Macroinvertebrate Colonization and Assemblages Associated with Aquatic Macrophytes in a Newly Created Urban Floodway Ecosystem, Dallas, Tx

A study of macroinvertebrate colonization and assemblages, including secondary productivity of the familiar bluet damselfly or Enallagma civile Hagen (Odonata: Coenagrionidae), associated with the aquatic macrophytes Heteranthera dubia (Jacq.) MacMill. (water stargrass) and Potamogeton nodosus Poir. (American pondweed) was conducted at the Dallas Floodway Extension Trinity River Project (DFE) Lower Chain of Wetlands (LCOW), Dallas, TX, from September 2010 through November 2011. Macroinvertebrate abundance, taxa richness, Simpson's index of diversity, and Simpson's evenness from the two macrophytes and from three different wetland cells of varying construction completion dates, water sources, and native aquatic vegetation establishment were analyzed along with basic water quality metrics (temperature °C, pH, dissolved oxygen mg/L, and conductivity µs/cm). E. civile nymphs were separated into five developmental classes for secondary productivity estimations between macrophytes and wetland cell types. Mean annual secondary productivity in the DFE LCOW among two macrophytes of E. civile was 1392.90 ash-free dry weight mg/m²/yr, standing stock biomass was 136.77 AFDW mg/m2/yr, cohort production / biomass (P/B) ratio was calculated to be 4.30 / yr and the annual production / biomass (P/B) ratio was 10.18 /yr.

Xanthomonas albilineans est l'agent causal de l'échaudure des feuilles, une des principales maladies bactériennes de la canne à sucre dont l'impact peut être très important lorsque des variétés sensibles sont infectées au champ. Les mécanismes qui régissent les interactions entre cet agent pathogène et la canne à sucre sont encore très peu connus. Les objectifs de ce travail étaient (i) d'identifier des déterminants moléculaires impliqués dans la survie épiphyte de X. albilineans et (ii) de préciser la localisation de la bactérie dans les tissus de la canne à sucre. Parmi les facteurs étudiés, les polysaccharides de surface et une protéine de la membrane externe (XaOmpA1) de X. albilineans s'avèrent indispensables pour la survie épiphyte de cet agent pathogène. En revanche, la molécule signal diffusible DSF et les métabolites secondaires codés par les gènes NRPS (« Non Ribosomal Peptide Synthetases ») ne sont pas requis pour l'installation de la bactérie en surface des feuilles, au moins en l'absence d'autres microorganismes compétiteurs. Toutefois, la colonisation optimale de la phyllosphère de la canne à sucre nécessite la présence d'un système DSF/RpfGC intact. Dans la deuxième partie de ce travail, nous avons vérifié la localisation in planta de X. albilineans par microscopie confocale, immunocytochimie, et microscopie électronique à transmission. Les observations microscopiques réalisées ont permis de montrer que X. albilineans n'est pas limitée au xylème de la canne à sucre, comme on le considérait jusqu'à présent. Bien au contraire, cette bactérie est capable de quitter le système vasculaire de sa plante hôte et de pénétrer dans d'autres types cellulaires, notamment les cellules du parenchyme non vasculaire. Il s'agit là, à notre connaissance, d'un nouveau mécanisme de colonisation d'une plante par une bactérie phytopathogène qui reste à décrypter. / Xanthomonas albilineans is the causal agent of leaf scald, a lethal disease of sugarcane that can significantly impact infected susceptible varieties in the field. The mechanisms that govern the interactions between this bacterial pathogen and its host plant are not well known. The objectives of this study were (i) to identify molecular factors involved in epiphytic survival of X. albilineans and (ii) to verify the localization of X. albilineans in sugarcane tissues. Among the studied factors, surface polysaccharides and an outer-membrane protein (XaOmpA1) of X. albilineans were crucial for epiphytic survival of this pathogen. Secondary metabolites synthesized by non-ribosomal peptide synthetases and the diffusible signal factor DSF were not critical for survival of X. albilineans on the sugarcane leaf surface, at least in absence of competing microorganisms. However, an intact DSF/RpfGC system was necessary for optimal colonization of the phyllosphere. In the second part of this study, we verified in planta localization of X. albilineans by confocal microscopy, immunochemistry and transmission electron microscopy. Microscopic observations allowed us to show that X. albilineans is not a xylem limited bacterium as it was believed until now. This pathogen is able to invade numerous cellular types including vascular and non-vascular parenchyma cells. To our knowledge, this is a novel invasion strategy of a plant pathogenic bacterium that has not previously been described, and that remains to be deciphered.

Amostragem sustentável de bromélias nativas para estudos ambientais em unidades de conservação / Sustainable sampling of native bromeliads for environmental studies in conservation units

O Estado de São Paulo é caracterizado por um mosaico de paisagens formado por fisionomias vegetais como a floresta de restinga, a floresta ombrófila densa, a floresta estacional semidecidual e o cerradão, representadas, respectivamente, pelas unidades de conservação Parque Estadual Ilha do Cardoso, Parque Estadual Carlos Botelho, Estação Ecológica dos Caetetus e Estação Ecológica de Assis. Mesmo protegidas legalmente, essas áreas ocorrem, na maior parte, em regiões bastante alteradas pela ação antrópica. As epífitas, especialmente as bromélias, respondem diretamente a essas intervenções, sendo interessantes para diversos estudos ambientais. As bromélias apresentam estratégias diferenciadas para a absorção de elementos químicos a partir da serrapilheira depositada no tanque (bromélia-tanque) ou diretamente da atmosfera (bromélia atmosférica). Entretanto, o emprego dessas plantas para estudos de qualidade ambiental é comprometido por etapas da análise química, principalmente relativas à amostragem e à contaminação da superfície das folhas com material particulado. De acordo com o procedimento usual, a amostragem envolve a retirada de diversas plantas, acarretando possíveis danos aos ecossistemas. Por essa razão, a proposta desse trabalho consiste em uma metodologia inovadora de amostragem denominada sustentável, em que são retirados discos de folhas das bromélias sem a necessidade de remoção das rosetas das áreas estudadas. Considerando as diferentes formações vegetacionais, a diversidade de bromeliáceas, as características nutricionais das plantas epifíticas e a problemática de amostragem de bromélias nativas em unidades de conservação, este presente trabalho teve como objetivos específicos (I) a avaliação da composição química de bromélias nos diversos tipos vegetacionais, (II) a avaliação da composição química das diferentes espécies de bromélias, (III) a avaliação da contribuição dos modos nutricionais do tipo tanque e atmosférico e dos modos de crescimento epifítico e terrícola para a composição química de folhas de bromélias (IV) a avaliação do efeito da lavagem sobre a composição química das folhas, (V) a comparação dos resultados da amostragem sustentável com aqueles da amostragem usual e (VI) a avaliação da variabilidade da composição química dos diferentes terços das folhas e terços da planta. A análise por ativação neutrônica instrumental (INAA) foi empregada por permitir a determinação simultânea de diversos elementos químicos em elevado nível metrológico. Para cada estudo realizado, as amostras tiveram preparo específico, permitindo a análise por INAA. As bromélias apresentaram diferenças quanto à composição química entre as unidades de conservação, embora não tenha sido verificada um padrão único de distribuição dos elementos químicos, corroborando composição química inerente de cada espécie. Bromélias-tanque mostraram maiores concentrações dos elementos químicos essenciais. As folhas das plantas estudadas apresentaram concentrações levemente maiores no modo epifítico quando comparadas com aquelas crescendo sobre o solo. Os resultados obtidos demostraram a necessidade de um procedimento de lavagem para minimizar a influência de material particulado sobre as concentrações de elementos químicos nas folhas. A nova amostragem sustentável promoveu resultados comparáveis, principalmente para o terço inferior, com a amostragem usual. Ainda, aumentou-se a confiabilidade dos resultados dada a redução do número de etapas do preparo de amostras proporcionado por esta metodologia de amostragem / The State of São Paulo, Brazil, is characterized by a mosaic of landscapes formed by diverse vegetational types as restinga forest, dense ombrophilous forest, seasonal semideciduous forest and savanna woody, respectively represented by the conservation units Parque Estadual Ilha do Cardoso, Parque Estadual Carlos Botelho, Estação Ecológica dos Caetetus and Estação Ecológica de Assis. These units are legally protected, although occurring mostly in areas highly modified by anthropogenic impacts. Epiphytes, especially bromeliads, directly respond to these impacts, being interesting for environmental studies. Bromeliads have different strategies for the uptake of chemical elements from the litterfall deposited in the tank (tank bromeliad) or from the atmosphere (atmospheric bromeliad). Many steps of the chemical analysis affect the application of these plants for environmental quality studies, including sampling and leaf surface contamination with particulate matter. According to the usual procedure, sampling depends on the removal of several rosettes, which may cause impacts on the ecosystem. Therefore, the proposal of this work consists in a novel sampling methodology, named \"sustainable\", in which small disks of bromeliad leaves are collected without removing rosettes from the studied areas. Considering the different vegetation, the bromeliad diversity, the nutritional characteristics of epiphytic plants and the problem of sampling native bromeliads in conservation units, this work aimed at (I) the evaluation of chemical composition of bromeliads from diverse vegetational types, (II) the evaluation of chemical composition of different bromeliad species, (III) the evaluation of the contribution of tank and atmospheric nutritional modes, as well as epiphytic and terrestrial growth modes, for the chemical composition of bromeliads, (IV) the assessment of washing effects on leaf chemical composition, (V) the comparison of sustainable sampling results with those obtained from usual sampling and (VI) the assessment of chemical composition variability at different leaf and plant positions. Instrumental neutron activation analysis (INAA) was employed, allowing the simultaneous determination of diverse chemical elements at a high metrological level. For each study, samples were prepared accordingly before being analyzed by INAA. The bromeliads showed differences in chemical composition among the conservation units. There was no uniform distribution pattern of chemical elements among the bromeliad species, corroborating an inherent species chemical composition. Tank bromeliads showed apparently high efficiency in the uptake of essential chemical elements. The studied plants showed concentrations slightly higher in bromeliads growing in epiphytic mode when compared to those growing on soil. The results demonstrated the need for the washing procedure to minimize the particulate matter influence on the concentrations of chemical elements. The new sustainable sampling provided results comparable to the usual sampling procedure, especially for the bottom plant position. Further, the reliability of the results was increased due to the reduction of steps for sample preparation provided by this sampling methodology

Bactérias diazotróficas em Guzmania monostachia (Bromeliaceae): identificação, sinalização e colonização dos tecidos foliares / Diazotrophic bacteria in Guzmania monostachia (Bromeliaceae): identification, signaling and leaf tissue colonization

As bromélias habitam os mais diferentes ambientes sendo que muitas são encontradas como epífitas. Essas últimas estão sujeitas a condições de disponibilidade de água e nutrientes com intermitência. Elas conseguem sobreviver a essas circunstâncias por serem dotadas de diversas adaptações morfológicas e fisiológicas. A bromélia tanque Guzmania monostachia tem sido bastante estudada por possuir uma grande plasticidade fotossintética, porém, pouco é conhecido a respeito de outras possíveis adaptações como a interação com micro-organismos. Sendo assim, o presente trabalho isolou bactérias fixadoras de nitrogênio (diazotróficas) tanto da parte externa (epifíticas) quanto da parte interna (endofíticas) de diferentes porções (ápice, mediana e base) de folhas coletadas tanto no ambiente natural quanto na casa de vegetação. As bactérias foram selecionadas por meio do ensaio de redução de acetileno (ARA) e também pelo uso de quatro diferentes meios de cultura que não contém fonte de nitrogênio reduzido. As linhagens isoladas foram identificadas por meio do gene 16sRNA. Dois isolados Pseudomonas sp. e Burkholderia sp. foram escolhidos para serem marcados com um gene de fluorescência verde (GFP) e foram então inoculados (separadamente) em plantas de G. monostachia cultivadas em casa de vegetação. A colonização dos tecidos foliares foi monitorada com auxílio de um microscópio confocal. Além disso, foram estimadas as densidades populacionais epifíticas e endofíticas em diferentes grupos foliares (jovens e intermediários) e as folhas do grupo intermediário por serem maiores e totalmente expandidas foram divididas em porções (apical, mediana e basal). Também foram pesquisadas as seguintes moléculas descritas como importantes na interação entre planta e micro-organismo: óxido nítrico (NO), ácido salicílico (SA), etileno (ET) e ácido-indol-3-acético (IAA). Como resultados, a maioria das linhagens bacterianas foram classificadas como pertencentes ao grupo Proteobacteria, mas também foram encontrados isolados gram positivos pertencentes aos grupos Actinobacteria e Firmicutes. As bactérias endofíticas foram isoladas somente da porção basal foliar (tanto das folhas originadas do meio ambiente quanto das folhas originadas da casa de vegetação). Cabe ressaltar que após a inoculação de ambas bactérias marcadas com GFP, foi observado elas no interior dos tricomas foliares (estruturas presentes principalmente na base foliar). Após 20 horas da inoculação, ambas bactérias já foram visualizadas no interior da epiderme das folhas. Após 5 dias as bactérias foram se espalhando para regiões mais distantes do tricoma e também foram observadas no parênquima. Após 10 dias a bactéria Pseudomonas sp. foi encontrada nas paredes dos vasos condutores. Foram re-isoladas bactérias epifíticas e endofíticas da mediana e da base foliar, mas não da porção apical. Após 10 dias as bactérias foram isoladas como endofíticas somente da base. Essa porção não apresentou diferenças nas populações epifíticas e endofíticas. O NO aumentou nas folhas jovens e na base das intermediárias em um curto período de tempo após a inoculação. Aparentemente, ambas as bactérias não dispararam a via do SA. De acordo com os resultados aqui presentes, ambas as bactérias não pareceram ser prejudiciais à G. monostachia. Além disso, o presente trabalho mostra fortes evidencias de que as bactérias entram nos tecidos foliares por meio dos tricomas na base foliar e permanecem nessa porção, que é precisamente a mais importante para absorção de nutrientes / Bromeliads inhabit different environments and many are found as epiphytes. These plants are often subjected to periods of water and nutrient shortage. For their survival, epiphytic bromeliads are endowed with different morphological and physiological adaptations. Guzmania monostachia is a tank-bromeliad that has been extensively studied because of its great photosynthetic plasticity. However, little is known about other possible survival adaptations, such as beneficial interactions with microorganisms. Here, we isolated nitrogen fixing (diazotrophc) bacteria from both the outside (epiphytic) and the inside (endophytic) of different leaf portions (apex, middle and base), collected in natural environment and in greenhouse-cultivated plants. The bacteria were selected using the acetylene reduction assay (ARA) and four different media that do not contain reduced nitrogen source. The strains were identified by 16S rRNA. Two isolates, Pseudomonas sp. and Burkholderia sp. has been chosen to be tagged with green fluorescent protein (GFP) and then inoculated in G. monostachia plants cultivated in greenhouse. The colonization of the leaf tissues was monitored with the aid of confocal microscopy and also we estimate the external and internal bacterial population densities in different leaf groups (younger and expanded) and portions (apex, middle and base). In addition, we studied some important molecules in plant-microbe interactions: nitric oxide (NO), salicylic acid (SA), ethylene (ET) and indol-3-acetic acid (IAA). As a result, most of the isolated strains belong to the Proteobacteria group, but gram positive strains were also found belonging to the Firmicutes and Actinobacteria group. The endophytic bacteria were isolated only from the basal portion (both from leaves of natural environment and from leaves of greenhouse cultivated plants). Interestingly, after the inoculation of both bacteria tagged with GFP they were visualized entering by trichomes present mainly in the basal portion. Twenty hours after the inoculation, the bacteria were visualized inside the epidermis of the leaves. After five days, the bacteria were detected in the parenchyma and, ten days after the inoculation Pseudomonas sp. was found on the vessels walls. It was possible to re-isolate epiphytic and endophytic bacteria from the base and middle portions, but not from the apex. After 10 days the endophytic bacteria were found only in the base. The base did not show differences between epiphytic and endophytic populations. NO increased in a short time after the inoculation in the younger leaves and in the basal portion of intermediate leaves. Apparently, the SA pathway was not triggered by any of the bacteria used. According to these results, the bacteria tested do not seem to be harmful to the plant. Furthermore, we strongly suggest that they enter through the trichomes on the leaf base and remain in this portion, which is precisely the most important for the absorption of nutrients

Factors Influencing Epiphytic Lichen Communities in Aspen-Associated Forests of the Bear River Range, Idaho and Utah

In western North America, quaking aspen (Populus tremuloides Michx.) is the most common hardwood in montane landscapes. Fire suppression, grazing, wildlife management practices, and climate patterns of the past century are some of the threats to aspen coverage in this region. Researchers are concerned that aspen-dependent species may be losing habitat, thereby threatening their long-term local and regional viability. Though lichens have a rich history as air pollution indicators, I believe that they may also be useful as a metric of community diversity associated with habitat change. To date, few studies have specifically examined the status of aspen's epiphytic lichen community in the Rocky Mountains. A preliminary study was conducted using 10 transect-based plots to assess lichen species substrate preferences between aspen and various conifer species and to gain basic knowledge of species diversity. Following this work, I established 47 plots in the Bear River Range of northern Utah and southern Idaho to evaluate the effects of forest succession on epiphytic macrolichen communities. Plots were located in a narrow elevational belt (2,134-2,438 m) to minimize the known covariant effects of elevation and moisture on lichen communities. Results show increasing lichen diversity and a decrease in aspen-dependent species as aspen forests succeed to conifer cover types. The interactive roles of stand aspect, basal area and cover of dominant trees, stand age, aspen bark scars, and recent tree damage were examined in relation to these trends. An aspen index score was developed based on lichens showing an affinity for aspen habitat. I present a landscape-level multivariate analysis of short and long-term factors influencing epiphytic lichen communities in aspen forests. Nonrnetric multidimensional scaling (NMS) ordination stressed the importance of succession and local air pollution sources in shaping lichen communities. I also investigated the role of historic human intrusions and climate on aspen forests and aspen dependent epiphytic lichens at the landscape-level. Implications of this work include 1) realization of nitrogen impacts on ecosystems, 2) the potential for using lichens as bioindicators for monitoring aspen stand health, and 3) suggestions for working with natural disturbance regimes to minimize human impacts on aspen and associated species.

Despite their universality, much remains unknown about their basic ecology and interactions with higher organisms. To address this gap, this thesis firstly examines the bacterial communities associated with three co-occurring marine eukaryote hosts from temperate Australia: the demosponge Cymbastela concentrica, the subtidal red macroalga Delisea pulchra and the intertidal green macroalga Ulva australis. Molecular characterisation of the bacterial communities was undertaken using 16S rRNA gene library analysis to compare within-host (alpha) and between-host (beta) diversity for the three microbial communities. This study highlights the potentially substantial contribution host-associated microorganisms could have on marine microbial diversity. The remaining focus for this thesis was on the bacterial community associated with D. pulchra. This alga produces a suite of biologically active secondary metabolites (furanones) that non-toxically inhibit acyl homoserine lactone (AHL)-driven quorum sensing in bacteria, affecting a range of phenotypes including colonisation and virulence traits. The ecology of D. pulchra???s epiphytic bacteria was investigated using a mechanistic approach to explain bacterial colonisation patterns. In particular, concepts and models of ecological succession founded in eukaryote ecology were investigated. The thesis concludes with a study investigating the effect of furanones and elevated temperature on bacteria-induced disease and thallus bleaching of D. pulchra. In the presence of furanones colonisation and infection of two Roseobacter isolates from D. pulchra???s epiphytic bacterial community were inhibited. Ruegeria strain R11 was demonstrated to have temperature regulated virulence, which caused thallus bleaching in furanone-free algae. The implications of elevated sea temperatures resulting from global warming for algal health are discussed.

Comunidades fúngicas endofítica, epifítica e rizosférica em diferentes ecossistemas / Endophytic, epiphitic and rhizospheric fungi communities in different ecosystems

O presente trabalho teve por objetivo avaliar o efeito de três ecossistemas sobre, a ocorrência de fungos rizosféricos, endofíticos e epifíticos. As áreas de estudo estão localizadas em uma propriedade rural, situada na cidade de Venâncio Aires, RS. Os fungos endofíticos e epifíticos foram isolados de amostras de raízes de guajuvira, (Patagonula americana L.), coletadas na mata (área 1), de uva-do-japão (Hovenia dulcis Thunb.), na área intermediária (área 2) e de fumo ou de milho, na lavoura (área 3) e, os fungos rizosféricos isolados de solo coletado junto as raízes dos vegetais citados. As amostras foram coletadas nos meses de janeiro, maio, setembro e novembro de 2004 e 2005. Os fungos foram identificados segundo características morfológicas com auxílio de chaves de identificação. As áreas 2 e 3 foram mais similares com relação aos fungos rizosféricos e epifíticos, enquanto, que, as áreas 1 e 2 foram mais similares com relação aos fungos endofíticos. A diversidade dos fungos isolados das três áreas não diferiu ao longo dos dois anos de coleta. A presença de Fusarium spp., em vegetais sem sintomas de doença indica que, as mesmas podem ser raças avirulentas ou patógenos latentes em equilíbrio com o hospedeiro e o ambiente. A presença de fungos antagonistas, principalmente Trichoderma spp. também, são responsáveis por esse equilíbrio, o qual ocorre nas três áreas. / The present paper aimed at assessing the effect of three ecosystems, on the occurrence of rhizospheric, endophytic and epiphytic fungi. The study areas are located in a farm in Venâncio Aires, Rio Grande do Sul. Samples were collected in the months of January, May, September and November 2004 and 2005. The endophytic and epiphytic fungi were isolated from samples of guajayvi roots (Patagonula americana L.), picked in the forest (area 1), of Japanese raisin tree (Hovenia dulcis Thunb.), in the intermediate area (area 2), and tobacco or corn, in the cultivated area (area 3) and rhizospheric fungi isolated from soil. The fungi were identified according to morphological characteristics, with the help of identification keys. Areas 2 and 3 were more alike concerning rhizospheric and epiphytic fungi; Areas 1 and 2, in turn, were more alike concerning endophytic fungi. The diversity of the isolated fungi from these three areas did not change during the two years of collection. The abundance of fungi with phytopathogenic potential such as the Fusarium spp. and Macrophomina phaseolina, mostly as endophytic in area 3 shows the influence of the host on them. The presence of antagonistic fungi, principally Trichoderma spp. also, are responsable for this equilibrium, which occured in the three areas.

O presente trabalho teve por objetivo avaliar o efeito de três ecossistemas sobre, a ocorrência de fungos rizosféricos, endofíticos e epifíticos. As áreas de estudo estão localizadas em uma propriedade rural, situada na cidade de Venâncio Aires, RS. Os fungos endofíticos e epifíticos foram isolados de amostras de raízes de guajuvira, (Patagonula americana L.), coletadas na mata (área 1), de uva-do-japão (Hovenia dulcis Thunb.), na área intermediária (área 2) e de fumo ou de milho, na lavoura (área 3) e, os fungos rizosféricos isolados de solo coletado junto as raízes dos vegetais citados. As amostras foram coletadas nos meses de janeiro, maio, setembro e novembro de 2004 e 2005. Os fungos foram identificados segundo características morfológicas com auxílio de chaves de identificação. As áreas 2 e 3 foram mais similares com relação aos fungos rizosféricos e epifíticos, enquanto, que, as áreas 1 e 2 foram mais similares com relação aos fungos endofíticos. A diversidade dos fungos isolados das três áreas não diferiu ao longo dos dois anos de coleta. A presença de Fusarium spp., em vegetais sem sintomas de doença indica que, as mesmas podem ser raças avirulentas ou patógenos latentes em equilíbrio com o hospedeiro e o ambiente. A presença de fungos antagonistas, principalmente Trichoderma spp. também, são responsáveis por esse equilíbrio, o qual ocorre nas três áreas. / The present paper aimed at assessing the effect of three ecosystems, on the occurrence of rhizospheric, endophytic and epiphytic fungi.
