A síntese de heat shock proteins (HSP) é umas das formas de se avaliar a severidade do trauma sofrido pela polpa dental em consequência de um estresse térmico. Esta pesquisa avaliou in vitro danos celulares produzidos pelo estresse térmico gerado sobre a cultura de células macrófagos RAW 264.7 devido ao uso de fotopolimerizadores LED – Valo Led (Ultradent Products Inc) e Bluephase G2 (Ivoclar Vivadent Ltda.) – de alta irradiância. As células foram irradiadas com os LED diretamente sobre a base da placa de cultivo celular de 24 poços sem ou com interposição de dentina e resina gerando os seguintes grupos: G1 – grupo controle; G2 – Valo; G3 – Valo e dentina; G4 – Valo e dentina e resina; G5 – Bluephase; G6 – Bluephase e dentina; G7 – Bluephase e dentina e resina. Avaliou-se o metabolismo celular - pelo ensaio de MTT-, a produção de óxido nítrico (NO), a morfologia celular pela microscopia eletrônica de varredura (MEV) e análise da expressão gênica da HSP 70 pelo teste de Polymerase Chain Reaction em tempo real (PCR) de cada grupo. Os dados de MTT e NO foram avaliados pelos testes não-paramétricos (Kruskal-Wallis e Mann-Whitney) e os de PCR por testes paramétricos (ANOVA e Tukey). Resultados: os grupos G2 e G3 resultaram nos menores valores para metabolismo celular quando comparados com o G1 (p<0.05); os grupos G4, G6 e G7 apresentaram valores de metabolismo celulares estatisticamente superiores ao G1; a produção de óxido nítrico foi baixa para todos os grupos, porém o grupo G1 e o G7 apresentaram valores estatisticamente superiores aos demais grupos (p<0.05); os grupos G2, G3 e G5 apresentaram células com menor número de prolongamentos entre elas e presença de restos/fragmentos celulares evidenciando morte celular; os grupos G2 e G5 foram os únicos que apresentaram dados estatisticamente superiores quando comparado com o grupo G1 para a expressão gênica da proteína HSP 70. Os resultados sugerem que a irradiação com os LED induz vias de sinalização para reparar o tecido celular danificado. Contudo, estudos adicionais são necessários para melhor evidenciação dos resultados obtidos, visto que este é um estudo in vitro. / Vital tooth restorations involve operative care to preserve pulp integrity, since raising the pulp temperature can result in cellular damage. The heat shock protein synthesis (HSP) is known as a way to evaluate the severity of the trauma suffered by the dental pulp as a result of thermal stress. The present study evaluated in vitro cell damage caused by thermal stress generated on RAW 264.7 cell culture due to the use of high irradiance LED - Valo Led (Ultradent Products Inc) and Bluephase G2 (Ivoclar Vivadent Ltda.). The cells were irradiated with the LED units directly on the base of the 24-well cell culture plate without or with dentin and/or resin interposition generating the following groups: G1 - control group; G2-Valo; G3 - Valo and dentin; G4 - Valo and dentin and resin; G5-Bluephase; G6 - Bluephase and dentin; G7 - Bluephase and dentin and resin. Samples from each group were used to evaluate cell metabolism by the MTT assay, nitric oxide (NO) production, cell morphology by scanning electron microscopy (SEM) and analysis of the gene expression of HSP 70 by the real time Polymerase Chain Reaction (PCR) test. MTT and NO data were evaluated by non-parametric (Kruskal-Wallis and Mann-Whitney) and RT-PCR data by parametric tests (ANOVA and Tukey). According to the results, it was possible to observe that the G2 and G3 groups resulted in the lowest values for cellular metabolism when compared to the G1 (p <0.05). The G4, G6 and G7 groups had statistically higher values of cellular metabolism than the G1. The production of nitric oxide was low for all groups, and the G1, as well as G7, presented statistically higher values than the other groups (p <0.05). In the analysis of the cell morphology, the G2, G3 and G5 groups presented cells with less number of extensions between them and presence of cell debris/fragments evidencing cell death. Analysis of the gene expression of HSP 70 demonstrated that the G2 and G5 groups were the only ones that presented statistically superior data when compared to the control group for the gene expression of the HSP 70 protein. The results suggest that the irradiation with the LED induce signaling pathways to repair damaged cell tissue. However, additional studies are needed to better demonstrate the results obtained, since this is an in vitro study.
