Na realidade, a principal finalidade do discurso desencannado é desconstruir toda a seriedade do fazer publicitário tradicional, provocando, assim, o riso em seus destinatários. Para que tais análises fossem, de fato, empreendidas, recorreu-se a alguns pressupostos da Teoria Semiolinguística de Análise do Discurso, cunhada por Patrick Charaudeau, como é o caso dos conceitos de sujeitos do ato de linguagem, Processo de Semiotização do Mundo, Contrato de Comunicação, Visadas Discursivas e Competências Comunicacionais, haja vista que todos esses anúncios só são possíveis dentro de um novo universo criado na e pela linguagem, posto que subvertem o quadro de referência tradicional ao qual se reportam os sujeitos do ato de linguagem, principalmente, no que tange às visadas discursivas, escolhidas para fazer parte do discurso desencannado. As noções de ethos e pathos também foram resgatadas, afinal, a imagem que o sujeito enunciador quer passar de si no discurso, bem como as estratégias de patemização que escolhe para fazer parte desse discurso, são fundamentais na compreensão e na interpretação dos textos desencannados. Recorreu-se, ainda, aos estudos de alguns autores sobre publicidade canônica para que se pudesse estabelecer quais das estratégias foram utilizadas, quais, no entanto, foram deixadas de lado e quais, por sua vez, foram subvertidas. Por fim, como a intenção primeira do sujeito enunciador do Desencannes é fazer seu destinatário rir, tornou-se preciso explorar as concepções de humor de Freud (1996), Bergson (2001), Bakhtin (2013) e Almeida (1999) a fim de que se pudesse entender por que e como o riso foi produzido. Assim sendo, torna-se possível estabelecer que as peças publicitárias do Desencannes são, na realidade, publicidades às avessas, já que não têm a intenção de vender uma marca, mas, sim, de colocar em xeque toda a rigidez do discurso publicitário canônico, afirmando que publicidade “boa de verdade” é aquela que faz seu destinatário rir e não aquela que se prende a paradigmas e modelos engessados / The present work aims at analizing some fictional works of publicity, available in a comedy website named Desencannes, which discusses how the referred works subvert the catchy strategies used in the mainstream publicity, as well as how they use other ones. In fact, the main aim of the Desencannes speech is to deconstruct the serious tone of the traditional publicity and make the audience laugh. In order to make the analisis we used the Theory of the Semiolinguistic Speech Analisis, coined by Patrick Charaudeau, for instance, using its concepts of language act subjects, World Semiotization Process, Communication Contract, Discursive Target and Communication Competencies, given the fact that these advertisements are only possible in the new linguistic universe created by the language and in it because they subvert the traditional reference board to which the subjects of the linguistic act refer, especially in relation to the discursive targets, chosen to be part of the desencannado speech. The notions of ethos and pathos have also been used, as the image wished to be transpassed by the subject, as well as the patemization strategies chosen to be part of this speech, are fundamental in the comprehension and interpretation of the desencannados texts. We have also used some works about canonical publicity so as to establish which strategies have been used, the others that have been put aside, and finally the subverted ones. Finally, because the intentions of the Desencannes subject is to make the audience laugh, it was necessary to exploit the concepts of humor of Freud (1996), Bergson (2001), Bakhtin (2013) and Almeida (1999) to understand how laughter is produced. Therefore it is possible to establish that the advertisements of the Desencannes are, in fact, inverted, as there is no intention of selling a product but to put into question the rigidity of the canonical publicist speech, showing that “good publicity” is the one that makes the audience laugh and not the ones stuck to paradigms and models

Att koppla samman nätverk över geografiskt olika platser har blivit allt vanligare i dagens samhälle. Rapporten kommer belysa olika WAN-lösningar för att göra detta möjligt. De olika lösningar som kommer belysas i denna rapport är att hyra egen fiber, lager2-länk, MPLS och även VPN. Det kommer göras jämförelser mellan dessa lösningar när det gäller kostnad, säkerhet, hastighet och tillförlitlighet. Detta har gjorts med vetenskapliga artiklar och intervjuer men rapporten innefattar även en praktisk tillämpling i form av ett experiment. / To connect networks across geographically diverse sites has become increasingly common in todays society. This report will highlight the various WAN solutions to make this possible. The different solutions that will be highlighted in this report is rent own fiber , Layer 2 link, MPLS , and VPN. There will be comparisons between these solutions in terms of cost, safety , speed and reliability. This has been done with scientific articles and interviews , but the report also includes a practical laboration.

Etude du système Xer au travers de la transmission verticale et horizontale de l'information génétique chez les bactéries / Study the Xer system through the vertical and horizontal genetic transmission in bacteria

Les génomes bactériens sont le siège de deux phénomènes de transferts de l'information génétique: les transferts verticaux et horizontaux. Leur coéxistence ce qui implique une délicate balance entre maintenance et instabilité des génomes. L'information génétique des bactéries est généralement portée par des réplicons circulaires : chromosomes et plasmides. Un des sérieux désavantages des réplicons circulaires est leur grande sensibilité aux réarrangements causés par recombinaison homologue. Un nombre impair de crossing-over pendant ou après la réplication de ces réplicons entraine la formation de molécules dimériques. Ces dimères correspondent à une fusion covalente des deux copies du réplicon, non-résolus les dimères ne ségrégeront pas correctement au moment de la division cellulaire. La résolution des formes multimériques des plasmides et chromosomes circulaires est médiée par un mécanisme de recombinaison spécifique de site efficace et extrêmement contrôlé : le système Xer. Les mécanismes de résolution des dimères de chromosome et de plasmide sont différents. De plus, bien que le système de résolution de dimères de chromosome soit contrôlé dans le temps et dans l'espace, chez de nombreuses bactéries il est détourné de son rôle principal par des éléments génétiques mobiles appelés IMEXs (Integrative Mobile Elements exploiting Xer). Le système Xer est alors impliqué dans les transferts verticaux et horizontaux de gènes, illustrant comment une même machine moléculaire peut intervenir dans plusieurs processus biologiques. Pour cela, des acteurs externes différents vont jouer un rôle clé dans le contrôle d'une machine Xer centrale et ainsi apporter une diversité de mécanismes, chacun dédié à une processus biologique propre. Afin de mieux comprendre les contrôles différentiels du système Xer, je me suis intéressé aux éléments génétiques mobiles. Via l'étude de la stabilité intra-chromosomique des IMEXs et de la résolution des dimères de certains grands plasmides, j'ai pu montrer que FtsK, la protéine activatrice de la résolution des dimères de chromosome, est impliquée dans la stabilisation des éléments acquis par transferts horizontaux de gènes. / The genetic information of bacteria is generally carried by circular replicons: chromosomes and plasmids. One of the serious disadvantages of circular replicons is their high sensitivity to rearrangements caused by homologous recombination. An odd number of crossing-over, during or after the replication of these replicons, results in the formation of dimeric molecules. These dimers correspond to a covalent fusion between the two copies of the replicon. If they are not resolved, the dimers will not segregate properly at the time of cell division. The resolution of multimeric forms of circular plasmids and chromosomes is mediated by an efficient and highly controlled site-specific recombination mechanism: the Xer system. The mechanisms of resolution of the chromosome and plasmid dimers are different. In addition, even if the chromosome dimer resolution system is controlled in time and space, in many bacteria it is hijacked by mobile genetic elements called IMEXs (Integrative Mobile Elements Exploiting Xer). The Xer system is then involved in the vertical and horizontal transfer of genes, illustrating how the same molecular machine can intervene in several biological processes. For this, different external actors will play a key role in controlling a central Xer machine and thus bring a variety of mechanisms, each dedicated to a specific biological process. In order to better understand the differential controls of the Xer system, I focused on mobile genetic elements. By studying the intra-chromosomal stability of IMEXs and the resolution of large plasmids dimers, I was able to show that FtsK is involved in the stabilization of the acquired mobile genetic elements.

Conserving spaces of memory and heritage: the complexities, challenges and politics of the stone wall project on bluestone quarry at Robben Island

Magister Artium - MA / This thesis is a critical study of a conservation project on restoration of a Stone Wall at Bluestone Quarry on Robben Island, a world heritage site. The Stone Wall was built by the ex-political prisoners, in the early 1960s, as part of their hard labour. The thesis mainly focuses on the contestations that arose during the twelve year period of the project (2002 to 2014) among the stakeholders that included the ex-political prisoners, the environmentalists, the heritage managers and South African Heritage Resource Agency. Central to this study was the question, when a restoration project of a significant heritage site is informed by oral history and memories how are the concerns of diverse range of interest groups addressed and resolved? The thesis is grounded in the theoretical frameworks of sites of memory, heritage and conservation. The study involved both archival research and oral history as its research methodologies. The thesis shows that during the restoration project of the Stone Wall, the proposed designs had impacts on authenticity and biodiversity of the site. The various stakeholders that were involved debated and sought ways to influence decisions in resolving these impacts. Where necessary compromises were made. The thesis argues that during the project, oral history and memory work, and by extension the ex-political prisoners, had a significant role in influencing some of the important decisions. Among other things, the thesis seeks to provide a critical understanding of issues of heritage and conservation management on sites that are of cultural/historical significance.

Organization management challenges of national heritage institutions in South Africa : a case study of the Robben Island Museum (RIM)

Masters in Public Administration - MPA / In January 1997, Robben Island Museum (RIM) opened its doors to the public as a tourist destination. This was done without any formal management structure or strategic planning, to cope not only with its mandate as the first National Museum of the new democratic South Africa, but also to meet the demand of the great interest of visitors to see this prison where the icon of the liberation struggle, Nelson Mandela was incarcerated for 27 years. This operational take-off without formal administrative management structure and strategic planning was understandable given the social and political transitional climate in the country. The effect thereof as the old adage states ‘failure to plan is planning for failure’ seems to hold true for RIM who is struggling to become a well-managed national heritage site. RIM over the years have been beset with governance and management challenges and enjoyed continual bad publicity. The study’s aim was to bring an understanding of what the major governance and managerial challenges were and to make recommendations to address those. The study was guided by a central research question namely What were the administrative and management challenges of RIM and in what ways did it impact on the operational effectiveness of RIM as a national museum and World heritage site. A number of findings emanated from the study. A key one was that clarity of roles and responsibilities of the council and the management is required. Furthermore, the vision and mission of RIM must inform the formulation of strategies to give effect thereto. This in turn must be the base for the development of an appropriate organisational structure to implement the strategies. A number of recommendation stems from the research. An important one was that appointment must be on the basis of ‘fit for purpose’. This means that staff with the right skills and capacity should be appointed as a matter of priority. In conclusion, the findings of the study could be used by other heritage organisations faced with similar organisational challenges. / Robben Island Museum Training and Development Staff Bursary

Hurricane Preparedness in a Construction Site: a Framework to Assess the Construction Companies’ Current Practices

Civil infrastructure construction sites including incomplete structures and unsecured resources are among the most vulnerable environments to hurricane conditions. Hurricane driven damages cause disruption of construction sites and considerable schedule delays, and thus negatively impact the efficiency of the construction projects. This research aims to study current best practices on securing construction sites from hurricane conditions and evaluate the performance of the preparedness plan. To do that, first, an interview-survey is conducted with key project personnel from multiple construction companies. Then, the insight from the interviews serves to do a Micro and Macro Environmental Analysis. For further analysis, a Balanced Scorecard is used to suggest metrics to measure and improve the performance of the Hurricane Preparedness Plan. The findings from this research improves the organizational processes and enhances the assessment of disaster preparedness, which ultimately generates new and highly specific knowledge on disaster mitigation and preparedness guidelines for construction sites.

This diploma thesis deals with automatic task processing, especially with automatic processing of forms on web pages. The thesis throws light on the theoretical and practical matter. At the beginning of this thesis are described the reasons, why this topic were chosen. Also there are mentioned the possible uses of application, realized on this principles. Foremost simplification of retrieval of data with standardized (but general) format from common users, which could be used for data mining process. For creating this kind of application is described concepts of its implementation, including description of problematic parts and their possible solution. In this part is mentioned algorithm, which deals with security limitation of current web browsers -- so-called cross-site scripting. One part of this thesis is description of current commercial and non-commercial solutions, which at least partly fulfills the demands of the application. At the conclusion is analyzed functionality of created application and proposed advancements and improvements for creating new versions of application
