradient Theory

Knowledge regarding carbon dioxide (CO2)-brine interfacial tension (IFT) is important for petroleum industry and Carbon Capture and Storage (CCS) strategies. In petroleum industry, CO2-brine IFT is especially importance for CO2 – based enhanced oil recovery strategy as it affects phase behavior and fluid transport in porous media. CCS which involves storing CO2 in geological storage sites also requires understanding regarding CO2-brine IFT as this parameter affects CO2 quantity that could be securely stored in the storage site. Several methods have been used to compute CO2-brine interfacial tension. One of the methods employed is by using Density Gradient Theory (DGT) approach. In DGT model, IFT is computed based on the component density distribution across the interface. However, current model is only applicable for modelling low to medium ionic strength solution. This limitation is due to the model only considers the increase of IFT due to the changes of bulk phases properties and does not account for ion distribution at interface. In this study, a new modelling strategy to compute CO2-brine IFT based on DGT was proposed. In the proposed model, ion distribution across interface was accounted for by separating the interface to two sections. The saddle point of tangent plane distance where ( ) was defined as the boundary separating the two sections of the interface. Electrolyte is assumed to be present only in the second section which is connected to the bulk liquid phase side. Numerical simulations were performed using the proposed approach for single and mixed salt solutions for three salts (NaCl, KCl, and CaCl2), for temperature (298 K to 443 K), pressure (2 MPa to 70 MPa), and ionic strength (0.085 mol·kg-1 to 15 mol·kg-1). The simulation result shows that the tuned model was able to predict with good accuracy CO2-brine IFT for all studied cases. Comparison with current DGT model showed that the proposed approach yields better match with the experiment data. In this study, the thermodynamic properties were computed using Cubic Plus Association (CPA) equation of state, and the electrolyte contribution was accounted for by adding Debye-Huckel activity coefficient in the thermodynamic properties computation.

Towards monodisperse Silicon Nanocrystals: density gradient centrifugation applied on commercial gold nanoparticles

The application of silicon nanocrystals as non-toxic bio-labels and downconverters requires their uniform size distribution in order to minimize the inhomogeneous broadening of the photoluminescence peak. In this thesis, we set the basis for their size-separation via the density-gradient centrifugation method. To be more precise, we successfully apply this technique to separate 5 and 10 nm gold nanoparticles from an ensemble by using an engineered medium layer stack. In addition, we explain how atomic force microscopy is used to measure the size of the nanoparticles, with a particular attentionon the removal of unwanted solvent-related effects. As a future plan, we will implement the technique for the size-separation of silicon nanocrystals.

Evaluación espermática de semen de ovino tratado por la técnica de gradiente de densidad

La finalidad del presente estudio fue evaluar en 2 tiempos la calidad espermática de muestras de semen de ovino tratadas por la técnica de gradiente de densidad con respecto a muestras no tratadas. Se utilizó un total de 102 eyaculados de carnero que presentaron en promedio un volumen de 1.12 ± 0.34 ml, color blanquecino, aspecto cremoso, pH de 6.73 ± 0.25, concentración inicial de 3.54 x 109 ± 5.30 x 108 esp/ml y motilidad masal alrededor del grado 4. Posteriormente, cada eyaculado se dividió en un grupo control y experimental de volúmenes iguales. El semen del grupo control fue diluido con Triladyl® y el experimental, tratado con la técnica de gradiente de densidad. La tasa de recuperación espermática después del tratamiento fue del 31.50 ± 10.89%. Se encontró un aumento significativo (p<0.05) en la motilidad individual progresiva, el porcentaje de espermatozoides vivos y de espermatozoides con membrana intacta en las muestras tratadas con respecto al grupo control (92.07 ± 1.81% vs. 85.74 ± 1.75%, 78.42 ± 5.13% vs. 75.19 ± 4.59%, 78.55 ± 4.34% vs. 73.37 ± 4.48%, respectivamente); así como una disminución significativa (p<0.05) en el porcentaje de espermatozoides anormales en las muestras del grupo experimental con respecto al grupo control (8.41 ± 1.33% vs. 9.94 ± 1.73%). El resto de muestras de ambos grupos fue distribuido en pajillas de 0.25 ml y enfriadas hasta alcanzar los 5°C para su refrigeración por 24 horas. Las muestras tratadas con gradiente de densidad presentaron un aumento significativos (p<0.05), con respecto al grupo control, en motilidad individual progresiva, el porcentaje de espermatozoides vivos y el porcentaje de espermatozoides con membrana intacta (86.94 ± 3.14% vs. 82.44 ± 2.26%, 71.24 ± 4.11% vs. 68.82 ± 4.15%, 70.87 ± 3.11% vs. 67.98 ± 4.42%, respectivamente). En conclusión, la técnica de gradiente favoreció la obtención de un mayor número de espermatozoides vivos, de mejor motilidad y con membrana intacta, así como la disminución del número de espermatozoides anormales tanto en muestras frescas como refrigeradas. The purpose of the present study was to assess at 2 evaluation times the sperm quality from ram semen treated with the density gradient technique and to compare it against non-treated ram semen. A total of 102 ejaculates were evaluated. The mean macroscopic parameters were: volume of 1.12 ± 0.34 ml, creamy white color, creamy-dense appearance, pH of 6.73 ± 0.25, initial concentration of 3.54 x 109 ± 5.30 x 108 spz/ml and mass motility around grade 4. Then, each ejaculate was divided into two equal volume groups: the control and experimental group. For the control group, the sample was diluted in Triladyl®. The experimental group was treated with the density gradient technique. The recovery rate after treatment was 31.50 ± 10.89%.The results showed that, at zero hour, samples treated undergo a significant increase (p <0.05), compared with the control group, in individual progressive motility, percentage of living spermatozoa and the percentage of spermatozoa with intact membrane (92.07 ± 1.81% vs. 85.74 ± 1.75%, 78.42 ± 5.13% vs. 75.19 ± 4.59%, 78.55 ± 4.34% vs. 73.37 ± 4.48%, respectively). There was also a significant decrease (p <0.05) in the percentage of abnormal in samples from the experimental group compared to control group spermatozoa (8.41 ± 1.33% vs. 9.94 ± 1.73%). The remaining samples, from both the control and experimental group, were distributed in 0.25 ml straws and cooled to 5°C for 24 hours refrigeration. Then, the samples were evaluated. After 24 hours at 5°C, the samples treated with the density gradient technique showed significantly greater values (p <0.05), compared with the control group, in individual progressive motility, percentage of living spermatozoa and the percentage of spermatozoa with intact membrane (86.94 ± 3.14% vs. 82.44 ± 2.26%, 71.24 ± 4.11% vs. 68.82 ± 4.15%, 70.87 ± 3.11% vs. 67.98 ± 4.42%, respectively). In conclusion, in the present study, the density gradient technique allowed to obtain better a greater number of living spermatozoa with better motility, intact membrane and with lower abnormalities in fresh and refrigerated samples.

O presente estudo foi desenvolvido para verificar o efeito de diferentes forças de centrifugação na seleção espermática por gradientes de Percoll ® , através de avaliação de parâmetros de qualidade espermática e posterior desenvolvimento e qualidade de embriões bovinos produzidos in vitro (PIV). Parâmetros de estresse oxidativo também foram avaliados para verificar uma possível relação com os achados na avaliação de esperma. No experimento 1, amostras de sêmen de quatro touros foram homogeneizadas e submetidas a centrifugação em gradientes descontínuo de Percoll ® (30,60 e 90%) em diferentes forças: F1 (9000 X g), F2 (6500 X g), F3 (4500 X g) e F4 (2200 X g). No Experimento 2, as amostras de sêmen de cada touro foram processadas individualmente e centrifugadas em: F1 (9000 X g) e F4 (2200 X g). Todas as amostras espermáticas foram avaliadas quanto a concentração, motilidade, vigor, morfologia, espécies reativas de oxigênio (EROs) e integridade da membrana plasmática, sendo no experimento 2 avaliados ainda a peroxidação lipídica, defesas antioxidantes e desenvolvimento embrionário. Não foi observada diferença na concentração espermática nos sêmens submetidos a diferentes forças centrifugação. No Experimento 1, a percentagem de espermatozoides móveis foi superior (p <0,05) após centrifugação em F3 e F4 e a produção de EROs de F1 foi superior (p <0,05) em comparação com outras forças. No Experimento 2, quando o sêmen de cada touro foi processado individualmente, não foram observadas diferenças significativas nos parâmetros de qualidade espermática, peroxidação lipídica, defesas antioxidantes, taxa de clivagem e tempo médio da primeira clivagem entre F1 e F4. No entanto, o aumento da força de centrifugação reduziu a taxa de penetração e a fertilização normal (P<0,05). Este trabalho demonstrou pela primeira vez que a força de centrifugação de 2200 X g aumentou a penetração e a taxa de fecundação em relação à força de centrifugação habitual (9000 X g) utilizadas na separação de espermatozoides por gradientes descontínuos Percoll ® em bovinos. Estes resultados sugerem que esta força de centrifugação pode ser utilizada com sucesso na PIV de embriões bovinos, uma vez que não reduz a recuperação de espermatozoides e aumenta a taxa de fecundação. / Centrifugation by Percoll® gradient is the most widely used method in the preparation of bull sperm for the purpose of in vitro fertilization, however at the moment, no scientific study determined the best centrifugation speed to remove seminal plasma and other constituents, i.e. the speed at which the loss of sperm cells is minimized and where the spermatozoa in the pellet still remain functional. The present study was designed to examine the efficiency of different centrifugation forces in sperm separation Percoll by methods evaluating sperm quality parameters and subsequent development and quality of bovine in vitro production (IVP) embryos. Additionally, we evaluated oxidative stress parameters to verify a possible relationship with the findings in the sperm evaluation. In Experiment 1, the semen samples from each bull were pooled and submitted to centrifugation in discontinuous gradients Percoll (30,60 and 90%) at different forces: F1 (9000 X g), F2 (6500 X g), F3 (4500 X g) and F4 (2200 X g). In Experiment 2, the semen samples from each bull were done separately and submitted to: F1 (9000 X g) and F4 (2200 X g). All sperm samples were evaluated to the concentration, motility, vigor, morphology, reactive oxygen species (ROS) and integrity of the plasma membrane, and in experiment 2 also were evaluated lipid peroxidation, antioxidants assays and embryo development. No difference was observed in the concentration of sperm submitted to different centrifugation forces. In Experiment 1, the total percentage of motile sperm was increased (p < 0.05) after centrifugation in F3 and F4 and the ROS production to F1 was superior (p < 0.05) compared to other forces. In Experiment 2, when the bull semen was processed individually, no significant difference was observed in the sperm quality parameters assessment, lipid peroxidation, antioxidants assays, cleavage rate and average time of the first cleavage between F1 and F4, sires or interaction between them after Percoll. However, the increased force centrifugation reduced the rate of penetration and normal fertilization (P < 0.05). This work demonstrated for the first time that 2200 X g centrifugation force enhanced the penetration and fertilization rates in relation to usual centrifugation force (9000 X g) using sperm separation by discontinuous Percoll gradients in bovine. These findings suggest that this centrifugation force could be used with successful in the bovine IVP embryo since it does not reduce sperm recuperation and increases the fecundation rate.

Density-Based Separations in Aqueous Multiphase Systems: Tools for Biological Research and Low-Cost Diagnostics

Cells often exist in heterogeneous mixtures. Density provides a property to separate several types of cells from the mixed sample in which they originate. Density-based separation methods provide a standard method to quickly separate or enrich specific populations of cells, such as lymphocytes from whole blood. This dissertation explores the use of aqueous multiphase systems (AMPS) as self-forming step-gradients in density for the separation of cells. AMPS were first discovered over a hundred years ago as aqueous two-phase systems. Density as a tool to separate cells is at least as old. Despite this long history, the work in this thesis is the first work to use AMPS to perform density-based separations on cells. This combination provides a powerful technique to separate cells and enable new testing at the point-of-care. Chapter 1 provides a short overview of aqueous multiphase systems and density-based separations of cells. Chapter 2 describes the process of taking technology, including AMPS, from a demonstration in a laboratory to a large scale evaluation in a field setting. In Chapter 3 and Appendix I, AMPS provide a means to enrich reticulocytes from whole blood as a means to grow malaria parasites. Chapter 4 and Appendix II describe the development and proof-of-prinicple of a density-based diagnostic test for sickle cell disease (SCD) using AMPS. Chapter 5 and Appendix III detail the results of a large scale field evaluation of a rapid test for SCD using AMPS in Zambia. Demonstrations of AMPS for density- and size-based separations are provided in Appendices IV and V. Appendix VI demonstrates the general usefulness of density to separate crystal polymorphs with another density-based separation method: magnetic levitation in a paramagnetic fluid. High Performance Lipoprotein Profiling for Cardiovascular Risk Assessment

With the severity of cardiovascular disease (CVD) and the related mortality rate to this disease, new methods are necessary for risk assessment and treatment prior to the onset of the disease. The current paradigm in CVD risk assessment has shifted towards the multivariate approach over the individual use of traditional risk factors or lipid measurements. Through a combination of analytical techniques and multivariate statistical analysis, a novel method of cardiovascular risk assessment was developed. The analytical techniques employed include density gradient ultracentrifugation (DGU) and matrix assisted laser desorption ionization mass spectrometry (MALDI-MS) applied to human serum. These techniques provided detailed information about the characterization of the lipoproteins and their structural components, specifically the apolipoproteins belonging to high density lipoproteins (HDL). This information when combined with multivariate statistical analysis provided a method that accurately identified the presence of CVD in clinical studies between cohorts of subjects that had been previously diagnosed with CVD and cohorts of subjects that had been identified as healthy controls (CTRL) based on a clear angiography. The lipoprotein density profiles were divided into subclasses based on their density and measured using a fluorescent probe to tag the lipoprotein particles. Use of multiple ethylenediaminetetraacetic acid (EDTA) based solutes allowed for the manipulation of the density gradient formation in order to separate the lipoproteins by specific density ranges in order to achieve better baseline separation of the profiles. Application of the integrated fluorescence intensities for each subclass of lipoprotein to linear discriminant analysis/sliced inverse regression (LDA/SIR) and quadratic discriminant analysis (QDA) yielded an advanced and accurate form of risk assessment for CVD. This method was found to be highly accurate as well as identify potential atherogenic lipoprotein subclasses through studying the LDA/SIR prediction equation generated. It was also shown that the LDA/SIR equation could be used to monitor medical treatment and lifestyle change for their effects on the risk assessment model. Further study into the atherogenicity of HDL through analysis of the apolipoproteins using MALDI-MS led to identification of potential risk factors that could be added to the statistical analyses. These risk factors included mass differences in the Apolipoprotein A-I (Apo A-I) and Apolipoprotein C-I (Apo C-I) between CVD and CTRL samples as well as the presence of specific mass peaks related to Apolipoprotein A-II (Apo A-II) that were primarily found in the CVD samples. These differences, in addition to the lipoprotein density profile data, were found to increase the potential accuracy of CVD risk assessment. The combination of these methods has shown great potential in the assessment of CVD risk as well as the ability to increase researchers' understanding of the nature of VD and how to treat it.
