Abstract Incidence of esophageal adenocarcinoma is rising rapidly in Western countries. The main risk factor for esophageal adenocarcinoma is Barrett’s esophagus. Barrett’s esophagus results from long-term gastroesophageal reflux disease. The gastrointestinal tract is colonized by bacteria, fungi and viruses forming the alimentary tract microbiome. Microbiome transformation is involved in pathogenesis of alimentary tract cancer and also in the development of Barrett’s metaplasia. Toll-like receptors (TLR) are molecules of the innate immune system and they are involved in bacterial and viral recognition and regulation of immune functions in the host and cancer cells. This thesis examined the effect of alimentary tract microbiome and cancer- on the function of TLRs in normal gastrointestinal epithelial cells. An additional focus of the thesis was also to assess the carcinogenetic effect of TLRs 1–9 in Barrett’s esophagus metaplasia – dysplasia – carcinoma sequence. Study material consisted of: a patient cohort, organ donors, conventional and germ-free mice. TLRs are expressed also in a “microbe-free” gut. There were significant differences in all TLRs between small- and large intestine of conventional mice and in humans. In germ-free mice that difference was not observed. Normal tissue sampled adjacent to the tumors of cancer patients can be used as controls in immunohistochemical TLR studies in gastrointestinal cancer Clinical data indicate that TLRs linearly increase toward dysplasia in Barrett’s esophagus. High cytoplasmic and nuclear TLR4 expression and TLR1 and 8 nuclear immunoreactivity in esophageal adenocarcinoma are associated with metastatic disease and poor prognosis. Based on our results, bacteria seem to downregulate TLR expression of the intestine. TLRs 1–9 apparently have a role in malignant progression of Barrett’s dysplasia. TLR1, TLR4 and TLR8 may represent a novel therapeutic target in esophageal adenocarcinoma. / Tiivistelmä Ruokatorven adenokarsinooma on länsimaissa nopeasti yleistyvä syöpätyyppi. Tämän syöpätyypin tärkein riskitekijä on Barrettin ruokatorvi, joka kehittyy pitkään jatkuneen gastroesofageaalisen refluksitaudin pohjalta. Ruuansulatuskanavassa on suuri määrä bakteereja, sieniä ja viruksia, jotka muodostavat yhdessä ruuansulatuskanavan mikrobiomin. Normaalin mikrobiomin muutokset ovat yhteydessä usean eri ruuansulatuskanavan syövän patogeneesiin ja myös Barrettin ruokatorven muodostumiseen. Tollin kaltaiset reseptorit ovat luontaisen immuniteetin molekyylejä, jotka osallistuvat bakteerien ja virusten tunnistukseen ja sääntelevät immuunivastetta sekä normaalitilanteessa että syövissä. Väitöskirjassa tutkitaan ruuansulatuskanavan mikrobiomin ja syövän vaikutuksia normaalien epiteelisolujen TLR:ien toimintaan. Lisäksi selvitetään TLR:ien karsinogeneettisiä vaikutuksia Barrettin ruokatorven metaplasia- dysplasia -adenokarsinoomasekvenssissä. Tutkimusmateriaalina käytetään potilaskohortista ja elinluovutuksista peräisin olevia potilasnäytteitä sekä normaalien ja bakteerittomien hiirien näytteitä. Tuloksemme osoittavat, että TLR:t ilmentyvät myös bakteerittomassa ruuansulatuskanavassa, ja TLR:en ilmentyminen oli merkittävästi voimakkaampaa ohutsuolessa kuin paksusuolessa normaaleilla hiirillä ja ihmisillä. Tätä eroa ei havaittu bakteerittomilla hiirillä. Ruuansulatuskanavan syöpien viereistä ja sen altistamaa tervettä kudosta voidaan käyttää terveenä kontrollina immunohistokemiallisissa TLR- tutkimuksissa. Kliinisessä aineistossa TLR:ien ilmentyminen kasvaa lineaarisesti kohti dysplasiaa Barrettin ruokatorvessa. TLR4:n korkea ilmentyminen solulimassa ja tumassa sekä TLR8:n ilmentyminen tumassa ovat yhteydessä metastaattiseen tautiin ja huonoon ennusteeseen. Tulosten perusteella bakteerit näyttävät heikentävän TLR:ien toimintaa suolistossa. Lisäksi kaikilla tutkituilla TLR:illä (1–9) näyttää olevan osuutta Barrettin dysplasian etenemisessä kohti syöpää. TLR1, TLR4 ja TLR8 ovat mahdollisia terapeuttisia kohteita ruokatorven adenokarsinoomassa.

Genetic background of spontaneous preterm birth and lung diseases in preterm infants:studies of potential susceptibility genes and polymorphisms

Abstract Each year in Finland, approximately 5.7% of infants are born preterm, i.e., before 37 completed weeks of gestation. Preterm birth is a major cause of mortality and several neonatal morbidities, especially the respiratory diseases. Infants born very preterm (<32 wk) are at higher risk of developing a chronic lung disease called bronchopulmonary dysplasia (BPD). The genetic factors predisposing to spontaneous preterm birth (SPTB) and BPD are incompletely known. The aims of this thesis project were to identify genetic factors that affect susceptibility to SPTB and BPD. Genetic case-control association studies were performed in mothers and infants of northern Finnish origin (SPTB study), or in multiple populations of very preterm infants of Finnish or European origin (BPD study). The candidate genes were selected based on their proposed roles in inflammation which is involved in both SPTB and BPD susceptibility. Additionally, the aim was to study the possible functional role of polymorphisms in the gene encoding surfactant protein B (SP-B) that have been shown previously to associate with pulmonary function. An association between Met31Thr polymorphisms in the gene encoding SP-D (SFTPD) and SPTB infants was found. The other collectin genes that were studied, encoding SP-A and mannose-binding lectin, did not associate with SPTB in mothers or infants. An intronic polymorphism in the gene encoding Kit ligand (KITLG) was associated with the risk of BPD in the northern Finnish and in the combined population that originated from Finland, Canada and Hungary. The role of KITLG in BPD was further supported by biomarker data, which showed higher concentrations of Kit ligand at the time of birth in infants that later developed BPD. The genes encoding interleukin 6 (IL-6), its receptors, IL-10, tumor necrosis factor alpha or glucocorticoid receptor did not associate with BPD susceptibility. Finally, a genetic variant 131Thr in the gene encoding SP-B (SFTPB) was associated with lower SP-B levels in vivo and delayed secretion in vitro. To date, there is no effective method to prevent SPTB, and especially the extremely preterm infants are at an increased risk of developing serious respiratory diseases. Better understanding of the mechanisms underlying both SPTB and BPD could help in the successful prediction of risk groups as well as in the design of new preventive and treatment strategies. / Tiivistelmä Noin 5,7 % lapsista syntyy Suomessa ennenaikaisesti, eli ennen kuin raskaus on kestänyt täydet 37 viikkoa. Ennenaikainen syntymä altistaa vastasyntyneen lapsen vakaville pitkäaikaissairauksille. Erityisesti hyvin pienillä keskosilla, jotka ovat syntyneet ennen 32. raskausviikkoa, on suurempi riski sairastua vakavaan hengitysvaikeuteen eli bronkopulmonaaliseen dysplasiaan, joka tunnetaan myös nimellä BPD-tauti. Perinnölliset tekijät vaikuttavat niin spontaanin ennenaikaisen syntymän (SEAS) kuin BPD-taudinkin taustalla, mutta nämä tekijät tunnetaan huonosti. Tässä väitöskirjatyössä pyrittiin tunnistamaan perinnöllisiä tekijöitä, jotka vaikuttavat SEAS:in ja BPD-taudin taustalla. Perinnöllisen taustan selvittämisessä ehdokasgeenien sisältämien muuntelevien kohtien esiintyvyyttä verrattiin terveiden verrokkien ja tautitapausten välillä. SEAS-tutkimuksessa tutkimusväestö koostui suomalaisista äideistä ja heidän lapsistaan. BPD-tutkimuksessa oli mukana hyvin ennenaikaisesti syntyneitä lapsia Suomesta, Kanadasta ja Unkarista. Tämän lisäksi kokeellisten tutkimusten avulla tutkittiin aiemmin keuhkosairauksiin liittyneen geenin muuntelevien kohtien osuutta sen koodaaman surfaktanttiproteiini (SP) B:n toiminnassa. Tutkimuksissa havaittiin SP-D:tä koodaavan geenin Met31Thr-polymorfismin olevan mahdollinen riskitekijä SEAS:lle lapsilla, mutta se ei selittänyt SEAS-riskiä äideissä. SP-A:ta ja mannoosia sitovaa lektiiniä koodaavilla geeneillä ei ollut yhteyttä SEAS-riskiin. Kit-ligandia koodaavan geenin intronissa sijaitseva polymorfismi selitti BPD-tautiriskiä pohjoissuomalaisessa sekä yhdistetyssä tutkimusväestössä. Lisäksi lapsilla, jotka myöhemmin sairastuivat BPD-tautiin, havaittiin suurempia Kit-ligandipitoisuuksia syntymähetkellä. Interleukiini 6:ta (IL-6), sen reseptoreita, IL-10:ta, tuumorinekroosifaktori-alfaa tai glukokortikoidireseptoria koodaavien geenien polymorfismien ja BPD-taudin välillä ei ollut yhteyttä. SP-B:tä koodaavan geenin Ile131Thr-polymorfismin Thr-variaatio liittyi alhaisempaan SP-B:n pitoisuuteen lapsivedessä sekä hidastuneeseen proteiinin tuottoon kokeellisessa solumallissa. Tulokset antavat uutta tietoa SEAS:n ja BPD-taudin perinnöllisestä taustasta. Tämä tieto voi auttaa synnytyksen käynnistymiseen sekä BPD-alttiuteen johtavien biologisten mekanismien selvittämisessä ja uusien hoitokeinojen kehittämisessä.

The Beneficial Effects of Hypercapnia, and the Detrimental Effects of Peroxynitrite, in Chronic Neonatal Lung Injury

Bronchopulmonary dysplasia (BPD) is a chronic neonatal lung injury (CNLI) affecting infants of < 32 weeks gestation, which has a significant associated morbidity and mortality. The hallmarks of BPD as seen in the current era are arrested alveologenesis and parenchymal thickening. Those most severely affected may develop pulmonary hypertension which worsens the prognosis. No effective preventive therapy exists. Generation of damaging reactive oxygen species is implicated in its development. The more recently recognized reactive nitrogen species may also contribute to this disease. Thus, there is considerable interest in preventive antioxidant therapies, but results to date have not been promising. Newborn rats, exposed to 60% O2 for 14 days, develop a parenchymal injury and pulmonary hypertension that resembles the morphological features of human BPD. Previous studies have shown that following exposure to 60% O2, a pulmonary influx of neutrophils is followed by that of macrophages. Inhibiting the influx of neutrophils prevents the generation of reactive oxygen species, while simultaneously enhancing postnatal lung growth. Other interventions have shown that development of pulmonary hypertension is dependent upon increases in both 8-isoprostane and its downstream regulator of vascular tone, endothelin-1. Gentler ventilation strategies, incorporated to minimize induction of stretch-mediated pro-inflammatory cytokines, have shown benefits of permissive hypercapnia in adult lung injury. Multicentre clinical trials of permissive hypercapnia in neonates have not shown benefit. Therapeutic hypercapnia has been demonstrated to have a protective effect of PaCO2 in both acute studies of ventilator-induced and ischemia-reperfusion injuries in animal models. In the studies reported herein, therapeutic hypercapnia was found to completely protect against CNLI and attenuate 60% O2-induced macrophage-derived protein nitration. The likely nitrating agent was macrophage-derived peroxynitrite. The critical role of peroxynitrite, in the development of chronic neonatal lung injury in this model, was confirmed using a peroxynitrite decomposition catalyst. This protected against the impairments of alveolarization and of pulmonary vascularization induced by 60% O2. These results suggest a more significant role for reactive nitrogen species than previously recognized. Finally, preliminary evidence is presented supporting a role for neutrophil-derived elastase in initiating the macrophage influx in the lungs, required for peroxynitrite generation, during 60% O2-mediated injury.

Posturálně lokomoční vývoj u dětí s vývojovou dysplazií kyčelních kloubů / Postural locomotar development in children with developmental dysplasia of the hip

The aim was to highlight the context of developmental dysplasia therapy with postural hip locomotor development for children aged 4 weeks to 12 months. General examination was carried out in 9 children with disabilities in hip joints, degrees IIa- III. without contractures of adductors. We investigated spontaneous activity, and spontaneous reactivity. We compared the clinical picture of hip joints with the ultrasound image. Investigation of spontaneous activity was done by aspection. The test of spontaneous reactivity was taken using video and photos. Experimental results of our study showed the incidence of pathology in the spontaneous activity of 88.89 % children. Spontaneous reactivity was pathology in all cases, with various tests are discussed in the work. Ultrasound findings correlated with clinical manifestation in 66,67 %, but we cannot say that the proportion of patients will generally be higher than 60 %. Consumo alimentar e concentrações séricas de micronutrientes: associação com lesões neoplásicas e câncer cervical / Diet and serum micronutrientes: association with cervical neoplasia and cancer

O câncer cervical é o segundo câncer mais comum entre as mulheres em todo o mundo. A infecção por Papilomavirus (HPV) do tipo oncogênico é causa necessária. Estudos internacionais sugerem importante papel de carotenóides e tocoferóis séricos e dietéticos na redução do risco para lesões precursoras, mas os resultados dos estudos prévios são inconsistentes. Indivíduos e métodos: O presente estudo de casos e controles de base hospitalar conduzido na cidade de São Paulo analisou a associação entre concentrações séricas de carotenóides (licopeno, &#946;-caroteno), tocoferóis (&#945;- e &#947;-), consumo alimentar e casos incidentes, com confirmação histopatológica, de neoplasia intraepitelial cervical (NIC) graus 1,2,3 e câncer cervical. O estudo incluiu 453 mulheres do grupo controle (sem lesões de colo uterino) e 4 grupos de casos (NIC1, n=140; NIC2, n=126; NIC3, n=231; câncer cervical, n=108) recrutadas em dois hospitais públicos de referência entre 2003 e 2005. Resultados. A concentração sérica de licopeno foi inversamente associada à NIC1, NIC3 e câncer cervical com os seguintes Odds Ratio (OR) (intervalo de confiança de 95%), respectivamente: 0,53 (0,27-1,00; p de tendência=0,05); 0,48 (0,22-1,04; p de tendência=0,05) e 0,18 (0,06-0,52; p de tendência=0,002) quando comparado o maior com o menor tercil após ajuste por variáveis de confusão e grupos de HPV. O maior tercil de &#946;-caroteno sérico foi inversamente associado ao câncer quando comparado ao menor: OR ajustado = 0,36 (0,13-1,03: p de tendência=0,04). Maiores concentrações de &#945;- e &#947;-tocoferóis foram inversamente associadas a NIC3 com redução do risco em 50%, após ajuste por covariáveis e HPV. Maior tercil de consumo do grupo de folhas verde-escuras, legumes e frutas de cor alaranjada ou amarela escura foi inversamente associado ao risco para NIC3: OR ajustado comparado ao menor foi 0,52 (0,27-1,00; p de tendência=0,05). Conclusão: Os resultados do presente estudo sugerem que maiores concentrações séricas de antioxidantes e o consumo de alimentos ricos em carotenóides podem reduzir o risco para NIC3 e câncer cervical em mulheres brasileiras. / Cervical cancer is the second most common cancer in women worldwide. Human papillomavirus (HPV) infection has been recognized to be a necessary cause for cervical cancer. Serum concentrations and dietary carotenoids and tocopherols have been associated with the risk for cervical dysplasia, but results from previous studies were not consistent. Subjects and methods: In this hospital-based case-control study conducted in São Paulo city, Brazil, we evaluated the association of serum carotenoids (&#946;-carotene, lycopene), tocopherols (&#945;- and &#947;-), and dietary intakes with the risk of incident histologically confirmed cervical intraepithelial neoplasia (CIN) grades 1, 2, 3, and cervical cancer. The sample included 453 controls and four groups of cases (CIN1, n= 140; CIN2, n= 126; CIN3, n= 231; invasive cancer, n=108) recruited from two major public hospital-based clinics between 2003 and 2005. Results. Concentrations of serum lycopene were negatively associated with CIN1, CIN3 and cancer, with odds ratio (OR) (95% confidence interval) for the highest compared with the lowest tertile of 0.53 (0.27-1.00, p trend=0.05) for CIN1, 0.48 (0.22-1.04, p trend=0.05) for CIN2 and 0.18 (0.06-0.52, p trend= 0.002) for cervical cancer adjusted for confounding variables and HPV status. The adjusted OR for cancer of the highest versus the lowest tertile of serum &#946;-carotene was 0.36 (0.13-1.03, p trend= 0.04). Increasing concentrations of serum &#945;- and &#947;-tocopherols were associated with 50% decreasing risk of CIN3, for the women in the highest compared with the lowest quartile after adjusting for confounding variables and HPV. Increased dietary intakes of dark green and deep yellow vegetables and fruits were inversely associated with CIN3 among women in the highest compared to the lowest tertile (adjusted OR= 0.52, 95%CI = 0.27-1.00; p trend=0.05). Conclusion: These results support the evidence that high serum levels of antioxidants as well as dietary intakes may reduce the risk for CIN3 and invasive cancer in Brazilian women.

Caracterização clínico-histopatológica e avaliação terapêutica de fotoprotetor nas queilites actínicas / Clinical histopathological features and therapeutic evaluation of lip sunscreen on actinic cheilitis

A queilite actínica (QA) é uma doença que afeta a semimucosa labial, principalmente inferior, causada pela exposição crônica à radiação solar ultravioleta (UV). É uma lesão potencialmente maligna extremamente frequente e considerada precursora da grande maioria dos cânceres de lábio, em pacientes não fumantes. A etiologia da QA permite medidas preventivas de fotoproteção solar, porém grande parte da população não as utiliza em função do desconhecimento do risco potencial e falta de informação. De outro lado se acredita que o dano causado ao lábio é irreversível e que a adoção de medidas fotoprotetoras apenas limita o dano e que quadros de displasia intensa deveriam ser tratados de forma cirúrgica radical. O objetivo principal deste estudo foi avaliar o efeito do uso de fotoprotetor labial, na forma de bastão, sobre parâmetros clínicos de severidade e grau de displasia histológica por meio de acompanhamento clínico e investigações microscópicas sucessivas. Foram selecionados pacientes que procuraram a Clínica de Diagnóstico Oral da FOUSP e apresentavam sinais clínicos sugestivos de QA. Os pacientes foram orientados quanto às condições de participação no trabalho de pesquisa e assinaram termo de consentimento livre e esclarecido. Após a confirmação de QA, em sequência ao procedimento de biópsia e análise histopatológica, os pacientes foram efetivamente incluídos no grupo de pesquisa. O local da biópsia incisional foi escolhido a partir do exame clínico e teste do azul de toluidina. Os pacientes foram orientados quanto ao uso do fotoprotetor e de barreiras físicas, sendo acompanhados a cada três meses para reavaliação clínica e reforço das medidas de fotoproteção. Após pelo menos seis meses de uso do fotoprotetor uma nova biópsia incisional foi realizada para avaliação histopatológica do quadro. Os espécimes de biópsia dos dois momentos foram analisados inicialmente por diversos patologistas (via rotina clínica-laboratório) e posteriormente por uma única patologista, mais especializada nessa área, cega em relação aos resultados anteriores. Vinte pacientes compuseram a casuística do trabalho, 12 homens e 8 mulheres, com idade média de 63 anos. De acordo com a análise histopatológica realizada via rotina do laboratório de patologia cirúrgica, dos 20 pacientes, dez apresentaram melhora das displasias e do quadro clínico; quatro permaneceram com as mesmas displasias, porém com melhora do quadro clínico; seis apresentaram piora do quadro histológico, porém houve melhora do quadro clínico em quatro deles. A análise realizada pela única patologista produziu resultados bastante diversos, registrando piora dos graus de displasia do primeiro para o segundo momento. Concluímos que a utilização do fotoprotetor produz melhora dos sinais clínicos exibidos pelos pacientes com QA e que os parâmetros clínicos não correspondem aos sinais de displasia histopatológica. Trata-se de doença ainda pouco reconhecida e valorizada tanto pela população quanto pelos profissionais da área de saúde com responsabilidade sobre essa área anatômica e estudos adicionais são requeridos para melhor compreensão da doença, normalização dos critérios de displasia microscópica e estabelecimento de características preditivas em relação à evolução carcinomatosa da doença. / Actinic cheilitis (AC) is an injury that mainly affects the lower lip vermilion, and it is caused by chronic exposure to ultraviolet light. It is a very frequent potentially malignant disorder and it is considered the precursor of most lip cancers, in non-smoker patients. The AC etiology provides preventive measures of sun light protection, but most people do not use it because are unaware of the potential risks and lacking knowledge. On the other hand it is believed that the damaged caused to the lip is irreversible and the introduction of photoprotective measures just contain the damage and cases of severe dysplasia should be treated with radical surgery. The main purpose of this study was to evaluate the use of sunscreen lip balm in stick form, under clinical parameters of severity and degree of histological dysplasia by clinical follow-up and sequential microscopic researches. Patients selected were those who sought Clínica de Diagnóstico Oral in FOUSP and presented suggested signs of AC were selected. They were informed about the research terms of participation as well as signed a free and informed consent term. After confirm the diagnosis of AC with biopsy procedure and histopathological analysis, patients were included in the research group. The incisional biopsy site was selected by clinical exam and toluidine blue test. Patients were instructed about the use of sunscreen lip balm and physical protection, all of them had follow up appointments every three months to clinical evaluation and reinforcement of the use the use of lip sunscreen. After at least six months of lip sunscreen use, a new incisional biopsy was performed for histopathological evaluation. The biopsy specimens of the two procedures were initially analyzed by several pathologists (via laboratory routine) and later by a single blinded pathologist, more specialized in this field. Twenty patients were included in the sample of the study, 12 men and 8 women with a mean age of 63 years. According to histopathological analysis by laboratory routine, ten out of the twenty patients showed improvement in dysplasia and clinical signs; four remained with the same dysplasias, but with improvement of clinical signs; six had worsening results in histopathological analysis, however four of them presented improvement of clinical signs. The analysis conducted by the single pathologist produced very different results, registering worsening degrees of dysplasia from the first to the second procedure. In conclusion the use of sunscreen lip balm produces improvement of clinical signs on the patients with AC as well as the clinical parameters do not match with the signs of histopathological dysplasia. It is a poorly recognized and undervalued disease by both population and healthy professionals with responsibility for this anatomic area. Additional studies are required for better understanding of the disease, standardization of the criteria of microscopic dysplasia and establishment of predictive features to the progression of carcinoma.

O objetivo da pesquisa foi avaliar os efeitos da terapia de ondas de choque extracorpórea em pacientes com osteoartrose coxofemoral secundária a displasia. Foram analisados 30 cães (Grupo I) com auxílio da plataforma de baropodometria, portadores de osteoartrose bilateral da articulação coxofemoral, que receberam como tratamento ondas de choque radiais somente em uma das articulações acometidas de modo aleatório que foi comparado ao membro contralateral com o acompanhamento de 3 meses, 18 animais (Grupo II) com displasia coxofemoral e osteoartrose bilateral medicados com condroprotetor e também comparado ao banco de 20 animais hígidos (Grupo III). O protocolo de tratamento envolveu três sessões de 2000 pulsos cada com frequência de 10 Hz de 2-3,4 bars, com intervalo de 7 dias, e a aplicação realizada sob sedação e curta anestesia. A avaliação mensal quantitativa do apoio dos membros e da eficácia do tratamento foi realizada com análise cinética utilizando sistema de baropodometria, mensurando-se o pico de força vertical (PFV) e o impulso vertical (IV).O grupo I e II tiveram em média valores do PFV e IV inferiores ao grupo III. No grupo I, observou-se um aumento em média de 10,6% do PFV e de 10,4% do IV do membro tratado, assim como uma melhora na simetria. O grupo II apresentou um aumento em média do membro pélvico esquerdo de 9% do IV, porém houve piora de 11% na simetria deste mesmo parâmetro. Os dados da análise cinética no membro tratado do grupo I atingiram em média valores próximos, mas ainda inferiores, ao grupo III de animais hígidos. Estas avaliações verificaram a eficácia das ondas de choque como tratamento conservativo de cães portadores de osteoartrose secundária a displasia, sugerindo efeitos benéficos de forma quantitativa para uma melhor locomoção. / The objective of this research was evaluate the extracorporeal shockwave therapy (ESWT) effects in patients with hip osteoarthritis secondary to hip dysplasia. Thirty dogs (Group I) with bilateral hip osteoarthritis were evaluated trough pressure walkway system. They had received unilateral radial shockwaves for treatment in random selection. The treated limb was compared with contra lateral limb during a follow up of 3 months. A comparison with the treated group with eighteen dogs under conservative treatment (Group II) and twenty healthy dogs (Group III) was also performed. Protocol treatment included three sessions of 2000 pulses each in a frequency of 10Hz with a pressure between 2 and 3.4 bar performed weekly using chemical restraint and short general anesthesia. Monthly assessment with kinetic analysis had done it with pressure walkway system. The peak of vertical force (PVF) and vertical impulse (VI) were measured. Group III kinetic analysis mean values were higher than groups I and II. Our results of group I suggest 10.6% of improvement in the treated limb in the PVF and 10.4% in the VI. In this group was also detected an improvement in the symmetry. The group II showed an improvement of 9% of VI only in the left pelvic limb but the symmetry decreased in 11% in this parameter. The kinetic analysis data of group I reached mean values of the treated limb close to healthy dogs of group III but remains slightly lower. These evaluations verified the efficacy of ESWT suggesting benefic quantitative effects in dog\'s locomotion.
